RESUMO

Anthracosilicosis (AS), a prevalent form of pneumoconiosis among coal miners, results from the accumulation of carbon and silica in the lungs from inhaled coal dust. This study investigated genotoxic effects and certain cytokine genes polymorphic variants in Russian coal miners with АS. Peripheral leukocytes were sampled from 129 patients with AS confirmed by X-ray and tissue biopsy and from 164 asymptomatic coal miners. Four single-nucleotide polymorphisms were genotyped in the extracted DNA samples: IL1ß T-511C (rs16944), IL6 C-174G (rs1800795), IL12b A1188C (rs3212227) and VEGFA C634G (rs2010963). Genotoxic effects were assessed by the analysis of chromosome aberrations in cultured peripheral lymphocytes. The mean frequency of chromatid-type aberrations and chromosome-type aberrations, namely, chromatid-type breaks and dicentric chromosomes, was found to be higher in AS patients [3.70 (95% confidence interval {CI}, 3.29-4.10) and 0.28 (95% CI, 0.17-0.38)] compared to the control group [2.41 (95% CI, 2.00-2.82) and 0.09 (95% CI, 0.03-0.15)], respectively. IL1ß gene T/T genotype (rs16944) was associated with AS [17.83% in AS patients against 4.35% in healthy donors, odds ratio = 4.77 (1.88-12.15), P < 0.01]. A significant increase in the level of certain chromosome interchanges among AS donors is of interest because such effects are typical for radiation damage and caused by acute oxidative stress. IL1ß T allele probably may be considered as an AS susceptibility factor among coal miners.

In this paper, the lysozyme (LSZ) adsorption impact on the silica suspension stability was established. In other words, the stabilization/destabilization mechanism of the SiO2/LSZ system was explained based on the adsorption, electrokinetic and stability measurement results. Lysozyme adsorbs on the silica surface in the whole pH range. This process contributes to the changes in silica surface charge and zeta potential values. The lysozyme addition influences the system stability too. At pH 7.6 and 9, a large decrease in the silica suspension stability was found. It is connected with the neutralization of solid negative charge by the positively charged macromolecules. As a result, large aggregates can be formed, which is highly desirable in the silica removal procedure.

This work represents a first systematic approach to the size-based elemental quantification and size estimation of metal(loid) oxide nanoparticles such as silica (SiO2) in a real food matrix using asymmetric flow field-flow fractionation coupled online with inductively coupled plasma mass spectrometry (ICP-MS) and multi-angle light scattering (MALS) and offline with transmission electron microscopy (TEM) with energy-dispersive X-ray analysis (EDAX). Coffee creamer was selected as the model sample since it is known to contain silica as well as metal oxides such as titania at the milligramme per kilogramme levels. Optimisation of sample preparation conditions such as matrix-to-solvent ratio, defatting with organic solvents and sonication time that may affect nanoparticle size and size distribution in suspensions was investigated. Special attention was paid to the selection of conditions that minimise particle transformation during sample preparation and analysis. The coffee creamer matrix components were found to stabilise food grade SiO2 particles in comparison with water suspensions whilst no significant effect of defatting using hexane was found. The use of sample preparation procedures that mimic food cooking in real life was also investigated regarding their effect on particle size and particle size distribution of silica nanoparticles in the investigated food matrix; no significant effect of the water temperature ranging from ambient temperature to 60 °C was observed. Field-flow fractionation coupled to inductively coupled plasma-mass spectrometry (FFF-ICP-MS) analysis of extracts of both unspiked coffee creamer and coffee creamer spiked with food grade silicon dioxide, using different approaches for size estimation, enabled determination of SiO2 size-based speciation. Element-specific detection by ICP-MS and post-FFF calibration with elemental calibration standards was used to determine the elemental composition of size fractions separated online by FFF. Quantitative data on mass balance is provided for the size-based speciation of the investigated inorganic nano-objects in the complex matrix. The combination of FFF with offline fractionation by filtration and with detection by ICP-MS and TEM/EDAX has been proven essential to provide reliable information of nanoparticle size in the complex food matrix.

The two marine inorganic polymers, biosilica (BS), enzymatically synthesized from ortho-silicate, and polyphosphate (polyP), a likewise enzymatically synthesized polymer consisting of 10 to >100 phosphate residues linked by high-energy phosphoanhydride bonds, have previously been shown to display a morphogenetic effect on osteoblasts. In the present study, the effect of these polymers on the differential differentiation of human multipotent stromal cells (hMSC), mesenchymal stem cells, that had been encapsulated into beads of the biocompatible plant polymer alginate, was studied. The differentiation of the hMSCs in the alginate beads was directed either to the osteogenic cell lineage by exposure to an osteogenic medium (mineralization activation cocktail; differentiation into osteoblasts) or to the chondrogenic cell lineage by incubating in chondrocyte differentiation medium (triggering chondrocyte maturation). Both biosilica and polyP, applied as Ca²âº salts, were found to induce an increased mineralization in osteogenic cells; these inorganic polymers display also morphogenetic potential. The effects were substantiated by gene expression studies, which revealed that biosilica and polyP strongly and significantly increase the expression of bone morphogenetic protein 2 (BMP-2) and alkaline phosphatase (ALP) in osteogenic cells, which was significantly more pronounced in osteogenic versus chondrogenic cells. A differential effect of the two polymers was seen on the expression of the two collagen types, I and II. While collagen Type I is highly expressed in osteogenic cells, but not in chondrogenic cells after exposure to biosilica or polyP, the upregulation of the steady-state level of collagen Type II transcripts in chondrogenic cells is comparably stronger than in osteogenic cells. It is concluded that the two polymers, biosilica and polyP, are morphogenetically active additives for the otherwise biologically inert alginate polymer. It is proposed that alginate, supplemented with polyP and/or biosilica, is a suitable biomaterial that promotes the growth and differentiation of hMSCs and might be beneficial for application in 3D tissue printing of hMSCs and for the delivery of hMSCs in fractures, surgically created during distraction osteogenesis.

The use of coagulation and Fenton oxidation was studied for total organic carbon (TOC) and silica removal from steam-assisted gravity drainage (SAGD) water at 800C and two different concentrations replicating the stream feeding the warm lime softening unit having 675 mg/L TOC and 350 mg/L silica and the blowdown of the once through steam generator having 3700mg/L TOC and 2585 mg/L silica. Coagulation was carried out by the addition of FeCl3, Al(NO3)3 or Ca(NO3)2. The results showed that Fe(III) salt outperformed Al(III) and Ca(II) salts. A two-stage addition of 2.5 g FeCl3 per g TOC intermediated by a filtration unit resulted in approximately 72% TOC removal and more than 80% silica removal while maintaining low solid waste. Comparing results pertaining to coagulant concentration and final pH, it can be easily concluded that silica removal is governed by the resultant pH, whereas TOC removal was accomplished through surface neutralization and localized enmeshment coagulation. Fenton oxidation is proposed to further treat the filtrate obtained from the second stage Fe(III) coagulation. An additional 10% TOC removal could be achieved; at seven times lower H202 dose in the presence of Fe2+ or Fe0 reagent. Moreover, the advanced Fenton process resulted in high silica removal as a result of adsorption onto Fe(OH)3 precipitate, which formed at the equilibrium pH of the system.

The hybrid zero-valent-iron (hZVI) process is a novel chemical treatment process that has shown promise for removing heavy metals and nutrients from industrial wastewaters. In this study, a pilot-scale demonstration was conducted to continuously treat 3.8-7.6 L/min (1-2 gpm) of the flue-gas-desulfurization (FGD) wastewater at a coal-fired power plant for 5 months. In this paper, a spike test was conducted to evaluate performance of the hZVI process for removing selected toxic metals at artificially elevated concentrations. The results showed that a multiple-stage hZVI process could decrease selenate-Se from 22 mg/L to ~10 µg/L and dissolved Hg(2+) from 1.15 mg/L to ~10 ng/L. In addition, the process simultaneously removed a broad spectrum of heavy metals such as As(III), As(V), Cr(VI), Cd(II), Pb(II) and Cu(II) from mg/L to near or sub-ppb (µg/L) level after a single-stage treatment. The process consumed about 0.3 kg ZVI per 1 m(3) FGD wastewater treated at a cost of about US$0.6/m(3). Solid waste production and energy consumption were reasonably low. The successful pilot study demonstrated that the hZVI technology can be a low-cost, high-performance treatment platform for solving some of the toughest heavy metal water problems.

In the present paper, for the first time the ability of the porous biosilica originated from three marine diatom strains of 'Nanofrustulum spp.' viz. N. wachnickianum (SZCZCH193), N. shiloi (SZCZM1342), N. cf. shiloi (SZCZP1809), to eliminate MB from aqueous solutions was investigated. The highest biomass was achieved under silicate enrichment for N. wachnickianum and N. shiloi (0.98 g L-1 DW and 0.93 g L-1 DW respectively), and under 15 °C for N. cf. shiloi (2.2 g L-1 DW). The siliceous skeletons of the strains were purified with hydrogen peroxide and characterized by SEM, EDS, the N2 adsorption/desorption, XRD, TGA, and ATR-FTIR. The porous biosilica (20 mg DW) obtained from the strains i.e. SZCZCH193, SZCZM1342, SZCZP1809, showed efficiency in 77.6%, 96.8%, and 98.1% of 14 mg L-1 MB removal under pH 7 for 180 min, and the maximum adsorption capacity was calculated as 8.39, 19.02, and 15.17 mg g-1, respectively. Additionally, it was possible to increase the MB removal efficiency in alkaline (pH = 11) conditions up to 99.08% for SZCZP1809 after 120 min. Modelling revealed that the adsorption of MB follows Pseudo-first order, Bangham's pore diffusion and Sips isotherm models.

A filtration-aided acid cleaning method was used to collect biosilica structures from a diatom culture medium, natural seawater, or water bloom. Cell extraction, acid cleaning, and acid removal were all performed on a polytetrafluoroethylene (PTFE) filter cloth, significantly improving the treatment capacity and efficiency of the traditional acid wash method. Five typical diatoms were cultivated in the laboratory for acid cleaning. Different growth speeds were introduced, and different process parameters for acid cleaning were utilized. After the acid cleaning, biosilica structures were collected from the frustules of diatoms using different methods. Girdle bands and valves of Coscinodiscus sp. were separated by floating of the valves. Central spines of Ditylum brightwellii and valves of Skeletonema costatum were separately collected by settling or filtration. Rod-like frustules, such as those of Bacillaris paradoxa, are not suitable for large quantities of acid wash. The silica structures were observed and tested using an AFM-calibrated glass needle to determine their elasticity. Elasticity tests showed that ringent girdle bands are more flexible than complete ones (Coscinodiscus sp.) and that both long-chain clusters of Nitzschia palea and central spines of D. brightwellii have certain elasticities. The required pressure for deforming or breaking the biosilica structures of diatoms was also determined.

In the 8th edition of Japan's Specifications and Standards for Food Additives, the purity test for silicone resins requires the determination of the refractive index and kinetic viscosity of the extracted silicone oil, and allows for only a limited amount of silicon dioxide. In the purity test, carbon tetrachloride is used to separate the silicone oil and silicon dioxide. To exclude carbon tetrachloride, methods were developed for separating the silicone oil and silicon dioxide from silicone resin, which use hexane and 10% n-dodecylbenzenesulfonic acid in hexane. For silicone oil, the measured refractive index and kinetic viscosity of the silicone oil obtained from the hexane extract were shown to be equivalent to those of the intact silicone oil. In regard to silicon dioxide, it was confirmed that, following the separation with 10% n-dodecylbenzenesulfonic acid in hexane, the level of silicon dioxide in silicone resin can be accurately determined. Therefore, in this study, we developed a method for testing the purity of silicone resins without the use of carbon tetrachloride, which is a harmful reagent.

Rice husk (RH) agro-waste was used as a raw material for synthesizing mesoporous molecular sieves, MCM-41. The Fe-MCM-41 was prepared by the hydrothermal technique (HT), resulting in a higher surface area and crystallinity than when prepared under ambient conditions. In addition, a hexagonal structure was clearly seen with hydrothermal technique (HT) preparation. The adsorption of arsenate by HT-Fe-MCM-41 was investigated. The factors studied affecting arsenate adsorption capacity were ferric content in MCM-41, contact time, pH of solution, and initial arsenate concentration. It was found that HT-Fe-MCM-41 at the Si/Fe mole ratio of 10 gave the highest adsorption capacity. Arsenate adsorption reached equilibrium within 4 h. The adsorption capacity of HT-Fe-MCM-41 (Si/Fe = 10) was affected by the initial pH value and the initial arsenate concentration. The adsorption capacity was highest at pH 3 and decreased thereafter with increases in the pH of solution value. The Langmuir model fit the arsenate adsorption isotherm well. The maximum adsorption capacity for arsenate was 1,111 microg g(-1).

In this study the effective utilization of two types of municipal solid waste incinerator (MSWI) ashes, namely air-cooled ash (ACS) and water-cooled ash (WCS) samples obtained from a municipal solid waste incineration plant, was examined by applying zeolitic material synthesis and silica extraction. The influence of the experimental conditions including the ratio of sample : NaOH solution, the reaction temperature and time, and the concentration of NaOH solution were investigated. The results for the 25 experimental trials can be summarized as: (1) the formation of tobermorite and/or pectolite-1A as a major component in most conditions; (2) the synthesis of hydroxycancrinite as a major phase at 200 degrees C; (3) a dramatic increase in the extracted SiO(2) yield at 1 : 30 value of sample : NaOH ratio and 200 degrees C, even at short reaction times; and (4) relatively high SiO(2) yields for WCS ashes rather than ACS ashes. An increase in the reaction time improved the quantity of synthesized zeolitic materials. The reaction temperature determined the type of zeolite. An increase in the NaOH concentration can be an essential factor to improve zeolitic material synthesis, but it significantly reduced the yield of SiO(2) extraction. In conclusion, suitable conditions for obtaining both SiO(2) extraction and synthesized zeolites from the ashes of the incinerated solid waste materials should be: 200 degrees C reaction temperature; a 1 : 30 (g : mL) value for the sample : NaOH ratio; 2 mol L(-1) NaOH concentration; and a reaction time of more than 24 h.

Regulatory agencies approve automatic samplers containing peristaltic pumps as a sample collection method for stormwater characterization and for treatment-device evaluation. Autosampler performance, as discussed in the limited available literature, can vary across the entire particle size range typically found in stormwater from different source areas and outfalls-reasonably consistent performance for particle sizes < 250 microm, but much less consistency for particles > 250 microm. Therefore, a series of experiments was undertaken to quantify the upper range of consistent particle capture that may occur with sampling stormwater suspended sediment and particulate-bound pollutants. These experiments, based on triplicate sampling at each experimental condition, found that peristaltic pump autosamplers commonly used in stormwater monitoring could not repeatedly and effectively capture particles > 250 microm from a simulated stormwater whose particles have a specific gravity of 2.65. It was expected that the effective size for autosamplers would be correspondingly larger for particles having smaller specific gravities. The height of the sampler had no influence on particle recovery up to a height of 2.5 m, with slightly decreasing recoveries of large particles occurring at greater heights, as a result of reduced sampler intake velocity. Therefore, to characterize the solids across the entire size range and specific gravities that may occur in stormwater runoff, autosamplers should be deployed in conjunction with bedload and floatables sampling.

RESUMO

OBJECTIVE: To study the chemical constituents of Polygala telephioides. METHOD: The compounds were isolated and purified on macroporous resin, silica gel, Sephadex LH-20, Chromatorex ODS column chromatograph and the structures were determined based on the spectral and chemical evidences. RESULT: Four compounds were obtained and characterized as telephiose G, telephiose D, isomangiferin, quescetin 3-O-beta-D-glucopyranoside, respectively. CONCLUSION: Compounds 2-4 were obtained from this plant for the first time and the compound 2 (telephiose G) was a new compound.

The increasingly ubiquitous release of emerging refractory pollutants into water is a serious concern due to associated risks. In this study, mesoporous hierarchical BiOBr/Fe3O4@SiO2-a solvothermally synthesized visible-light-driven magnetic photocatalyst-not only exhibited fast kinetics (t1/2 = 8.7 min) in the photocatalytic degradation of ibuprofen in water but also achieved almost complete mineralization over a prolonged irradiation of 6 h. Various reactive species, including O2¯, OH, and H2O2, were detected, while the scavenging experiments revealed that eCB--mediated reactions and direct-hole oxidation are the major degradation routes. The magnetically recycled BiOBr/Fe3O4@SiO2 maintained ∼80% of its initial photocatalytic activity even after five consecutive cycles. The typically copresent wastewater constituents, including NOM and anions, inhibited the photocatalytic performance to varying extents, and hence necessitated an increase in the photocatalyst dosage to achieve complete ibuprofen degradation in real sewage. Based on the findings of batch experiments, the process was scaled up by developing a 5 L prototype photocatalytic reactor integrated with an electromagnetic separation unit. The results of prototype photocatalytic experiments were comparable to those of batch experiments, and an electromagnetic separation efficiency of ∼99% was achievable in 5 min.

Silica has been extracted from rice husks via a simple hydrothermal process and functionalized with triethoxy(octyl)silane -OCTES (Octyl-SiO2) and (3-aminopropyl)triethoxysilane - 3-APTES (Amino-SiO2), with the aim of using it as support to immobilize lipase from Thermomyces lanuginosus (TLL) via adsorption. The supports have been characterized by particle size distribution and elemental analyses, XRD, TGA, SEM, AFM and N2 physisorption so as to confirm their functionalization. Effect of pH, temperature, initial protein loading and contact time on the adsorption process has been systematically evaluated. Maximum immobilized protein loading of 12.3 ± 0.1 mg/g for Amino-SiO2 (5 mM buffer sodium acetate at pH 4.0, 25 °C and initial protein loading of 20 mg/g) and 21.9 ± 0.1 mg/g for Octyl-SiO2 (5 mM buffer sodium acetate at pH 5.0, 25 °C and initial protein loading of 30 mg/g) was observed. However, these biocatalysts presented similar catalytic activity in olive oil emulsion hydrolysis (between 630 and 645 U/g). TLL adsorption was a spontaneous process involving physisorption. Experimental data on Octyl-SiO2 and Amino-SiO2 adsorption were well-fitted to the Langmuir isotherm model. It was also investigated whether these biocatalysts could synthesize cetyl esters via esterification reaction. Thus, it was found that cetyl stearate synthesis required 100-110 min of reaction time to attain maximum conversion percentage (around 94%). Ester productivity of immobilized TLL on Amino-SiO2 was 1.3-3.1 times higher than Octyl-SiO2.

Seed extracts from Moringa oleifera are of wide interest for use in water purification where they can play an important role in flocculation; they also have potential as anti-microbial agents. Previous work has focused on the crude protein extract. Here we describe the detailed biophysical characterization of individual proteins from these seeds. The results provide new insights relating to the active compounds involved. One fraction, designated Mo-CBP3, has been characterized at a molecular level using a range of biochemical and biophysical techniques including liquid chromatography, X-ray diffraction, mass spectrometry, and neutron reflection. The interfacial behavior is of particular interest in considering water purification applications and interactions with both charged (e.g. silica) and uncharged (alumina) surfaces were studied. The reflection studies show that, in marked contrast to the crude extract, only a single layer of the purified Mo-CBP3 binds to a silica interface and that there is no binding to an alumina interface. These observations are consistent with the crystallographic structure of Mo-CBP3-4, which is one of the main isoforms of the Mo-CBP3 fraction. The results are put in context of previous studies of the properties of the crude extract. This work shows possible routes to development of separation processes that would be based on the specific properties of individual proteins.

Bone fractures characterize an important event in the medical healthcare, being related to traumas, aging, and diseases. In critical conditions, such as extensive bone loss and osteoporosis, the tissue restoration may be compromised and culminate in a non-union consolidation. In this context, the osteogenic properties of biomaterials with a natural origin have gained prominence. Particularly, marine sponges are promising organisms that can be exploited as biomaterials for bone grafts. Thus, the objectives of this study were to study the physicochemical and morphological properties of biosilica (BS) from sponges by using scanning electron microscopy, Fourier-transform infrared, X-ray diffraction (SEM, FTIR and XRD respectively), mineralization, and pH. In addition, tests on an osteoblast precursor cell line (MC3T3-E1) were performed to investigate its cytotoxicity and proliferation in presence of BS. Bioglass (BG) was used as gold standard material for comparison purposes. Sponge BS was obtained, and this fact was proven by SEM, FTIR, and XRD analysis. Calcium assay showed a progressive release of this ion from day 7 and a more balanced pH for BS was maintained compared to BG. Cytotoxicity assay indicated that BS had a positive influence on MC3T3-E1 cells viability and qRT-PCR showed that this material stimulated Runx2 and BMP4 gene expressions. Taken together, the results indicate a potential use of sponge biosilica for tissue engineering applications.

Hybrid silica monoliths functionalized with octyl groups and dedicated to chromatographic separations in the reversed-phase mode were directly synthesized within capillaries according to the protocol described by Yan et al. [L.J. Yan, Q.H. Zhang, Y.Q. Feng, W.B. Zhang, T. Li, L.H. Zhang, Y.K. Zhang, J. Chromatogr. A 1121 (2006) 92]. Although these monoliths allowed reaching high efficiencies in capillary electrochromatography (CEC), serious limitations prohibited their application in nano-liquid chromatography (nano-LC). Such limitations observed as poor performances in the nano-LC mode and the lack of reproducibility of the synthesis were related to the longitudinal morphological inhomogeneities of the hybrid material along the capillary. Thus, several modifications were conducted in the synthesis protocol in order to improve the resulting morphology of the monolith making it suitable for nano-LC separations. The influence of several critical parameters (such as the addition temperature of the basic catalyst and the hydrolysis duration) on the textural and chromatographic properties had been extensively studied. It was found that a decrease (i.e. 0 degrees C) of the temperature addition of the basic catalyst associated with a shorter hydrolysis duration (1h instead of 6h) allowed (i) delaying the gelation time and consequently facilitating the capillary filling step, (ii) increasing the structural homogeneity of the hybrid monoliths, i.e. their chromatographic performances in nano-liquid chromatography also (iii) greatly improving the reproducibility of the synthesis within the capillary without impairing the material's carbon load, i.e. the incorporation of the less hydrolysable C(8) precursor. The resulting hybrid monoliths afforded retention factors comparable to that previously obtained for C(18) grafted silica monoliths and efficiencies that are the best ever recorded in nano-LC with hybrid monoliths and that are close to the ones achieved with grafted silica monoliths. In fact, this modified protocol allowed a significant improvement of the performances in nano-LC which could be observed by the decrease of the mean value of H(min) going from 123 microm (Yan's protocol) to 24 microm (modified protocol) for a same length of capillary (l = 8.5 cm). In addition, the reproducibility of the synthesis was greatly improved through a factor six of reduction on the calculated standard deviation of these efficiencies. The high permeability and longitudinal homogeneity of the synthesized monolith allowed increasing the capillary length (for example, a 75-cm capillary was conveniently filled with hybrid silica monolith) and the column could be eluted at a very low backpressure leading to chromatographic performances up to 40,000 plates. Finally, the good efficiencies in the nano-LC mode combined with the excellent performances already present in the CEC mode led to fast (less than 1 min) and high efficient separations in the pressurized capillary electrochromatography (p-CEC) mode.

The potential of kenaf (Hibiscus cannabinus L.) for phytoremediation of lead (Pb) on sand tailings was investigated. A pot experiment employing factorial design with two main effects of fertilizer and lead was conducted in a nursery using sand tailings from an ex-tin mine as the growing medium. Results showed that Pb was found in the root, stem, and seed capsule of kenaf but not in the leaf. Application of organic fertilizer promoted greater biomass yield as well as higher accumulation capacity of Pb. In Pb-spiked treatments, roots accumulated more than 85% of total plant Pb which implies that kenaf root can be an important sink for bioavailable Pb. Scanning transmission electron microscope (STEM) X-ray microanalysis confirmed that electron-dense deposits located along cell walls of kenaf roots were Pb precipitates. The ability of kenaf to tolerate Pb and avoid phytotoxicity could be attributed to the immobilization of Pb in the roots and hence the restriction of upward movement (translocation factor < 1). With the application of fertilizer, kenaf was also found to have higher biomass and subsequently higher bioaccumulation capacity, indicating its suitability for phytoremediation of Pb-contaminated site.
