Many workers can be exposed simultaneously to heat and volatile chemicals. In a controlled human exposure study, it was observed that an increase in ambient temperature was associated with increased blood concentrations for acetone and toluene. Based on the expected changes in physiological parameters that occur with an increase in ambient temperature, we aimed to develop a PBPK model for acetone and toluene that could account for the impact of temperature on the kinetics of these solvents. Changes in temperature-dependent physiological parameters (i.e. blood flows, cardiac output, alveolar ventilation) based on recent measurements in volunteers were introduced in the PBPK models to simulate observed blood concentrations for different temperature exposure conditions. Because initial simulations did not adequately predict solvent kinetics at any temperature, the most sensitive parameter (alveolar ventilation; Qp) was, therefore, optimized on experimental acetone blood concentrations to obtain a relationship with temperature. The new temperature-dependent Qp relationship gave Qp values consistent with the literature and estimated a mean increase of 19% at 30 °C (wet bulb globe temperature) compared to 21 °C. The integration of a new temperature-dependent Qp relationship in the PBPK toluene model yielded adequate simulations of the experimental data for toluene in blood, exhaled air and urine. With further validation with other solvents, the temperature-dependant PBPK model could be a useful tool to better assess the risks of simultaneous exposure to volatile chemicals and heat stress and interpret biomonitoring data in workers as well as in the general population. BACKGROUND: Worldwide, lung cancer is the second leading cause of cancer death in women. The present study explored associations between occupational exposures that are prevalent among women, and lung cancer. METHODS: Data from 10 case-control studies of lung cancer from Europe, Canada, and New Zealand conducted between 1988 and 2008 were combined. Lifetime occupational history and information on nonoccupational factors including smoking were available for 3040 incident lung cancer cases and 4187 controls. We linked each reported job to the Canadian Job-Exposure Matrix (CANJEM), which provided estimates of probability, intensity, and frequency of exposure to each selected agent in each job. For this analysis, we selected 15 agents (cleaning agents, biocides, cotton dust, synthetic fibers, formaldehyde, cooking fumes, organic solvents, cellulose, polycyclic aromatic hydrocarbons from petroleum, ammonia, metallic dust, alkanes C18+, iron compounds, isopropanol, and calcium carbonate) that had lifetime exposure prevalence of at least 5% in the combined study population. For each agent, we estimated lung cancer risk in each study center for ever-exposure, by duration of exposure, and by cumulative exposure, using separate logistic regression models adjusted for smoking and other covariates. We then estimated the meta-odds ratios using random-effects meta-analysis. RESULTS AND CONCLUSIONS: None of the agents assessed showed consistent and compelling associations with lung cancer among women. The following agents showed elevated odds ratio in some analyses: metallic dust, iron compounds, isopropanol, and organic solvents. Future research into occupational lung cancer risk factors among women should prioritize these agents.

In recent years, accumulating evidence supports that occupational exposure to solvents is associated with an increased incidence of Parkinson's disease (PD) among workers. The neurotoxic effects of 1-bromopropane (1-BP), a widely used new-type solvent, are well-established, yet data on its relationship with the etiology of PD remain limited. Simultaneously, high-fat consumption in modern society is recognized as a significant risk factor for PD. However, whether there is a synergistic effect between a high-fat diet and 1-BP exposure remains unclear. In this study, adult C57BL/6 mice were fed either a chow or a high-fat diet for 18 weeks prior to 12-week 1-BP treatment. Subsequent neurobehavioral and neuropathological examinations were conducted to assess the effects of 1-BP exposure on parkinsonian pathology. The results demonstrated that 1-BP exposure produced obvious neurobehavioral abnormalities and dopaminergic degeneration in the nigral region of mice. Importantly, a high-fat diet further exacerbated the impact of 1-BP on motor and cognitive abnormalities in mice. Mechanistic investigation revealed that mitochondrial damage and mtDNA release induced by 1-BP and high-fat diet activate NLRP3 and cGAS-STING pathway- mediated neuroinflammatory response, and ultimately lead to necroptosis of dopaminergic neurons. In summary, our study unveils a potential link between chronic 1-BP exposure and PD-like pathology with motor and no-motor defects in experimental animals, and long-term high-fat diet can further promote 1-BP neurotoxicity, which underscores the pivotal role of environmental factors in the etiology of PD.

Bis-(2-Chloroisopropyl) ether (BCIPE) was used as a solvent for fats, greases, paint, varnish removers, and in spotting and cleaning solutions. However, BCIPE has not been commercially manufactured or used for numerous years. In experimental animal studies, BCIPE is moderately toxic following acute oral, dermal, and inhalation routes of exposure. BCIPE is a severe eye irritant but not a dermal irritant or dermal sensitizer. BCIPE was not genotoxic or mutagenic in in vitro and in vivo assays; it was not toxic in a 3-generation reproductive dietary study in rats. Short-term, repeated inhalation and oral exposure in rats produced increased liver and kidney weights and congestion; dermal exposure in rabbits did not produce any observable adverse effects. BCIPE did not produce a statistically significant increase in tumors in two different 2-year dietary studies in mice and rats. In mice, technical grade BCIPE produced increased incidences of alveolar/bronchiolar adenomas in females, hepatocellular carcinomas in males, and a low incidence of forestomach hyperplasia (in both sexes at the high-dose). Further investigation with technical grade BCIPE concluded that these effects were species- and dose-specific with limited, if any, relevance to humans. The NOAEL of 400 ppm (15 mg/kg/day) from the 2-year dietary study in female rats was considered the point of departure for the health-based WEEL derivation. After adjustment for duration of exposure, interindividual variability, and intraindividual variability, an 8-h time-weighted average (TWA) WEEL value of 3 ppm (21 mg/m3) was derived. This exposure limit is expected to provide a significant margin of safety against any potential adverse health effects in workers.

BACKGROUND: The laundry and dry cleaning industries are critical for maintaining cleanliness and hygiene in our daily lives. However, they have also been identified as sources of hazardous chemical exposure for workers, leading to potentially severe health implications. Despite mounting evidence that solvents like perchloroethylene and trichloroethylene are carcinogenic, they remain commonly used in the industry. Additionally, while alternative solvents are increasingly being utilized in response to indications of adverse health and environmental effects, there remains a significant gap in our understanding of the potential risks associated with exposure to these new agents. METHODS: This study aims to identify gaps in the literature concerning worker exposure to contemporary toxic chemicals in the laundry and dry cleaning industry and their associated carcinogenic risks. A scoping review of peer-reviewed publications from 2012 to 2022 was conducted to achieve this objective, focusing on studies that detailed chemical exposures, sampling methods, and workers within the laundry and dry cleaning sector. RESULTS: In this scoping review, 12 relevant papers were assessed. A majority (66%) examined perchloroethylene exposure, with one notable finding revealing that biomarkers from dry cleaners had significant micronuclei frequency and DNA damage, even when exposed to PCE at levels below occupational exposure limits. Similarly, another study supported these results, finding an increase in early DNA damage among exposed workers. Separate studies on TCE and benzene presented varied exposure levels and health risks, raising concern due to their IARC Group 1 carcinogen classification. Information on alternative solvents was limited, highlighting gaps in health outcome data, exposure guidelines, and carcinogenic classifications. CONCLUSION: Research on health outcomes, specifically carcinogenicity from solvent exposure in dry cleaning, is limited, with 66% of studies not monitoring health implications, particularly for emerging solvents. Further, findings indicated potential DNA damage from perchloroethylene, even below set occupational limits, emphasizing the need to reevaluate safety limits. As alternative solvents like butylal and high-flashpoint hydrocarbons become more prevalent, investigations into the effects of their exposure are necessary to safeguard workers' health. In this discussion paper, we describe the history of the science and societal action resulting in the mitigation of neurotoxic disorders from exposure to organic solvents at the workplaces in Sweden. When alkyd paints were introduced in large scale in construction painting in the 1960s and 1970s, Scandinavian unions voiced increasing concern as members reported symptoms like headache and vertigo, supported by participatory studies and case studies. Although acute and chronic neurotoxic effects were established for some specific solvents such as carbon disulphide, this was not the case for those used in the new paints. Union advocacy promoted formal epidemiological studies, providing increasing evidence for chronic neurotoxicity at levels far below current occupational exposure levels. The results were widely disseminated and accepted and led to concerted action with preventive measures, most importantly substitution of the organic solvents in paints for indoor use, but also drastic reductions in occupational exposure limits. The findings also resulted in funding of further research on solvent toxicity and the establishment of expert groups to advice authorities on occupational standards for exposure to chemicals. The substitution strategy was subsequently adopted in many other countries and occupational exposure limits were lowered, although several years or even decades later. While the societal context in Sweden was unique in many ways, we conclude that there are lessons to be learned from this preventive success when addressing current challenges.

Deep eutectic solvents (DESs) are preferable in terms of starting materials, storage and synthesis, simplicity, and component material affordability. In several industries ranging from chemical, electrochemical, biological, biotechnology, material science, etc., DES has demonstrated remarkable potential. Despite all these accomplishments, the safety issue with DES must be adequately addressed. Different DES interacts with the cellular membranes differently. It is not possible to classify all DES as easily biodegradable. By expanding the current understanding of the toxicity and biodegradation of DES, interactions between organisms and cellular membranes can be linked. The DES toxicity profile varies according to their concentration, the nature of the individual components, and how they interact with living things. Therefore, the results of this review can serve as a baseline for DES development in the future.

A comparative toxicity of widely applied organic solvents (methanol, ethanol, n-propanol, i-propanol, n-butanol, 2-butanol, i-butanol, t-butanol, 3-methoxy-3-methylbutanol-1 (MMB), ethylene glycol, diethylene glycol, 2-methoxyethanol, 2-ethoxyethanol, glycerol, ethyl acetate, acetonitrile, benzene, dioxane, dimethylformamide, dimethylacetamide, dimethylsulfoxide, 2-pyrrolidone, and N-methyl-2-pyrrolidone) and surfactants (PEG 300, PEG 6000, Tween 20, Tween 80, miramistin, and Cremophor EL) was studied using a sea urchin embryo model. Sea urchin embryo morphological alterations caused by the tested chemicals were described. The tested molecules affected P. lividus embryo development in a concentration-dependent manner. The observed phenotypic anomalies ranged from developmental delay and retardation of plutei growth to formation of aberrant blastules and gastrules, cleavage alteration/arrest, and embryo mortality. Discernible morphological defects were found after embryo exposure with common pharmaceutical ingredients, such as glycerol, Tween 80, and Cremophor EL. In general, solvents were less toxic than surfactants. PEG 6000 PEG 300, DMSO, ethanol, and methanol were identified as the most tolerable compounds with minimum effective concentration (MEC) values of 3.0-7.92 mg/mL. Previously reported MEC value of Pluronic F127 (4.0 mg/mL) fell within the same concentration range. Toxic effects of methanol, ethanol, DMSO, 2-methoxyethanol, 2-ethoxyethanol, Tween 20, and Tween 80 on P. lividus embryos correlated well with their toxicity obtained using other cell and animal models. The sea urchin embryos could be considered as an appropriate test system for toxicity assessment of solvents and surfactants for their further application as solubilizers of hydrophobic molecules in conventional in vitro cell-based assays and in vivo mammalian models. Nevertheless, to avoid adverse effect of a solubilizing agent in ecotoxicological and biological experiments, the preliminary assessment of its toxicity on a chosen test model would be beneficial.

The safety of propylene glycol (PG) and vegetable glycerin (VG) as solvents in electronic cigarette liquid has received increasing attention and discussion. However, the conclusions derived from toxicity assessments conducted through animal experiments and traditional in vitro methodologies have consistently been contentious. This study constructed an original real-time aerosol exposure system, centered around a self-designed microfluidic bionic-lung chip, to assess the biological effects following exposure to aerosols from different solvents (PG, PG/VG mixture alone and PG/VG mixture in combination with nicotine) on BEAS-2B cells. The study aimed to investigate the impact of aerosols from different solvents on gene expression profiles, intracellular biomarkers (i.e., reactive oxygen species content, nitric oxide content, and caspase-3/7 activity), and extracellular biomarkers (i.e., IL-6, IL-8, TNF-α, and malondialdehyde) of BEAS-2B cells on-chip. Transcriptome analyses suggest that ribosomal function could serve as a potential target for the impact of aerosols derived from various solvents on the biological responses of BEAS-2B cells on-chip. And the results showed that aerosols of PG/VG mixtures had significantly less effect on intracellular and extracellular biomarkers in BEAS-2B cells than aerosols of PG, whereas increasing nicotine levels might elevate these effects of aerosol from PG/VG mixture.

The heterogeneity and complexity of solvent-extracted organic matter associated with PM2.5 (SEOM-PM2.5) is well known; however, there is scarce information on its biological effects in human cells. This work aimed to evaluate the effect of SEOM-PM2.5 collected in northern Mexico City during the cold-dry season (November 2017) on NL-20 cells, a human bronchial epithelial cell line. The SEOM obtained accounted for 15.5% of the PM2.5 mass and contained 21 polycyclic aromatic hydrocarbons (PAHs). The cell viability decreased following exposure to SEOM-PM2.5, and there were noticeable morphological changes such as increased cell size and the presence of cytoplasmic vesicles in cells treated with 5-40 µg/mL SEOM-PM2.5. Exposure to 5 µg/mL SEOM-PM2.5 led to several alterations compared with the control cells, including the induction of double-stranded DNA breaks based (p < 0.001); nuclear fragmentation and an increased mitotic index (p < 0.05); 53BP1 staining, a marker of DNA repair by non-homologous end-joining (p < 0.001); increased BiP protein expression; and reduced ATF6, IRE1α, and PERK gene expression. Conversely, when exposed to 40 µg/mL SEOM-PM2.5, the cells showed an increase in reactive oxygen species formation (p < 0.001), BiP protein expression (p < 0.05), and PERK gene expression (p < 0.05), indicating endoplasmic reticulum stress. Our data suggest concentration-dependent toxicological effects of SEOM-PM2.5 on NL-20 cells, including genotoxicity, genomic instability, and endoplasmic reticulum stress.

Apis mellifera was used as a model species for ecotoxicological testing. In the present study, we tested the effects of acetone (0.1% in feed), a solvent commonly used to dissolve pesticides, on bees exposed at different developmental stages (larval and/or adult). Moreover, we explored the potential effect of in vitro larval rearing, a commonly used technique for accurately monitoring worker exposure at the larval stage, by combining acetone exposure and treatment conditions (in vitro larval rearing vs. in vivo larval rearing). We then analyzed the life-history traits of the experimental bees using radio frequency identification technology over three sessions (May, June, and August) to assess the potential seasonal dependence of the solvent effects. Our results highlight the substantial influence of in vitro larval rearing on the life cycle of bees, with a 47.7% decrease in life span, a decrease of 0.9 days in the age at first exit, an increase of 57.3% in the loss rate at first exit, and a decrease of 40.6% in foraging tenure. We did not observe any effect of exposure to acetone at the larval stage on the capacities of bees reared in vitro. Conversely, acetone exposure at the adult stage reduced the bee life span by 21.8% to 60%, decreased the age at first exit by 1.12 to 4.34 days, and reduced the foraging tenure by 30% to 37.7%. Interestingly, we found a significant effect of season on acetone exposure, suggesting that interference with the life-history traits of honey bees is dependent on season. These findings suggest improved integration of long-term monitoring for assessing sublethal responses in bees following exposure to chemicals during both the larval and adult stages. El uso de inhalantes como drogas de abuso es una práctica de fácil acceso en menores y jóvenes tanto en países de bajos ingresos como en los de altos ingresos. Dentro de las complicaciones que se pueden presentar se encuentra el neumotórax espontáneo, que se ha relacionado tanto con el daño en el parénquima pulmonar producido por la sustancia y con la técnica de inhalación y/o exposición generando barotrauma. Se presenta un reporte de caso de un hombre joven que inhaló pegante y desarrolló un neumotórax espontáneo completo del pulmón izquierdo. Requirió intervención quirúrgica con colocación de sonda a tórax y su evolución fue satisfactoria. Esta complicación se ha reportado en la literatura con mayor frecuencia en hombres y con el uso de cocaína. Sólo se identificó otro caso similar en el contexto de inhalación de pegante con tolueno en sus componentes. Debido a los pocos reportes encontrados respecto al uso de sustancias inhalantes y al tolueno, es importante resaltar el hallazgo a fin de contribuir al inicio de futuras investigaciones a mayor escala. Inhalants as drugs of abuse is an accessible practice in minors and young people in low-income countries as well as high-income ones. Spontaneous pneumothorax is a possible complication, which has been related both to damage of the lung parenchyma produced by the substance and inhalation and/or exposure technique making barotrauma. A case report of young man who inhaled glue with toluene and developed a complete spontaneous pneumothorax of the left lung is presented. He required surgical intervention with placement of a chest tube and his evolution was satisfactory. This complication has been reported in the literature more frequently in men and with the use of cocaine. Only one other similar case report was identified in the context of inhalation of glue with toluene in its components. Due to the few reports found regarding use of inhalant substances and toluene, it is important to highlight the finding to contribute to the initiation of future research on a larger scale. (AU)

Abstract Introduction: Car painters are routinely exposed to organic solvents classified as carcinogenic and mutagenic substances. Objective: To characterize the population susceptibility and evaluate the genotoxic effects of exposure to organic solvents. Methods: A cross-sectional study comparing a group of car painters exposed to organic solvents with a non-exposed group. CYP2E1 polymorphisms and the presence of micronuclei in lymphocytes were determined. Results: One hundred twenty-two workers participated in the study: 62 who worked in car paint shops and were exposed to solvents, and 60 who were not exposed. There were statistically significant differences between the two groups regarding micronucleated cells and nucleoplasmic bridges frequencies (p=0.042 and p=0.046, respectively; exact likelihood ratio). Significant differences were found at the interaction between the CYP2E1 genotype c1c1 and occupational exposure to solvents, with higher frequencies of micronuclei (p= 0.013) and micronucleated cells (p= 0.015). However, when the frequencies of micronuclei, micronucleated cells and nucleoplasmic bridges in the exposure group were compared between the c1c1 and c2c2/c1c2 allele groups of the CYP2E1 polymorphism, statistically significant differences were found. Conclusions: This study confirms that when workers with CYP2E1 polymorphisms, specifically the c1c1 genotype, are exposed to organic solvents, they are more likely to have somatic cell mutations, a condition associated with increased susceptibility to diseases such as cancer


Resumen Introducción: Los pintores de vehículos automotores están rutinariamente expuestos a agentes como los solventes orgánicos, capaces de producir efectos mutágenos y carcinógenos. Objetivo: Caracterizar la susceptibilidad poblacional y evaluar los efectos genotóxicos debidos a la exposición a solventes orgánicos. Métodos: Estudio de corte transversal que comparó a un grupo de pintores de carros expuestos a solven tes orgánicos con un grupo de personas no expuestas. Fueron determinados tanto los polimorfismos de CYP2E1 como la presencia de micronúcleos en linfocitos. Resultados: Participaron 122 personas, 62 trabajadores de talleres de pintura de autos expuestos a solventes y 60 personas no expuestas. Con relación al cuestionario Q 16, 32% de los expuestos refirieron síntomas sugestivos de neurotoxicidad. Las frecuencias de células micronucleadas y de puentes nucleoplásmicos fueron significativamente mayores en los expuestos que en los no expuestos: p= 0.042 y p= 0.046, respectivamente, Razón de verosimilitud exacta). Fueron halladas diferencias significativas en la interacción de CYP2E1 (c1c1) y la exposición ocupacional a solventes, con mayores frecuencias de micronúcleos (p= 0.013) y de células micronucleadas (p= 0.015). Conclusiones: Este estudio reafirma que los trabajadores expuestos a solventes orgánicos con polimorfismos de CYP2E1, específicamente con genotipo c1c1, tienen mayor probabilidad de presentar mutaciones en las células somáticas, condición asociada con una mayor susceptibilidad a enfermedades como el cáncer

Abstract: Objective: To assess neuropsychological performance among workers at a paint factory in Mexico City. Materials and methods: During 2004 and 2005 we assessed the neuropsychological performance of 208 workers who were exposed to organic solvents at a paint factory, mainly toluene and xylene. We categorized workers into low and high exposure groups using a cumulative index for toluene, based on times spent in different tasks. We evaluated cognitive and motor functions with 13 neuropsychological tests. Results: We found lower attention, longer time to complete the test β=5.5 (R2=12.3%), and a lower score in the motor-cognitive test β=-15.7 (R2=19.5%) in the high exposure group through multiple linear regression model analysis, with adjustment for age and education. Conclusion: Our results are similar to the ones reported in the literature, but the effects are less severe, probably due to lower exposure to organic solvents.


Resumen: Objetivo: Evaluar el desempeño neuropsicológico de los trabajadores de una fábrica de pintura en la Ciudad de México. Material y métodos: Se evaluó el desempeño neuropsicológico de 208 trabajadores expuestos a disolventes orgánicos, principalmente tolueno y xileno, en una fábrica de pintura en la Ciudad de México durante 2004 y 2005. Se categorizaron en grupos de baja y alta exposición con un índice acumulado de tolueno con base en el tiempo empleado por actividad. Se evaluaron funciones motoras y cognitivas con 13 pruebas neuropsicológicas. Resultados: Se registró un tiempo más largo para completar la prueba β=5.5 (R2=12.3%) y una puntuación baja en la prueba motor-cognitiva de β=-15.7 (R2=19.5%) en el grupo de alta exposición en los modelos de regresión lineal múltiple, ajustados por confusores. Conclusión: Los presentes resultados son similares a los reportados en la literatura, aunque los efectos son menos graves, probablemente debido a la baja exposición.

Resumen Introducción. La exposición a solventes orgánicos y pinturas se ha asociado con efectos genotóxicos y mayor riesgo de neoplasias. Sin embargo, aún no se ha caracterizado bien el tipo de daño que esta exposición induce en el ADN humano, ni los mecanismos por los cuales se genera. Uno de los grupos con mayor exposición a dichos solventes y pinturas son los pintores de automóviles del sector informal que trabajan sin adecuadas prácticas de seguridad ocupacional. Objetivo. Determinar el daño oxidativo y por metilación del ADN de linfocitos de pintores de automóviles expuestos a solventes orgánicos y pinturas. Materiales y métodos. Se analizaron linfocitos aislados de sangre periférica de 62 pintores y 62 sujetos no expuestos mediante el ensayo cometa de gran eficiencia acoplado a las enzimas Fpg y AlkA. Las categorías de daño en el ADN evaluadas fueron el daño basal (sin enzimas), el daño oxidativo y el daño por metilación, y el parámetro de medición, el porcentaje de ADN en la cola. Resultados. El porcentaje de ADN en la cola fue mayor en el grupo expuesto con respecto al no expuesto (p<0,05). En el grupo expuesto, dicho porcentaje fue mayor en la categoría de daño oxidativo comparado con la del basal (16,50 Vs. 12,87; p<0,001), en tanto que en el daño por metilación no se encontraron diferencias significativas (14,00 Vs. 12,87; p>0,05). Conclusión. La exposición a solventes orgánicos y pinturas se asoció con el aumento de las lesiones oxidativas del ADN de los linfocitos de pintores de automóviles, tales como la producción de 8-oxo-2'-desoxiguanosina (8-oxodG) y otros productos formamidopirimidina, los cuales se consideran considerablemente mutagénicos.


Abstract Introduction: The exposure to organic solvents and paints has been associated with genotoxicity and a greater risk of neoplasms. However, the type of DNA damage induced in humans by the exposure to these compounds, which would help explain the mechanisms of their genotoxicity, is still not fully characterized. Due to inadequate practices of occupational safety, car painters in the informal sector are a highly exposed group to organic solvents and paints. Objective: To identify the oxidative and methylating damage in the DNA of lymphocytes of car painters exposed to organic solvents and paints. Materials and methods: Isolated peripheral blood lymphocytes from 62 painters and 62 unexposed subjects were analyzed by the modified high-throughput comet assay with the Fpg and AlkA enzymes. The categories used for the evaluation of the DNA damage were basal damage (without enzymes), oxidative and methylating damage. The measurement parameter used to establish the damage was the percentage of DNA in the tail. Results: The percentage of DNA in the tail was higher in the exposed group compared to the unexposed group (p<0.05). In the exposed group, this percentage was higher in the oxidative damage category than the baseline (16.50 vs. 12.87; p<0.001), whereas methylating damage did not show significant differences (14.00 vs. 12.87; p>0.05). Conclusion: In this study, exposure to organic solvents and paints was associated with an increase in oxidative lesions in the DNA of car painters' lymphocytes, such as the production of 8-oxodG and other formamidopyrimidine products which are considered highly mutagenic.

Abstract Recently, there has been a lot of interest in the utilization of rhodococci in the bioremediation of petroleum contaminated environments. This study investigates the response of Rhodococcus erythropolis IBBPo1 cells to 1% organic solvents (alkanes, aromatics). A combination of microbiology, biochemical, and molecular approaches were used to examine cell adaptation mechanisms likely to be pursued by this strain after 1% organic solvent exposure. R. erythropolis IBBPo1 was found to utilize 1% alkanes (cyclohexane, n-hexane, n-decane) and aromatics (toluene, styrene, ethylbenzene) as the sole carbon source. Modifications in cell viability, cell morphology, membrane permeability, lipid profile, carotenoid pigments profile and 16S rRNA gene were revealed in R. erythropolis IBBPo1 cells grown 1 and 24 h on minimal medium in the presence of 1% alkanes (cyclohexane, n-hexane, n-decane) and aromatics (toluene, styrene, ethylbenzene). Due to its environmental origin and its metabolic potential, R. erythropolis IBBPo1 is an excellent candidate for the bioremediation of soils contaminated with crude oils and other toxic compounds. Moreover, the carotenoid pigments produced by this nonpathogenic Gram-positive bacterium have a variety of other potential applications.

OBJECTIVE: To compare hearing performance relating to the peripheral and central auditory system between solvent-exposed and non-exposed workers. METHODS: Forty-eight workers exposed to a mixture of solvents and 48 non-exposed control subjects of matched age, gender and educational level were selected to participate in the study. The evaluation procedures included: pure-tone audiometry (500 - 8,000 Hz), to investigate the peripheral auditory system; the Random Gap Detection test, to assess the central auditory system; and the Amsterdam Inventory for Auditory Disability and Handicap, to investigate subjects' self-reported hearing performance in daily-life activities. A Student t test and analyses of covariance (ANCOVA) were computed to determine possible significant differences between solvent-exposed and non-exposed subjects for the hearing level, Random Gap Detection test and Amsterdam Inventory for Auditory Disability and Handicap. Pearson correlations among the three measures were also calculated. RESULTS: Solvent-exposed subjects exhibited significantly poorer hearing thresholds for the right ear than non-exposed subjects. Also, solvent-exposed subjects exhibited poorer results for the Random Gap Detection test and self-reported poorer listening performance than non-exposed subjects. Results of the Amsterdam Inventory for Auditory Disability and Handicap were significantly correlated with the binaural average of subject pure-tone thresholds and Random Gap Detection test performance. CONCLUSIONS: Solvent exposure is associated with poorer hearing performance in daily life activities that relate to the function of the peripheral and central auditory system.


OBJETIVO: Comparar o desempenho das atividades diárias relacionadas a funções do sistema auditivo periférico e central entre trabalhadores expostos e não expostos a solventes. MÉTODOS: Participaram do estudo 96 trabalhadores, sendo 48 expostos a solventes e 48 não expostos, pareados por escolaridade, idade e sexo. Os procedimentos de avaliação incluíram: audiometria de tons puros (500 a 8.000 Hz), para avaliar o sistema auditivo periférico; teste de Random Gap Detection, para avaliar o sistema auditivo central; e o Amsterdam Inventory for Auditory Disability and Handicap, para estudar a performance em atividades da vida diária que envolvem a audição, por meio de autoavaliação. Teste t de student e a análise de covariância (ANCOVA) foram utilizados. Foram calculadas as correlações de Pearson entre os resultados dos três testes. RESULTADOS: Sujeitos expostos a solventes tiveram limiares auditivos significativamente piores na orelha direita que os sujeitos não expostos. Apresentaram também resultados significativamente piores no teste de Random Gap Detection e funcionamento autorreportado significativamente mais alterado que os sujeitos não expostos. Foi observada correlação significativa entre os resultados do Amsterdam Inventory for Auditory Disability and Handicap e a média binaural dos limiares auditivos e com os resultados do teste de Random Gap Detection. CONCLUSÕES: A exposição a solventes está associada com as dificuldades da vida diária relacionadas com as funções do sistema auditivo periférico e central.

CONCLUSIONES: La exposición a solventes está asociada con las dificultades de la vida diaria relacionadas con las funciones del sistema auditivo periférico y central.

Introdução: O xilol é necessário para a diafanização de amostras citopatológicas. Pode trazer problemas de ordem ocupacional, proteção ao meio ambiente e de custos. Objetivo:Avaliar o desempenho de várias concentrações da mistura verniz/xilol em relação à diafanização e conservação em amostras de citologia coradas com a técnica dePapanicolaou (CP). Material e métodos: Foram avaliadas 75 lâminas de raspado bucal (RB) e 8.773 esfregaços cérvico-vaginais (CV). As lâminas foram coradas pela CP, com a retirada das etapas de álcool/xilol e xilol e foram secas à temperatura ambiente porvinte minutos antes da montagem com lamínula. A montagem foi realizada com soluções de verniz/xilol de concentrações 75/25%, 70/30%, 60/40%, 50/50% e 40/60%. As lâminas foram distribuídas de forma aleatória aos profissionais para avaliação da técnica, sendo conceituadas como boas, regulares ou ruins e reavaliadas após nove meses dearquivamento. A concentração de verniz/xilol que apresentou melhor qualidade final nas lâminas de RB foi aplicada também nos CV. Resultados: A concentração de 75/25% deverniz/xilol apresentou melhor desempenho para as duas situações, em RB e CV. Após nove meses, a qualidade foi mantida. Conclusão: A concentração de 75/25% apresentou melhor resultado.

Introdução: Milhões de pessoas são expostas diariamente aos solventes. Inúmeros solventes podem causar intoxicação aguda. Menos clara é a associação da exposição crônica e em baixas doses poder produzir alteração neurológica. Diversas atividades ocupacionais estão envolvidas com a sua exposição. Condição bastante diferente é o seu uso inadvertido com intuito alucinógeno. Método: Trata-se de revisão atualizada da literatura a partir de pesquisa na base de dados MEDLINE e LILACS. Também utilizou-se livros e documentos publicados em formato eletrônico.Discussão: Distúrbios neuromusculares, distúrbios do movimento, alterações cognitivo-comportamentais e neurofisiológicas têm sido relacionados a inúmeros agentes tóxicos nos últimos anos. A proposta deste artigo é revisar os principais distúrbios neurológicos associados à exposição crônica por solventes orgânicos. A fim de facilitar a abordagem inicial no atendimento ambulatorial aos distúrbios neurotoxicológicos foram confeccionadas tabelas descrevendo os principais agentes tóxicos, as fontes de exposição envolvidas e suas principais manifestações neurológicas. Conclusão: Tolueno, bissulfeto de carbono e n-Hexano são alguns solventes envolvidos nos distúrbios neurotoxicológicos, Contudo, fica evidente nesta revisão que são necessários novos estudos a fim de determinar a real associação destes e outros solventes nos distúrbios crônicos do sistema nervoso central e periférico.


Introduction: Millions of peoples are exposed to solvents every day. Most solvents cause acute intoxication. Less evident is the association of chronic exposure and in low doses in producing neurological disorders. Innumerable occupational activities are involved in the exposure to solvents. Their inadvertent use with hallucinogenous intention is an entirely different condition. Method: The method consists of an updated review of the literature based on research in the MEDLINE and LILACS databases, as well as books and documents published online. Discussion: Neuromuscular disorders, movement disorders, cognitive-behavioral and neurophysiological changes have been attributed to innumerable toxic agents in recent years. This article proposes to review the main neurological disorders associated with chronic exposure to organic solvents. To facilitate the initial approach to treatment of neurotoxicological disorders of outpatient, tables were devised to describe the main toxic agents, the sources of exposure involved and their main neurological manifestations. Conclusions: Toluene, carbon bisulfate and n-hexane are some of the solvents involved in neurotoxicological disorders. However, this review reveals the need for new studies to determine the real association of these and other solvents in chronic disorders of the central and peripheral nervous systems.

No âmbito da saúde ocupacional, as exposições a agentes químicos recebem cada vez mais atenção. Pela análise de bioindicadores, se faz possível identificar e quantificar substâncias nocivas à saúde e seus efeitos. Trabalhadores de postos de revenda de combustíveis veiculares (PRCV) estão expostos a uma mistura de hidrocarbonetos policíclicos aromáticos (HPAs) proveniente da gasolina, e nesta mistura encontra-se o benzeno, composto hematotóxico e carcinogênico, sem níveis seguros no ambiente, fato que corrobora para a importância de ações em vigilância ocupacional nesta área. O presente estudo avaliou os efeitos genotóxicos, através do teste de Aberrações cromossômicas (ACs), de uma exposição ocupacional aos vapores de gasolina em frentistas da Zona Oeste do Rio de Janeiro. O projeto central foi aprovado pelo Comitê de Ética em Pesquisa da Ensp/Fiocruz (CAAE17438013.5.0000.5240, parecer 434.418). 39 trabalhadores de seis PRCV da área programática 5.3 (bairros de Santa Cruz, Paciência e Sepetiba) responderam a dois questionários sobre fatores socioeconômicos, processo de trabalho e histórico de saúde; amostras de sangue e urina foram coletadas para determinação de hemograma, análises bioquímicas, ACs e ácido trans, trans-mucônico urinário (ATTM). Os voluntários não apresentaram histórico de doenças de possível associação com a exposição, assim os resultados de hemograma e bioquímica, em valores médios, ficaram dentro das faixas de normalidade. Os processos de trabalho e hábitos dos trabalhadores foram avaliados e não apresentaram relação com as ACs. (...) Os resultados sugerem que o teste de ACs não deve ser utilizado de forma isolada para avaliar exposições a baixas doses de benzeno por sua inespecificidade, mas ainda representa uma ferramenta útil na avaliação de dano precoce à saúde humana...


On the Occupational Health field, exposure to chemical agents is getting more attention, since technologies’ advances allow identification of health early effects caused by such exposures. By the use of bioindicator’s analysis, it is possible to identify and quantify harmful substances and their effects. Gas station workers are exposed to a hidrocarbon’s mixture from gasoline, in which can be found benzene, carcinogenic and hematotoxic substance, with no safe levels on environment, reinforcing the need of occupational vigilance actions. This study evaluated the genotoxic effecs, using the chromosomal aberration’s test (CAs), of an occupational exposure to gasoline fumes in gas station workers from Rio de Janeiro’s West Side. The main project was approved by Ensp/Fiocruz Research’s Ethics Committee (CAAE 17438013.5.0000.5240, case 434.418). Thirty-nine workers from six gas stations from the districts of Santa Cruz, Paciência and Sepetiba answered questionnaires concerning socio-demographic, work and health questions; blood and urine samples were taken to assess CBC, biochemical factors, trans, trans-muconic acid (TTMA) and chromosomal aberrations. The subjects did not report history of diseases that could be related to the exposure, so the results of CBC and biochemical factors were at normality ranges. Work processes and the subjects’habits were assessed and were not associated to CAs. TTMA measurement and statistical evaluation through ANOVA showed a correlation between this metabolite and smoking habit (...), and the high concentration gets along with literature. (...) The results suggest chromosomal aberrations test shall not be used as the only mechanism to evaluate benzene exposure at lower doses for its inespecificity, but the test is, for it self, a useful feature for early human health damage evaluation...
