الملخص

Objective::To observe the clinical efficacy of modified Shenling Baizhusan on stroke-associated pneumonia (SAP) with lung-Qi deficiency syndrome and its regulatory effect on inflammatory factors and immune function. Method::One hundred and ten patients were randomly divided into control group (55 cases) and observation group (55 cases) by random number table. Patients in control group got piperacillin sodium and tazobactam sodium for slow intravenous injection, 4.5 g/time, 3 times/days, for 7-10 days, and new antibiotic was chose by drug outcomes, ambroxol hydrochloride injection for low intravenous infusion, 30 mg/time, 2 times/days, for 10 days, and nutritional support and symptomatic comprehensive treatment. In addition to the therapy in control group, patients in observation group were added with modified Shenling Baizhusan, 1 dose/day, for 10 days. And scores of pulmonary infection (CPIS), time of CPIS<6 time of disappearance of cough, time of recovery of temperature, time of recovery of leukocyte, and time of disappearance of lung rales were recorded. And before and after treatment, lung-Qi deficiency syndromes were scored, and levels of procalcitonin (PCT), γ-interferon (IFN-γ), tumor necrosis factor-α (TNF-α), high-sensitivity C-reactive protein (hs-CRP), immunoglobulin A (IgA), immunoglobulin G (IgG), immunoglobulin M (IgM), T Lymphocyte subsets (CD4+, CD8+ and CD4+ /CD8+) were all detected. Result::By rank sum test, the clinical effect in control group was better than that in control group (Z=2.106, P<0.05). Scores of CPIS and lung-Qi deficiency syndromes were lower than those in control group (P<0.01). And time of CPIS<6, time of disappearance of cough, time of recovery of temperature, time of recovery of leukocyte, and time of disappearance of lung moist rales were all shorter than those in control group (P<0.01). And levels of PCT, TNF-α, hs-CRP and CD8+ were lower than those in control group (P<0.01, P<0.05), while levels of IFN-γ, IgA, IgM, CD4+ and CD4+ /CD8+ were all higher than those in control group (P<0.01, P<0.05). Conclusion::In addition to the comprehensive anti-infection therapy, modified Shenling Baizhusan can control the degree of illness, alleviate symptoms, regulate the expressions of inflammatory factors, increase the immune function of the body, and improve the comprehensive efficacy.

<p><b>OBJECTIVE</b>To investigate the effect of 2-phenoxyethanol on potency of Sabin inactivated poliomyelitis vaccine (IPV).</p><p><b>METHODS</b>Sabin IPV samples containing 5 mg or 7 mg 2-phenoxyethanol each dosage respectively were placed separately at 4 degrees C, 37 degrees C for 2 days and 7 days. D-antigen contents were tested with ELISA method. Then neutralizing antibodies in mice and guinea pigs were detected. The safety experiment was performed according to unusual toxicity test of China requirement for biological product.</p><p><b>RESULTS</b>After addition of 2-phenoxyethanol, the I, II, and III D-antigen contents of Sabin IPV did not change. The antibody levels in mice and guinea pigs were not different between experimental group and control group. Animals were safe during observation period.</p><p><b>CONCLUSION</b>2-Phenoxyethanol had no effect on potency and safety of Sabin IPV. It can be used as antiseptic for Sabin IPV.</p>

<p><b>BACKGROUND</b>To study preparation of polyvalent DNA vaccine and the control of multiple gene expression.</p><p><b>METHODS</b>A bicistronic vector pcDNA3.0BA was constructed from pcDNA3.0. HCV PC154 gene and HBV preS2S gene were inserted into this vector to form bicistronic expression construct pcDNA3.0BAPC154S2S and monocistronic expression construct pcDNA3.0BAPC154 or pcDNA3.0BAS2S. These plasmids were transiently expressed in COS-7 cells and injected into muscles of BALB/c mice.</p><p><b>RESULTS</b>pcDNA3.0BA contains two cistronic units, which can co-express two kinds of genes, with the first immunogen gene and the second gene serving as additional immunogen or as modulator for the immune responses. HBV surface Ag and HCV core Ag were coexpressed in vitro. The antibody responses and lymphoproliferation to antigens were similar between bicistronic and monocistronic expression construct in mice.</p><p><b>CONCLUSION</b>pcDNA3.0BA is a novel vector, which can coexpress two proteins and elicit polyvalent immune responses.</p>

<p><b>OBJECTIVE</b>To observe the effects of diethlhexyl phthalate (DEHP) on lipid peroxidation and the life span in Drosophila melanogaster.</p><p><b>METHODS</b>Fed Drosophila with the concentration 0.20% DEHP of exposure after 0, 14, 28 days, the activity of total superoxide dismutase (SOD), CuZn-SOD and the concentration of malondialdehyde were determined. At the same time, the longevity test was carried out to examine the effect of DEHP on the Drosophila's lifespan.</p><p><b>RESULTS</b>The lifespan of Drosophila was shortened in a dose of DEHP exposed groups. The indexes of mean life span (MLS), 50% lethal time and mean maximum life span in three DEHP-treated groups (concentration of 0.05%, 0.10% and 0.20%) were lower than those of the controlled group respectively (P < 0.01 or P < 0.05). The MLS of both Drosophila sexes were reduced from the control of 64 days and 59 days to the test 60 days-52 days and 54 days-49 days respectively. DEHP decreased the activity of SOD (P < 0.01 or P < 0.05), and lead to a time-dependent relation and an increase in the concentration of malondialdehyde (P < 0.01 or P < 0.05) in the DEHP-exposed Drosophila groups.</p><p><b>CONCLUSION</b>DEHP might promote the process of lipid peroxidation and shorten the life span in Drosophila melanogaster. It should be one of the reasons in the senescence of Drosophila.</p>
