India is the second largest producer of fruits in the world with different fruit growing zones viz., temperate, arid and semi-arid, subtropical, and tropical zones. In this present era of climate change, fruit growers are facing environment-related problems such as sudden changes in temperature, irregular and heavy rainfall, and soil-related problems like compaction, salinity, alkalinity, and acidity. In addition to this, pest and disease incidence also play a major role in limiting fruit production. Objectives of rootstock breeding includes resistance or tolerance to biotic stress, adaptability to soil and environmental conditions, dwarfism, precocity in bearing, high yield and good fruit quality. The purpose of rootstock breeding varies with crops and geographical locations. Developing rootstocks resistant or tolerant to biotic stresses in apple (fire blight and woolly apple aphid), citrus (root rot, nematodes and viral diseases), grape (Phylloxera spp. and nematode) and mango (mango fruit fly and stone weevil) for specific tree characters (dwarfing, canopy management) and horticultural traits (yield and quality) are the important aspects in rootstock breeding. The reported rootstocks which are impactful in fruit cultivation like apple (MM-111), grape (Dogridge, 99-R, 110-R and 1103-P), and mango (Turpentine, Deorakhio and Olour). The use of rootstocks which are tolerant or adaptable to adverse climatic situations and biotic stress can be an alternative option for scientists and fruit growers to face the challenges encountered by the fruit industry.

Fennel plant (Foeniculum vulgare, Mill.) is one of most widely used plants in food and medicine. Our study aimed to show the response of fennel plants from Egypt and China planted in Egypt to foliar application of benzyladenine (BA) at various concentrations; control, 50, 100 and 150 ppm. The obtained results indicated that treated fennel plants with BA resulted in a significant increase in the vegetative growth, number of umbels/plant, fruit, yield and oil yield per plant and feddan as well as main components, total phenolic contents and antioxidant activity of essential oils compared to untreated plants. In general, the application of benzyladenine was more effective on Egyptian fennel plants than that of Chinese plants. Egyptian fennel plants gave higher values of growth, fruit yield, oil percentage and oil yield than Chinese plants. However, the major components, TPC and antioxidant activity (DPPH %) had higher values in Chinese essential oil than Egyptian essential oil. The highest values of all studied parameters were obtained by using BA at 100 ppm. GC–MS analysis of the volatile oils showed that the major components were trans-anethole followed by fenshone, ?- pinene and D-limonene, respectively for both essential oils. 100 ppm of BA gave the highest values of trans-anethole content and radical scavenging activities of oils. Therefore, we concluded that spraying fennel plants with 100 ppm BA can be used for obtaining higher vegetative growth, fruit and quantity and quality of oil.

Plants belonging to the genus Hypericum L. are widely used in traditional folk medicine due to their hypericin and pseudohypericin contents. In this study, we investigated the biological activity, phenolic and elemental content of methanol and acetone extracts of sheepskin grass, Hypericum heterophyllum Vent. a species that that lacks both hypericin and pseudohypericin. The total antioxidant status of the extracts was determined by commercial kits. Antibacterial effect of extracts was investigated on seven bacterial strains. Cytotoxic effects of the extracts on lung cancer cell lines were determined by MTT (3-4,5-dimethyl-thiazolyl-2,5-diphenyltetrazolium bromide) method. Phenolic content was determined by LC-ESI-MS/MS. Elements in the plant were determined by ICP-OES. The total phenolic content and antioxidant status of the species’ methanol extract were found to be higher than the acetone extract. Both of the extracts at a concentration of 20% have an antibacterial effect, especially the antibacterial effect of acetone extract. It was determined that acetone extract has an anticarcinogenic impact depending on the dose. Chlorogenic acid, miquelianin and isoquercitrin are the most abundant flavonoids in methanol and acetone extract. The plant contains Ca, K, and Mg elements in high concentrations. The phenolic substances and elements in H. heterophyllum, widely used in our country, have been presented for the first time in the literature. Besides, it can be said that the plant has antioxidant, anticarcinogen, and antimicrobial activities due to the crucial flavonoids and elements it contains

Introduction: Spice consumption is one of the globally recognized healthy nutritional practices. Most spices contain phenolic compounds that may prevent or prolong the onset of non-communicable diseases. The harvesting, processing and preparation procedures of such spices may influence the phenolic amount extracted and eventual biological availability. Literature about how extraction period and spice form affects total phenolic content yield in water infusions is scanty. Aim: This study determined the effect of spice form and length of extraction time on the total phenolic content (TPC) yield of the selected Ugandan spices infused in water. Methods: Samples of Ocimum gratissimum, Allium sativum, Cymbopogon citratus and Zingiber officinale, were collected in triplicates from Kanungu, Bushenyi and Lugazi Districts, in Uganda. Fresh and dry samples of these spices were infused in hot water for four minutes and 40 minutes and sieved with Whatman paper, No. 1. Phenolic content was measured with a spectrophotometer at Makerere University, Biochemistry Department, following Folin-ciocalteu method, using gallic acid as the reference standard. Results were analyzed using GraphPad Prism 8.0.1 software. Results: Higher TPC yield was generally observed in dry samples compared to the flesh ones and 40-minute extracts of both fresh and dry samples also had higher TPC content compared to the four minutes ones. The highest TPC yield was observed in Cymbopogon citratus (12.21±0.75 mg GAE/g) among the dry samples and Ocimum gratissimum (10.02±2.45 mg GAE/g) among the fresh samples, extracted for 40 minutes. Conclusion: Longer extraction time and sample dryness maximize TPC yield. Ocimum gratissimum and Cymbopogon citratus may benefit consumers by improving their antioxidant status.

Abstract Phoenix dactylifera L. has traditionally been used as a medicine in many cultures. The aim of this study was to evaluate the nutritional properties, aromatic compounds, total phenolic content and the antioxidant activity of ten ripe date fruit varieties grown in Tunisia. Sugar profiles were analyzed by high performance liquid chromatography, while fatty acid compounds were detected by gas chromatography and aromatic compounds were analyzed by GC-Electron Impact Mass Spectroscopy. Total phenolic contents were measured using colorimetric methods, whereas antioxidant capacities were evaluated in vitro using DPPH and ABTS radicals. It has been found that total sugars are the predominant component in all date varieties, followed by moisture, along with moderate amounts of proteins, ash, and fats. Multivariate tests based on the volatile compounds profile showed significant differences among varieties. Between the sixty-two volatile compounds detected, alcohols, aldehydes and unsaturated hydrocarbons constituted the main chemical classes. The date varieties exhibited strong antioxidant potential that correlated with phenolic content. In conclusion date varieties can play a major role in human nutrition and health because of their wide range of valuable nutritional components and natural antioxidants that could potentially be considered as a functional food ingredient.

Abstract The present study was conducted to assess the phenolic content, and antibacterial and antioxidant activities of Lathyrus L. species. The extraction of phenolic compounds from whole seeds, seed coat and cotyledon of Lathyrus hierosolymitanus Boiss. and Lathyrus annuus L. seeds was performed employing different solvents. Total phenolic content (TPC) was measured by Folin- Ciocalteau assay, while the antioxidant activity was determined by DPPH radical scavenging activity, and reducing power assay. It was found that TPC of extracts ranged from 0.12 mg to 6.53 mg GAE/gdw. For each solvent, seed coat extracts were generally observed to render higher TPC and antioxidant activities. There was a correlation between TPC and antioxidant activity. In addition, all extracts were also examined for their antimicrobial activity against Bacillus cereus, Escherichia coli and Pseudomonas aeruginosa. Methanol extracts showed the highest antibacterial activity which is consistent with TPC, but there was no correlation between TPC and antibacterial activity. Solvents were observed to have effects on gallic acid, caffeic acid, and epicatechin extractions. HPLC analysis results of extracts confirmed methanol and ethanol as preferred solvents for phenolic extraction from Lathyrus sp. Phenolic content in the extracts could be suggested to contribute to their antioxidant and antibacterial activity.

@#Literature has consistently reported that horticultural wastes including leaves, skin, stones and seeds contain substantial amounts of bioactive compounds. Therefore, this study aims to evaluate antioxidant activity, Total Phenolic Content (TPC) and colour parameters in avocado, banana, and papaya leaves. Antioxidant activity of the leaves was determined using Trolox Equivalent Antioxidant Capacity (TEAC) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assays, TPC was evaluated using Folin-Ciocalteu assay whereas the colour parameters were analysed with a colour picker software. Data analysis was carried out using SPSS version 25.0 of triplicate determinations. Mean differences among the fruit leaves extracts were determined using One-way ANOVA, while the correlations between the studied components were by the Pearson’s Correlation Coefficient test. The TEAC values were in the range of 332.30 ± 18.04 µg Trolox/g D.W. (avocado leaves) to 12217.71 ± 18.04 µg Trolox/g D.W. (banana leaves) while the DPPH radical scavenging activity was from 10.07 ± 3.89% (banana leaves) to 86.70 ± 0.26 % (avocado leaves). Besides, TPC was from 871.33 ± 38.35 µg GAE/g D.W. (papaya leaves) to 1199.08 ± 6.00 µg GAE/g D.W. (avocado leaves). The hue values were from 19º in avocado leaves extract to 37º in banana leaves extract. Results from Pearson’s Correlation Coefficient test revealed that there were no significant correlations between the studied assays. Avocado leaves had the highest DPPH radical scavenging activity and TPC among the three extracts. Findings derived from the present study could be exploited in nutraceuticals formulation.

Abstract In Peru, Caulerpa filiformis is a marine algae listed as an invasive species. For years, its distribution has been considered to be in the north coast (Isla Lobos de Afuera and Piura) until a recent report of its distribution in the central coast (Ancash, Lima, and Ica). The present investigation aims to determine the main groups of secondary metabolites, total phenol content, and antioxidant activity of the methanolic extract of C. filiformis from Sechura Bay (Piura) and Paracas Bay (Ica). The main chemical groups were determined through phytochemical screening, the content of phenols by the Folin-Ciocalteu method, and antioxidant activity by the ABTS method (2,2-azinobis-[3-ethylbenzothiazoline-6-sulfonic acid]) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH). The phytochemical screening of the methanolic extract of C. filiformis from Sechura Bay and Paracas Bay revealed the presence of carbohydrates, polyphenols, tannins, flavonoids, lipids, alkaloids, steroids, and triterpenes for both extracts. The total phenol content of the extract of C. filiformis from Sechura Bay (39.31 ± 0.39 mg of AGE/g extract) was significantly higher (p < 0.05) than that from Paracas Bay (18.78 ± 0.31 mg of AGE/g extract). In the ABTS and DPPH assays, the antioxidant capacity of the Sechura C. filiformis extract (IC50 = 3.49 ± 0.01 and 2.18 ± 0.02 mg/mL) was significantly higher (p < 0.05) than that of the Paracas C. filiformis extract (IC50 = 6.41 ± 0.02 and 2.42 ± 0.04 mg /mL). These findings suggest that the methanolic extract of C. filiformis is a source of secondary metabolites with an antioxidant potential.

Objective: To determine total phenolic and flavonoids contents and also quantify vindoline and rutin in different morphotypes of Catharanthus roseus using High-performance liquid chromatography (HPLC) method. Methods: Total flavonoids content (TFC) was determined by Aluminium chloride colorimetric and total phenolic content (TPC) was estimated by Folin-Ciocalteu reagent assay. The chromatographic separation was done by using a C18 column at room temperature and eluted with a mobile phase consisting of a mixture of phosphate buffer (pH=5.8) and acetonitrile at a flow rate of 1.0 ml/minute and detection was carried out at 254 nm. Results: TPC and TFC content was found highest in Cr00DP and lowest in Cr00WFSRE. Results also showed that the purple morphotypes Cr00DP gives more vindoline (0.3 mg/g) and rutin (18.57 mg/g) concentration compared to the pink morphotype Cr00PFRE contained 18.3 mg/g rutin and 0.2 mg/g vindoline. White morphotypes contained 0.383 mg/g rutin and 0.004 mg/g vindoline which was significantly less as compared to purple and pink morphotypes. Conclusion: The plant has significant number of alkaloids and flavonoids. The obtained outcomes from different morphotypes are thus significant for the purpose of vindoline and rutin isolation from Catharanthus roseus plant. These isolated bioactive phytoconstituents are a good candidate for further pharmacological and clinical study.

ABSTRACT The ethanolic extract of Melipona beecheii Bennett 1831, propolis from Yucatán Mexico was evaluated in vitro for the determination of its phenolic compound content, antioxidant capacity and antifungal activity. The results were compared against those of the ethanolic extract of Apis mellifera propolis. The total phenolic content, flavonoid content and flavanones-dihydroflavonols content were assessed by colorimetric methods. The antifungal activity was assessed in vitro against Candida albicans. For the ethanolic extract of M. beecheii propolis; total phenolic content, was 263.25 ± 8.78 µg/ml, total flavonoid content was 768 ± 204 µg/ml and flavanones-dihydroflavonols content was 335.42 ± 15.08 µg/ml. For antioxidant activity assessed as DPPH scavenging and iron reducing power ethanolic extract of M. beecheii propolis reported IC50 of 32.47 and 1.60 µg/ml of gallic acid equivalent respectively. Regarding antifungal activity against C. albicans, the minimal inhibitory concentration and minimal fungicidal concentration for ethanolic extract of M. beecheii propolis were 1.62 ± 0.33 and 2.50 ± 0.22 µg/ml of dry extract; For both minimal inhibitory concentration and minimal fungicidal concentration, ethanolic extract of M. beecheii propolis required 30% less concentration of dry extract than ethanol extract of A. mellifera propolis to exert the same antifungal actions against C. albicans. To the best of our knowledge, this is the first report about the flavonoid and flavanones-dihydroflavonols content of M. beecheii propolis. Due to the lack of information available about the stingless bee's honeycomb products, the study and conservation of endemic honeybees should remain as an active focus of research.

Aims: Salinity is one of the major abiotic stress that negatively affects plant growth in germination and early seedling stages. Salinity has becoming a serious problem as most of the parts of worldwide lands were affected by high salt concentration. Therefore, the effects of salinity ranging from 0 mM, 25 mM, 50 mM and 75 mM Sodium chloride (NaCl) concentrations on germination and early seedling growth of water spinach and their salt tolerance mechanism. Study Design: Completely Randomized Design (CRD). Place and Duration of Study: This study was conducted at Tissue Culture Laboratory, Department of Biology, Faculty of Science in University Putra Malaysia (UPM) from June to August, 2018. Methodology: In order to study the effects of salinity on water spinach, several parameters have been taken into account for measurement which include water uptake percentage, germination percentage, germination index, mean germination time, relative injury rate, seed vigor, seedling height reduction, hypocotyl and radicle length, seedling biomass, salt tolerance, total phenolic content and total flavonoids contents. Results: The results obtained showed that salinity adversely reduced water uptake efficiency, seed vigor, hypocotyl and radicle length, total phenolic content and total flavonoids content of water spinach. The seedling height reduction of water spinach increased significantly in relative to increasing salinity. However, seeds treated in mild salt concentration at 25 mM of NaCl showed an increment of germination percentage and germination index. Conclusion: Salt tolerance of water spinach increased as the response towards increasing salinity.

Phytochemical analysis, antimicrobial and antioxidant properties of the leaf extract and fractions of Sabicea brevipes were studied. Theleaveswere defatted and the marc extracted with methanol. The extractwas further purified by solvent-solvent partitioning using n-hexane, ethyl acetate and n-butanol to obtain the three solvent fractions. They were screened for phenolics, flavonoids, tannins, saponins, terpenoids, glycosides, and steroids. Total phenolics, flavonoids and tannins were determined quantitatively. The antimicrobial test was screened in vitroby agar diffusion method. Analysis of variance (ANOVA) was used to test for significant difference at p ≤ 0.05 in all study groups. The methanol extract exhibited the most significant amount of phenolics (110.78 ± 1.06 mg GAE/g) while ethyl acetate fraction had the least total phenolics content (50.55 ± 2.91 mg GAE/g). The same trend was observed for the total flavonoids content whereas the methanol extractmeasured (418.40 ± 14.03 mg QE/g) while ethyl acetate fraction had 192.40 ± 3.06 mg QE/g. Total tannins contents were: methanol extract (102.22 ± 7.58 mg GAE/g) and ethyl acetate (27.33 ± 0.77 mg GAE/g). The antioxidant results showed that the methanol extract had the highest DPPH free radical scavenging ability (93.69%) with half maximal inhibitory concentration (IC50) of 0.601± 0.02 and also highest ferric ion reducing power (50.381 ± 1.56 μmol Fe2+/g). Also, the methanol extract showed high total antioxidant capacity (96.79 ± 0.31 mg AAE) and IC50of 0.798± 0.01. The antimicrobial results revealed that the methanol extract showed better activity against Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, Staphylococcus aureus, andCandida albicans than the solvent fractions at concentrations of 200, 100, 50, 25, 12.5, mg/mL having various inhibition zone diameters (IZDs). The methanol extract and fractions of S. brevipescompared favourably in terms of zone of inhibition and minimum inhibition concentration (MIC) with the standard drug disc (Gentamycin and Ketoconazole) against the tested microorganisms. The MIC of the extract and solvent fractions ranged from 6.31 mg/mL to 50.12 mg/mL. The continual use of the extract of Sabicea brevipesin preventing oxidative stress and in the treatment of common infection is justified by these results.

Nelumbo nucifera Gaertn. (Nymphaeaceae) is commonly called lotus and its leaves are widely been used as functional ingredients due to its antioxidant activity. For maximum efficacy, optimized extraction condition was established using response surface methodology. The high F-values, low p-values and insignificant p-value for lack-of-fit supported the fitness of the model and yielded the second-order polynomial regression for the antioxidant activity. The optimized extract was obtained by the extraction of 1 g of lotus leaves with 40 mL of 50% MeOH at 10.0℃, which exerted 70.1% antioxidant activity. Close correlation between phenolic content and antioxidant activity suggested phenolic compounds as active constituents of lotus leaves. In addition, comparison of different parts of lotus demonstrated the most potent antioxidant activity of flowers, followed by leaves and roots. Taken together, these results provide useful information about lotus leaves for the development as antioxidant ingredients. In addition, flowers and roots as well as leaves are suggested as good sources for antioxidant activity.

OBJECTIVE@#This research aimed to evaluate the protective effects of bioactive compounds such as phenolic acids, flavonoids, and tannins present in four species extracted with methanol.@*METHODS@#The total phenolic content of the methanolic extracts was measured spectrophotometrically. The effect of the extracts on cell viability in U266 cells was measured. The effects of extracts on free radical scavenging were assessed by the DPPH test and FRAP assay. Antibacterial effects of the natural products in this report were investigated by using the disc diffusion method.@*RESULTS@#Our results clearly demonstrated that the methanolic extracts were characterized by a high amount of phenolic compounds. It has been speculated that ME-TA and ME-TAl exhibit a significant (P < 0.05) and dose-dependent antiradical potential. The exposure of cells to high doses of extracts almost completely suppressed cell growth in vitro. ME-TA and ME-TAl showed significant cytotoxic effects at a concentration of 100 μg/mL in the U266 cell line. ME-TAl and ME-CF inhibited the growth of B. subtilis and S. aureus, respectively, to the same extent as 10 μg/μL of chloramphenicol at a concentration of 1 mg/mL.@*CONCLUSION@#Overall, these results suggest that plants used in traditional medicine have a novel application as free radical scavengers, bacterial inhibitors and tumor suppressors.
