ABSTRACT

miRNAs are important regulators of plant gene expression. There are few studies on the regulation of miRNAs in Lonicera edulis. We used high-throughput sequencing technology to analyse miRNAs in L. edulis, aiming to identify miRNAs and elucidate their function in L. edulis. In the present study, we employed the high-throughput sequencing technology to profile miRNAs in L. edulis. A total of 51,819,072 small RNA tags with sizes ranging from 18 to 30 nt were obtained, indicating that L. edulis have a large and diverse small RNA population. Bioinformatic analysis identified 507 mature miRNAs, and 16 predicted novel miRNAs that are likely to be unique to L. edulis. Three miRNAs related to anthocyanin biosynthesis were locked by gene ontology (GO) analysis and target gene analysis. The selected three miRNAs are relatively high in the expression of L. edulis. Some of the previous studies have studied these types of miRNAs involved in the anthocyanin metabolism pathway in fruits. Among them, expression profiles of three conserved miRNAs were validated by stem loop qRT-PCR. Further, the potential target genes of conserved and novel miRNAs were predicted and subjected to GO annotation. Enrichment analysis of the GO-represented biological processes and molecular functions revealed that these target genes were potentially involved in a wide range of metabolic pathways and developmental processes. In particular, different families of miRNAs can directly or indirectly regulate anthocyanin biosynthesis. In recent years, the research on miRNAs has become more and more clear, but the research on miRNAs involved in the regulation of anthocyanin synthesis of L. edulis is still lagging. This study provides a useful resource for further elucidation of the functional roles of miRNAs during fruit development and ripening

ABSTRACT

Objective To explore the operational methods and clinical effect of laparoscopic repair of ab -dominal incision hernia .Methods The clinical data of 10 cases with abdominal giant incisional hernia undergone laparoscopic repair were retrospectively analyzed .Results All the cases were successful .Operative time ranged from 85 to 180 minutes, with an average of 150 minutes. The bleeding was 40-120mL during the operation .Postoperative hospital stay was 6-14(average 8) days.3 cases developed pain in operation area after operation and were better after 1-2 months. The seroma was observed in 1 case witch cured by puncture treatment .No incisional infection ,bleeding, intestinal fistula, intestinal obstruction ,or death was observed during the follow-up.In the follow-up after 4 months to 4 years,no recurrence was occurred .Conclusion The laparoscopic repair of abdominal giant incision hernia using composite patch is a safe and effective method ,which is worthy of clinical application .

ABSTRACT

Objective To determine the diagnostic value of the change of different parts of electrocardiogram in treadmill exercise test (TET) for coronary heart disease (CHD).MethodsFrom Jan.2006 to Mar.2011,445 patients with CHD underwent treadmill exercise test and coronary angiography (CAG) in our hospital. We analyzed retrospectively the diagnostic value of the change of different parts of electrocardiogram in TET test. Results ( 1 ) There were 200 cases who had positive results during treadmill exercise test and 150 cases of them had been diagnosed of CHD by coronary angiography; The other 245 cases had negative results during treadmill exercise test and 39 cases of them had been diagnosed as CHD by coronary angiography. The sensitivity and specificity rates of treadmill exercise test in diagnosis of CHD were 79.36%( 150/189 ) and 80.47% ( 206/256 ) respectively. ( 2 ) Of the 200 positive cases tested by coronary angiography,150 cases were diagnosed of CHD,including 22,58 and 70 cases with limb, chest and combined chest and limb leads positive results respectively. The positive rates for the groups of combined chest and limb leads and limb ST leads were significantly higher than that of the chest leads only group ( x2:6.34,3.93 ; P <0.05).(3)R wave amplitude of the CAG negative group after exercise (17.54 ± 2.52)mm was significantly lower than the CAG positive group ( 19.42 ± 3.46 )mm ( t =6.33,P < 0.05 ).( 4 ) For the 200 cases with positive TET,there were no significant statistical difference ( P > 0.05 ) on CAG positive rate between T wave normalization group [ 73.3% (44/60) ] with and TET positive group [ 75.0% (105/140) ].But these two groups had significantly higher ( x2:80.21,132.82 ; P < 0.05 ) CAG positive rates than TET negative group [ 15.9% (39/245) ].ConclusionThe treadmill exercise test is valuable in noninvasive diagnosis of CHD.The severity of CAG can be preliminarily estimated by analyzing the pattern of the change of different parts of electrocardiogram

ABSTRACT

Object To study the regular change of total flavonoids in Erigeron breviscapus (Vant.) Hand-Mazz (TFE) in different parts of the plant, different individual plant and population from different habitat. Methods Samples of E. breviscapus from eight districts around Qiubei, Kunming and at altitude on Cangshan mountain, Dali were collected and their flavonoids determined. Results TFE showed different values in samples from different origin, with the tendency to show an increase with increase of altitude. The aerial parts including stem, leaf, and flower were higher than that of the underground root.E. breviscapus grown in the same ecological conditions also showed different TFE values from individual to individual. Conclusion Variation in TFE was influenced by the combined effects of genotype and ecological conditions.
