ABSTRACT

SARS-CoV-2 Omicron variant (B.1.1.529) was first discovered in South Africa in November 2021 and has since become a mainstream strain worldwide. Omicron variant was defined as the fifth "variant of concern (VOC)" by World Health Organization on November 26, 2021. This paper illustrates the mutation trends of Omicron variants in terms of SARS-CoV-2 genome and protein structure as well as nucleic acid site mutations and amino acid site mutations, describes the features of Omicron mutation sites in terms of lineage comparison among the VOCs and Omicron sublineages, and further highlights the influences of Omicron site mutations from the aspects of immune escape, virulence and transmission ability. Moreover, this paper also reviews the development of direct antiviral agents, antibodies and vaccines, aiming to provide reference for further investigation.

ABSTRACT

Major challenges for cancer treatment are how to effectively eliminate primary tumor and sufficiently induce immunogenic cell death (ICD) to provoke a robust immune response for metastasis control. Here, a self-assembled cascade bioreactor was developed to improve cancer treatment with enhanced tumor penetration and synergistic therapy of starvation, chemodynamic (CDT) and photothermal therapy. Ultrasmall FeS-GOx nanodots were synthesized with glucose oxidase (GOx) as template and induced by paclitaxel (PTX) to form self-assembling FeS-GOx@PTX (FGP)

ABSTRACT

Objective:To evaluate the effect of virtual painless ward management based on mobile internet on pain management of patients after craniotomy. Methods:From May to October, 2019, 117 patients who accepted virtual painless ward management based on mobile Internet after craniotomy were selected as painless ward group. From December, 2018 to April, 2019, 117 patients previously hospitalized in the same department after craniotomy were selected as control group. The regression equation was established to compare the Numerical Rating Scale (NRS), nursing satisfaction and anxiety rate between two groups, and the general data, such as age, were taken as independent variables into the regression equation. Partial regression coefficient (B), odds ratio (OR) and 95% confidence interval (CI) of the painless ward management after adjustment compared with that of the non-painless ward management were calculated. Results:Multivariate regression analysis showed that, compared with the control group, the score of NRS within four days significantly decreased (B = -2.700, 95%CI -3.167 to -2.232, P < 0.001), the nursing satisfaction significantly increased (B = 0.542, 95%CI 0.289 to 0.795, P < 0.001), and the anxiety rate significantly decreased (B = -2.119, OR = 0.120, 95%CI 0.053 to 0.271, P < 0.001) in the painless ward group. However, the improvement of the anxiety rate might not completely depend on the decrease of NRS score (B = -0.112, OR = 0.894, 95%CI 0.727 to 1.100, P > 0.05). Conclusion:The procedure of virtual painless ward based on mobile internet could obviously improve the management level of analgesia for patients after craniotomy.

ABSTRACT

OBJECTIVE@#To obtain the subtypes of the clinical hypertension population based on symptoms and to explore the relationship between hypertension and comorbidities.@*METHODS@#The data set was collected from the Chinese medicine (CM) electronic medical records of 33,458 hypertension inpatients in the Affiliated Hospital of Shandong University of Traditional Chinese Medicine between July 2014 and May 2017. Then, a hypertension disease comorbidity network (HDCN) was built to investigate the complicated associations between hypertension and their comorbidities. Moreover, a hypertension patient similarity network (HPSN) was constructed with patients' shared symptoms, and 7 main hypertension patient subgroups were identified from HPSN with a community detection method to exhibit the characteristics of clinical phenotypes and molecular mechanisms. In addition, the significant symptoms, diseases, CM syndromes and pathways of each main patient subgroup were obtained by enrichment analysis.@*RESULTS@#The significant symptoms and diseases of these patient subgroups were associated with different damaged target organs of hypertension. Additionally, the specific phenotypic features (symptoms, diseases, and CM syndromes) were consistent with specific molecular features (pathways) in the same patient subgroup.@*CONCLUSION@#The utility and comprehensiveness of disease classification based on community detection of patient networks using shared CM symptom phenotypes showed the importance of hypertension patient subgroups.

ABSTRACT

Objective:To assess the performance of liver and spleen stiffness measured by MR elastography (MRE) and their combined model in diagnosing liver fibrosis. Methods:From November 2018 to November 2019, 104 patients with chronic liver disease were prospectively enrolled in Beijing Friendship Hospital, all patients underwent MRE scans. Liver and spleen stiffness were measured from MRE elastograms. Liver biopsy was used to identify fibrosis stage (F0—F4). The differences among different fibrosis stages were analyzed by one-way ANOVA or independent samples t test. The Spearman rank correlation analysis was used to evaluate the correlation with fibrosis stages. Liver and spleen stiffness combined model was established by logistic regression. The ROC curve was used to evaluate the performance of the liver, spleen stiffness and combined model in staging fibrosis (≥F1), significant fibrosis (≥F2), advanced fibrosis (≥F3), and cirrhosis (F4).The area under the ROC curve(AUC) was compared using Delong test. Results:The liver and spleen stiffness both showed significant differences among the 5 fibrosis stages ( F=64.058, 32.890, both P<0.001). The liver and spleen stiffness were positively associated with fibrosis stage ( r s=0.89, 0.69, both P<0.001). The AUC of liver stiffness in staging ≥F1, ≥F2, ≥F3 were 0. 91, 0.97, 0.93, respectively. The corresponding AUCs of the spleen stiffness were 0.81, 0.82, 0.85, respectively, which were statistically lower than those of liver stiffness ( Z=2.283, 4.085, 2.314, P=0.022,<0.001, 0.021). In diagnosing F4, the AUCs of liver and spleen stiffness were both 0.95. The AUCs of the liver and spleen combined model were 0.92, 0.97, 0.93, 0.96 in diagnosing ≥F1, ≥F2, ≥F3 and F4, with no significantly differences from liver stiffness (all P>0.05). Conclusions:The liver stiffness measured with MRE have better diagnostic performance than spleen stiffness in staging fibrosis. Parameters combined model slightly improves diagnostic value but without significant difference with liver stiffness in staging early fibrosis. Spleen stiffness evaluation is feasible in detecting cirrhosis.

ABSTRACT

SUMMARY OBJECTIVE This study aimed to propose a co-expression-network (CEN) based gene functional inference by extending the "Guilt by Association" (GBA) principle to predict candidate gene functions for type 1 diabetes mellitus (T1DM). METHODS Firstly, transcriptome data of T1DM were retrieved from the genomics data repository for differentially expressed gene (DEGs) analysis, and a weighted differential CEN was generated. The area under the receiver operating characteristics curve (AUC) was chosen to determine the performance metric for each Gene Ontology (GO) term. Differential expression analysis identified 325 DEGs in T1DM, and co-expression analysis generated a differential CEN of edge weight > 0.8. RESULTS A total of 282 GO annotations with DEGs > 20 remained for functional inference. By calculating the multifunctionality score of genes, gene function inference was performed to identify the optimal gene functions for T1DM based on the optimal ranking gene list. Considering an AUC > 0.7, six optimal gene functions for T1DM were identified, such as regulation of immune system process and receptor activity. CONCLUSIONS CEN-based gene functional inference by extending the GBA principle predicted 6 optimal gene functions for T1DM. The results may be potential paths for therapeutic or preventive treatments of T1DM.

ABSTRACT

BACKGROUND@#Pulsed radiofrequency (PRF) is a minimally invasive interventional technique that provides a novel and effective treatment strategy for neuropathic pain (NP). PRF is advantageous because it does not damage nerves and avoids sensory loss after treatment. At present, animal studies have demonstrated that PRF is safe and effective for relieving the NP associated with sciatic nerve damage in rats with chronic constriction injury (CCI). However, the mechanism through which this effect occurs is unknown. An increasing body of evidence shows that the expression of the P2X ligand-gated ion channel 3 (P2X3) receptor is closely related to NP; this study was to investigate whether the expression of this receptor is involved in NP relief due to PRF.@*METHODS@#A total of 36 healthy adult male Sprague-Dawley (SD) rats were randomly divided into three groups: Sham group, CCI group, and PRF group. The right sciatic nerve was ligated in CCI group and PRF group to establish a CCI model; the right sciatic nerve was separated but not ligated in Sham group. On day 14 after the operation, PRF was administered to the ligated sciatic nerve in PRF group (42°C, 45 V, 2 min). A non-live electrode was placed at the exposed sciatic nerve for the rats in Sham and CCI groups. The hindpaw withdrawal threshold (HWT) and thermal withdrawal latency (TWL) were measured at the right hindpaw at different time points before and after PRF or sham therapy. On day 28 after treatment, the dorsal root ganglion (DRG) and spinal dorsal horn of the right L4-6 were harvested from each group to determine the mRNA and protein levels of the P2X3 receptor.@*RESULTS@#On day 28 after PRF treatment, the HWT (8.33 ± 0.67 g vs. 3.62 ± 0.48 g) and TWL (25.42 ± 1.90 s vs. 15.10 ± 1.71 s) were significantly higher in PRF group as compared to CCI group (P < 0.05). The mRNA expression of the P2X3 receptor in the DRG in PRF group was 23.7% lower than that in CCI group (P < 0.05), in the spinal dorsal horns in PRF group was 22.7% lower than that in CCI group (P < 0.05). The protein expression of the P2X3 receptor in the DRG in PRF group was 27.8% lower than that in CCI group (P < 0.05), in the spinal dorsal horns in PRF group was 35.6% lower than that in CCI group (P < 0.05).@*CONCLUSION@#PRF possibly reduces NP in CCI rats by inhibiting the expression of the P2X3 receptor in the L4-6 DRG and spinal dorsal horns.

ABSTRACT

@#Objective To investigate the long-term effect of pulsed radiofrequency on the expression of calcitonin gene-related peptide (CGRP) in dorsal root ganglion of rats with chronic constriction injury. Methods A total of 36 Sprague-Dawley rats were randomly divided into sham group (n=12), model group (n=12) and treatment group (n=12). The sciatic nerve was ligated in the model group and the treatment group, and the treatment group was treated with pulsed radiofrequency on the injured nerve for 120 seconds. Hindpaw withdrawl threshold (HWT) was measured before treatment, and seven days, 14 days, 21 days and 28 days after treatment. After HWT, the right L4-6 dorsal root ganglions were tested the CGRP expression using RT-qPCR and ELISA. Results HWT gradually increased in the treatment group after treatment, and was higher than that in the model group since seven days after treatment (P<0.05), similar to that of the sham group 21 days and 28 days after treatment (P>0.05). The transcription and translation levels of CGRP were both significantly lower in the treatment group than in the model group (P<0.01).Conclusion Pulsed radiofrequency might relieve the neuropathic pain through reducing CGRP expression in dorsal root ganglion in a long term.

ABSTRACT

BACKGROUND: With the continuous development of mesenchymal stem cell therapy, it has been reported that stem cell therapy is likely to cause the occurrence and development of tumors. OBJECTIVE: To investigate the potential risks of hepatocellular carcinoma (HCC) in patients with hepatitis B cirrhosis after receiving human umbilical cord mesenchymal stem cells (hUC-MSCs) transplantation. METHODS: The study collected the information of patients with hepatitis B cirrhosis treated with hUC-MSCs, admitted at the Infectious Disease Department of the 105thHospital of PLA from January 2011 to December 2013. The following investigation lasted 36 months. The follow-up was terminated at the time of diagnostic confirmation. The risk factors that may affect the occurrence of HCC were analyzed by univariate Logistic and multivariate unconditional Logistic regression analyses. RESULTS AND CONCLUSION: (1) A total of 386 patients were followed up, including 171 patients who received hUC-MSCs transplantation as the observation group and 215 patients only given general internal medicine treatment as the control group. (2) At the follow-up of 12 months, the incidence of HCC in the observation group was significantly higher than that in the control group (P < 0.05). At the follow-up of 36 months, the incidence of HCC was 11.7% in the observation group and 9.8% in the control group (P > 0.05). (3) Univariate Logistic regression analysis showed that the HCC patients had higher age, alpha-fetoprotein (AFP), alpha-fetoprotein variants (AFP-L3), AFP-L3 ratio (AFP-L3%), and Golgi glycoprotein 73 (GP73) than those with no HCC in both control and observation groups (P < 0.05). Multivariate unconditional Logistic regression analysis showed that only APF-L3% was an independent risk factor for HCC in patients with hepatitis B cirrhosis undergoing hUC-MSCs transplantation. Overall, hUC-MSCs transplantation does not increase the HCC incidence in patients with hepatitis B cirrhosis within 3 years, but it may lead to an early onset of HCC. AFP-L3% can be used as an early predictor of HCC in patients with hepatitis B cirrhosis undergoing hUC-MSCs transplantation.

ABSTRACT

Objective To observe the hindpaw withdrawal threshold (HWT), and the ultrastructure and expression of glia cell line-de-rived neurotrophic factor (GDNF) in sciatic nerve (SN) in chronic constriction injury (CCI) rats after pulsed radiofrequency (PRF). Meth-ods 30 Sprague-Dawley rats were divided into sham modeling-sham treating (SS) group, CCI-Sham treating (CS) group and CCI-PRF (CP) group. The right SNs of the rats in the CS and CP groups were ligated, and it was separated without ligation in the SS group. The CP group accepted PRF at the ligation 14 days after modeling, while the electrodes were placed without electricity in the SS and CS groups. Their HWT was measured before and 1, 7, 14 days after modeling, and 1, 7, 14 days after treatment. The right SN of ligation was observed under electron microscope 14 days after treatment, meanwhile, the GDNF expression was determined with enzyme-linked immunosorbent assay (ELISA). Results HWT was significantly shorter in the CS and CP groups than in the SS group after modeling, and it increased in the CP group 14 days after treatment compared with that of the CS group (P<0.01). The degeneration of SN significantly improved in the CP group compared with the CS group, while the expression of GDNF increased compared with that in the CS and SS groups (P<0.01). Conclusion PRF could relieve the CCI-induced neuropathic pain by upregulating the GDNF expression in the SN to prevent the SN from injury.

The results shows that the contents of rutin, quercetion 3-O-[4-O-(α-L-rhamnosyl )-β-D-glucoside] and kaempferolis were 120.9, 114.0, 222.1 μg x g(-1). In this paper, the contents of 24 kinds of chemical components of ginkgo pollen were determinated by UPLC-TQ-MS for the first time. This method is simple and quick, which will be benefit for recycling utilization of ginkgo pollen.

ABSTRACT

Objective To explore the distribution characteristics and clinical typing methods in distal crevasses of Stanford B aortic dissection. Methods Review of the cases in the People's Hospital of Xinjiang Urgur Autonomous Region from 2010 January to 2013 June were diagnosis of Stanford type B aortic dissection with computed tomographic angiography data,Observed its distal crevasses distribution and statistical its number, then summarizes the distribution characteristics of the distal crevasses and further put forward a method of clinical typing. Results Refer to 115 cases with Stanford type B aortic dissection computed tomographic angiography data, including 101 cases with distal crevasses (87.83%) and a total of 240 distal crevasses,an average of 2.37 per case. Conclusions The distal crevasses more often appear in the area involving visceral artery, combined with its different in distribution characteristics and processing methods,we put forward the classification method, namely:Type Ⅰ:the distal crevasses are located in the zone of the thoracic artery; Type Ⅱ:the distal crevasses are close to the visceral artery or involvement it; Type Ⅲ:the distal crevasses are lower than the renal artery, not involving the visceral artery; Type Ⅳ:the distal crevasses are located in the zone of the iliac artery.

ABSTRACT

Objective To explore the treatment principle of distal crevasses in aortic dissection. Methods According to the different treatment principles of distal crevasses in aortic dissection to divide the patients into two groups:following the treatment principle group:take the principle of from near to far, endovascular repair of distal crevasses,if not deal with the crevasses involving visceral arteries,do not handle the distal crevasses; unfollowing the treatment principle group:do not deal with the crevasses involving visceral arteries, endovascular repair of it's distal crevasses).Analyze the postoperative discomfort (chest and back pain, lower back pain) incidence, postoperative distal crevasses happening again and false lumen changes respectively. Results Fifty-three patients were included in the standard,in which follow the treatment principle group has 37 cases, and do not follow the treatment principle group has 16 patients. There were no significance statistically in complained of postoperative discomfort and postoperative distal crevasses happening again(P > 0.05).And there was statistically significant in the false lumen (P < 0.05).Conclusions The prognosis of patients in the following the principle treatment is better than that of unfollowing the principle group. To treat the distal crevasses in aortic dissection should be took the principle of from near to far,if not deal with the crevasses involving visceral arteries, don't handle the distal crevasses also.

ABSTRACT

Objective: To analyze the nutritional ingredient in the pollen of Gingko biloba and provide the theoretical basis for the comprehensive utilization of pollen of G. biloba. Methods: The nucleosides and dissociative amino acids in the pollen of G. biloba were detected by UPLC-TQM. The content of nucleosides in pollen of G. biloba was calculated with the help of UPLC-TQ/MS, and the mineral elements in the pollen of G. biloba were determined by ICP-AES. Results: In pollen of G. biloba there were 24 abundant dissociative amino acids accounting for 62.316 mg/g totally. In this number, dissociative essential amino acids accounted for 6.23 mg/g with the highest content of 17.11 mg/g aspartic acid (ASP). Meanwhile, a small quantity of taurine, approximately 0.24 mg/g, has been tested out. Furthermore, there were 16 kinds of nucleosides in the pollen of G. biloba. Among those nucleosides, guanosine made up for the highest percent with 251.99 μg/g while other nucleosides accounted for a low percent. In pollen of G. biloba, K is the most abundant with 26.301 mg/g, followed by Ca with 7.076 mg/g, Mg with 5.361 mg/g, and Fe with 2.545 mg/g. As to the heavy mental elements, as and Cd were not found in it. Conclusion: From above analysis, pollen of G. biloba can provide the theoretical basis for the development of health-care food with pollen of G. biloba.
