ABSTRACT

Objective To compare the effects of losartan and irbesartan on blood pressure,blood uric acid,insulin sensitivity and insulin resistance in female hypertensive patients complicated with hyperuricemia.Methods From August 2015 to December 2017,100 females of hypertension complicated with hyperuricemia who hospitalized in the Third People's Hospital of Ningxia were enrolled in the study.All the patients were divided into two groups according to the random digital table,with 50 cases in each group.The observation group was treated with losartan,and the control group was treated with irbesartan for 8 weeks.The total effective rate and adverse reactions were compared between the two groups.Also the changes of blood pressure,serum uric acid,fasting glucose (FBG),fasting insulin (FINS),high sensitivity C-reactive protein (hs-CRP),insulin sensitivity index (ISI) and insulin resistance index (HOMA-IR) were compared between the two groups after treatment.Results There was no statistically significant difference in the total effective rate between the two groups [92.0% (46/50) vs.90.0% (45/50)] (P > 0.05).Before treatment,there were no statistically significant differences in blood pressure,serum uric acid,FBG,FINS,hs-CRP,ISI and HOMA-IR between the two groups (all P > 0.05).After treatment,the systolic blood pressure and diastolic blood pressure in the two groups were (133.09 ± 10.11) mmHg vs.(131.54 ± 11.01) mmHg and (82.76 ± 6.23) mmHg vs.(83.75 ± 6.88) mmHg,which were lower than those before treatment (observation group:t =19.742,10.606,control group:t =18.925,-9.956,all P < 0.05).But there were no statistically significant differences between the two groups (all P > 0.05).After treatment,the serum uric acid in the observation group was lower than that in the control group [(387.21 ± 25.56) μmol/L vs.(429.67 ± 27.44) μmol/L] (t =8.006,P < 0.05).The hs-CRP,FINS,HOMA-IR,ISI in the observation group were (4.92 ± 1.02) rmg/L,(15.92 ± 3.01) mU/L,(1.71 ± 0.24),(1.047 ± 0.095),which in the control group were (4.54 ± 1.00) mg/L,(17.23 ± 3.20) mU/L,(1.65 ± 0.27),(1.140 ± 0.083).After treatment,the hs-CRP,FINS,HOMA-IR in the two groups were decreased (all P < 0.05),while ISI was increased (P < 0.05).Furthermore,the improvement of FINS,ISI and HOMA-IR in the observation group was better than those in the control group (t =2.109,-5.213,3.191,all P < 0.05).Conclusion Both losartan and irbesartan can improve clinical symptoms,lower blood pressure and improve insulin resistance in female hypertension patients complicated with hyperuricemia,and losartan is more effective than irbesartan.

ABSTRACT

OBJECTIVE:To investigate clinical efficacy and safety of Miao medicine Jinyin huashi granules combined with western medicine in the treatment of chronic calculous cholecystitis(CCC). METHODS:A total of 120 CCC patients in our hospi-tal during Jan. 2014-Jan. 2016 were randomly divided into control group and observation group,with 60 cases in each group. Con-trol group was given 50% Magnesium sulfate solution 10 mL orally before meal,tid;amoxicillin 0.5 g orally,tid+Racanisodamine tablets 10 mg,tid+Compound dantong tablets 1 slice,tid,after meal. Observation group was additionally treated with Miao medi-cine Jinyin huashi granules 15 g,tid,on the basis of control group. Both groups were treated for consecutive 4 weeks. Clinical effi-cacies,the improvement of upper abdominal pain,nausea and greasy,calculus were observed in 2 groups. The thickness of gall-bladder,serum levels of IL-2 and IL-5,mRNA and protein expression of CYP7A1 and B-UCT were compared between 2 groups before and after treatment. The occurrence of ADR was recorded in 2 groups. RESULTS:Total response rate of observation group was 96.67%,which was significantly higher than 88.33% of control group,with statistical significance (P<0.05). One d and one week after treatment,the improvement rates of upper abdominal pain were 63.33% and 81.67% in observation group, which were significantly than 36.67% and 50.00% of control group,with statistical significance(P<0.05). There was no statisti-cal significance in the improvement rate of nausea or greasy after treatment between 2 groups(P>0.05). The stone-free rate of ob-servation group was 33.33% and significantly higher than 11.67% of control group,with statistical significance(P<0.05). Before treatment,there was no statistical significance in the thickness of gallbladder wall,serum levels of IL-2 or IL-15,mRNA and pro-tein expression of CYP7A1 or B-UCT between 2 groups(P>0.05). After treatment,the thickness of gallbladder wall,serum lev-els of IL-2 and IL-15 were all decreased significantly in 2 groups,while mRNA and protein expression of CYP7A1 and B-UCT were increased significantly;observation group was significantly better than control group,with statistical significance (P<0.05). There was no statistical significance in the incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:Miao medicine Jinyin huashi granules combined with western medicine show significant therapeutic efficacy for CCC,can effectively improve right upper quadrant pain,nausea and greasy,decrease serum levels of IL-2 and IL-5 and up-regulate mRNA and protein expression of CYP7A1 and B-UCT with good safety.

[ ABSTRACT] AIM:To investigate the protective effect of naringin ( Nar) on the injury of human umbilical vein endothelial cells ( HUVECs) induced by 33 mmol/L high glucose ( HG) and to explore its possible mechanisms.METH-ODS:The injury model was established by treating HUVECs with HG medium for the indicated time (6, 12, 24, 48 and 72 h) , and then the levels of NO, eNOS and p-eNOS were detected, respectively.The effects of Nar on high glucose-in-duced endothelial cell injury were observed.HUVECs were treated with Nar at concentrations of 5, 10, 25, 50 and 100 mg/L for 6 h, 12 h, 24 h, 36 h and 48 h.The levels of NO in the supernatants were measured.The effects of Nar on HG-injured HUVECs were explored by treating the cells with 10 μmol/L of LY294002, a PI3K inhibitor, or 0.5 μmol/L of AKT inhibitorⅣ, an AKT inhibitor, and then the levels of NO, PI3K, AKT, eNOS and their phosphorylated proteins were determined by Western blot.RESULTS:Nar at concentration of 50 mg/L significantly attenuated the injury of endothelial cells induced by high glucose ( P<0.01) , and the protective effects of Nar were abolished by pretreating with the inhibitor of PI3K or AKT (P<0.01).CONCLUSION: Nar protects endothelial cells against the injury induced by high glucose through PI3K/AKT/eNOS pathway.

ABSTRACT

Objective To detect the effect of a new bioactive bone screws composed of nanohydroxyapatite/polyamid 66/glass fiber (n-HA/PA66/GF) biomaterial on biological behavior of osteoblasts with a view to a later clinical application of the screws.Methods Mice osteoblasts were co-cultured with n-HA/PA66/GF biomaterial or its extract.Cell growth was detected under a microscope,cell proliferation using MTT assay,apoptosis by flow cytometry,osteocalcin by ELISA method,cell migration using Transwell invasion assay,cell adhesion and growth by scanning electron microscope,and cytoskeleton and actin filament distribution using immunofluorescene.Results Direct contact test demonstrated n-HA/PA66/GF material had no obvious cytotoxicity to osteoblasts.Extract of n-HA/PA66/GF material stimulated osteoblast proliferative activity over time with absorbance value of 0.96 ± 0.14,1.54 ± 0.15,and 2.39 ±0.27 respectively after 2,4,and 6 hours of coculture (P < 0.05).The n-HA/PA66/GF material induced more osteoblasts to entering S period without obvious influence on apoptosis rate and promoted osteocalcin secretion.Migrated cells in medium supplied with n-HA/PA66/GF material or FBS was 8.73 ± 3.26 and 9.47 ± 3.29 in each visual field,but there was no significant difference (P > 0.05).Osteoblasts had a regular form on the surface of n-HA/PA66/GF material,closely adhered to the surface,and reproduced and aggregated with each other to form stratified cell layers.However,nHA/PA66/GF material exhibited no obvious influence on distribution of cytoskeleton and actin filament.Conclusion The new n-HA/PA66/GF screws has excellent cytocompatibility with positive regulatory effect on cell growth,proliferation,secretion,adhesion,cycle,and osteocalcin secretion.

ABSTRACT

Objective To explore the clinical significance of changes of cell immune function in patients with oral cancer before and after operation.Methods 20 healthy volunteers(control group) and 20 patients with oral squamous cell carcinoma who pathologically confirmed after operation (observation group) were included in this study.The peripheral blood T lymphocyte subsets and the levels of cytokines in Th1 cells were detected before and after surgery.Results Compared with the control group,the percentage of preoperative serum CD3+,CD4+,CD8+,CD5+ T lymphocyte and CD4/CD8 ratio in the observation group were decreased significantly (t =7.908,7.282,2.428,4.360,4.329,P < O.05).In the observation group,the percentage of CD3+,CD4+,CDs+,CD5+ T lymphocyte and CD4/CD8 ratio after operation were significantly improved than before surgery (t =3.994,6.159,2.401,2.446,2.102,all P <0.05),but CD4+ T lymphocytes and the ratio of CD4/CD8 were still significantly lower than the control group(t =2.741,3.133,all P < 0.05).In the observation group,the preoperative levels of cytokines secreted by T lymphocytes were significantly lower than those in the control group (t =3.175,2.747,43.000,3.137,16.160,46.750,all P < 0.05).After operation,the levels of cytokines secreted by T lymphocytes were improved.Conclusion Patients with oral cancer have immune suppression,which manifest with the decrease of Th cells,the increase of suppressor T cells and the decrease of CD4/CD8 ratio.The immune function of the patients is improved after operation.

ABSTRACT

<p><b>OBJECTIVE</b>To study the microstructure of the Ag-nHA-nTiO2/PA66 membrane and investigate its biocompatibility.</p><p><b>METHODS</b>The microstructure of Ag-nHA-nTiO2PA66 membrane and e-polytetra fluoroethylene (e-PTFE) membrane were observed by light microscope and scanning electron microscope(SEM). MG63 osteoblast-like cells were cultured on the two kinds of membrane and blank group. The cell proliferation was checked by methyl thiazolyl tetrazolium (MTT) method and alkaline phosphatase (ALP) activity was detected by enzyme linked immunosorbent assay (ELISA). The adhesion and proliferation of the cells on the two kinds of membrane was observed by SEM.</p><p><b>RESULTS</b>The Ag-nHA-nTiO2/PA66 membrane was composed of the obverse face and the opposite face. The obverse face was porous and the opposite face was smooth. Microstructures of the obverse and the opposite face of the e-PTFE membrane were same. The e-PTFE membrane showed many tiny lined cracks in elliptic structure. MTT assay and ALP measurement showed that there were no significant difference between each of the two membrane groups and the blank (P > 0.05). The adhesion and proliferation of cells on the Ag-nHA-nTiO2/PA66 membrane were better than the e-PTFE membrane.</p><p><b>CONCLUSION</b>Ag-nHA-nTiO2/PA66 membrane has no negative effects on the growth of osteoblast-like cells. Ag-nHA-nTiO/PA66 membrane is biocompatible and its microstructure is appropriate as a guided bone regeneration materials.</p>

ABSTRACT

Bacterial infection after implantation of bone tissue engineering scaffolds is still a serious clinical problem. Ag-nano-hydroxyapatite/polyamide66 (Ag-nHA/PA66) antibacterial composite scaffold were prepared with phase-inversion method in this study. The scaffolds were mineralized in saturated calcium phosphate solution at 37 degrees C for 1 day. The microstructure and the newly formed nano-apatite deposition on the scaffolds before and after mineralization were observed using scanning electron microscopy (SEM). In order to investigate the release behaviors of Ag+, the Ag-nHA/PA66 scaffolds were immersed into 5 ml PBS at 37 degrees C for a different period between 3 h and 168 h before and after mineralization. Then the samples were cultured with E. coli (8099) to test the antibacterial effect of the scaffolds. The results showed that, after mineralization, Ag-nHA/PA66 porous scaffolds still possessed a good inter-connection and a new apatite layer was formed on the surface of the scaffolds. The average macropore size was 626.61 +/- 141.94 microm, the porosity was 76.89 +/- 8.21% and the compressive strength was 2.94 +/- 1.12 MPa. All these physical parameters had no significant difference from those of the un-mineralized scaffolds. The Ag+ release of the scaffolds with and without mineralization was fast within 1 day and then kept slow and stable after 1 day. The antibacterial test confirmed that after mineralization the scaffolds had good antibacterial effects on E. coli.

ABSTRACT

Objective To discuss oral cavity nursing in senile diabetic patients with oral disease.Methods 80 senile diabetic patients were offered diet control and treatment for lowering blood glucose with simultaneous oral nursing for 4 weeks.The blood glucose,blood lipid before and after nursing were observed.Results The total effective rate reached 91.3% after oral nursing,blood glucose and blood lipid greatly lowered after nursing.Conclusions Prevention of oral disease is important for preventing diabetic complications,appropriate oral nursing contributes to prevention and treatment of diabetes mellitus complicated with oral disease.

ABSTRACT

@#Objective To investigate the protective and cure effects of dexamethasone on bleomycin-induced pulmonary fibrosis.Methods32 rats were randomly divided into the control group (C-group, n=8), injury group (I-group, n=12) and dexamethasone group (D-group, n=12). The acute pulmonary model was established by intratracheal injection of bleomycin with rats of the I-group and D-group; while rats of the C-group injected with distilled water. After that, rats of the D-group were injected with dexamethasone sodium phosphate in intraperitoneal every day, those of the C-group and I-group were injected with saline. The animals were killed on the 3rd, 7th, 14th, and 27th days after treatment, and tests of bronchoalveolar lavage fluid (BALF), total lung collagen content and lung tissue processing were performed.ResultsPathological evidence of the I-group rats demonstrated that the alveolar compartment companied with massive inflammatory cell invasion and a number of myofibroblast proliferation became more thick. However, lung injury in the D-group rats got better than that in the I-group. Neutrophil percentage achieved peak in both I-group and D-group on the 7th day. But the neutrophil ratio in the D-group was significantly lower than that of the I-group on the 7th day ( P<0.05) and the 14th day ( P<0.01). Total lung collagen content achieved peak on the 14th day both in I-group and D-group, but that of the I-group was significantly higher than that of the C-group ( P<0.01) and the D-group was significantly lower than the I-group ( P<0.01).ConclusionDexamethasone plays a protective and cure role in lung fibrosis by efficiently inhibiting the gather and invasion of neutropils and restraining the increase of collagen secreted by proliferous fibroblasts.

BACKGROUND:Contraction of three-dimensional collagen gels has been used as a model for the contraction which characterizes both normal wound healing and the development of fibrosis in the tissue. Several factors and cytokines,such as tumor necrosis factor alpha (TNF-α), interleukin-1, prostaglandin E2 and insulin have been proved to play important roles in collagen remodeling in vitro as well as serum extravasation during the fibrotic progress.OBJECTIVE: To observe extracellular collagen matrix contraction and apoptosis of fetal lung fibroblasts in TNF-α,interleukin-1, insulin, prostaglandin E2, albumin and globum under three-dimensional culture, and investigate the effects of cytokines, insuin, serum and serum protein on the remodeling and fibrotic formation of lung tissue.DESIGN: A randomized controlled experiment.SETTING: Department of Respiratory Medicine, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA.MATERIALS: The experiment was carried out The experiments were carried out in the respiratory laboratory of Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from August 2005 to January 2006. Human fetal lung fibroblasts (American Type Culture Collection), Dulbecco's modified Eagle's medium (DMEM) and fetal bovine serum, insulin, transforming growth factor (TGF) (R&D), prostaglandin E2, type Ⅰ collagen was extracted from rat-tail tendons.METHODS: In order to investigate the effect of initial collagen concentration in the gels on the contractility of fibroblasts,the appropriate amount of collagen was mixed with distilled water, four fold-concentrated DMEM, and cells were suspended so that the mass concentration was 0.75-2.0 g/L, with a physiological ionic strength and the desired cell concentration. In order to investigate the effect of cell number in the gels on the contraction, the cellular concentration fibroblasts in the gels were prepared to 0.2×107-4×107 L-1. The areas of floating gels were measured daily and the contraction was calculated by contrasting the initial size (% of initial area). Different serum concentrations (0.01%-0.5%)in the medium were prepared, the serum albumin (0.1%) or globulin (0.1%) were added to the serum-free culture medium to observe the gel contraction. TGF (10 mg/L), interleukin-1 (10 mg/L), insulin (1 mg/L) and prostaglandin E2 (0.1 μmol/L) were added to observe the effects of cytokines and insulin on fibroblasts-mediated collagen gel contraction.The DNA content and cellular survivability in gels in collagen were determined.MAIN OUTCOME MEASURES: Effect of lung fibroblasts on collagen contraction with or without the presence of cytokines in three-dimensional culture; Effect of collagen with different concentration on the proliferation and apoptosis.RESULTS: ① Collagen gel contraction showed a dependence on the number of fibroblasts. ② Collagen gel contraction was augmented by increasing serum concentration in culture medium, and albumin increased the contraction dramatically. ③TGF and insulin significantly increased the contraction, whereas prostaglandin E2 and interleukin-1 significantly inhibited the gel contraction. ④ The lower the initial collagen concentration was, the more gel contracted and smaller final size were observed, and cell apoptosis increased.CONCLUSION: During the fibrotic process, fibroblast population migrated into the injured lung tissue may play an important role in the development of pulmonary fibrosis and serum infiltrating into injury lung tissue may play an role in stimulating the fibrotic progress. Infiltrating fluids and edema result in the dilution of the collagen concentration in the pulmonary interstitial which may lead to stronger contraction and serious fibrosis. In the dense fibrosis area, cells were hard to survive. In consequence, the final structure of fibrotic lung could not been changed and lung fibrosis progressed.

ABSTRACT

Background and purpose:Endostatin can effectively inhibit angiogenesis,it could slow down tumor growth in heterogenic transplantation model by given ES systematically.This study was to evaluate the inhibition of endostatin on tumor growth and metastasis of lung cancer of the thoracic cavity implant model in nude mouse.Methods:H460 cells were inoculated subcutaneously into the thoracic cavity of nude mouse,and the mice were randomized into ES group(ESG)and control group(CG).At the fi rst day,the ESG was given 20 mg/kg of endostatin s.c.qd,the CG was treated daily s.c.with equal volumes of PBS for 20 days.The weight of the animal and the transplantation tumor,mice life span,MMP2 of blood were observed.Results:The rate of tumor formation was 100%,and the thoracic cavity implant models gradually had metastasis.The transplantation tumor weights were(1.54?0.14)in ESG group that was less than that(0.9?0.3)in CG group(t=-3.163,P=0.005);the comparison of the change value of weight on day 1 and day 21 between CG(1.9?2.8)and ESG(-0.8?2.8)was statistically different(t=-2.156,P=0.045).The average survival time of the ESG was longer than that of the CG.Conclusion:The results demonstrate that endostatin could inhibit the growth and metastasis of H460 cell in athymic mouse,and one of the important mechanisms involves was inhibition of the MMPs expression.The thoracic cavity implant model was easier to obtain metastasis,and provided a reliable research tool for the study of endostatin inhibition on tumor growth and metastasis of lung cancer.
