ABSTRACT
Objective: To analyze the risk factors for complications of endoscopic full-thickness resection (EFTR) of upper gastrointestinal submucosal tumors (SMTs). Methods: This was a retrospective observational study. The indications for EFTR included: (1) SMTs originating from the muscularis propria layer and growing out of the cavity or infiltrating the deep part of the muscularis propria layer; (2) SMTs diameter <5 cm; and (3) tumor identified as closely adherent to the serous layer during endoscopic submucosal dissection or endoscopic mucosal resection. This study included patients with SMTs originating from the muscularis propria layer in upper digestive tract, diagnosed preoperatively by endoscopic ultrasonography or computed tomography, who were successfully treated with EFTR. Those with incomplete clinical data were excluded. The clinical data of 154 patients with upper gastrointestinal SMTs who underwent EFTR at the Department of Gastroenterology, First Affiliated Hospital of Soochow University from January 2016 to January 2022 were retrospectively analyzed. Post-EFTR complications (such as delayed perforation, delayed bleeding, and postoperative infection, including electrocoagulation syndrome) were monitored and the risk factors for them were analyzed. Results: Among the 154 study patients, 33 (21.4%) developed complications, including delayed bleeding in three (1.9%), delayed perforation in two (1.3%), and postoperative infection in 28 (18.2%). One patient with bleeding was classified as having a major complication (hospitalized for more than 10 days because of complication). According to univariate analysis, complication was associated with tumor diameter >15 mm, operation time >90 minutes, defect closure method(purse string suture), and diameter of resected specimen ≥20 mm (all P<0.05). Multivariate logistic regression analysis showed that operation time >90 minutes (OR=6.252, 95%CI: 2.530-15.446, P<0.001) and tumor diameter >15 mm (OR=4.843, 95%CI: 1.985-11.817, P=0.001) were independent risk factors for complications after EFTR in patients with upper gastrointestinal SMTs. The independent risk factors for postoperative infection in these patients were operation time>90 minutes (OR=4.993, 95%CI:1.964-12.694, P=0.001) and purse string suture (OR=7.142, 95%CI: 1.953-26.123, P=0.003). Conclusion: Patients with upper gastrointestinal SMTs undergoing EFTR with tumor diameter >15 mm or operation time >90 minutes have a significantly increased risk of postoperative complications. Postoperative monitoring is important for these patients with SMTs.

ABSTRACT
ObjectiveTo conduct phylogenetic analysis of internal transcribed spacer 2 (ITS2) and chloroplast gene segments including psbA-trnH, rbcL, and matK of Sophora japonica cv. jinhuai resource samples from different geographical sources, and to explore the genetic diversity of S. japonica cv. jinhuai. MethodPolymerase chain reaction (PCR) method was used to amplify the nucleic acid sequences of ITS2, psbA-trnH, rbcL, and matK of S. japonica cv. jinhuai. Neighbor joining (NJ) method was used to construct phylogenetic trees, and Kimura 2-Parameter (K2P) model was used to calculate the genetic distance of different samples. MEGA and BIOEDIT softwares were applied for mutiple alignment and analysis of ITS2, psbA-trnH, rbcL, and matK sequences of S. japonica cv. jinhuai. ResultThe lengths of ITS2 sequence were 278-279 bp. The lengths of psbA-trnH were 289 bp. The lengths of rbcL sequence were 673 bp. The lengths of matK sequences were 786-792 bp. There were 3 mutation points in ITS2 and psbA-trnH, no mutation point in rbcL, and 13 mutation points in matK. The samples of S. japonica cv. jinhuai were clustered into two groups based on the phylogenetic tree constructed by ITS2 sequences. The sample of seedling tree in Baibao was clustered into one group, while the other 25 samples were clustered into another group. For the psbA-trnH sequence, the success rate of PCR amplification of 28 samples of S. japonica cv. jinhuai was 100%. The 28 samples of S. japonica cv. jinhuai were clustered into three groups based on the clustering results of psbA-trnH sequence. The sample of seedling tree in Shaoshui was clustered into one group. The five samples of grafting tree and seedling tree in Miaotou, grafting trees in Jiantang, Wenqiao, and Daxu, and seeding tree in Xianshui were clustered into one group. The sample of seedling tree in Xianshui was clustered into one group, while the other 25 samples were clustered into another group. The clustering results of the rbcL sequence of S. japonica cv. jinhuai could not distinguish 28 resource samples. The phylogenetic tree constructed by the combined sequence of ITS2+psbA-trnH+rbcL+matK divided S. japonica cv. jinhuai resource samples into 4 groups. The 13 samples of seedling trees in Qiyang, Daoxian, Miaotou, Shaoshui, Shitang, Xianshui, Jiantang, and Xiangli, and grafting trees in Qiyang, Miaotou, Yongsui, Wenqiao, and Yangtang were clustered into one group. The sample of seedling tree in Wenqiao was clustered into one group. The sample of seedling tree in Daxu was clustered into one group. The remaining samples were clustered into another group. ConclusionPhylogenetic and mutation analysis provide the theoretic foundation to investigate the evolution of the resources of S. japonica cv. jinhuai, and evaluate their genuineness. The results of mutation points can be used to identify the related S. japonica cv. jinhuai resources. The findings of this study show that the combination of different gene sequences has an optimal effect on plant identification.
ABSTRACT
The paper introduces professor MA Hui-fang's experience in age-related macular degeneration treated with acupuncture at Jingming (BL 1). It is believed that the basic pathogenesis of this disease refers to liver and kidney insufficiency and weakness of spleen qi. Based on the academic ideas of yang qi and meridian-collateral system, the treatment principle is proposed as "invigorating yang qi, replenishing the liver and kidney and nourishing the spleen and stomach". Regarding acupoint selection, Jingming (BL 1) is specially used, combined with the 4 front-mu points (Zhongwan [CV 12], Guanyuan [CV 4] and bilateral Tianshu [ST 25]), as well as the empirical points for eye diseases (Jingming [BL 1], Baihui [GV 20] and Zulinqi [GB 41]). Concerning to needling technique, shuci (transport needling), fenci (intermuscular needling) and yuandaoci (distal needling) are dominated. Eventually, a set of unique therapeutic method has been accumulated through professor MA Hui-fang's clinical practice in treatment of age-related macular degeneration.

ABSTRACT
OBJECTIVE: To explore the reversal effect and molecular mechanism of shengqi fuzheng injection(SQFZ) on pancreatic cancer multidrug resistance(MDR) cells. METHODS: Patu8988 was exposured to fluorouracil(5-Fu) to obtain stable 5-Fu-resistant Patu8988/5-Fu cell lines. MTT assay was used to detect the inhibition of SQFZ on cell proliferation and the resistance index of Patu8988/5-Fu and reversal index of SQFZ were calculated. Flow cytometry(FCM) was employed to measure the accumulation of Rho123 in cells. The mRNA expression of MDR1, Bcl-2 and Bax and presence of Bcl-2, Bax and P-gp protein were analyzed by quantitative real-time PCR and Western blot methods respectively. RESULTS: SQFZ at the concentration of 5, 10, 20 μL·mL-1 obviously reversed MDR of Patu8988/5-Fu cells, with the reversal index of 1.5, 2.3 and 3.4 respectively. It increased Rho123 accumulation in a dose-dependent manner in MDR cells(r=0.979, P<0.05). The study revealed that SQFZ downregulated the expressions of MDR1 and Bcl2 gene and upregulated the expression of Bax gene. Furthermore, SQFZ decreased the expression of P-gp and Bcl-2 protein, and increased the expression of Bax protein in Patu8988/5-Fu cells. CONCLUSION: SQFZ can reverse multidrug resistance of Patu8988/FU cells, likely by modulating gene transcription and protein expression of MDRl, Bcl-2 and Bax.

<p><b>OBJECTIVE</b>To study the effect of Mps1 on BRAFWT/MEK/ERK pathway in the presence of wild type BRAF or BRAFV600E in melanoma.</p><p><b>METHODS</b>Melanoma cells harboring BRAFWT genotype were transfected either with pBabe-puro-GST-BRAF-WT and/or pBabe-puro-GFP-Mps1-WT or pBabe-puro-GST-BRAFV600E and/or pBabe-puro-GFP-Mps1-WT, followed by Western blot to detect Mps1 and p-ERK expression. The melanoma cells harboring BRAFWT and BRAFV600E genotype were infected with pSUPER-Mps1 retrovirus to knockdown the endogenous Mps1 protein, followed by Western blot to detect Mps1 and p-ERK expression. Meanwhile, melanoma cells harboring BRAFV600E genotype were infected with pBabe-puro-GFP-Mps1 and Western blot was performed to detect Mps1 and p-ERK expression.</p><p><b>RESULTS</b>In melanoma cells harboring BRAFWT genotype and transfected with pBabe-puro-GST-BRAF-WT and pBabe-puro-GFP-Mps1-WT, phospho-ERK levels were notably reduced as compared to either negative control or empty vector. However, cells transfected with pBabe-puro-GST-BRAFV600E and pBabe-puro-GFP-Mps1-WT, phospho-ERK levels did not change significantly compared with either negative control or empty vector. Knockout of Mps1 in BRAF wild-type cell lines led to an increased ERK activity. However, there was no significant change of ERK activity in BRAFV600E cell lines in the absence of Mps1. The expression of p-ERK in BRAFV600E mutant cell lines infected with pBabe-puro-GFP-Mps1-WT did not show any significant difference from either negative control or empty vector.</p><p><b>CONCLUSIONS</b>Based on these findings, it suggests that there exists an auto-regulatory negative feedback loop between the Mps1 kinase and BRAFWT/ERK signaling. Oncogenic BRAFV600E abrogates the regulatory negative feedback loop of Mps1 on the MAPK pathway.</p>

ABSTRACT
The study of neuronal activity with low frequency has shown an increasing interest for its greater stability and reliability recent years. One challenge in analyzing this kind of activity is to find similarities and differences between signals efficiently and effectively. The traditional analysis methods, such as short-time Fourier transform, are easily obscured by background noises and often involve a large number of parameters. Therefore, this paper introduces a novel time-frequency analysis method based on wavelet transformation and half-ellipsoid modeling to extract instantaneous frequency and instantaneous phase information. This method overcomes some shortcomings of conventional time-frequency analysis. In this method, wavelet transformation is used to provide high-level representations of raw signals, and parsimonious half-ellipsoid models are used to extract changes in time domain and frequency domain of neural recordings. The method was validated to local field potentials (LFPs) of olfactory bulb of anesthetized rats during three different odor stimuli. The results suggested that this method could detect odor-relevant features from olfactory signals with large variability. The Odors then were classified with support vector machine (SVM) algorithm and the classification accuracy reached 79.4%.

ER-α36, a novel variant of ER-α, is expressed in breast, uterus, digestive tract, respiratory tract etc. The aim of the present study was to investigate the distribution and expression of ER-α36 in the central nervous system (CNS). Here, we comparatively analyzed the expression pattern of ER-α36 in the hippocampus and cortex of neonatal (1-day-old) and adult (12-week-old) Sprague-Dawley (SD) rats by using immunohistochemistry/immunocytochemistry analysis and Western blot. The results showed that ER-α36 was expressed both in hippocampus and cortex of adult rats, but mainly distributed in pyramidal neurons. ER-α36 was mainly located on the cytomembrane of hippocampal and cortical neurons from neonatal rats. Compared with the cortical neurons, the hippocampal neurons showed lower ER-α36 protein expression in the neonatal rats, but exhibited higher level of ER-α36 in the adult rats. Furthermore, the adult rats showed higher levels of ER-α36 expression in both hippocampus and cortex compared with the neonatal rats. These results suggest that ER-α36 might be involved in the regulation of membrane-initiated estrogen signaling throughout the postnatal development of diverse brain regions, and thus will be a potential target for the treatment of degenerative diseases in nervous system.

ABSTRACT
Objective To investigate the reproductive endocrine status of women with epilepsy at childbearing age and to systematically analyze the clinical features of reproductive endocrine disorders, especially polycystic ovarian syndrome (PCOS),to facilitate early detection and timely intervention. Methods In this study, scoring of anthropometry and physical signs, menstrul assessment, examination of sex hormone and pelvic ultrasound in women with epilepsy at childbearing age were performed, and the data such as overweight, central obesity,oligo/amenorrhea,luteinizing hormone (LH)/follicule-stimulating hormone (FSH),hyperandrogenism and polycystic ovary (PCO) were collected. The characteristics of their reproductive endocrine hormone disorders were analyzed statistically. Results The age of these patients was (22. 5 ± 7.0 ) years, and women younger than 30 years old and at their peak fertility accounted for 84. 89%. The prevalence rate of PCOS in women with epilepsy at childbearing age (12. 75% ) was significantly higher than that of ordinary women at childbearing age (7.2%) in China.Highly specific indicators for PCOS were hyperandrogenism (100%),LH/FSH > 2 (93%) and oligo/amenorrhea (90%),whilst the highly sensitive indicators for PCOS were PCO (92%), oligo/amenorrhea (85%) and hyperandrogenism (54%). This study revealed statistically significant difference in LH,LH/FSH and testosterone (T) between PCOS group (LH: (10.24 ± 6.92) IU/L; LH/FSH;(2.20 ± 1.16);T: ( 1.07 ± 0. 35) ng/ml) and non-PCOS group ( LH: (4. 16 ± 2.62 ) IU/L; LH/FSH:( 0. 87 ± 0. 56 );T: (0. 46 ±0. 25) ng/ml,t = -3. 899,-4. 240 and -4. 918 respectively, all P <0. 01 ). Conclusions Hormone indices are objective indicators for the diagnosis of PCOS. In clinical practice, attention should be paid to height, weight, abodominal circumference, menstrul history and ultrasound examination of the ovary in women with epilepsy. When reproductive endocrine hormone disorders are suspected from clinical features, the sex hormones (T,LH,and FSH ) should be checked to allow timely detection and early interventions.

ABSTRACT
It was found that Ag nanoparticles (NPs) could enhance the chemiluminescence (CL) intensity of luminol- potassium ferricyanide system. On the basis of enhancement effect,a flow injection method was developed for the determination of terbutaline sulfate. The structure and shape of Ag NPs were characterized by transmission electron microscopy. The UV-Vis absorption spectra and chemiluminescence spectra suggested that new luminophor was not formed after Ag NPs introducing in luminol-potassium ferricyanide chemiluminescence system. A possible mechanism of Ag NPs strengthening on luminol-potassium ferricyanide CL reaction was also discussed. The effect of concentration of luminol, potassium ferricyanide, sodium hydroxide and Ag NPs on CL reaction was investigated respectively. In the optimum conditions, the linear range was 1.0×10~(-9) -2.0 ×10~(-5) g/L(r =0.9935) and the detection limit was 1×10~(-10) g/L. The relative standard deviation (RSD) was 3.6% for 1. 0 ×10~(-6) g/L terbutaline sulfate (n = 11 ). The recommended method has been successfully applied to the determination of terbutaline sulfate tablets and the recovery was between 98. 5% - 102. 5% ,moreover the results were almost identical with the same results of Pharmacopoeia method.
