ABSTRACT

@#AIM: To analysis the epidemiological and clinical characteristics of ocular trauma in Shaanxi and surrounding areas, and provide research support for the formulation of prevention and treatment measures for ocular trauma. METHODS: Retrospective study. The general data and clinical data of 1 723 unilateral ocular trauma patients(1 723 eyes)admitted to our hospital from January 2 018 to April 2 020 were collected to establish the ocular trauma database and analyzed.RESULTS: Among the 1 723 cases(1 723 eyes)of ocular trauma patients, young and middle-aged patients(20-59 years old)accounted for the majority(1 149 eyes, 66.69%)and mainly were male patients(1 392 eyes, 80.79%), and the vast majority of patients came from rural areas(1 270 eyes, 73.71%)and engaged in manual labor(1 288 eyes, 74.75%). The main causes of ocular trauma were blunt tools(511 eyes, 29.66%), sharp objects(423 eyes, 24.55%), splashing metal(non-metal)foreign bodies(299 eyes, 17.35%), falling(183 eyes, 10.62%), <i>etc</i>. The main places of injury were workplaces(633 eyes, 36.74%), farming places(474 eyes, 27.51%), residential homes(302 eyes, 17.53%), public places(248 eyes, 14.39%), <i>etc</i>. The main type of ocular trauma was open globe injury(1 311 eyes, 76.09%), and the main therapy was surgical treatment(1 638 eyes, 95.07%). Logistic analysis found that age, post-injury visit time, types of ocular trauma, complications of ocular trauma, history of ocular trauma were all important factors influencing prognosis of ocular trauma patients(<i>P</i><0.05). CONCLUSION: Ocular trauma in Shaanxi and surrounding areas has characteristics of age, sex, place of residence, and occupation. For special occupations and special populations, timely targeted health education and prevention measures should be taken to reduce the incidence of ocular trauma. In addition, the timely treatment of ocular trauma can help improve the prognosis of patients.

ABSTRACT

To explore the pharmacogenomic markers that affect the platinum-based chemotherapy response in non-small-cell lung carcinoma (NSCLC), we performed a two-cohort of genome-wide association studies (GWAS), including 34 for WES-based and 433 for microarray-based analyses, as well as two independent validation cohorts. After integrating the results of two studies, the genetic variations related to the platinum-based chemotherapy response were further determined by fine-mapping in 838 samples, and their potential functional impact were investigated by eQTL analysis and in vitro cell experiments. We found that a total of 68 variations were significant at P < 1 × 10-3 in cohort 1 discovery stage, of which 3 SNPs were verified in 262 independent samples. A total of 541 SNPs were significant at P < 1 × 10-4 in cohort 2 discovery stage, of which 8 SNPs were verified in 347 independent samples. Comparing the validated SNPs in two GWAS, ADCY1 gene was verified in both independent studies. The results of fine-mapping showed that the G allele carriers of ADCY1 rs2280496 and C allele carriers of rs189178649 were more likely to be resistant to platinum-based chemotherapy. In conclusion, our study found that rs2280496 and rs189178649 in ADCY1 gene were associated the sensitivity of platinum-based chemotherapy in NSCLC patients.

ABSTRACT

This study aims to systematically evaluate the effect of oral Chinese patent medicines on hypertension with network Meta-analysis. Randomized controlled trials on the treatment of hypertension with oral Chinese patent medicine combined with conventional western medicine were retrieved from China National Knowledge Infrastructure(CNKI), Wanfang, VIP, SinoMed, PubMed, EMbase, and Cochrane Library(from establishment of the database to August 2021). Two researchers independently screened the articles, extracted the data, and evaluated article quality. Then R 4.1.0 was employed for data analysis. Finally, 195 eligible articles were screened out, involving 22 546 patients and 18 oral Chinese patent medicines. The results of the network Meta-analysis are as follows. In terms of reducing systolic blood pressure(SBP) and diastolic blood pressure(DBP), Xuesaitong, Qiangli Dingxuan Tablets, Songling Xuemaikang Capsules combined with conventional western medicine are superior. In improving blood lipids, the overall effects of Xinmaitong Capsules, Compound Xueshuantong Capsules, Ginkgo Folium preparations, Yindan Xinnaotong Soft Capsules, and Naoxintong Capsules combined with conventional western medicine are outstanding. In terms of regulating endothelial function, Yindan Xinnaotong Soft Capsules, Xinmaitong Capsules, Zhenju Jiangya Tablets, Compound Danshen Dripping Pills, Xuesaitong with conventional western medicine have certain advantages. As for the safety, the incidence of adverse reactions of conventional western medicine combined with oral Chinese patent medicines is lower than that of conventional western medicine alone. In summary, compared with conventional western medicine alone, the 18 oral Chinese patent medicines combined with conventional western medicine in the treatment of hypertension show advantages in improving blood pressure, blood lipids, and endothelial function. Among them, Xuesaitong, Qiangli Dingxuan Tablets, and Songling Xuemaikang Capsules may be the best oral Chinese patent medicines for lowering blood pressure. The conclusion needs to be further verified by more high-quality studies.

ABSTRACT

Food wastes are rich in nutrients and can be used for producing useful chemicals through biotransformation. Some oleaginous microorganisms can use food wastes to produce lipids and high value-added metabolites such as polyunsaturated fatty acids, squalene, and carotenoids. This not only reduces the production cost, but also improves the economic value of the products, thus has large potential for commercial production. This review summarized the advances in food waste treatment, with a focus on the lipid production by oleaginous microorganisms using food wastes. Moreover, challenges and future directions were prospected with the aim to provide a useful reference for related researchers.

ABSTRACT

The current clinical evidence and underlying mechanisms of acupuncture and moxibustion in the treatment of irritable bowel syndrome (IBS) were summarized, so as to better optimize clinical treatment. The relevant articles of acupuncture and moxibustion in the treatment of IBS in recent years were retrieved and summarized. We found that the clinical efficacy of acupuncture and moxibustion in the treatment of IBS was relatively reliable. However, the mutual relationships among various mechanisms of action such as abnormal gastrointestinal motility, high visceral sensitivity, intestinal microenvironment disorders, and abnormal intestinal-brain interactions need to be further explored. The authors believe that in-depth explorations of the bidirectional regulation of "gut-brain axis", the law of changes in the abundance and diversity of intestinal flora, and the establishment of a more ideal animal model of TCM syndrome differentiation are useful ideas for subsequent research.

ABSTRACT

OBJECTIVE@#To investigate the effect of tumor necrosis factor death receptor (DR) 4 demethylation to the proliferation and apoptosis of myeloid leukemia K562 cells.@*METHODS@#The logarithmic phase of K562 cells were treated by desitabine (DCA) at 0, 0.8, 1.6 and 3.2 μmol/L, and the cells were divided into control group, DCA low dose group, DCA medium dose group and DCA high dose group respectively. The cells in control group were treated by tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) 0.5 μg/ml for 24 h, and the cells were divided into TRAIL group. The cells in DCA high dose group were treated by TRAIL 0.5 μg/ml for 24 h, and were divided into DCA high dose + TRAIL group. Methylation-specific polymerase chain reaction (MS-PCR) was used to measure the methylation status of the DR4 gene promoter in the control group and DCA low, medium and high dose groups. Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) and Western blot were used to determine the relative expression of DR4 mRNA and protein in the control group and DCA low, medium and high dose groups. Dime- thylthiazole (MTT) method was used to determine the inhibition rate of cell proliferation of the cells in control group, DCA high dose group, TRAIL group, DCA high dose + TRAIL group. Flow cytometry was used to determine the apoptotic rate of the cells in control group, DCA high dose group, TRAIL group, DCA high dose + TRAIL group.@*RESULTS@#The cells in the control group were methylation-positive, the brightness of the methylation bands of the cells in the DCA low, medium, and high dose groups was gradually decreased to disappear, and the DCA high dose group showed negative for methylation. The relative expression of DR4 mRNA and protein in the control group, DCA low, medium and high dose groups was increased sequentially (r=0.624, 0.704). The inhibition rate of cell proliferation of the cells in the control group, DCA high dose group, TRAIL group, DCA high dose + TRAIL group was increased sequentially (r=0.653, 0.754, 0.709, 0.725) at 24, 48 and 72 h.@*CONCLUSION@#DCA can reverse the methylation level of DR4 gene promoter in ML K562 cells and up-regulate the expression of DR4, which may enhance the proliferation inhibition and apoptosis promotion effects of TRAIL on K562 cells.

ABSTRACT

The ability of tumor cells to sustain continuous proliferation is one of the major characteristics of cancer. The activation of oncogenes and the mutation or inactivation of tumor suppressor genes ensure the rapid proliferation of tumor cells. The PI3K-Akt-mTOR axis is one of the most frequently modified signaling pathways whose activation sustains cancer growth. Unsurprisingly, it is also one of the most commonly attempted targets for cancer therapy. FK506 binding protein 8 (FKBP8) is an intrinsic inhibitor of mTOR kinase that also exerts an anti-apoptotic function. We aimed to explain these contradictory aspects of FKBP8 in cancer by identifying a "switch" type regulator. We identified through immunoprecipitation-mass spectrometry-based proteomic analysis that the mitochondrial protein prohibitin 1 (PHB1) specifically interacts with FKBP8. Furthermore, the downregulation of PHB1 inhibited the proliferation of ovarian cancer cells and the mTOR signaling pathway, whereas the FKBP8 level in the mitochondria was substantially reduced. Moreover, concomitant with these changes, the interaction between FKBP8 and mTOR substantially increased in the absence of PHB1. Collectively, our finding highlights PHB1 as a potential regulator of FKBP8 because of its subcellular localization and mTOR regulating role.

ABSTRACT

Objective:To observe the effect of modified Shengjiangsan on renal fibrosis in rats with membranous nephropathy (MN) and to explore the mechanism of its complications of renal fibrosis. Method:Rats were injected with cationized bovine serum albumin(C-BSA)in the tail vein to establish a rat model of membranous nephropathy. The normal group,model group,modified Shengjiangsan group (27.3 g·kg<sup>-1</sup>)and benazepril group(10 mg·kg<sup>-1</sup>)were established in this study. Each group was given corresponding dosage of the drug once a day for 4 weeks of continuous intervention. After the administration,we observed the pathological changes of rat kidneys by the technology of Masson staining, silverhexylamine iodate (PASM) staining, transmission electron microscopy (TEM), immunofluorescence technology (IF) was used to detect immunoglobulin(Ig)G deposition in rat kidneys. The levels of interleukin-1<italic>β</italic> (IL-1<italic>β</italic>), interleukin-6 (IL-6), tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>) in rat serum were detected by enzyme-linked immunosorbent assay (ELISA) method. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and immunohistochemistry (IHC) were used to detect the mRNA and protein expression levels of monocyte chemotactic protein 1 (MCP-1), intercellular adhesion molecule 1 (ICAM-1), nuclear factor kappa B (NF-<italic>κ</italic>B), Toll-like receptor 4 (TLR4), plasminogen activator inhibitor 1 (PAI-1), transforming growth factor-<italic>β</italic><sub>1</sub> (TGF-<italic>β</italic><sub>1</sub>), <italic>α</italic>-smooth muscle actin (<italic>α</italic>-SMΑ) and type Ⅳ Collagen (Collagen Ⅳ) in rat kidney tissues. Result:Compared with normal group, the kidney tissue of the model group was obviously fibrotic, the serum levels of IL-1<italic>β</italic>, IL-6, and TNF-<italic>α</italic> were significantly increased(<italic>P</italic><0.05), and the expressions of MCP-1, ICAM-1, NF-<italic>κ</italic>B, TLR4, PAI-1, TGF-<italic>β</italic><sub>1</sub>, <italic>α</italic>-SMA and Collagen Ⅳ mRNA and protein in kidney tissue were significantly increased(<italic>P</italic><0.05). Compared with model group, modified Shengjiangsan and benazepril significantly improved renal fibrosis in rats, reduced the levels of IL-1<italic>β</italic>, IL-6, and TNF-<italic>α</italic> in the serum of MN rats(<italic>P</italic><0.05), down-regulated MCP-1, ICAM-1, NF-<italic>κ</italic>B, TLR4, PAI-1, TGF-<italic>β</italic><sub>1</sub>, <italic>α</italic>-SMA and Collagen Ⅳ mRNA and protein expression in kidney tissue(<italic>P</italic><0.05). Conclusion:Modified Shengjiangsan can reduce the release and expression of inflammatory factors by down-regulating the TLR4/NF-<italic>κ</italic>B signaling pathway, inhibit renal fibrosis, and reduce renal damage in MN rats.

ABSTRACT

Objective:To study the quality inspection standard of seeds of <italic>Belamcanda chinensis</italic> and establish the quality grading standard of seeds of <italic>B. chinensis</italic>. Method:The purity of seeds of <italic>B. chinensis</italic> from different producing areas was analyzed,and the 1 000-grain weight,water content,viability and germination rate of seeds of <italic>B. chinensis</italic> with different diameters were determined after diameter classification. K-means was used for cluster analysis to preliminarily formulate the quality grading standard of seeds of <italic>B. chinensis</italic>. Result:There were obvious regional differences in the size of seeds of <italic>B. chinensis</italic>. The particle size of seeds of <italic>B. chinensis</italic> of Xiaochang,Dawu and Huanggang Academy of Agricultural Sciences in Hubei Province was larger and mostly between 4.5 mm to 5.5 mm. The seeds of<italic> B. chinensis </italic>from Tuanfeng,Qujing,Yunnan,and Anguo,Hebei province had smaller particle sizes than those from the other three producing areas,which were mostly between 3.5 mm to 5.0 mm. Arils in seeds of <italic>B. chinensis</italic> were removed by 10% NaClO to effectively break seed dormancy and significantly improve the seed germination rate. On this basis,the seed diameter was taken as the main grading index,and the seed moisture content,viability,germination rate and purity were taken as important reference indexes. The quality of seeds of <italic>B. chinensis</italic> was preliminarily divided into three grades,grade Ⅰ:seed diameter≥5.5 mm,moisture content≤10%,viability≥94%,germination rate≥60%,and cleanliness≥93%;grade Ⅱ:seed diameter≥4.5 mm,moisture content≤10%,viability≥90%,germination rate≥55%,and cleanliness≥85%;and grade Ⅲ:seed diameter≥3.5 mm,moisture content≤10%,viability≥84%,germination rate≥45%,cleanliness≥80%. Conclusion:In this study,the quality grading standard of seeds of <italic>B. chinensis</italic> was preliminarily established to provide reference for the quality evaluation of seeds of <italic>B. chinensis</italic> and the breeding of improved varieties. In addition,the maturity and the storage time of seeds of <italic>B. chinensis</italic> have a greater impact on the quality of seeds,so it is recommended to select fully mature(dark black) seeds and new seeds for production.

ABSTRACT

Objective:To observe the effect of Yiqiyangyin Huoxuetongluo prescription on janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) signaling pathway and cell apoptosis in rats with diabetic nephropathy (DN), and to explore the mechanism of its intervention in DN. Method:A total of 100 SD rats were randomly divided into an experimental group (<italic>n</italic>=80) and a normal group (<italic>n</italic>=20). The DN model was induced by high-sugar and high-fat diet combined with intraperitoneal injection of streptozotocin (STZ) in the experimental group, and confirmed by the pathological changes of kidney tissues in rats (three in each group) observed under light and electron microscopes. The model rats were randomly divided into a model group (normal saline, equal volume), low-, medium-, and high-dose (5.775, 11.550, and 23.100 g·kg<sup>-1</sup>) Yiqiyangyin Huoxuetongluo prescription groups, and an irbesartan group (irbesartan tablets, 0.016 g·kg<sup>-1</sup>). After drug intervention (<italic>i.g</italic>., once a day for 16 consecutive weeks), the 24-hour urine total protein (UTP), serum total cholesterol (TC), triglyceride (TG), creatinine (SCr), and blood urea nitrogen (BUN) levels of the rats were measured. Western blot was used to detect the protein expression of JAK2/STAT3 signaling pathway. Immunohistochemistry was used to determine the protein expression of B-cell lymphoma 2 (Bcl-2), Bcl-2-associated X protein (Bax), zonula occludens-1 (ZO-1), and actinin-4 in rat kidney tissues. Result:Compared with the normal group, the model group exhibited elevated UTP, serum TC, TG, BUN, and SCr levels (<italic>P</italic><0.05), severe pathological damage of rat kidney tissues, up-regulated expression of phospho-JAK2 (p-JAK2), phospho-STAT3 (p-STAT3), and Bax, increased renal cell apoptosis, and diminished expression of Bcl-2, ZO-1, and actinin-4 (<italic>P</italic><0.05). Compared with the model group, the Yiqiyangyin Huoxuetongluo prescription group and the irbesartan group showed dwindled UTP, serum TC, TG, BUN, and SCr levels (<italic>P</italic><0.05), relieved pathological damage of rat kidney tissues, down-regulated p-JAK2, p-STAT3, and Bax expression, and up-regulated expression of Bcl-2, ZO-1, and actinin-4 (<italic>P</italic><0.05). Conclusion:Yiqiyangyin Huoxuetongluo prescription can reduce renal cell apoptosis and improve the prognosis of DN by inhibiting the activation of JAK2/STAT3 signaling pathway.

ABSTRACT

Objective:To observe the effects of Yishen Tongluo prescription (YTP) on autophagy-related proteins in rats with membranous nephropathy (MN) and explore its possible molecular mechanism in protecting the kidney. Method:Twenty of 80 Sprague-Dawley (SD) rats were randomly selected as the normal control, and the rest rats were pre-immunized and injected with cationized bovine serum albumin (C-BSA) through the tail vein to induce MN. The SD rats that were successfully modeled were randomized into the model group, benazepril hydrochloride group (10 mg·kg<sup>-1</sup>), and low- (6.61g·kg<sup>-1</sup>), medium- (13.22 g·kg<sup>-1</sup>), and high-dose (26.44 g·kg<sup>-1</sup>) YTP groups, and administered with the corresponding drugs by gavage, once a day, for four consecutive weeks. Then the changes in such quantitative indicators as plasma albumin (ALB), triglyceride (TG), total cholesterol (TC), serum creatinine (SCr), blood urea nitrogen (BUN), and 24-hour urinary total protein (UTP) were detected, followed by hematoxylin and eosin (HE) staining, Masson's trichrome staining, and periodic Schiff-methenamine (PASM) staining for observing the pathological changes in kidney under the transmission electron microscope (TEM). The deposition of immunoglobulin G (IgG) and complement 3 (C3) in the glomerulus was detected by fluorescence immunoassay. The expression levels of autophagy marker proteins Beclin-1, microtubule-associated protein light chain 3Ⅱ (LC3Ⅱ), and p62 were measured by immunohistochemistry (IHC), and those of related proteins in the adenosine monophosphate-activated protein kinase / mechanisic target of rapamycin/Unc-51-like kinase 1 (AMPK/mTOR/ULK1) signaling pathway were determined by Western blot assy. Result:Compared with the normal group, the model group exhibited significantly increased UTP (<italic>P</italic><0.01) and serum TG and TC (<italic>P</italic><0.01), decreased ALB (<italic>P</italic><0.01), disordered glomerular structure, enlarged volume, thickened basement membrane, vacuolated renal tubules, excessively deposited collagen fibers and fuchsinophilic proteins, extensively fused podocyte foot processes, and diffusely deposited IgG and C3 in glomerular capillary loops. Besides, the expression levels of Beclin-1, LC3II, and phosphorylated AMPK (p-AMPK) decreased (<italic>P</italic><0.01), while those of p62, phosphorylated mTOR (p-mTOR), and phosphorylated ULK1 (p-ULK1) increased (<italic>P</italic><0.01). The comparison with the model group revealed that the TG, TC, and UTP levels in the low-, medium-, and high-dose YTP groups and the benazepril hydrochloride group were reduced to varying degrees (<italic>P</italic><0.05, <italic>P</italic><0.01), whereas the ALB level was increased (<italic>P</italic><0.01). There was no statistically significant difference in SCr or BUN level. The pathological damages were alleviated. The expression levels of Beclin-1, LC3Ⅱ, and p-AMPK were up-regulated (<italic>P</italic><0.05, <italic>P</italic><0.01), while those of p62, p-mTOR, and p-ULK1 were down-regulated (<italic>P</italic><0.05, <italic>P</italic><0.01). Conclusion:YTP protects the kidney of rats with MN possibly by regulating related proteins in the AMPK/mTOR/ULK1 signaling pathway and activating the autophagy.

ABSTRACT

Objective:To observe the clinical efficacy as well as anti-inflammatory and anti-fibrosis mechanism of Yishen Tongluo decoction in the treatment of chronic glomerulonephritis (CGN) with spleen and kidney Qi deficiency and blood stasis syndrome. Method:According to the random number table method, one hundred and twenty patients were divided into control group (60 cases) and observation group (60 cases). The two groups were given oral valsartan capsules, 160 mg/time, 1 time/day, and dipyridamole tablets orally, 50 mg/time, 3 times/day. Patients in control group additionally took Wubi Shanyao pills orally, 9 g/time, 2 times/day, while patients in observation group additionally took Yishen Tongluo decoction orally, 1 dose/day. Both groups were treated continuously for four months. Before and after treatment, 24 h urine total protein (24 h UTP), creatinine (SCr), urea nitrogen (BUN), serum cystatin C (CysC), tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>), interleukin-6 (IL-6), IL-17, interferon-<italic>γ</italic> (IFN-<italic>γ</italic>), transforming growth factor-<italic>β</italic><sub>1</sub> (TGF-<italic>β</italic><sub>1</sub>), connective tissue growth factor (CTGF), matrix metalloproteinase-9 (MMP-9), tissue inhibitor of metalloproteinases (TMP-1) and hypoxia inducible factor-1<italic>α </italic>(HIF-1<italic>α</italic>) levels were detected in both groups. The scores of spleen and kidney Qi deficiency and blood stasis syndrome were graded. Urine protein, and urine red blood cells (urinary RBC) were monitored. Result:The 24 h UTP, SCr, BUN and CysC levels of the observation group were lower than those of the control group (<italic>P</italic><0.01). The score of spleen and kidney Qi deficiency and blood stasis syndrome in the observation group was lower than that in the control group (<italic>P</italic><0.01). The levels of TNF-<italic>α</italic>, IL-6, IL-17 and IFN-<italic>γ </italic>in the observation group were lower than those in the control group (<italic>P</italic><0.01). The levels of TGF-<italic>β</italic><sub>1</sub>, CTGF, TMP-1 and HIF-1<italic>α</italic> in the observation group were lower than those in the control group (<italic>P</italic><0.01). While MMP-9 level was higher than that in control group (<italic>P</italic><0.01). Conclusion:On the basis of conventional Western medicine treatment, Yishen Tongluo decoction in the treatment of CGN patients with spleen and kidney Qi deficiency and blood stasis syndrome can reduce proteinuria, and improve traditional Chinese medicine (TCM) syndromes, with anti-inflammatory and anti-kidney fibrosis effects. Thereby, it plays a role in protecting renal function and delaying the malignant progression of renal function, with high clinical efficacy and value of clinical use.

ABSTRACT

Objective:To investigate the intervention effect of modified Shengjiangsan on hypoxia-inducible factor-1<italic>α </italic>(HIF-1<italic>α</italic>)/nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4) signaling pathway in membranous nephropathy (MN) rats and to explore its mechanism to reduce oxidative stress and apoptosis in renal tissues. Method:Cationized bovine serum albumin (C-BSA) was injected into the tail vein of rats to replicate the MN model. Rats were randomly divided into a model group, a modified Shengjiangsan group, and a benazepril group after modeling, and administered by gavage once a day accordingly. At the end of the 4<sup>th</sup> week, the 24-h urine total protein (UTP), urea nitrogen (BUN), and serum creatinine (SCr) levels of each group were detected. Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of superoxide dismutase (SOD), malondialdehyde (MDA), and reactive oxygen species (ROS) in renal tissues of rats. In situ end labeling(TUNEL) staining was used to detect the cell apoptosis rate. The mRNA and protein expression levels of HIF-1<italic>α</italic> and NOX4 were detected by real-time fluorescence-based quantitative polymerase chain reaction(Real-time PCR)and Western blot, respectively. The immunohistochemistry method was used to detect the protein expression levels of B-cell lymphomas -2 (Bcl-2), B-cell lymphomas xl (Bcl-xl), Bcl-2 associated X protein (Bax), Bcl-2 cell death regulator antibody (Bim). Result:Compared with the normal group, the model group showed increased UTP (<italic>P</italic><0.05), decreased SOD, elevated MDA and ROS (<italic>P</italic><0.05), up-regulated mRNA and protein expression of HIF-1<italic>α</italic> and NOX4 (<italic>P</italic><0.05), enhanced protein expression of Bax and Bim, declining protein expression of Bcl-xl and Bcl-2 (<italic>P</italic><0.05), and increased cell apoptosis in renal tissues. Compared with the model group, the modified Shengjiangsan group and the benazepril group displayed declining UTP (<italic>P</italic><0.05), up-regulated SOD, decreased MDA and ROS (<italic>P</italic><0.05), down-regulated mRNA and protein expression of HIF-1<italic>α</italic> and NOX4 (<italic>P</italic><0.05), diminished protein expression of Bax and Bim, elevated protein expression of Bcl-xl and Bcl-2 (<italic>P</italic><0.05), and reduced cell apoptosis in renal tissues (<italic>P</italic><0.05). Conclusion:The protective effect of modified Shengjiangsan on the kidney is presumedly achieved by reducing the oxidative stress and apoptosis in renal tissues of MN rats via inhibiting the HIF-1<italic>α</italic>/NOX4 signaling pathway.

ABSTRACT

Objective:To observe the activation of microglia in hippocampus of depressed and anxious mice induced by maternal separation with acute restraint stress and the expression of interleukin-1<italic>β</italic>(IL-1<italic>β</italic>),interleukin-6(IL-6),tumor necrosis factor-<italic>α</italic>(TNF-<italic>α</italic>), investigating the mechanism of Wenyang Jieyu prescription in treating anxiety and depression. Method:Eighty four male C57BL offspring were randomly divided into control group, acute restraint stress group and model group on postnatal day 0(PD0). Maternal separation combined with acute restraint stress was used to prepare anxious and depressed model mice, dividing the model mice into model group, Wenyang, Jieyu, Wenyang Jieyu and fluoxetine group according to random number table method. During the period of PD21-PD90, the control, acute restraint stress and model mice were fed with normal diet, with the other groups fed with corresponding medicine mixed diet. The Wenyang, Jieyu and Wenyang Jieyu groups were given 5.85, 12.03 and 16.71 g·kg<sup>-1</sup>·d<sup>-1</sup> respectively. The fluoxetine group was given 2.60 mg·kg<sup>-1</sup>·d<sup>-1</sup>. Open field, zero maze test and social interaction tests were used to evaluate the anxiety and depression of model mice. The expression of Iba-1 in hippocampal microglia was detected by immunohistochemistry(IHC). The mRNA expression of IL-1<italic>β</italic>, IL-6, TNF-<italic>α</italic>, Iba-1 and glucocorticoid receptor(GR)were detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Result:Compared with the control group, total movement distance and time spent in central zone in 5 min of the model mice significantly decreased(<italic>P</italic><0.01), time spent in opened arm and total movement distance decreased significantly(<italic>P</italic><0.05,<italic>P</italic><0.01), investigation time during testing and training increased significantly(<italic>P</italic><0.01). The expression of Iba-1 protein and mRNA,IL-1<italic>β</italic>,IL-6,TNF-<italic>α</italic> mRNA significantly increased(<italic>P</italic><0.01), the expression levels of GR mRNA significantly decreased(<italic>P</italic><0.01). The result of IHC staining showed that microglia were over activated. Compared with the model group, total movement distance and time spent in central zone in 5 min of mice in the Wenyang Jieyu and fluoxetine group significantly increased(<italic>P</italic><0.05,<italic>P</italic><0.01).Time spent in opened arm significantly increased(<italic>P</italic><0.01). Investigation time during testing and training significantly decreased(<italic>P</italic><0.05,<italic>P</italic><0.01). IHC staining showed that microglia recovered partly in both groups. Conclusion:The comprehensive curative effect and pharmacological action of Wenyang Jieyu prescription were better than Wenyang prescription and Jieyu prescription. Wenyang Jieyu prescription can treat anxiety and depression in maternal separation and acute restraint stress mice, its possible mechanism may be related to the decreased activation of microglia, down-regulation of IL-1<italic>β</italic>,IL-6,TNF-<italic>α</italic> expression and up-regulation of GR expression.

ABSTRACT

Objective:The purpose of this article was to observe the effect of modified Shengjiangsan on podocyte apoptosis in membranous nephropathy (MN) rats, to explore the molecular mechanism of its treatment of MN and to provide experimental basis for its clinical application. Method:The MN rat model was established by injection of cationic bovine serum albumin into the tail vein of rats. The successfully modeled rats were then randomly divided into model group (equal volume of normal saline), modified Shengjiangsan group (27.3 g·kg<sup>-1</sup>) and benazepril group (10 mg·kg<sup>-1</sup>), with corresponding drug dosage once a day for 4 weeks of continuous intervention. After drug administration, the 24-hour urine protein (UTP) was detected. Real time fluorescent quantitative polymerase chain reaction (Real-time PCR) and immunohistochemical (IHC) methods were used to detect Podocalyxin, Nephrin, Podocin, Synaptopodin mRNA and protein expression levels in rat kidney tissue. terninal-deoxynucleoitidyl transferase medsated nick and labeling (TUNEL) method was used to detect cell apoptosis rate in rat kidney tissue, and Western blot was used to detect Notch1, Hes1, B lymphoblastoma-2 (Bcl-2) associated X protein (Bax), and Bcl-2 protein expression levels in rat kidney tissue. Result:Compared with the normal group, UTP in the model group increased significantly, renal tissue cell apoptosis increased significantly, podocyte marker proteins podocalyxin, Nephrin, Podocin, Synaptopodin mRNA and protein expression levels decreased significantly, and Notch1, Hes1, Bax protein expression increased significantly, and Bcl-2 protein expression was significantly reduced(<italic>P</italic><0.05). Compared with the model group, UTP levels in MN rats were significantly reduced in modified Shengjiangsan and benazepril groups, with reduced rate of renal cell apoptosis, increased mRNA and protein expression levels of podocalyxin, Nephrin, Podocin, and Synaptopodin in renal tissue, decreased Notch1, Hes1, Bax protein expression, and increased Bcl-2 protein expression(<italic>P</italic><0.05). Conclusion:Modified Shengjiangsan can inhibit the Notch signaling pathway, reduce the apoptosis of rat kidney tissue podocytes, and reduce the kidney injury of MN rats.

ABSTRACT

The present study evaluates different processing and drying methods and investigates their effects on the chemical components in Paeoniae Radix Alba via content determination. The fresh medicinal materials of Paeoniae Radix Alba collected from Bozhou of Anhui province were processed(boiled and peeled) and dried(hot air-dried, infrared-dried, and microwave-dried) at different temperatures(40, 50, 60 and 70 ℃), and the 11 components(monoterpene glycosides, polyphenols, tannin, and benzoic acid) in Paeoniae Radix Alba were determined by ultra-performance liquid chromatography coupled to triple quadrupole with electrospray tandem mass spectrometry(UPLC-TQ-MS). Then the compounds in processed and dried samples were analyzed by partial least squares discriminant analysis(PLS-DA) and orthogonal partial least squares discriminant analysis(OPLS-DA), and the contribution rates of differential components were evaluated by variable important in projection(VIP). The results indicated that the samples obtained by different processing and drying methods could be distinguished. Albiflorin, gallic acid, 1,2,3,4,6-pentakis-O-galloyl-β-D-glucose, and benzoic acid were the common differential components in boiled Paeoniae Radix Alba. Benzoic acid was the common differential component in peeled Paeoniae Radix Alba. Gallic acid was the common differential component in Paeoniae Radix Alba dried by different methods. The samples could not be distinguished after drying at different temperatures due to the lack of common differential components. This study is expected to provide a reference for the selection of processing and drying methods and the optimization of processing parameters.

ABSTRACT

OBJECTIVE@#To compare the effect of moxibustion combined with basic treatment and simple basic treatment on the clinical symptoms, renal function and hypercoagulable state in patients with idiopathic membranous nephropathy (IMN) of low to medium risk with spleen-kidney deficiency and blood stasis.@*METHODS@#A total of 60 patients with IMN of low to medium risk with spleen-kidney deficiency and blood stasis were randomized into an observation group (30 cases, 2 cases dropped off) and a control group (30 cases, 1 case dropped off). In the control group, the conventional basic treatment of anti-hypertension, regulating blood lipid and anti-coagulation was adopted. On the basis of the control group, moxibustion was applied at Shenshu (BL 23), Pishu (BL 20), Guanyuan (CV 4), Zusanli (ST 36) and Sanyinjiao (SP 6) in the observation group, once a day, 5 days a week continuously with 2 day interval. The treatment of 6 months was required in the both groups. Before treatment and 3 and 6 months into treatment, the total TCM syndrome score, the renal function indexes (24-hour urinary protein quantity [UTP], albumin [ALB], urea nitrogen [BUN] and creatinine [Scr]), the blood coagulation indexes (fibrinogen [FIB], D-Dimer [D-D], p-selection and von Willebrand factor [vWF]), total cholesterol (TC) and triacylglycerol (TG) levels were observed, and the therapeutic efficacy was evaluated on 3 and 6 months into treatment in the two groups.@*RESULTS@#The effective rates of 3 and 6 months into treatment were 78.6% (22/28) and 89.3% (25/28) in the observation group, which were higher than 62.1% (18/29) and 75.9% (22/29) in the control group respectively (@*CONCLUSION@#Moxibustion combined with basic treatment can effectively improve the clinical symptoms, renal function and renal microcirculation in patients with idiopathic membranous nephropathy of low to medium risk with spleen-kidney deficiency and blood stasis, the therapeutic effect is superior to the simple basic treatment.

@#AIM: To analyze the risk factors of corneal edema after phacoemulsification and intraocular lens implantation in patients with cataract. <p>METHODS: The clinical data of 623 patients with cataract(957 eyes)who underwent phacoemulsification and intraocular lens implantation in our hospital between March 2018 and September 2020 were retrospectively analyzed. Patients with corneal edema observed at any time within the first three days after surgery were included in corneal edema group(82 cases, 82 eyes), and patients without corneal edema were enrolled as non-corneal edema group(541 cases, 875 eyes). The preoperative, intraoperative and postoperative related indexes of the two groups were collected and included in Logistic regression model to screen the high-risk factors. <p>RESULTS: Logistics regression analysis showed that age(<i>OR</i>=1.162), Emery lens nuclear hardness grading(<i>OR</i>=1.301)and perfusion time(<i>OR</i>=1.172)were independent risk factors for corneal edema after phacoemulsification and intraocular lens implantation, and the anterior chamber depth(<i>OR</i>=0.651)were independent protective factors avoiding occurrence(<i>P</i><0.05).<p>CONCLUSION:The factors involved in corneal edema after phacoemulsification and intraocular lens implantation are complicated in patients with cataract. It is necessary to be vigilant for high-risk patients in clinical practice and take preventive measures during perioperative period so as to promote rapid recovery of prognosis and visual function.

ABSTRACT

Objective To detect the expression of transforming growth factor-β1 (TGF-β1), p38MAPK and bone morphogenetic protein-7 (BMP-7) protein in the liver specimens of patients with hepatic alveolar echinococcosis, and to investigate the potential role of TGF-β1, p38MAPK and BMP-7 protein in hepatic fibrosis caused by hepatic alveolar echinococcosis. Methods A total of 20 patients with hepatic alveolar echinococcosis were enrolled as study subjects, and hepatic specimens were sampled from the sites within 0.5 cm (Group A) and 0.5 to 1.5 cm from hepatic alveolar echinococcosis lesions (Group B), while normal liver specimens sampled from the sites 2 cm and greater from hepatic alveolar echinococcosis lesions served as controls (Group C). The fibrosis of liver specimens was pathological examined using HE and Masson staining, and the expression of TGF-β1, p38MAPK and BMP-7 protein was quantified in liver tissues using Western blotting. The associations of TGF-β1, p38MAPK and BMP-7 protein expression with hepatic fibrosis were assessed. Results HE staining showed the malaligned structure of hepatocytes and destruction of the structure of hepatic lobules at various degrees in liver specimens in groups A and B, with hepatocyte degeneration, atrophy and necrosis, hyperplasia of fibrous tissues and eosinophilic granulocyte infiltration seen, while no abnormal pathological alterations of liver tissues, normal hepatocyte structure and morphology and uniform size, no malaligned structure of hepatocytes, clear structure of hepatic lobules, no or mild hepatocyte degeneration or necrosis, and no eosinophilic granulocyte infiltration were seen in Group C. Masson staining showed that there was hyperplasia of multiple fibrous connective tissues in the liver portal areas in groups A and B, with fibrosis seen in hepatic lobules, while no obvious pathological changes were seen in Group C. There were significant differences seen in TGF-β1 (P < 0.001), p38MAPK (P < 0.01) and BMP-7 protein (P < 0.05) expression in liver tissues in groups A, B and C, and higher TGF-β1, p38MAPK and BMP-7 protein expression was quantified in groups A and B than in Group C (all P values < 0.05), while greater TGF-β1, p38MAPK and BMP-7 protein expression was detected in Group B than in Group C (all P values < 0.05). The expression of TGF-β1, p38MAPK and BMP-7 protein correlated positively with the severity of hepatic fibrosis (r = 0.866, 0.702 and 0.801, all P values < 0.05), and there were significant differences in TGF-β1 (F = 72.580, P < 0.01), p38MAPK (χ2 = 31.705, P < 0.01) and BMP-7 protein expression (χ2 = 48.388, P < 0.01) among liver tissues with different degrees of fibrosis. The TGF-β1 protein expression correlated positively with p38MAPK and BMP-7 protein expression (r = 0.607 and 0.702, both P values < 0.001), and the BMP-7 protein expression also correlated positively with p38MAPK protein expression (r = 0.456, P < 0.001). Conclusion The interaction among TGF-β1, p38MAPK and BMP-7 jointly participates in the development of hepatic fibrosis induced hepatic alveolar echinococcosis.
