ABSTRACT

OBJECTIVE@#To study the role of microRNA-17-5p (miR-17-5p) in the pathogenesis of pediatric nephrotic syndrome (NS) and its effect on renal podocyte apoptosis via the activin A (ActA)/Smads pathway.@*METHODS@#An analysis was performed on 55 children with NS (NS group) who were admitted from March 2018 to March 2019. Fifty healthy children who underwent physical examination during the same period of time were enrolled as the control group. The mRNA expression of miR-17-5p in peripheral blood was measured and compared between the two groups. Human renal podocytes were transfected with antisense oligonucleotide recombinant plasmid containing miR-17-5p (inhibition group) or control vector containing nonsense random sequence (negative control group), and untreated human renal podocytes were used as the blank group. These groups were compared in terms of cell apoptosis and the mRNA and protein expression of miR-17-5p, ActA, and Smads after transfection.@*RESULTS@#The NS group had a significantly higher level of miR-17-5p in peripheral blood than the control group (P<0.001). Compared with the blank and negative control groups, the inhibition group had significantly lower apoptosis rate and relative mRNA expression of miR-17-5p and significantly higher relative mRNA and protein expression of ActA, Smad2, and Smad3 (P<0.001).@*CONCLUSIONS@#There is an increase in the content of miR-17-5p in peripheral blood in children with NS. Low expression of miR-17-5p can inhibit the apoptosis of human renal podocytes, which may be associated with the upregulation of the mRNA and protein expression of ActA, Smad2 and Smad3.

ABSTRACT

Three new mycophenolic acid derivatives, penicacids E-G (1-3), together with three known analogues, mycophenolic acid (4), 4'-hydroxy-mycophenolic acid (5) and mycophenolic methyl ester (6), were isolated from a marine-derived fungus Penicillium parvum HDN17-478 from a South China Sea marine sediment sample. The structures of compounds 1-3 were elucidated by HRMS, NMR, and Mosher's method. Among them, compounds 1 and 2 were the first examples of mycophenolic acid analogs with a double bond at C-3'/C-4' position. The cytotoxicity of 1-6 was evaluated against the HCT-116, BEL-7402, MGC-803, SH-SY5Y, HO-8910 and HL-60 cell lines, and compounds 4 and 6 showed potent cytotoxicity with IC

ABSTRACT

@#Abstract:Objective To develop a colloidal gold immunochromatographic strip ⁃ based method for the rapid detection of Zika virus(ZIKV)NS1 antigen. Methods The gold nanoparticles modified with the anti⁃ZIKV NS1 monoclonal antibody as the detection probe were coated on the glass ⁃fiber pad. The anti ⁃ZIKV NS1 monoclonal antibody and the goat anti ⁃mouse polyclonal antibody were immobilized on a nitrocellulose membrane as the test line and the control line,respectively. In order to achieve critical results,the ratio of the optical density (OD)of the test line to that of the control line was compared. Serial diluted ZIKV NS1 standard antigen was applied to evaluate sensitivity of the immunoassay. The culture supernatant and serum samples for arboviruses(ZIKV,Dengue virus, Japanese encephalitis virus and Chikungunya virus) were utilized to demonstrate the specificity of the method. Results The detection result could read by naked eyes within 20 minutes. The visual cut ⁃off level for the test strip was achieved at 100 ng/mL of the Zika virus NS1 standard antigen. No cross⁃reactions with Dengue virus,Japanese encephalitis virus and Chikungunya virus were observed. The strip could remain good stability within 36 weeks whether stored in 4 ℃ or room temperature(22-25 ℃). Conclusion Apart from stability, the method was convenient,rapid and specific for ZIKV NS1 antigen,which showed a promising potential in the point of care test and the screening test.

ABSTRACT

Objective To investigate the association of smoking status with incident cardiovascular disease (CVD) and its subtypes among the middle-aged and older male populations. Methods This study included 13 940 males from Dongfeng-Tongji (DFTJ) cohort who were free of coronary heart disease (CHD), stroke, cancer or severely abnormal electrocardiogram (ECG) at baseline. All participants completed baseline questionnaires, physical examinations, clinical biochemical tests and blood sample collection. Cox proportional hazard models were used to estimate the hazard ratios (HRs) and 95% confident intervals (CI) for the association analyses. Results Compared with never smokers, current smokers had significant higher risks of CVD, CHD and stroke, the adjusted HRs of current smokers who smoked for more than 40 pack-years were 1.49 (95% CI: 1.32-1.68, Ptrend=0.001), 1.40 (95% CI: 1.22-1.62, Ptrend=0.026) and 1.59 (95% CI: 1.26-2.00, Ptrend=0.029) for CVD, CHD and stroke, respectively; and the adjusted HRs of current smokers who started smoking before 20 years old were 1.29 (95% CI: 1.06-1.58, Ptrend=0.007) and 1.30 (95% CI: 1.03-1.64, Ptrend=0.010) for CVD and CHD, respectively. Former smokers who had quitted smoking for 10 or more years had significant lower risks of CVD (HR: 0.80, 95% CI: 0.71-0.91, Ptrend=0.017) and stroke (HR: 0.65, 95% CI: 0.50-0.84, Ptrend=0.207) when comparing to current smokers. Conclusions Smoking is significantly associated with higher risks of CVD, CHD and stroke, and greater amount of smoking and earlier age at smoking initiation are associated with a higher risk of CVD. Smoking cessation can reduce the risk of CVD.

; A i m To explore the effects of tetramethylpyrazine (T M P) on renal oxidative stress in immune-mediated bone marrow failure (B M F) C57 mice. Methods C57 mice were randomly divided into normal control group, total body irradiation group (T B I), model group, TMP low dose group (5 g • L " 1), T M P medium dose group (10 g • L " 1), and T M P high dose group (2 0 g • L " 1) . The B M F model was established by total body irradiation with 5. 0 Gy X-ray and l y m - phocyte infusion. The peripheral blood triline cells, bone marrow nucleated cells, bone marrow pathology, kidney pathology and oxidative stress indexes were observed 14 days later. Results Bone marrow pathology of the model group showed that the bone marrow nucleated cells decreased and the peripheral blood triline cells decreased significantly, indicating that the i m - mune-mediated B M F model was successfully established. Renal pathology of model group showed tubular edema, and the oxidative stress index M DA increased, and the antioxidant stress indicators GSH, SOD, GSHPx decreased, indicating that there was renal oxidative stress damage in BMF mice. After treatment with TMP, there was no obvious abnormality in renal pathology, the oxidative stress index MDA of the kidney decreased and the antioxidant stress index GSH, SOD increased. Immunohistochemistry showed that the expression of HIF-1 a decreased and VEGF existence was observed. Conclusions TMP can alleviate renal oxidative stress injury in immune-mediated BMF mice, which may be related to the regulation of HIF-1 oc/ VEGF expression.

ABSTRACT

<p><b>OBJECTIVE</b>To study the value of transesophageal atrial pacing (TEAP) in neonates with tachyarrhythmia.</p><p><b>METHODS</b>The clinical data of 26 neonates with tachyarrhythmia who underwent TEAP electrophysiological examination or cardioversion were collected.</p><p><b>RESULTS</b>Of the 26 neonates, 15(58%) were diagnosed with atrioventricular reentrant tachycardia, 3(12%) were diagnosed with sinus tachycardia, 3(12%) were diagnosed with ventricular tachycardia, 2(8%) were diagnosed with fast/slow atrioventricular nodal reentrant tachycardia, 2(8%) were diagnosed with atrial tachycardia, and 1(4%) was diagnosed with sinus tachycardia with ventricular preexcitation. Overdrive suppression was performed for 22 neonates, among whom 18 achieved successful cardioversion, and 2 with atrial tachycardia and 2 with ventricular tachycardia failed to restore sinus rhythm.</p><p><b>CONCLUSIONS</b>TEAP is helpful to the diagnosis of tachyarrhythmia in neonates and can bring about a high rate of cardioversion success.</p>

ABSTRACT

Objective To investigate the relationship between resilience, cognitive emotion regulation strategies and fatigue reaction of navy soldiers. Methods A total of 846 soldiers were selected froma navy troop by convenience sampling method, and were tested by cognitive emotion regulation questionnaire-Chinese version (CERQ-C), Connor-Davidson resilience scale (CD-RISC) andmultidimensional fatigue inventory scale (MFI-20). SPSS 21.0 software were used to analyze the relationship between the variables. Results Non-adaptive emotion regulation strategy was positively correlatedwith fatigue reaction in the soldiers (P<0.01), and the three dimensions of resilience werenegatively correlated with fatigue reaction in the soldiers (P<0.01). Both resilience and cognitive emotion regulation strategies could independently explain the variance of fatigue reaction of the soldiers, in which self-improvement (β=-0.206, P < 0.05) and adaptive strategies (β=-0.229, P<0.01) had significant negative predictive effects on fatigue reaction of soldiers, while non-adaptive strategies (β=-0.406, P<0.01) had significant positive predictive effectson fatigue reaction. The mediating effective value of resilience on adaptive strategies and fatigue reaction of soldiers was 0.53 X (-0.36)=-0.19, z=7.02, and on non-adaptive strategies and fatigue reaction of soldiers was (-0.30)X(-0.36)=0.108, z=5.12. Conclusion Cognitive emotion regulation strategieshas direct effect on fatigue reaction of navy soldiers, and it also has indirect influence on the fatigue reaction through the mediating effect of resilience.

ABSTRACT

Objective: To observe pilots with asymptomatic cerebral infarction (ACI) for their short-term prognosis. Methods: Twenty-two pilots who were diagnosed having ACI by magnetic resonance imaging were enrolled in this study. When they returned to the hospital for regular reexamination, the number of ACI foci, the incidents of acute cerebrovascular disease and grounding of aircraft for any reasons were recorded. According to whether the ACI lesions increased, the patients were divided into two groups. The risk factors for cerebral vascular disease were compared between the two groups. Results: All the patients were followed for 6-42 months (mean 14.18±8.55 months), and 6(18.18%) patients were found to have increase of lesions. No neurological deficit was seen in the two groups, although 3 flight crews were grounded for non-ACI reason. Age and flight time showed statistically significant differences between the lesions increased group and no lesion increased group. Conclusions: The number of ACI foci of some pilots may increase, which is affected by age and flight time. Further investigations on the long-term prognosis and the impacts on flight are needed.

ABSTRACT

<p><b>BACKGROUND</b>Spinal anesthesia was considered as a reasonable anesthetic option in severe preeclampsia when cesarean delivery is indicated, and there is no indwelling epidural catheter or contraindication to spinal anesthesia. However, the ideal dose of intrathecal bupivacaine has not been quantified for cesarean delivery for severe preeclamptic patients. This study aimed to determine the ED 50 and ED 95 of intrathecal bupivacaine for severely preeclamptic patients undergoing elective cesarean delivery.</p><p><b>METHODS</b>Two hundred severely preeclamptic patients are undergoing elective cesarean delivery under combined spinal-epidural anesthesia enrolled in this randomized, double-blinded, dose-ranging study. Patients received 4 mg, 6 mg, 8 mg, or 10 mg intrathecal hyperbaric bupivacaine with 2.5 μg sufentanil. Successful spinal anesthesia was defined as a T6 sensory level achieved within 10 minutes after intrathecal drug administration and/or no epidural supplement was required during the cesarean section. The ED 50 and ED 95 were calculated with a logistic regression model.</p><p><b>RESULTS</b>ED 50 and ED 95 of intrathecal bupivacaine for successful spinal anesthesia were 5.67 mg (95% confidence interval [CI]: 5.20-6.10 mg) and 8.82 mg (95% CI: 8.14-9.87 mg) respectively. The incidence of hypotension in Group 8 mg and Group 10 mg was higher than that in Group 4 mg and Group 6 mg (P < 0.05). The sensory block was significantly different among groups 10 minutes after intrathecal injection (P < 0.05). The use of lidocaine in Group 4 mg was higher than that in other groups (P < 0.05). The use of phenylephrine in Group 8 mg and Group 10 mg was higher than that in the other two groups (P < 0.05). The lowest systolic blood pressure before the infant delivery of Group 8 mg and Group 10 mg was lower than the other two groups (P < 0.05). The satisfaction of muscle relaxation in Group 4 mg was lower than other groups (P < 0.05). There was no significant difference in patients' satisfaction and the newborns' Apgar score and the blood gas analysis of umbilical artery serum (P > 0.05).</p><p><b>CONCLUSION</b>Our study showed that the ED 50 and ED 95 of intrathecal bupivacaine for severely preeclamptic patients undergoing elective cesarean delivery were 5.67 mg and 8.82 mg, respectively. In addition, decreasing the dose of intrathecal bupivacaine could reduce the incidence of maternal hypotension.</p>

ABSTRACT

Background Most animal models of experimental autoimmune uveitis (EAU) are single attacked procedure,with a different feature from the natural course of human recurrent autoimmune uveitis.So establishing a recurrent EAU model is necessary for the clinical study on EAU.Objective This study was to establish the recurrent EAU model in rat and investigate the ocular inflammation and pathological manifestation and interleukin-17 (IL-17)expression in the eye.Methods T cells isolated from the spleen and draining lymph nodes of Lewis rats immunized with interphotoreceptor retinoid-binding protein (IRBP) 1177-1191 peptide fragments (R16) 10 days earlier were re-stimulated with R16 in vitro and injected into naive syngeneic rats to establish the recurrent EAU models,and the normal Lewis rats were used as controls.The eyes of model rats were then examined daily for clinical signs of uveitis by slit-lamp biomicroscopy and scored Caspi's criteria.The rats were sacrificed 1 month,2,3months after injection respectively,and the retinal sections were prepared for the pathological examination by hemotoxylin & eosin staining.Immunohistochemistry was performed to detect the expression of IL-17 in the retina.Results Adoptive transfer of R16-specific T cells to Lewis rats induced recurrent uveitis.The inflammatory scores on the fourth day,the sixth day,and the inflammatory response disappeared on the tenth day after injection.However,the inflammatory reaction occurred repeatedly 4 or 5 times in the 2-month duration after that,and the right and left eyes of a single recipient showed a different pattern of relapse,and the clinical manifestations of EAU was similar to the natural course to those of human autoimmune uveitis.In the retinal specimens of 1-,2-and 3-month group,the number of inflammatory cells was gradually decreased as the time lapse.Compared with the normal group,the thicknesses of the entire retina,outer nuclear layer and inner nuclear layer decreased with a significant difference among the 4 groups (F=20.46,288.40,4.43,all P=0.00).The number of RGCs in the normal group,1-,2-and 3-month group was 231.27 ± 15.36,225.36 ± 17.79,132.18 ±9.39 and 67.45 ± 11.90,respectively,showing a significant difference among them (F=68.94,P=0.00).Immunohistochemistry showed that the scores of the IL-17 expression in the rat retina were 0.64 ± 0.17,1.92 ± 0.19,1.17 ± 0.23 and 0.83 ± 0.23,showing statistically significant difference (F=64.10,P=0.00).Conclusions The stimulation of R16-specific T cells can induce recurrent EAU in Lewis rat.Th17 is involved in the disease course.

ABSTRACT

Based on a pair of specific primers, a 804-bp fragment was amplified from the plasmid pT-Cap containing Cap gene of Porcine Getah Virus(PGETV) and cloned into the prokaryotic expression vector pCold I which carried the His tag, this recombinant plasmid was then determined by enzyme digestion, PCR and DNA sequencing. This recombinant plasmid pCold-Cap was transformed into E. coli Rosetta 2, and PGETV Cap fusion protein was expressed through IPTG induction. The results showed that the Cap gene obtained efficient and soluble expression in Rosetta 2 induced by 0. Immol/L IPTG under 15"C for 24h, the expression quantity was 40. 2%. The product had a molecular mass about 32. 3kD as expected. The target protein was separated in gel slices and used to immunize Balb/c mice. The polyclonal antibody with high titer against Cap protein specifically analyzed by Western blot was obtained. The successful preparation of the polyclonal antibody laid the foundation for the further study on the detection and identification of PGETV.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the characteristics of particulate matter pollution in coke oven plant, so as to provide scientific data for establishing occupational exposure limits for coke oven emissions.</p><p><b>METHODS</b>Concentrations of CO, SO₂, BSM, BTEX (concentrations of benzene, toluene and xylene were determined in this study), PM₁₀, PM₂.₅, 16 selected PAHs in PM₁₀ and PM₂.₅ were determined in the work environment of a coke oven plant in Wuhan. The work environment was divided into the adjunct area, the bottom of, the side of and the top of coke oven.</p><p><b>RESULTS</b>The concentrations of CO, SO₂, BSM, BETX, PM₁₀, PM₂.₅, PAHs in PM₁₀ and PM₂.₅ were significantly related to working environmental categories, respectively, and were increasing as the adjunct area < bottom < side < top (P (trend) < 0.05). PM₁₀ was statistically significantly correlated with CO, SO₂, benzene, BTEX and BSM (0.705, 0.823, 0.664, 0.624 and 0.734, respectively). PM₂.₅ was statistically significantly correlated with CO, SO₂, benzene, BTEX and BSM (0.635, 0.916, 0. 680, 0.553 and 0.726, respectively). BSM was statistically significantly correlated with benzene (0.689). The ratios of PM₂.₅ to PM₁₀ between different work environments were not significantly different in one-way ANOVA (P > 0.05). The distribution of aromatic rings and the concentrations of total benzo[a] pyrene equivalents in PM₁₀ and PM₂.₅ were not statistically different between work environments.</p><p><b>CONCLUSION</b>The concentrations of particulate matter was related with other contents of coke oven emissions in coke work environment, and the contents and types of PAHs in PM₁₀ and PM₂.₅ were similar.</p>

BackgroundStudies determined that Th17 cells are important inflammatory cell group in experimental autoimmune uveoretinitis ( EAU ).Interleukin-17 ( IL-17 ),as a marker of Th17,is involved in the occurrence and development of EAU.Mesenchymal stem cells (MSCs) play an immunomodulatory role,mainly by inhibiting the expression of Th17 in a variety of self-autoimmune disease.This is one of the current research focuses.ObjectiveThe present study was to investigate the therapeutic effect of MSCs in EAU and their impact on IL-17 expression in the retina.MethodsMSCs were isolated from the bone marrow of the femurs from 10 SPF Wistar rats and cultured and passaged.The third to fifth generations of cells were used in this experiment.EAU models were induced in 48 6-8 week-old SPF Lewis rats by subcutaneous injection of interphotoreceptor retinoid binding protein (IRBP) R16 peptide emulsified in adjuvant.EAU rats were randomly assigned to the model control group and the MSCs group.MSCs suspension (5×106) of 1 ml was injected via the rat tail vein once a day for 3 consecutive days after immunization,and the same amount of PBS was injected in the model control group in the same manner.Six matched normal Lewis rats were used as the normal control group.The inflammatory response was clinically examined under the slit lamp biomicroscope daily,and the histopathological changes of the retina were examined by hematoxylin and eosin staining on days 9,12,15 and 20.The clinical and histopathological scoring was performed according to the Caspi criteria.Expression of the IL-17 protein in the retina was detected by immunohistochemistry on 9,12,15 and 20days following molding.The use of the animals complied with the Regulations for the Administration of Affairs Concerning Experimental Animals by Tianjin Municipality Science and Technology Commission.Results MSCs showed the fusiform in shape and vortex-like growth.Flow cytometry verified that presented the positive expression for CD29 and CD44 but absent expression for CD45 and CD34.The scores of the anterior segment were significantly lower ( U=2.815,P =0.005 ; U =2.768,P =0.006 ; U =2.900,P =0.004 ; U =2.855,P =0.004 ),and the retinal inflammation scores were lower in the MSCs group than the model control group at various time points ( U =2.345,P =0.019 ; U =2.559,P =0.011 ; U =2.166,P =0.030 ; U =2.373,P =0.018 ).Im mnunochemistry showed that the expressions levels of the IL-17 protein (A value) in the rat retina were 26.47±5.68,77.78± 9.65,47.02±6.68 and 26.59±5.94 in the MSCs group on days 9,12,15 and 20,and those in the control group were 45.34±4.63,105.95± 10.74,64.11 ±9.76 and 37.02±6.51,showing a significant reduction ( t =6.305,P =0.000 ; t =4.799,P =0.001 ; t=3.540,P=0.005;t=2.900,P=0.016). ConclusionsMSCs can inhibit the aggravation of EAU and suppress the expression of IL-17 in ocular tissue.

ABSTRACT

OBJECTIVE: To examine the effects of Trifolium pratense isoflavones (TPIF), the main active component of Trifolium pratense L., on ovariectomy-induced osteoporosis in rats. METHODS: A total of 60 six-month-old female rats were randomly assigned to a sham-operated group and five ovariectomized(OVX) groups, ie, OVX with vehicle(10 mL · kg-1 · d-1, ig), OVX with graded doses of TPIF(30, 60, and 90 mg · kg-1 · d-1, ig), and OVX with nilestriol(2.5 mg · kg-1 · week-1, ig). The animals were sacrificed after 12-week treatment. RESULTS: Compared to the OVX group, TPIF significantly inhibited the increase of body weight. In addition, TPIF administration significantly decreased serum levels of alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP), and increased serum level of estrogen. CONCLUSION: TPIF may be a potential alternative medicine for the prevention and treatment of postmenopausal osteoporosis. Its mechanism may be related to increasing estrogen level, inhibiting bone resorption, and promoting bone formation.

ABSTRACT

This study was purposed to explore the effects of hyperbaric oxygen (HBO) combined with adriamycin (ADM) on inducing apoptosis of multidrug resistant cells line K562/A02. The cell apoptosis and expression of caspase-3 activity were analyzed by flow cytometry and transmission electron microscopy; the expression levels of HIF-1α, BCL-2 and BAX mRNA were detected by quantitative real time PCR; the caspase 8 activity was determined by using caspase 8 kit; the expression level of P-gp was detected by Western blot. The results showed that the apoptosis rate of K562/A02 cells in combination group (0.2 MPa HBO + ADM) was higher than that in ADM group [(47.36 ± 3.87) % vs (28.51 ± 1.09) %], the difference was statistical significant (p < 0.05); the expression levels of HIF-1α mRNA, P-gp and BCL-2 in combination group were lower than those in ADM group, there were significant differences (p < 0.05); the activities of BAX, caspase 3 and caspase 8 proteins in combination group were higher than those in ADM group, the difference was statistical significant (p < 0.05). It is concluded that 0.2 MPa HBO combined with ADM can reverse the drug-resistance of K562/A02 cells to ADM, enhance the apoptosis rate of cells. The molecular mechanism may be related with down-regulation of P-gp and BCL-2 expression, and up-regulation of caspase-3 and caspase-8 activities by HIF-1α.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the change of electrocardiographic ischemic J wave in patients with coronary heart disease (CHD) underwent percutaneous coronary intervention (PCI).</p><p><b>METHODS</b>CHD patients (CHD, n = 162) underwent PCI in the hospital were divided into unstable angina pectoris (angina pectoris group, n = 108) and acute myocardial infarction group (AMI group, n = 54), 12-lead electrocardiogram was recorded within 24 hours before and after PCI. Patients were further grouped into left anterior descending artery group and right or circumflex coronary artery stenosis group. The incidence, amplitude and time course of J waves were measured and analyzed.</p><p><b>RESULTS</b>(1) The highest incidence of J wave appeared in II, III, aVF leads pre- and post-PCI in CHD groups (33.95%, 22.84%, 26.54% and 30.86%, 19.75%, 23.46%). Higher incidence of J wave in AMI group was evidenced in V(5), II, aVF, III, V(3) leads before PCI (33.33%, 27.78%, 16.67%, 12.96%, 11.11%) and in V(5), II, aVF, aVL, III leads after PCI (22.22%, 22.22%, 14.81%, 12.96%, 9.26%). (2) Before PCI, higher incidence of J wave was found in V(5), V(6) leads (25.78%) in left anterior descending artery group and in II, III, aVF leads (35.37%) in right or circumflex coronary artery stenosis group. After PCI, the higher incidence of J wave in left anterior descending artery group and right or circumflex coronary artery stenosis group was evidenced in II, III, aVF leads (21.09% and 35.37%, respectively). (3) The ischemic J wave' amplitude and time course in II, III, aVF, V(3), V(4) leads were significantly reduced post PCI in CHD patients.</p><p><b>CONCLUSIONS</b>Higher incidence of J wave was found in II, III, aVF in CHD patients. PCI is associated with reduced amplitude and time course of J wave suggesting PCI could improve ischemic J waves in CHD patients.</p>

ABSTRACT

Background Studies determined that rapamycin has not only the antibiosis but also suppressing the auto-immunology.The treating effect of rapamycin on experimental autoimmune uveoretinitis (EAU) is still concerned.Objective This work was to investigate the therapeutic effect of rapamycin on EAU and study the effect on the expression of inflammatory cytokines which were secreted by T lymphocyte subgroup in EAU.Methods EAU was induced in 20 SPF male Lewis rats by subcutaneous injection of interphotoreceptor retinoid binding protein (IRBP) R16 peptide emulsified in adjuvant.The rats were randomized into model control group and rapamycin injection group and 10 rats for each group.The rapamycin of 0.2 mg/( kg · d) 0.4 ml was intraperitoneally injected for the consecutive 7 days immediately after modeling,and the equal volume of normal saline solution was used at the same fashion in the model control group and 5 normal matched rats( normal control group).The ocular manifestation of the rats were examined under the slit lamp regularly.The retinal sections of the rats were prepared in the 14 days after modeling for the histopathological examination with hematoxylin and eosin staining.The ocular signs of inflammation and histopathological severity were scored based on the criteria of Caspi.Expression of interferon-γ (IFN-γ) and interleukin-17 (IL-17) in rat retinas were detected by immunohistochemistry.This experiment followed the Regulation for the Administration of Affair Concerning Experimental Animals by Tianjin Municipal Government.Results The scores of ocular signs elevated gradually from 6 through 12 days after modeling and slowly declined after that in the model control group.A same tendency was seen in the rapamycin injection group.The significant differences were seen in the scores of ocular signs between the two groups from 6 to 14 days( P<0.01 ).The disorder of retinal structure and infiltration of inflammatory cells were seen in the model control group,but the retina layers were normal in the rapamycin injection group.The pathological score was evidently declined in the rapamycin injection group ( 0.90 ± 0.45 ) in comparison with model control group( 3.30±0.48 ) ( t =16.541,P<0.01 ).The expressions of the IFN-γ and IL-17 in retina located in the outer nuclear layer,inner nuclear layer,internal plexiform layer and retinal ganglion cell layer with the weakened levels(A values) in rapamycin injection group compared with model control group,showing a considerably difference between them ( IFN-γ:21.16±4.23 vs 62.14 ±7.32; IL-17:49.86±6.59 vs 124.85 ±6.33 )(q=33.334,q=56.923,P<0.01 ).Conclusions Rapamycin down-regulates the expressions of IFN-γ and IL-17 in retina and further eliminates the inflammatory response in the rat with EAU by the suppression of Th1 and Th17 cells function in EAU.

ABSTRACT

<p><b>OBJECTIVE</b>The purpose of this article was to evaluate the influence of cobalt-chromium (Co-Cr) alloy as the material of inner crown on periodontal tissue through detecting the weight of diseased teeth and the concentration of aspartate transaminase (AST) and alkaline phosphatase (ALP) of the gingival crevicular fluid (GCF) after Co-Cr alloy ceramics repairing.</p><p><b>METHODS</b>In this study, thirty cases of clinical diseased teeth were chosen from thirty patients based on patients' consent. Each tooth conformed to the inclusion criteria. All of the thirty teeth were repaired with Co-Cr alloy ceramics according to the indications. Then GCF of each diseased tooth was collected and weighed at the time of the day before prosthesis, the first and third month after repairing respectively in order to detect the concentration of AST and ALP. Furthermore, comparative analysis for different periods was performed after the data statistics.</p><p><b>RESULTS</b>To the weight of GCF and the concentration of AST after the respective comparison of three indexes which was of different periods, numerous of each index increased in accordance with the rule of preoperation, postoperative first month, and postoperative third month (P < 0.05). To the concentration of ALP at three time points, the compared results showed that the preoperative numerous was less than that of third month after operation and there was significant difference between them (P < 0.05). To the weight of GCF and the concentration of AST and ALP, after the respective comparison of three indexes which was the diseased teeth and the contralateral teeth with the same name in the periods of pre-operation, all the differences had no statistical significance (P > 0.05), but all the differences had statistical significance in the periods of postoperative third month (P < 0.05).</p><p><b>CONCLUSION</b>During the next three months after operation, GCF weight, concentration of AST and ALP of diseased teeth was gradually increased after their Co-Cr alloy ceramics crown repairing. This increase is somewhat harmful to periodontal health.</p>

ABSTRACT

Objective To construct a voxel mouse model combining with Monte Carlo method used for radiation dose calculation.Methods A set of slice images obtained from a nude male mouse (28 g) was utilized, all slice images were registered,some organs or tissues were identified, segmented and filled with specific color, and finally the physical property was defined by MCNP application.Results A mouse model with a voxel size of 0.2 mm×0.2 mm×0.2 mm, consisting of 14 organs or tissues,was constructed, which could satisfy the requirement of precision for radiation dose distribution calculation and moderate computing time.Conclusion The voxel mouse model can be used to calculate the quantity of ionization radiation dosimetry in related areas including radiological medicine, nuclear medicine and space radiation medicine.

ABSTRACT

<p><b>OBJECTIVE</b>Ambulatory arterial stiffness index (AASI) has been recently proposed to reflect the dynamic relation between diastolic and systolic blood pressure throughout the whole day. The aim of our study was to investigate the change in AASI with advancing age and the correlation with 24 hours pulse pressure (24 h PP) in healthy individuals.</p><p><b>METHODS</b>246 healthy subjects [mean age (59.7 +/- 14.6) years, women 38.6%] underwent 24 hours ambulatory blood pressure monitoring (ABPM) in normal life style. The blood pressure recordings, heart rate (HR), mean arterial pressure (MAP) and pulse pressure (PP) were analyzed simultaneously by computer for every 30 minutes during 6:00 am-22:00 pm and every 60 minutes during 22:00 pm-6:00 am. Using all the blood pressure recordings, we plotted diastolic against systolic blood pressure from each individuals and calculated the regression slope. AASI was derived from 1 minus this regression slope.</p><p><b>RESULTS</b>In 246 healthy individuals, AASI increased with age. Among the healthy individuals, the 95th percentile of AASI was 0.56, the upper boundary of the 95% prediction interval of AASI in relation to age were 0.49 at 20 - 39 years, 0.59 at 40 - 59 years, 0.69 at 60 - 79 years, 0.79 at > or = 80 years. The correlation coefficient between AASI and 24 h PP was 0.497 (P < 0.01). AASI linearly increased with age in healthy individuals, whereas the relation between pulse pressure and age was curvilinear.</p><p><b>CONCLUSIONS</b>AASI as a index reflecting blood pressure relationship, manifested the corresponding change with advancing age. The correlation between AASI and traditional index 24 h PP indicated AASI as a new measure of arterial stiffness.</p>
