ABSTRACT

The chemical constituents in the volatile oil of Syringa oblata were identified using GC-MS and NIST database. TCMSP and SwissTargetPrediction were employed to predict the potential targets of the active components in S. oblata. Through Online Mendelian Inheritance in Man(OMIM), GeneCards, and Kyoto Encyclopedia of Genes and Genomes(KEGG), we screened out the targets related to the prevention or treatment of angina pectoris by the volatile oil of S. oblata, and then used DAVID 6.8 to annotate the gene ontology(GO) terms and KEGG pathways. The "active components-targets-pathways" network was constructed in Cytoscape 3.6.0, and the key active components and targets of S. oblata were verified by Discovery Studio 2016. Forty-six chemical constituents were identified from the volatile oil of S. oblata; 198 potential targets of the active components and 1 138 targets associated with angina pectoris were predicted. A total of 71 common targets were shared by the active components and the disease, including cytochrome P450 19 A1(CYP19 A1) and prostaglandin G/H synthase 2(PTGS2). The KEGG pathways involved include PPAR, JAK-STAT, TNF, Toll-like receptor and NOD-like receptor signaling pathways. The active components in the volatile oil of S. oblata may play anti-inflammatory and anti-apoptosis roles. This study provides a reliable clue for further explanation of the effective components and the functioning mechanism of S. oblata in the treatment of angina pectoris.

ABSTRACT

Objective:To explore the effects of Shenwei Ningyu pills (SNP), a new Chinese medicine for depression, on the immunoinflammatory response mediated by Toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88) signaling pathway in the hippocampus of rats exposed to chronic restraint stress (CRS). Method:Forty-four male Sprague Dawley rats were randomly enrolled into a normal group, a model group, an escitalopram group, and an SNP group. Except for the rats in the normal group, all rats were exposed to CRS and isolated rearing for 21 days continuously. Rats in the escitalopram group and the SNP group were administered with escitalopram (30 mg·kg-1) and SNP (18 mg·kg-1) one hour prior to CRS, respectively. The changes in body weight, sucrose preference index, horizontal movement scores, and vertical movement scores were observed by body weight assessment, sucrose preference test, and open field test. The expression of hippocampal TLR4 and MyD88 was detected by Western blot. The content of serum interleukin-1β (IL-1β), IL-10, and tumor necrosis factor-α (TNF-α) was detected by enzyme-linked immunosorbent assay (ELISA). Result:The results of the behavioral assessment showed that there was no significant difference in the changes of behavioral baselines among the groups before intervention. However, significant differences were found among the groups following different interventions. The body weight, sugar preference index, horizontal movement score, and vertical movement score of rats in the model group decreased after CRS for 21 days as compared with those in the normal group (P<0.01). The above indicators in the SNP group and the escitalopram group were higher than those in the model group (P<0.01), which indicated that SNP exerted an obvious antidepressant effect. The results of Western blot and ELISA showed that compared with the normal group, the model group showed elevated levels of hippocampal TLR4 and MyD88 and serum IL-1β and TNF-α (P˂0.01) and dwindled serum IL-10 (P˂0.01), while SNP and escitalopram reversed the conditions in the model group (P˂0.01) except for TNF-α. Conclusion:The present study indicated that the antidepressant effect of SNP was presumedly achieved by inhibiting the immunoinflammatory response mediated by the TLR4/Myd88 signaling pathway in CRS rats.

ABSTRACT

Objective:To investigate the pathogenic distribution of benign infantile convulsions with mild gastroenteritis(BICE)and explore the relevance of serum cytokines and BICE.Methods:Eighty BICE infants admitted in Inner Mongolia Maternal and Child Health Hospital from January 2017 to December 2019 were selected as BICE group, and 80 mild gastroenteritis infants without convulsion attack were selected as control group during the same period.Fluorometric real-time PCR was used to detect the pathogen of enterovirus.Serum cytokines including interleukin(IL)-6, tumor necrosis factor(TNF)-α, IL-8 and soluble interleukin-2 receptor(SIL-2R)were detected by using chemiluminescence method. The relevance of frequency and duration of convulsion in infants with BICE and the indicators above were analyzed. Results:Rotavirus infection was the main cause of BICE during the study period in this region.Among 80 cases in the BICE group, rotavirus positive infants accounted for 38.8%(31/80) and norovirus positive infants accounted for 10.0%(8/80). The levels of IL-6, IL-8 and SIL-2R in the BICE group were prominently higher than those in the control group( P<0.05); the difference of TNF-α level between two groups had no statistical significance( P>0.05). The levels of IL-6, TNF -α, IL-8, SIL-2R in the group with convulsion attack times ≥ 2 and convulsion duration ≥5 min were higher than those in the group with convulsion attack times<2 and convulsion duration<5 min( P<0.05). The frequency and duration of convulsions were positively correlated with the levels of IL-6, TNF -α, IL-8 and SIL-2R( P<0.05). Conclusion:Rotavirus is the main pathogen of BICE in this region. There is immunologic imbalance in children with BICE, especially the changes of IL-6 and SIL-2R levels, which may provide effective cytological and experimental data evidence for judging progression and prognosis of the disease.

ABSTRACT

Objective:⑴ To understand the pathogen distribution of mild gastroenteritis with benign infantile convulsion (BICE). ⑵ To analyze the levels of serum interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), peripheral blood leukocyte count (WBC) and hemoglobin (Hb) in children with BICE, and to explore whether they are related to the convulsion of BICE.Methods:From May 2017 to May 2018, 30 BICE children in the Department of Pediatrics of Inner Mongolia Autonomous Region Maternal and Child Health Hospital were selected as the observation group, while 30 mild gastroenteritis children without convulsion attack were selected as the control group. The detection of enterovirus uses colloidal gold method or fluorescence polymerase chain reaction (PCR) method. Serum IL-6 and TNF-α were detected by chemiluminescence. The levels of serum inflammatory factors, WBC and Hb were compared between the control group and the observation group, as well as between the children with different seizure times and duration.Results:⑴ The onset of BICE group was mainly in autumn and winter. The incidence rate of control group in summer was higher than that in BICE group, with statistically significant difference ( P<0.05). ⑵ In BICE group, rotavirus, norovirus and Coxsackie virus were 46.67%, 6.67% and 10%, respectively. ⑶ The levels of serum [IL-6 (16.37±11.97)pg/ml] and [TNF-α (11.96±7.10)pg/ml] in BICE group were higher than those in control group [IL-6 (10.30±6.95)pg/ml, TNF-α (6.95±4.97)pg/ml], with statistically significant difference ( P=0.019, P=0.003). ⑷ The levels of serum [IL-6 (24.83±14.34)pg/ml] and [TNF-α (16.93±6.73)pg/ml] in children with convulsion ≥ 2 times were higher than those in children with convulsion < 2 times [IL-6 (11.48±6.91)pg/ml, TNF-α (9.07±5.66)pg/ml], with statistically significant difference ( P=0.013, P=0.002). The levels of serum [IL-6 (27.84±14.42)pg/ml] and [TNF-α (18.00±6.74)pg/ml] in children with convulsion duration ≥ 5 min were higher than those in children with convulsion duration <5 min [IL-6: (12.20±7.76)pg/ml, TNF-α: (9.75±5.95)pg/ml], with statistically significant difference ( P=0.001, P=0.003). ⑸ Compared with the control group, there was no significant difference in WBC and Hb content of blood routine test in BICE group ( P>0.05). Conclusions:⑴ Rotavirus infection is the main pathogen of BICE in this region. ⑵ Children with BICE have a certain degree of immune regulation dysfunction. IL-6 and TNF-α may participate in the convulsion process of BICE. The higher the levels of serum IL-6 and TNF-α, the more severe the convulsion degree in children. ⑶ This study shows that WBC number and Hb content in blood routine have nothing to do with the occurrence of BICE.

ABSTRACT

In December 2019, an outbreak of viral pneumonia began in Wuhan, Hubei Province, which caused the spread of infectious pneumonia to a certain extent in China and neighboring countries and regions, and triggered the epidemic crisis. The coronavirus disease 2019(COVID-19) is an acute respiratory infectious disease listed as a B infectious disease, which is managed according to standards for A infectious disease. Traditional Chinese medicine and integrated traditional Chinese and Western medicine have played an active role in the prevention and control of this epidemic. China's ethnomedicine has recognized infectious diseases since ancient times, and formed a medical system including theory, therapies, formula and herbal medicines for such diseases. Since the outbreak of the COVID-19 epidemic, Tibet Autonomous Region, Qinghai Province, Inner Mongolia Autonomous Region, Xinjiang Uygur Autonomous Region and Chuxiong Autonomous Prefecture of Yunnan, Qiandongnan Autonomous Prefecture of Guizhou have issued the prevention and control programs for COVID-19 using Tibetan, Mongolian, Uygur, Yi and Miao medicines. These programs reflect the wisdom of ethnomedicine in preventing and treating diseases, which have successfully extracted prescriptions and preventive measures for the outbreak of the epidemic from their own medical theories and traditional experiences. In this paper, we summarized and explained the prescriptions and medicinal materials of ethnomedicine in these programs, and the origin of Tibetan medicine prescriptions and Mongolian medicine prescriptions in ancient books were studied. These become the common characteristics of medical prevention and treatment programs for ethnomedicine to formulate therapeutic programs under the guidance of traditional medicine theories, recommend prescriptions and prevention and treatment methods with characteristics of ethnomedicine, and focus on the conve-nience and standardization. However, strengthening the support of science and technology and the popularization to the public, and improving the participation of ethnomedicine in national public health services and the capacity-building to deal with sudden and critical diseases are key contents in the development of ethnomedicine in the future.

ABSTRACT

The mechanism of detoxification of Chebulae Fructus against Aconiti kusnezoffii radix toxicity, which was known as Mongolian medical theory, was studied by establishing network of active components-targets-pathways of detoxification and enrichment analysis of targets and pathways based on network pharmacology. Firstly, the targets of active components collected from TCMSP and TCM Database@Taiwan were obtained through SwissTargetPrediction compared with disease targets from OMIM, TTD, DiGSeE. Then, the target enrichment analysis of GO functional annotations and KEGG pathways and protein function were analyzed by Metascape, furthermore, the action between main active ingredients and targets was assessed by SystemsDock Web Site. At last, the Cytoscape was used to construct the network of active components-targets-pathways. In conclusion, there were 15 components and 40 targets related to the cardiotoxicity caused by Aconiti kusnezoffii radix. Furthermore, Chebulae Fructus could regulate cardiac function to detoxify the toxicity by Aconiti kusnezoffii radix through the biological process of negative regulation of blood vessel diameter, regulation of ion transport circulatory system process, muscle contraction inorganic ion homeostasis and the pathways of neuroactive ligand-receptor interaction, calcium signaling pathway, adrenergic signaling in cardiomyocytes, etc.

ABSTRACT

Objective To evaluate the reliability of sweep visual evoked potential (SVEP) for assessing the normal objective visual acuity in adult wild-type C57BL/6J mice. Methods A total of 6 wild C57BL/6J mice of either gender at 32-33 days of age were collected. After the mice were anesthetized, the recording electrode (a 2-mm diameter tungsten electrode) was placed at 3 mm lateral to lambda over the left cortex (contralateral to the stimulated right eyes) and advanced to 400 μm within the cortex, and the needle electrodes which placed in the forehead and tail served as the reference and the ground, accordingly.Left eyes (unstimulated eyes) were covered during the recording. The screen (mean luminance 25 cd · m-2) was placed 20 cm in front of the mice, thereby covering 100° (horizontal) × 82° (vertical) of the visual field. The mice were stimulated by vertically sinusoidal grating, which consisted of 11 spatial frequencies, stimulus contrast was 100%,and time frequency was 1 Hz;the entire sweep took 12 seconds (11 spatial frequencies + 1s pre-adaptation),and the values were averaged until the error bars for the data were stable. The VEP waveforms of visual cortex corresponding to the left and right eyes were recorded separately. At least 3-4 trials consisting of 20 events for different frequencies were averaged.Left and right eye were tested alternately. Then the coordinate system was established by using the spatial frequency logarithm as x-axis and the amplitude of the response obtained by the discrete Fourier analysis as y-axis,the objective visual acuity was obtained by extrapolation to zero amplitude of the linear regression through the 4 effective data points adjacent to noise. Results SVEP showed that the amplitude of N100 wave was maximal at a spatial frequency of 0.05 c · deg-1.With the increase of the stimulus spatial frequency progressively,SVEP decreased in amplitude and increased in latency. The cortical electrical signal could not be distinguished from noise at a spatial frequency of 0.65 c · deg-1.The average acuity of 6 mice was (0.56 ± 0.04) c · deg-1 for right eyes and (0.50 ± 0.04)c · deg-1 for left eyes respectively. There was no significant difference between the two eyes(P>0.05);The visual evoked potential acuity of 12 eyes was (0.53 ±0.03)c ·deg-1.Conclusion SVEP could be used to estimate the mouse VEP acuity, which was closed to the behavioral visual acuity. However, the VEP acuity was more objective and accurate as well as time-saving.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of electroacupuncture (EA) treatment on the expression of cyclooxygenase (COX) 2 and microglia in spinal cord by using rat model of neuropathic pain, and to probe into the relationship between COX 2 and microglia.</p><p><b>METHODS</b>The rats were randomly divided into 6 groups, including normal control group, model group, sham group, EA 1 group (distant acupoints + local acupoints), EA 2 group (local acupoints), and EA 3 group (distant acupoints). Thermal withdrawal latencies were evaluated at 1 day preoperatively and 3, 5 and 7 days postoperatively. At 7 days postoperatively, the spinal COX 2 mRNA was detected by reverse-transcription polymerase chain reaction. Double immunofluorescent staining technology was applied to screen and verify the relationship between altered COX 2 and microglia.</p><p><b>RESULTS</b>Compared with the model group, thermal withdrawal latencies increased after EA treatment (P<0.01). The expressions of COX 2 mRNA were up-regulated in spinal cord of rat on day 7 after surgery (P<0.05). Compared with the model group, EA stimulation (EA 1 and EA 2 groups) reversed the up-regulation of COX 2 mRNA expression (P<0.05). EA 1 and EA 2 groups might have better treatment effect compared with the EA 3 group. Fluorescent images displayed COX 2 and microglia expressed at common areas.</p><p><b>CONCLUSIONS</b>EA was effective in analgesic and anti-inflammatory. EA has decreased the expression of spinal COX 2 mRNA in the trend of the therapeutic effect of "distant acupoints + local acupoints", and "local acupoints" intervention may be superior to that of "distant acupoints" intervention. Microglia may be related to the formation of COX 2.</p>

ABSTRACT

This study was aimed to investigate the mechanism underlying the neuropsychiatric comorbidity in the process of "stress"-"inflammation"-"comorbidity",from the perspective of traditional Chinese medicine (TCM) basic theory in combination with our previous 20-year findings.Notably, the neural-psychiatric comorbidity between psychiatric disorders, including depression, schizophrenia and anxiety, and systemic diseases, such as ischemic stroke,Alzheimer's disease (AD) and asthma, have something in common in the pathophysiological mechanism. The stress-induced structural and functional changes in the brain, the stress-initiated diversely structural and neurobiological changes in neurocircuitry, and the stress-mediated neurochemical alterations in neurotransmitter are considered to be involved in the common pathophysiological mechanisms in the neural-psychiatric comorbidity, which initiates a cascade of physiological and psychological processes that contributes to the development of various types of neuropsychiatric disorders.Accordingly,it will be of great significance to investigate mechanisms underlying the neuropsychiatric comorbidity in the process of "stress"-"inflammation"-"comorbidity" under the guidance of the basic theory of "treating disease from the root" and "same treatment for different diseases" in TCM.

ABSTRACT

Cerebral malaria (CM), a severe neurological syndrome caused by Plasmodium falciparum infection, is a serious life-threatening disease with a high mortality. Survivors' persistent brain injury is manifested as long-term neurocognitive disorders. The main neuropathological feature of CM is the sequestration of parasited red blood cells (pRBCs) in cerebral microvessels. Other neuropathological features of CM include petechial hemorrhage in the brain parenchyma, annular hemorrhage, extensive brain endothelial cell activation, and focal endothelial cell injury and necrosis. However, its pathogenesis is still not clear. Currently, some studies have suggested that the pathogenesis of cerebral malaria mainly include pRBC adhesion, inflammatory reaction cascade, vascular leakage damage and brain hypoxia. Studies have shown that the biomarkers currently used as diagnostic and prognostic markers for CM include C-X-C motif chemokine ligand 10 (CXCL10), CXC chemokine ligand 4 (CXCL4), angiopoietin (Ang). In this paper, we systematically summarize the basic and clinical research for cerebral malaria in recent years and the latest literatures for drug studies, and focused on the advance of studies on cerebral malaria and its immunologic mechanism in the recent three years in the aspects of cytokines, immune cells, regulatory factors and biomarkers, so as to provide references for relevant studies.

The radix, leaf, flower and bud of raw medicinal materials and extraction of total alkaloids of Aconitum kusnezoffii Reichb.were all involved in this investigation. All the compositions from the samples were analyzed through fourier transform infrared spectroscopy (FTIR) combined with second derivative IR spectroscopy and two-dimensional IR correlation spectroscopy (2D-IR).It was found that the spectra of raw medicinal materials showed that the radix of A.kusnezoffii Reichb.featuring a large quantity of starch was the same as starch with the characteristic peaks at 1,155,1,070 and 1,019.The leaf, flower and bud contained the similar aromatic hydrocarbons (1,600),glycosides (1,050-1,070),while lipids were not clear. The characteristic peaks of the buds, flowers and leaves were all at 1,595 cm-1 (vibration of phenyl framework) and 1,262 cm-1 (=C-O).Therefore,it was suggested that the common compound of the three parts be diterpenoid alkaloids.Second derivative IR spectroscopy showed that the characteristic peaks of radix was stronger than those of the flower, leaf and bud at 1,712 cm-1 (C=O),which proved that the quantity of characteristic peaks in the radix was larger than those in the flower, leaf and bud.In addition, six autopeaks at 1,745,1,650,1,560 (the most strong),1,465,1,400,1,300 were detected from the radix. The similar autopeaks at 1,745,1,650,1,560 (the most strong),1,465,1,400,1,300 were found in the leaf, bud and flower. In conclusion,it was demonstrated that the macro-fingerprint infrared spectroscopic identification method provided a large quantity of the comprehensive information and entirely grasped the quality of A.kusnezoffii Reichb.Besides,FTIR and 2D-IR provided massive information of the integral structures of the radix, leaf, flower and bud of A.kusnezoffii Reichb.and verified the differences between the four parts of the herb in physical structure and the contents, laying a foundation for further systematic work.
