ABSTRACT

Two ent-kauranoids were isolated from the ethanol extraction of rhizomes of Canna generalis (Cannaceae), and were purified by various technologies, including silica gel and high performance liquid chromatography, and their structures were determined by modern spectroscopy techniques as (5R,8S,9S,10R,13R)-2-oxo-ent-kaur-15-en-17-oic acid (1) and (4R,5S,8S,9S,10S,13R)-19-hydroxy-ent-kaur-15-en-17-oic acid (2). Compound 1 is a new ent-kauranoid, and compound 2 is obtained from rhizomes of Canna generalis for the first time.

ABSTRACT

@#Eleven alkaloids were isolated from the twigs and leaves of Ervatamia pandacaqui using chromatographic methods of silica gel, Sephadex LH-20, ODS, and HPLC.Their structures were elucidated by physical,chemical and spectroscopic methods and determined as voacristine 7-hydroxyindolenine (1),iboxygaine (2), 19S-hydroxyibogamine (3), 3-oxotabersonine (4), perivine (5), pericyclivine (6), rhazinalinol (7), geissoschizol (8), 3, 14-dihydroolivacine (9), vallesamine (10), and conolobine A (11), respectively.All compounds were isolated from this plant for the first time.

ABSTRACT

Five monoterpenoid compounds(1-5) were isolated and purified from the acetone fraction of the aqueous extract of Zingiberis Rhizoma Recens by MCI, Sephadex LH-20, silica gel, semi-preparative HPLC, and TLC. Their structures were identified with multiple spectroscopical methods including 1 D-NMR, 2 D-NMR, and MS. The five compounds were identified as(2E,6Z)-8-hydroxy-2,6-dimethylocta-2,6-dien-1-yl-(E)-3-(4-hydroxy-3-methoxyphenyl) acrylate(1),(2E,6E)-8-hydroxy-3,7-dimethylocta-2,6-die-noic acid(2),(E)-1,8-dihydroxy-3,7-dimethyl-2-octenoic acid(3), linalyl-β-D-glucopyranoside(4), and β-D-glucopyranoside-(2E)-3,7-dimethyl-2,6-octadien-1-yl(5), respectively.Compound 1 was a new monoterpene ester, and compounds 4-5 were isolated from this plant for the first time.

ABSTRACT

Three diarylheptanoids were isolated from the n-butanol fraction of Zingiber officinale peel by MCI Gel CHP-20, Sephadex LH-20, ODS and semipreparative high performance liquid chromatography. Their structures were identified by MS and NMR spectroscopy techniques: (2S,2'S,3R,3'R,4R,4'R,6R,6'R)-6,6'-bis((S)-1-hydroxy-2-(4-hydroxyphenyl)ethyl)-2,2'-bis(4-hydroxy-3-methoxyphenyl)octahydro-2H,2'H-[3,3'-bipyran]-4,4'-diol (1), (E)-7-(4-hydroxy-3-methoxyphenyl)-1-(4-hydroxyphenyl)hept-4-en-3-one (2), and alpinin B (3). Compound 1 is a new compound, and compounds 2-3 were obtained from Zingiber officinale peel for the first time.

ABSTRACT

Chemical constituents of water extracts of Asplenium ruprechtii were investigated. Five compounds were isolated by silica gel, Sephadex LH-20 gel column chromatographies and preparative HPLC, and their structures were identified by various spectral analyses as aspleniumside G(1), trans-p-coumaric acid(2), trans-p-coumaric acid 4-O-β-D-glucoside(3), cis-p-coumaric acid 4-O-β-D-glucoside(4), and(E)-ferulic acid-4-O-β-D-glucoside(5). Among them, compound 1 is a new 9,19-cycloartane glycoside.

ABSTRACT

The wild resources of Paris polyphylla var. yunnanensis, a secondary endangered medicinal plant, are severely scarce. Introduction and cultivation can alleviate market demand. To screen phosphatolytic bacteria in the rhizosphere soil of P. polyphylla var. yunnanensis and provide data support for the development of high-efficiency microbial fertilizer, in this study, the dilution plate coating method was used to isolate and screen the phosphorus solubilizing bacteria with the ability of mineralizing organic phosphorus from the rhizosphere soil of wild and transplanted varieties of P. polyphylla var. yunnanensis in 10 different locations in Yunnan, Sichuan and Guizhou. After separation and purification, the phosphatolytic capacity was analyzed by qualitative and quantitative analysis. Combined with physiological and biochemical experiments, the strains were identified using 16 S rDNA sequencing analysis. Forty one strains were selected from the rhizosphere soil of P. polyphylla var. yunnanensis from 10 different habitats. Among them, 21 strains were obtained from the rhizosphere soil of the wild variety P. polyphylla var. yunnanensis and 20 strains were obtained from the rhizosphere soil of the transplanted variety. And significance analysis found that 41 organophosphate solubilizing strains had significant differences in their ability to solubilize phosphorus. The amount of phosphate solubilizing was 0.08-67.61 mg·L~(-1), the pH value was between 4.27 and 6.82. The phosphatolytic amount of strain Y3-5 was 67.61 mg·L~(-1), and the phosphorus increase amount was 57.57 mg·L~(-1). All 41 strains were identified as Gram-positive Bacillus. Combining physiological characteristic and phylogenetic trees, Bacillus mobilis Y3-5 was finally selected as the candidate rhizosphere phosphatolytic bacteria of P. polyphylla var. yunnanensis. The distribution of phosphorus solubilizing bacteria in the rhizosphere soil of P. polyphylla var. yunnanensis was different, and there were significant diffe-rences in phosphorus solubility. Organophosphate-dissolving strain Y3-5 is expected to be a candidate strain of P. polyphylla var. yunnanensis microbial fertilizer.

ABSTRACT

Objective::To identify the active anti-tumor constituents of Benzoinum according to observation of the anti-tumor effect of chemical constituents from Benzoinum in vitro. Method::The 95%ethanol extract of Benzoinum was systematically separated by silica gel column chromatography, medium pressure liquid preparation chromatography and preparation liquid chromatography, and their structures were identified by physicochemical property and spectral data. Anti-tumor activities of the compounds of Benzoinum were screened by in vitro cells including human hepatoma cells in vitro (HepG2), human lung cancer cells (A549), human cervical cancer cells (HeLa), human breast cancer cells (MCF-7) and human prostate cancer cells (PC-3). Result::Fifteen compounds were isolated from Benzoinum and identified as myricadiol(1), 3-keto-oleanonic acid(2), (4E)-1, 5-bis(4-hydroxyphenyl)-1-methoxy-2-(methoxy-methyl)-4-pentene(3a and 3b), (E)-p-coumaryl alcohol γ-Ο-methyl ether(4), sesamin(5), 5-(3″benzoyloxypropyl)-7-methoxy-2-(3′, 4′-methylenedioxy phenyl)-benzofuran(6), dibutyl phthalate(7), methyl 4-hydroxy-3-methoxybenzoate(8), p-hydroxybenzaldehyde(9), p-hydroxyacetophenone(10), acetovanillone(11), 3-oxo-olean-11, 13(18)-dien-28, 19β-olide(12), vanillin(13), benzoic acid(14), and siaresinolic acid(15). Compounds 1 to 11 were isolated from the resin of Styrax tonkinensis for the first time. A part of these compounds had good anti-tumor activities. Among them, compound 2, 12 showed a strongest activity. Conclusion::The chemical constituents of Benzoinum have good prospects for the development and application of anti-tumor drugs.

ABSTRACT

Objective: To investigate the chemical constituents in the the above-ground part of the plant Achyranthes bidentata Bl. in Amarathaceae. Methods: The raw materials were extracted by heating with 70% ethanol, and the extract was prepared by column chromatography on Diaion HP-20, MCI Gel CHP-20P, Toyopearl HW-40, Sephadex LH-20, ODS and silica gel, coupled with the semi-preparative HPLC, to isolate the chemical constituents. The isolated compounds were identified according to their MS, 1H NMR and 13C NMR data. Results: Twenty one compounds were isolated and identified: haploperoside A(1), sweroside(2), 1'-O-benzyl-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(3), 2-ethyl-3-methyl-maleimide-N-β-D-glucopyranoside(4), 3-eth- ylidene-4-methyl-2, 5-pyrrolidinedione(5), methyl-5-hydroxy-2-pyridinecarboxylate(6), pericampylinone A(7), syringaresinol(8), episyringaresinol(9), methyl 3, 4-dihydroxybenzoate(10), 4-hydroxyphenylacetone(11), 3, 4-dihydroxyphenylacetic acid methyl ester(12), 2-(4-hydroxyphenyl)-ethanol(13), homovanillyl alcohol(14), dihydrosinapyl alcohol(15), 4-hydroxybenzoic acid (16), caffeic acid(17), 3-(4-hydroxy-3, 5-dimethoxyphenyl)propane-1, 2-diol(18), methyl(E)-3-(4-hydroxyphenyl)acrylate (19), trans-ferulic acid(20)and vanillic acid(21). Conclusion: Compounds 1-7 and 9-19 were isolated from the title plant for the first time.

ABSTRACT

We separated and purified five chemical constituents of dried ginger by Diaion HP-20, Sephadex LH-20, silica gel and semi-preparative high performance liquid chromatography. Five gingerols were identified by physicochemical properties and MS and NMR spectroscopy techniques: 4-(2-butyl-6-methyl-4H-pyran-4-yl)-2-methoxyphenol (1), 4-(2-hexyl-6-methyl-4H-pyran-4-yl)-2-methoxyphenol (2), 1-(4-hydroxy-3-methoxyphenyl)tridecane-3,5-diol (3), [10]-gingerdiol (4) and 1-[1-(4-hydroxy-3-methoxy phenyl)-3-oxodecan-5-yl]pyrrolidin-2-one (5a, 5b). Compounds 1-3, 5a, 5b are new compounds.

ABSTRACT

Adding biological passivation agent during composting is one of the most effective ways to reduce the toxicity of heavy metals in contaminated livestock manure. To further improve biological passivation, we obtained a strain with high-heavy metal compounds tolerance to passivate heavy-metal contaminated manure and to characterize heavy-metal biosorption. High-tolerance microorganisms for lead and cadmium were isolated and screened from swine manure composting samples. The strain was identified by its morphology and molecular biology. After the influence of different pH, temperature and salt concentrations on growth of the strain were investigated, the optimal growth conditions were obtained for further analysis of its biosorption characteristics of lead and cadmium. The bacterium with tolerance to lead and cadmium termed SC19 was obtained, whose lead resistance was 600 mg/L and cadmium resistance was 120 mg/L. The isolate was further identified as Cedecea sp., and then its optimum pH was 7.0, temperature was 37 °C, and salt concentration was 0.5%. Lead removal was highest after 30 min of adsorption by the SC19 strain cultured for the stationary phase 36 h, and the maximum removal rate and biosorption capacity of lead were 60.7% and 329.13 mg/g, respectively. Meanwhile, cadmium removal was highest after 30 min of adsorption by the strain cultured for the logarithmic phase 8 h, and the maximum removal rate and biosorption capacity of cadmium were 51.0% and 126.19 mg/g, respectively. Fourier Transform InfraRed (FT-IR) results revealed that the biosorption process mainly happened on the surface of SC19 cell and many active groups on the cell surface could chelate the Pb²⁺ and Cd²⁺. By comprehensive comparison, it was showed that strain SC19 shared a certain capacity of Pb²⁺ and Cd²⁺ biosorption, and the bacterium provided precious microbial germplasm resources for biological passivation of heavy metal contaminated manure.

@#By silica gel column chromatography, solvent extraction and preparative high performance liquid chromatography (HPLC), four new related substance were isolated and purified from the mass production and preparation process of alogliptin benzoate. Then it was analyzed and confirmed by various spectrum identification methods such as nuclear magnetic resonance (NMR) spectroscopy, high-resolution mass spectrometry (HR-MS) and Fourier-transform infrared spectroscopy (FTIR) according to its physical and chemical properties. The chemical structures of the four related substances produced in each step of the synthesis process of alogliptin benzoate were determined, and they were named as impurities L, M, T, and V. These four related substances were new impurities which were found for the first time. The isolation and identification of these impurities are of great importance to the quality control of alogliptin benzoate, and the optimization of manufacturing process.

@#Six steroidal saponins were isolated from the n-butanol extract of 90% ethanol extract of the aerial parts of Paris polyphylla var. yunnanensis by silica gel for column chromatography, MCI column chromatography and preparative high performance liquid chromatography (HPLC). According to the physicochemical properties and spectral data, they were identified as 26-O-β-D-glucopyranosyl-kryptogenin-3-O-α-L-rhamnopyranosyl-(1→4)-α-L-rhamnopyranosy-(1→4)-[α-L-rhamnopyranosyl-(1→2)]-β-D-glucopyranoside (1), dioseptemloside G (2), polyphylloside III (3), chonglouoside SL-19 (4), protodioscin (5), chonglouoside SL-5 (6). Among these, compound 1 was a new compound, and compound 2 was obtained from Paris plants for the first time. The platelet aggregation activities and cytotoxicities of the above compounds were evaluated. The results showed that none of the isolated compounds showed significant platelet aggregation activity; compound 2 and 4 exhibited strong cytotoxicity against human colon adenocarcinoma cell line HT29.

OBJECTIVE: To study the chemical constituents of ethanol extract from Yao medicine Cissampelopsis spelaeicola. METHODS: The petroleum ether, ethyl acetate and n-butanol fraction from 75% ethanol extract of C. spelaeicola were isolated and purified by silica gel, SephadexLH-20 gel column and AB-8 macroporous resin column, etc. The structures of the compounds were analyzed and identified by physicochemical properties and spectral data (mass spectrometry, hydrogen spectrum, carbon spectrum). RESULTS & CONCLUSIONS: Twelve compounds were isolated and identified from 75% ethanol extract of C. spelaeicola. β-sitosterol(Ⅰ) and Stigmasterol(Ⅱ) were isolated from petroleum ether fraction; p-hydroxybenzoic acid(Ⅲ), β-daucossterol(Ⅳ), protocatechuic acid(Ⅴ), 6β-hydroxyeremophi-7(11)-en-12,8β-olide(Ⅵ), 10β-hydroxyeremophil-7(11)-en-8,12-olide(Ⅶ), 10β-hydroxyeremophi-7(11),8(9)-dien-8,12-olide(Ⅷ), Quercetin(Ⅸ), Hyperin(Ⅹ) and 4α-hydroxy- eudesman-11-ene(Ⅺ) were isolated from ethyl acetate fraction; quercetin-3-O-robinobioside(Ⅻ) was isolated from n-butanol fraction. Compounds Ⅰ-Ⅻ are isolated from C. spelaeicola for the first time. The study can lay material foundation for activity stady of C. spelaeicola.

ABSTRACT Objective: To investigate the secondary metabolites from the cultures of Streptomyces sp CSDX076. Methods: The compounds were isolated using column chromatography and RP-18 medium-pressure liquid chromatography. Their structures were elucidated by one-dimensional and two-dimensional nuclear magnetic resonance spectroscopic methods in combination with mass spectrometry experiments. Results: Four compounds were isolated from the cultures of Streptomyces sp CSDX076 and identified as aurantiamide benzoate, deoxytryptoquivaline, 2-acetyl-3,5-dihydroxyl-benzene acetic acid, and 2-acetyl-3,5-dihydroxyl-benzene ester. Conclusion: It was the first time that the four isolated compounds were obtained from the Streptomyces genus.


RESUMEN Objetivo: Investigar los metabolitos secundarios de los cultivos de Streptomyces sp CSDX076. Métodos: Los compuestos fueron aislados usando la cromatografía de columna y cromatografía líquida RP-18 de presión media. Sus estructuras fueron dilucidadas mediante métodos espectroscópicos de resonancia magnética nuclear unidimensional y bidimensional, combinados con experimentos de espectrometría de masa. Resultados: Cuatro compuestos de culturas de Streptomyces sp CSDX076 fueron aislados e identificados como benzoato de aurantiamida, deoxitriptoquivalina, ácido acético 2-acetil-3,5-dihidroxil-benzeno, y éster 2-acetil-3,5-dihidroxil-benzeno. Conclusión: Fue la primera vez que los cuatro compuestos aislados se obtuvieron del género Streptomyces.

AIM To study the chemical constituents from Pterocephalus hookeri (C.B.Clarke) Hoeck and their neuroprotection activities.METHODS The 95% ethanol extract from P.hookeri was isolated and purified by Sephadex LH-20,DM-130 macroporous resin,silica and HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The neuroprotection activities were studied by establishment of Parkinson's disease model of genetically modified zebrafish.RESULTS Nine compounds were isolated and identified as triplostoside A (1),cantleyoside (2),sylvestroside Ⅰ (3),sylvestroside Ⅲ dimethyl acetal (4),sylvestroside Ⅲ (5),laciniatoside Ⅰ-7-dibutyl acetal (6),laciniatoside Ⅰ (7),laciniatoside Ⅱ (8),sylvestroside Ⅳ (9).The n-BuOH fraction of P.hookeri had good performance on neuroprotection in hydrogen peroxide model.CONCLUSION Compound 4 is isolated from this plant for the first time.P.hookeri has neuroprotection activities.

AIM To study the chemical constituents of the secondary metabolites of endophytic fungi DL02 from Saposhnikovia divaricata (Turcz.) Schischk.and their biological activities.METHODS The ethyl acetate and methanol extract leavening from S.divaricata was isolated and purified by silica and Sephadex LH-20,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The antifungal and antibacterial activities were determined by the minimum inhibitory concentration method.RESULTS Seven compounds were isolated and identified as β-sitosterol (1),kojic acid (2),stigmasta-7,22-diene-3β,5α,6α-triol (3),uracil (4),allantoin (5),erythritol (6) and adenosine (7).Compound 4 had strong inhibitory effects on pseudomonas aeruginosa with the MIC values of 31.3 μg/mL.CONCLUSION Compounds 2-5 and 7 are isolated from this fungus for the first time.Compounds 4-5 have strong biological activities.

AIM To study chemical constitutes from Achyranthis bidentata Bl.and their effects on cells proliferation.METHODS The aqueous extract from dregs of a decoction of A.bidentata was isolated and purified by Diaion HP-20,Sephadex LH-20,ODS,silica and HPLC colum,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The effects on proliferation of endothelial cells were evaluated by MTT.RESULTS Ten compounds were isolated and identified as cynarasaponin A (1),ginsenoside R0 (2),chikusetsusaponin Ⅳ a (3),chikusetsusaponin Ⅳ a methyl ester (4),chikusetsusaponin Ⅳ a ethylester (5),chikusetsusaponin Ⅳ a butyl ester (6),chikusetsusaponin-1 (7),zingibroside R1 (8),3-O-β-D-glucuronic acid glycosides-oleanolic acid (9),3-O-β-D-glucuronic acid glycosides ethylester-oleanolic acid (10).Compounds 1,6,7,8 exhibited obviously inhibiting effects on HUVEC proliferation.CONCLUSION Compounds 1 and 10 are isolated from A.bidentata for the first time;compounds 1,6,7,8 have strong inhibiting effects on cells proliferation.

AIM To study the chemical constituents from the dregs of Euphorbia Alatavica Boiss.METHODS The acetone-methanol extract from the dregs of E.Alatavica was isolated and purified by silica,Sephadex LH-20 and preparative-HPLC column,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Nine compounds were isolated and identified as 8-hydroxy-3-methoxy-5H-pyrido [2,1-d] pyrazin-5-one (1),isoscopoletin (2),dihydroapigenin (3),β-carboline alkaloid (4),syringaresinol (5),pinoresinol (6),methylparaben (7),methyl-3,4-dihydroxybenzoate (8),trimethyl3,4-dehydrochebulate (9).CONCLUSION All the compounds are isolated from this plant for the first time,compound 1 and 4 are isolated from the genus Euphorbia for the first time.

AIM To study the chemical constituents from Dendrobium nobile Lindl..METHODS The 95% ethnol extract from D.nobile was isolated and purified by silica and Sephadex LH-20,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Eight compounds were isolated and identified as erianin (1),gigantol (2),moscatin (3),confusarin (4),4,5-dihydroxy-2-methoxy-9,10-dihydro-phenanthrene (5),coumarin (6),hexadecanoic acid,methyl ester (7),dibutyl phthalate (8).CONCLUSION Compounds 1,3,5,6,7,8 are isolated from this plant for the first time.
