ABSTRACT

BACKGROUND@#Syringomyelia is a progressive chronic disease that leads to nerve pain, sensory dissociation, and dyskinesia. Symptoms often do not improve after surgery. Stem cells have been widely explored for the treatment of nervous system diseases due to their immunoregulatory and neural replacement abilities. @*METHODS@#In this study, we used a rat model of syringomyelia characterized by focal dilatation of the central canal to explore an effective transplantation scheme and evaluate the effect of mesenchymal stem cells and induced neural stem cells for the treatment of syringomyelia. @*RESULTS@#The results showed that cell transplantation could not only promote syrinx shrinkage but also stimulate the proliferation of ependymal cells, and the effect of this result was related to the transplantation location. These reactions appeared only when the cells were transplanted into the cavity. Additionally, we discovered that cell transplantation transformed activated microglia into the M2 phenotype. IGF1-expressing M2 microglia may play a significant role in the repair of nerve pain. @*CONCLUSION@#Cell transplantation can promote cavity shrinkage and regulate the local inflammatory environment.Moreover, the proliferation of ependymal cells may indicate the activation of endogenous stem cells, which is important for the regeneration and repair of spinal cord injury.

ABSTRACT

Objective:To investigative the molecular mechanism of C-X-C chemokine receptor type 4 (CXCR4) antagonist AMD3100 in epileptic seizure.Methods:(1) Animal experiment: 36 adult male SD rats were randomly divided into control group (Con, n=12), epilepsy group (Epi, n=12), Epi+AMD3100 group ( n=12). Experimental epilepsy rat models in the Epi group were induced by intraperitoneal injection of pentrazole (PTZ, 40 mg/kg); rats in the Epi+AMD3100 group were given intraperitoneal injection of PTZ (40 mg/kg) 20 min after lateral intracerebroventricular injection of 5 μL (5 mg/mL) AMD3100; rats in the Con group were given intraperitoneal injection of normal saline. Racine grading was used to evaluate the levels of epileptic seizure and the latency of epileptic seizure was recorded in rats from each group. EEG was used to record the abnormal discharges of brain neurons in rats from each group. The content of γ -aminobutyric acid (GABA) in the hippocampus was detected by ELISA kit; γ -aminobutyric acid A receptor α1 subunit ( GABAAR α1) mRNA levels of hippocampal neurons in each group were detected by real-time fluorescent quantitative PCR (qRT-PCR). (2) Cell experiment: the hippocampal neurons from 1-d-old SD rats were primarily cultured; 7 d after cultivation, these cells were divided into control group, epilepsy group and AMD3100 group; the cellular epileptic models in the epileptic group were induced by magnesium-free external fluid; neurons in the AMD3100 group were cultured in magnesium-free external solution containing 10 nmol/L AMD3100 for 3 h, and then changed to Neurobasal medium for further culture; cells in the control group were cultured with Neurobasal medium. Whole cell patch clamp technique was used to detect the spontaneous inhibitory postsynaptic currents (sIPSCs) after AMD3100 (10 nmol/L) perfusion. Results:(1) Animal experiment: the seizure latency in Epi+AMD3100 group was significantly shorter than that in Epi group ([663.30±74.84] s vs. [164.40±17.20] s, t=6.490, P<0.001). The frequency of seizures>grading 4 in the Epi+AMD3100 group was significantly decreased as compared with that in the Epi group (3.75±0.39 vs. 9.00±0.73, t=4.680, P<0.001). ELISA results showed that GABA content in the 3 groups was significantly different ( F=17.850, P<0.001): that in the Epi group was significantly lower than that in the Con group, and that in the Epi+AMD3100 group was significantly higher than that in Epi group ( P<0.05). The qRT-PCR results showed that GABAAR α1 mRNA content among the 3 groups was significantly different ( F=14.400, P<0.001): that in the Epi group was significantly lower than that in the Con group, and that in the Epi+AMD3100 group was significantly higher than that in the Epi group ( P<0.05). EEG results showed that the discharge frequency of rats in the Epi+AMD3100 group was lower than that in Epi group; there was no significant difference in EEG power among the 3 groups ( F=3.220, P<0.001), but the EEG power in the Epi+AMD3100 group was lower than that in Epi group and control group. (2) Cell experiment: patch clamp technique showed that the average frequency and amplitude of sIPSCs in the 3 groups were statistically significant ( F=13.670, P<0.001; F=10.920, P<0.001). As compared with those in the control group and epilepsy group, the average frequency and amplitude of sIPSCs in AMD3100 group were significantly increased ( P<0.05). Conclusion:CXCR4 antagonist AMD3100 can reduce seizure frequency by enhancing inhibitory neurotransmission.

ABSTRACT

OBJECTIVE: To study the mechanism of wound healing promotion of Panax notoginseng-Bletilla striata gum sponge on diabetic foot ulcer (DFU) model rats. METHODS: Healthy SD rats were selected and given high-lipid and high-glucose diet, intraperitoneal injection of streptozotocin once to establish diabetes model. Neodymium-iron-boron magnet was used to press the back of rats to make ulcer wound then established DFU model. Totally 60 DFU model rats were randomly divided into group A(blank group, i.e. normal saline gauze group), group B(vaseline gauze group),group C (gelatin sponge group) and group D (P. notoginseng-B. striata gum sponge group), with 15 rats in each group. The rats were given corresponding gauze/sponge to cover the wound for intervention treatment, changing dressing once every 1-2 days. On the 3rd and 7th day after intervention, the wound healing of rats in each group was observed with naked eyes, and the wound healing rate was calculated. The wound margin tissue was collected to obtain HE staining section, and histopathological observation was conducted under microscope. mRNA expression of β-catenin, GSK-3β and Rspo3 in wound tissue were determined by RT-PCR. RESULTS: On the 3rd and 7th day after intervention, compared with group A, B, C, healing rate of group D was increased significantly (P<0.05); inflammatory cell infiltration, collagen deposition, capillary and granulation tissue growth in wound tissue increased significantly. The mRNA expression levels of β-catenin and Rspo3 all increased, and those of GSK-3β all decreased; except for the difference of β-catenin at the 3rd day and GSK-3β at the 7th day after intervention between group D and group C were not significant, the difference of other indicators was statistically significant (P<0.05). CONCLUSIONS: P. notoginseng-B. striata gum sponge can effectively promote the wound healing in DFU model rats, the mechanism of which may be associated with up-regulating the expression of β-catenin and Rspo3 mRNA and down-regulating the expression of GSK-3β mRNA.

ABSTRACT

Objective To investigate the value of circulating tumor cell(CTC)in non-small cell lung can-cer(NSCLC)and its relationship with clinicopathological features and prognosis. Methods This study totally in-cluded 59 patients with histologically confirmed diagnosis of NSCLC. CTCs were quantified in 7.5 mL peripheral blood samples collected prior to surgery using CellSearch system(Veridex). Statistical analysis of the relationship between CTC positive rate and clinical data,pathological characteristics and prognosis were performed. Results The CTC positive rate was 30.51% and it was significantly correlated with distant metastasis(P = 0.012),TNM stage(P = 0.002)and lymph node metastasis(P = 0.002). The 5-year overall survival rate in the CTCs positive group was significantly lower(22.22% vs. 68.29%,P = 0.005)and the cumulative recurrence was significantly higher(55.55% vs.29.27%,P=0.022)compared with the CTCs negative group.The univariate analysis indicated that the prognostic influencing factors included TNM stage,lymph node metastasis,distant metastasis and CTCs re-sults and the multivariate analysis showed that TNM stage,distant metastasis and CTCs positive were significantly correlated with overall survival and cumulative recurrence(P<0.05). Conclusion CTC is an independent prog-nostic indicator for NSCLC.

ABSTRACT

ObjectiveTo investigate the early diagnosis and treatment of pleuropulmonary blastoma in children. Meth-ods The progress of pleuropulmonary blastoma from type 1 to type 3 was retrospectively analyzed.Results The male infant had no obvious abnormality in chest X-ray at 18 days. At one year old, X-ray iflm of the chest showed cystic lesions near hilum of the left lung (about 5 cm × 4 cm × 4 cm). The congenital cystic adenomatoid malformation was considered and an operation resection was suggested. However, it had been rejected by his parents. At 3 years and 2 months old, chest X-ray and CT showed left lung had a solid cystic lesion, the volume was signiifcantly enlarged, the back side was solid and the upper part had a large gas cavity. Two months later, the patient had asthmatic suffocation. The chest CT showed the tumor was completely solid. Pleuropulmonary blastoma type 3 was considered. The patient received the conventional chemotherapy after operation excision, and was followed up for 1 year. No recurrence was observed after the treatment.Conclusions Type 1 pleuropulmonary blastoma and congenital cystic adenomatoid malformation are hard to be differentially diagnosed. Early diagnosis and treatment of type 1 pleuropulmo-nary blastoma can improve the prognosis.

BACKGROUND:Cone-beam computed tomography has been widely applied in the dentistry due to irreplaceable advantages. Among those advantages, linear measurement is one of the important ones. Further studies are suggest to explore the accuracy and repeatability of linear measurement for the whole skul , as wel as the effect of different resolution and scanning thickness on the linear measurement results. OBJECTIVE:To evaluate the accuracy of linear measurement using cone-beam computed tomography (Newtom VG) on the human skul , with two different voxel sizes. METHODS:In this study, 22 anatomic landmarks in four dry human skul s were marked and 11 linear measurements were obtained. These were considered to be the gold standard (real measurement). The skul s were scanned by cone-beam computed tomography (Newtom VG) at two voxel sizes:0.3 mm and 0.15 mm. Linear measurement was performed in axial and coronal planes. SPSS software version 17 was used for data analysis between radiographic measurement and real measurement. RESULTS AND CONCLUSION:The mean differences of real and radiographic measurements were-0.27 to 0.14 in four different sections, and statistical analysis showed no significant difference between linear measurements and gold standard (P>0.05). There was no significant between two different voxel sizes (P>0.05). Cone-beam computed tomography (Newtom VG) is highly accurate and reproducible in linear measurements in the axial and coronal planes. A cone-beam computed tomography scan with a larger voxel size (0.3 mm) is recommended, resulting in lower radiation dose and faster scan time.

ABSTRACT

Objective To establish a method for the determination of the antifungal component produced by Streptomyces NG-715.Methods The antifungal component was used as the reference component for UV spectrophotometry detemined at 339 nm. Results By spectrophotometry,a good linearity was established within 8μg/mL~20 μg/mL,and the average recovery rate was 99.98%(n=6 )with RSD being 0.1 1%.Conclusion The method is simple,rapid,accurate with good reproducibility for determining the antifungal component produced by Streptomyces NG-715.
