RESUMEN

Objective: To evaluate the immunogenicity, safety, and immune persistence of the sequential booster with the recombinant protein-based COVID-19 vaccine (CHO cell) in healthy people aged 18-84 years. Methods: An open-label, multi-center trial was conducted in October 2021. The eligible healthy individuals, aged 18-84 years who had completed primary immunization with the inactivated COVID-19 vaccine 3 to 9 months before, were recruited from Shangyu district of Shaoxing and Kaihua county of Quzhou, Zhejiang province. All participants were divided into three groups based on the differences in prime-boost intervals: Group A (3-4 months), Group B (5-6 months) and Group C (7-9 months), with 320 persons per group. All participants received the recombinant COVID-19 vaccine (CHO cell). Blood samples were collected before the vaccination and after receiving the booster at 14 days, 30 days, and 180 days for analysis of GMTs, antibody positivity rates, and seroconversion rates. All adverse events were collected within one month and serious adverse events were collected within six months. The incidences of adverse reactions were analyzed after the booster. Results: The age of 960 participants was (52.3±11.5) years old, and 47.4% were males (455). The GMTs of Groups B and C were 65.26 (54.51-78.12) and 60.97 (50.61-73.45) at 14 days after the booster, both higher than Group A's 44.79 (36.94-54.30) (P value<0.05). The GMTs of Groups B and C were 23.95 (20.18-28.42) and 27.98 (23.45-33.39) at 30 days after the booster, both higher than Group A's 15.71 (13.24-18.63) (P value <0.05). At 14 days after the booster, the antibody positivity rates in Groups A, B, and C were 91.69% (276/301), 94.38% (302/320), and 93.95% (295/314), respectively. The seroconversion rates in the three groups were 90.37% (272/301), 93.75% (300/320), and 93.31% (293/314), respectively. There was no significant difference among these rates in the three groups (all P values >0.05). At 30 days after the booster, antibody positivity rates in Groups A, B, and C were 79.60% (238/299), 87.74% (279/318), and 90.48% (285/315), respectively. The seroconversion rates in the three groups were 76.92% (230/299), 85.85% (273/318), and 88.25% (278/315), respectively. There was a significant difference among these rates in the three groups (all P values <0.001). During the sequential booster immunization, the incidence of adverse events in 960 participants was 15.31% (147/960), with rates of about 14.38% (46/320), 17.50% (56/320), and 14.06% (45/320) in Groups A, B, and C, respectively. The incidence of adverse reactions was 8.02% (77/960), with rates of about 7.50% (24/320), 6.88% (22/320), and 9.69% (31/320) in Groups A, B, and C, respectively. No serious adverse events related to the booster were reported. Conclusion: Healthy individuals aged 18-84 years, who had completed primary immunization with the inactivated COVID-19 vaccine 3 to 9 months before, have good immunogenicity and safety profiles following the sequential booster with the recombinant COVID-19 vaccine (CHO cell).

RESUMEN

The evaluation of coronary artery ischemia is of great significance in the diagnosis and treatment of coronary heart disease. In recent years, fractional flow reserve derived from computed tomographic angiography (FFRct) has been used to evaluate the diagnosis of coronary artery specific ischemia, which enriches the clinical management of patients with coronary artery disease. A number of studies have confirmed that FFRct has reliable diagnostic performance compared with invasive fraction flow reserve (FFR). Because of its non-invasive, repeatable measurement and other advantages, the application universality of FFRct is significantly higher than that of invasive examination. At the same time, the application scope of FFRct is also continuously expanding, and it is not limited to its position as a "gatekeeper" for invasive coronary angiography. It has good accuracy in the diagnosis of complex coronary artery disease, and can also be used to guide the choice of surgical strategies such as coronary artery bypass grafting and stent implantation.

Objective To study the expression level of cathepsin S in elderly patients with different degree of coronary heart disease(CHD),and to evaluate the correlation between cathepsin S level and the severity of CHD.Methods A total of 126 elderly patients with CHD were enrolled,including 36 cases with acute myocardial infarction(group AMI),48 cases with unstable angina(group UAP),and 42 cases with stable angina(group SAP).During the same period,40 healthy subjects were selected as controls.Venous blood was collected immediately after admission.Arterial blood was taken through radial/femoral artery sheath in AMI patients who underwent primary percutaneous coronary intervention,and coronary arterial blood was taken in AMI patients undergoing intracoronary thrombus aspiration.SYNTAX score was assessed in patients undergoing coronary angiography.Serum levels of high sensitivity C reactive protein(hs-CRP),matrix metalloproteinase-9 (MMP-9) and cathepsin S were determined in all specimens and compared between groups.Results Serum levels of cathepsin S,MMP-9 and hs-CRP were higher in CHD patients than in the control group(P<0.05).Among CHD patients,group AMI had the highest serum levels of cathepsin S,MMP-9 and hs-CRP,followed by group UAP and group SAP(F =106.830,197.035 and 310.442,all P =0.000).Spearman's rank correlation test suggested that cathepsin S level was positively correlated with serum levels of MMP-9 (r=0.816,P =0.000)and hs-CRP(r =0.827,P =0.000).SYNTAX score was positively correlated with serum levels of cathepsin S(r=0.581,P=0.000),MMP-9(r=0.511,P=0.000),and hs-CRP (r=0.557,P =0.000).Among the 24 patients with AMI who underwent intracoronary thrombus aspiration,the levels of cathepsin S,MMP-9 and hs-CRP were higher in coronary artery blood than in peripheral artery blood (t =217.288,3.177 and 681.479,all P =0.000).Cathepsin S and hs-CRP levels in peripheral arterial blood had positive correlations with those in coronary arterial blood respectively(r =0.962 and 0.494,P =0.000 and 0.014),but such correlation between in peripheral arterial blood versus in coronary artery blood was not found in MMP-9 levels (r =-0.188,P =0.380).Conclusions Serum cathepsin S level is higher in elderly CHD patients than in healthy people,increases along with the increased severity of CHD,and positively correlates with the degree of coronary stenosis.

RESUMEN

Three Chrysanthemum-chalcone-isomerase genes( CmCHI) were successfully cloned by PCR from the database of Chrysanthemum transcriptome and named CmCHI1,CmCHI2 and CmCHI3,respectively. Bioinformatics analysis showed that the base numbers of CmCHI1-3 open reading frame were 708,633 and 681 bp,encoding 235,210 and 226 amino acids,respectively. Three fusion proteins of about 30 kDa were successfully induced by prokaryotic expression technology,and the corresponding recombinant fusion proteins were isolated and purified by Ni-NTA resin column. Clustering analysis showed that the 3 CmCHI were homologous with Compositae plants,and CmCHI1 and CmCHI3 belonged to type Ⅰ CHI. CmCHI2 belongs to type Ⅳ CHI. Using β-actin as an internal reference gene,RT-qPCR was used to detect and analyze the expression of CmCHI1-3 genes in Hangju. The results showed that the expression levels of CmCHI1 and CmCHI3 were higher,while the expression levels of CmCHI2 were lower. It was concluded that CmCHI1 and CmCHI3 were the main chalcone isomerase genes involved in the synthesis of flavonoids in Hangju,and CmCHI2 was a helper gene. Flooding treatment significantly promoted the expression of CmCHI1 and CmCHI3 genes,but had no regulatory effect on CmCHI2. The above results provided a basis for further study of the molecular regulation mechanism of CHI gene in the metabolism of flavonoids in Hangju,which laid a foundation for improving the content of flavonoids in Hangju and finally improving the medicinal quality of Hangju.

The contents of 22 kinds of mineral elements in different parts of Changium smyrnioides and in the rhizosphere soil of 10 different populations were determined by microwave digestion-inductively coupled plasma atomic emission spectrometry (ICP-AES). The characteristics of mineral elements in the plants and the soil of main distribution area of Ch. smyrnioides was analyzed and the mechanism of the quality formation of Ch. smyrnioides was explored to provide the basis for the quality evaluation and cultivation regulation of Ch. smyrnioides.The results showed that the quality of soil environment was better in the main distribution area of Ch. smyrnioides, the content of trace elements was higher, the contents of P and K were lower and the contents of mineral elements in the soil of each distribution area was significantly different. The three elements of Se, P and K are significantly accumulated in the root of Ch. smyrnioides. There were significant differences in the total contents of mineral elements in the roots of Ch. smyrnioides in different producing areas. The contents of mineral elements in different parts of Ch. smyrnioides were significantly different. Ch.smyrnioides of the main distribution area of Ch. smyrnioides belonged to the safety level, the distribution of mineral elements in the plants can be used as an indicator of the quality of medicinal herbs.

Objective To evaluate the magnetic resonance imaging (MRI) findings in differential diagnosis between the adult reversible splenial lesion syndrome (RESLES) and ischemic infarction of the splenium of the corpus callosum (SCC). Methods The MRI findings and clinical data of 7 RESLES patients and 13 patients with ischemic infarction of SCC who were clinically diagnosed and treated in our center from May 2015 to June 2017 were analyzed retrospectively. The main MRI findings included location,morphology,signal intensity,maximum cross-sectional area,diffusion weighted imaging (DWI),and apparent diffusion coefficient (ADC) value. Results On the MRI findings of 7 RESLES patients (5 males and 2 females),the centers of all lesions of the SCC were located in the midline of SCC,the lesion shapes were round,ellipse,or spindle,and the distribution of the lesions was bilateral and symmetric as the center of the midline of SCC. The lesions were hyperintense on DWI,and the mean maximum cross-sectional area of lesions was (56.9±32.6) mm and the mean ADC value was (0.3963±0.0715) ×10 mm/s. On the review MRI,all the lesions disappeared (mean interval:10 days). On the MRI findings of 13 patients with ischemic infarction of SCC (10 males and 3 females),the lesions were irregular or patchy in shape and were almost laterally and asymmetrically distributed. The lesions were hyperintense on DWI,and the mean maximum cross-sectional area was (55.1±43.9) mm and the mean ADC value was (0.4978±0.0123) ×10 mm/s. The mean maximum cross-sectional area (t=0.096,P=0.925) and the ADC value (t=-1.988,P=0.062) were not significantly different between RESLES group and ischemic infarction of SCC group. Conclusions The location,morphology,and distribution of the SCC lesions and the co-existence of other lesions in the brain are helpful for the differential diagnosis between RESLES and ischemic infarction of SCC. However,the mean maximum cross-sectional area and the ADC value show no obvious difference between these two diseases.

Objective To investigate the relationship between the expression of plasma miRNA-195 in pa-tients with severe coronary artery stenosis and its coronary collateral circulation. Methods One hundred and nine-ty patients with selective coronary angiography in our hospital from March 2000 to May 2017 were enrolled,who were divided into poor collateral circulation group(n = 90)and good collateral circulation group(100 cases)ac-cording to the Rentrop classification.Plasma miRNA-195 and VEGF levels were compared between the two groups. The relationship between plasma miRNA-195 and VEGF was analyzed,and also the relationship between plasma miRNA-195 and Gensini points.Results(1)Plasma miRNA-195 and VEGF levels in good collateral circulation group were significantly higher than those in poor collateral circulation group(P<0.05).(2)There was a positive correlation between plasma miRNA-195 and VEGF in patients with coronary heart disease(r=0.628,P=0.022). (3)There was a positive correlation between plasma miRNA-195 and Gensini score in patients with coronary heart disease(r = 0.765,P = 0.013). Conclusion MiRNA-195 may be involved in the production of neovasculariza-tion by regulating VEGF in the formation of collateral circulation in patients with severe coronary artery stenosis, which can be used as a biomarker for predicting the formation of collateral circulation for clinical reference.

<p><b>OBJECTIVE</b>To study the correlation of apparent diffusion coefficient (ADC) measured by diffusion-weighted magnetic resonance imaging (MRI) with the molecular subtypes and biological prognostic factors of invasive breast cancer masses.</p><p><b>METHODS</b>Breast MRI data (including dynamic enhanced and diffusion-weighted imaging) were collected from 64 patients with pathologically confirmed invasive breast cancer masses (a total of 69 lesions). The mean ADC values of the lesions were calculated and their correlations were analyzed with the 5 molecular subtypes of invasive breast cancer and the biological prognostic factors including estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor 2 (HER2), and Ki-67 index.</p><p><b>RESULTS</b>The ADC values did not differ significantly among the 5 molecular subtypes of invasive breast cancer masses (P>0.05) or among lesions with different ER, PR, or HER2 status (P>0.05). The mean ADC values were significantly higher in Ki-67-positive lesions than in the negative lesions (P=0.023 and negatively correlated with the expressions of Ki-67 (r=-0.249).</p><p><b>CONCLUSION</b>ADC value can not be used to identify the molecular subtypes of invasive breast cancer masses or to evaluate the biological prognosis of the lesions, but its correlation with Ki-67 expression may help in prognostic evaluation and guiding clinical therapy of the tumors.</p>

Effects of different water temperature, stocking density and feeding cycle on growth, feeding and survival of Hirudo nipponica have been studied, six temperature gradients were set: 18, 22, 26, 30, 34 and 38 ℃, five stocking density gradients were set: 30, 60, 120, 180 and 240 leech/L, four feeding cycle gradients were set: 2, 5, 10 and 20 d, respectively. The results showed that there exists a significant regression relationship between water temperature and specific growth rate: y=-0.016 5x²+0.836 9x-6.847 5(R²=0.990 8)(P<0.05), a regression analysis indicated that specific growth rate reached the maximum (3.76) at 25.36 ℃. When water temperature was beyond 30 ℃, the survival rate significantly decreased as water temperature increased (P<0.05). The specific growth rate and survival rate decreased as stocking density increased. A linear relationship exists between the feeding cycle and the SGR: y=-0.094 1x+3.832 9(R²=0.992 7). From this study, it can be concluded that the optimal water temperature and stocking density for the growth of H. nipponica is 22-26 ℃ and 30-120 leech/L, respectively.

Objective To investigate the inducing factors of diabetic ketosis(DK)in type 2 diabetes mellitus( T2DM) patients for more effective prevention. Methods A total of 545 T2DM patients with DK who received treatment in Zhejiang Provincial Diabetes Prevention and Treatment Center from June 2011 to June 2015 were enrolled,which contains 297 newly- diagnosed ketosis - prone diabetic(KPD)patients(KPD group)and 248 T2DM patients with DK(T2DM group),the etiological factors and clinical characteristics were analyzed retrospectively. Results The leading causes of DK were long term nonintervetion(71. 71% )and infection(15. 15% )in KPD group,while unadjusted therapeutic regimen(39. 91% )and break - off(22. 98% )and infection(20. 97% )were the leading cause in T2DM group. Compared with T2DM group, patients in KPD group were younger(P ﹤ 0. 01),and had lower BMI index(P ﹤ 0. 01),higher HbA1c level(P ﹤ 0. 01), more serious ketosis patients(urine ketone≥ ﹢ ﹢ ,P ﹤ 0. 05)and more DKA patients(P ﹤ 0. 05). Conclusion T2DM patients with ketosis could not be ignored,especially the KPD patients. More attention should be paid to diabetes health education,and screening of high - risk group and long term follow - up in patients with T2DM are effective means to reduce the incidence of DK in T2DM.

Objective To discuss the efficacy of six ingredient rehmannia pill combined with metformin in treatment of type 2 diabetes with Qi-Yin deficiency.Methods 120 cases with type 2 diabetes were selected.They were divided into two groups randomly.The observation group (60 cases) took six ingredient rehmannia pill combined with metformin.The control group (60 cases) took metformin.The efficacy of interlocking intramedullary nail and fracture contusion capsule in treatment of open tibial fracutre was evaluated by fasting blood glucose,postprandial 2 h blood glucose,total symptom scores before and after treatment,adverse reaction during treatment.Results The effective rate of observation group was significantly higher than that of the control group (P < 0.05).Before treatment,fasting blood glucose and postprandial 2 h blood glucose level had no significant difference between two groups.After treatment,the blood glucose of two groups were decreased (P < 0.05).The fasting blood glucose and blood glucose 2 h after meal of observation group were lower than that of the control group (P < 0.05).Before treatment,total symptom scores had no significant difference.After treatment,the two groups of the total symptom scores were decreased significantly (P < 0.05),and the observation group the total symptom score lower than that of the control group (P < 0.05).During the treatment,the control group of 1 cases of hypoglycemia,the observation group of 1 cases of hypoglycemia,1 cases of mild nausea.The adverse reactions of two groups had no significant difference.Conclusion Six ingredient rehmannia pill combined with metformin had good effect on type 2 diabetes.It could significantly improve the symptoms of diabetes diagnosed by traditional Chinese medicine.The hypoglycemic effect was obvious with high drugy safety.It was worthy of clinical application.

Objective To investigate the protective effects of statins on renal function in patients with severe acute pancreatitis(SAP). Methods Fifty-seven SAP patients combined with AKI were divided into statins group(n = 27)and control group(n = 30). The level changes of amylase(AMY),blood uria nitrogen(BUN), creatinine(Cr),total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterin(LDL)and high density lipoprotein cholesterol(HDL) before and after statin treatment were observed in statins group and then all were compared with those in control group. Results There was no significant difference of AMY,BUN,Cr,TC,TG, LDL and HDL between 2 groups before treatment. After treatment,AMY,BUN,Cr,TC and LDL were significantly lower while HDL were significantly higher in two groups(P<0.05). However,AMY,BUN and Cr were far lower on 7th day and 14th day in statins group than those in control group(P<0.05). Simultaneously,TC and LDL were far lower while HDL were far higher 14thday in statins group when compared with those in control group(P < 0.05). Conclusions Statins have protective effects on renal function in patients with SAP ,and the mechanism is probably related to the inhibition of HMG-COA ,elimination of oxygen free radicals and anti-fibrosis.

Objective To explore the biological characteristics and biofilm formation of a strain of Klebsiella pneumoniae isolated from the stool of a patient with clinical inflammatory bowel disease .Methods The TH6 strain was identified as Klebsiella pneumoniae by 16S RNA, MALDI-TOF and automatic biochemical instrument .The morphology, growth rate, biofilm formation, capsid virulence factors and other biological characteristics were analyzed .Results and Conclusion The TH6 strain was short rod-shaped with a capsule under the transmission electron microscope , and was of K57 type according to the capsule serotype test .K.pneumonia TH6 grew rapidly under aerobic and anaerobic conditions , almost at twice the rate of standard strain ATCC2146, and reached the logarithmic growth phase at 2 h.The strain had multi-drug resistance.Biofilm formation was observed under a confocal laser scanning microscope (CLSM) at 4, 8, 12, 24, 48, 72, 96 and 120 h and the biofilm was formed at 72 h.The ability of fast growth , biofilm formation and other characteristics of K.pneumonia TH6 showed that this strain has a strong ability to adapt to the new environment is difficult to cure after colonization in vivo.

The effects of harvest and different processing methods on the anti-thrombin activity of Poecilobdella manillensis were respectively studied. The indicators included processing methods (vacuum freeze drying, fresh homogenate, drying under sunlight, freezing, scalding, baking under different temperatures), different parts (entire body, cephalon, pygidium, exudate) and body weights (≤10, 10-20, 20-30, 30-40, ≥40 g). The anti-thrombin activities of P. manillensis with different processing methods were evaluated by direct anti-thrombin titration. The results indicated that the processing methods significantly affected the anti-thrombin activities of P. manillensis. Among the 11 groups, the anti-thrombin activity of P. manillensis processed with vacuum freeze drying (1 303.56 U•g⁻¹) was significantly highest than the other groups (P<0.05), and that processed with baking under 90 ℃ (15.44 U•g⁻¹) was the lowest. The anti-thrombin activity of the cephalon of P. manillensis (226.42 U•g⁻¹) was the highest, and that of the pygidium (102.12 U•g⁻¹) was lowest; the anti-thrombin activities for different body weights were significantly different (P<0.05); and among the five groups, the body weight of ≤10 g (328.86 U•g⁻¹) was the highest (P<0.05), and the body weight of ≥40 g (87.71 U•g⁻¹) was the lowest. In conclusion, harvest and different processing methods had a significant impact on the anti-thrombin activities of P. manillensis. In the study, for the optimal processing method for P. manillensis, the body weight between 20-30 g is recommended, and the vacuum freeze drying is preferred, which is followed by the drying under sunlight.

The effect of water temperature, stocking density and feeding cycle on the growth of Poecilobdella manillensis juvenile was conducted P. manillensis was conducted respectively under different conditions: water temperatures(18, 22, 26, 30,34, 38 degrees C and CT), stocking density (75, 125, 200, 275, 350 individual/L) and feeding cycle(2, 4, 6, 8, 12, 16 d). After 30 days, survival rate, weight gain rate, specific growth rate were measured. There was a significant correlation between water temperature and specific growth rate (γ = -0.066x2 + 3.543 1x -38.09, R2 = 0.837 9). Based on the regression equation, the specific growth rate of P. manillensis achieved the maximum (9.461 4) at 26.84 degrees C. And the most optimal water temperature was 26-30 degrees C. Meanwhile, the survival rates of P. manillensis was 0 at 38 degrees C in 3 d. There was significant negative correlation between density and specific growth rate (γ = -0.005 7x + 9.197 3, R2 = 0.998 3) and between feeding cycle and specific growth rate (γ = -0.468 2x + 10.574, R2 = 0.998 8).

RESUMEN

Objective To study the effect of dopamine receptors on neurological and physiological activities. Methods Dopamine D1 receptor gene (DRD1) knockout mice and dopamine D3 receptor (DRD3) gene knockout mice were introduced, and double gene knockout mice were bred in our lab.Seven SPF male mice in each group were used in this experiment.The food intake, water intake, body weight gain for 24 hours were tested on the age of 30 d, 50 d, and 70 d and were compared with those of wild type mice.Results DRD1 gene and DRD3 gene showed significant effect on the body weight in mice in age of 21 day and 35 day, but at the age of 90 day, the differences became insignificant among the mice of various genetypes.Conclusions Dopamine may effect on the foraging and satiety in newborn mice through regulating the hypothalamic-pituitary-adrenal ( HPA ) axis activity, and finally leads to a reduced body weight gain in newborn mice and puppies during lactation.Furthermore, DRD1 gene and DRD3 gene may influence on body weight of newborn mice through regulating mothers’ lactation, lead to a lower body weight at ablactation, and compensatory increase of body weight after ablactation.Our results provide a substantial foundation for studying the function and interaction of DRD1 and DRD3 genes.

RESUMEN

The effect of sterilization methods on biological activity of fibronectin on the surface of biomaterials was elaborated in the present study. Sterile protein- modified biomaterials were fabricated by microfilter filtration and UV irradiation, respectively. UV irradiation altered the conformation of surface- adsorbed fibronectin and further affected the attachment, morphology and biological function of endothelial cells. However, microfilter filtration did not to change the normal conformation of fibronectin, or the proliferation and biological function of endothelial cells, indicating that microfilter filtration sterilization is the most suitable method for protein-substrate.

Objective To understand the awareness status of diabetic nephropathy( DN)and its influence factors among diabetes patients. Methods A total of 1236 cases of diabetes were investigated on the awareness of DN and Logistic regression was used to analyze its influence factors. Results The awareness rate of DN was 77. 67% and its influence factors were age,education level,monthly household income and available diabetic health education( OR=1. 15,1. 80, 1. 41and 0. 98 respectively,all P< 0. 05). Moreover,the awareness status of DN prevention was at a low level. Among the 9 methods to prevent DN,only had 4 methods the awareness rate of which was over 80%. Conclusion Health education on DN control and prevention among diabetes patients should be emphasized.
