RESUMEN

Objective To explore the effects of different acupoints, different target organs, and different interventions on acupoint efficacy based on ACU&MOX-DATA platform; To illustrate and visualize whether the above factors have the characteristics of"specific effect"or"common effect"of acupoint efficacy. Methods The multi-source heterogeneous data were integrated from the original omics data and public omics data. After standardization, differential gene analysis, disease pathology network analysis, and enrichment analysis were performed using Batch Search and Stimulation Mode modules in ACU&MOX-DATA platform under the conditions of different acupoints, different target organs, and different interventions. Results Under the same disease state and the same intervention, there were differences in effects among different acupoints;under the same disease state, the same acupoint and intervention, the responses produced by different target organs were not completely consistent;under the same disease state and acupoint, there were differences in effects among different intervention measures. Conclusion Based on the analysis of ACU&MOX-DATA platform,it is preliminary clear that acupoints, target organs, and interventions are the key factors affecting acupoint efficacy.Meanwhile, the above results have indicated that there are specific or common regulatory characteristics of acupoint efficacy.Applying ACU&MOX-DATA platform to analyze and visualize the critical scientific problems in the field of acupuncture and moxibustion can provide references for deepening acupoint cognition, guiding clinical acupoint selection, and improving clinical efficacy.

RESUMEN

Objective To observe the intervention effect of acupuncture at"Zusanli"(ST36)or"Neiguan"(PC6)on the ulcerative colitis(UC)model mice. And to analyze the similarities and differences of gene transcription expression between the diseased colon and the non-diseased heart after acupuncture intervention by using transcriptomics,to explore whether there are efficacy differences between the two acupoints. Methods The UC mouse model was established by inducing dextran sulfate sodium salt(DSS).After successful modeling, the mice were randomly divided into model group,Zusanli group, and Neiguan group, with 9 mice in each group.Zusanli group and Neiguan group intervened with acupuncture at"Zusanli"and"Neiguan",respectively,30 min each time, once a day,for 5 consecutive days. The control and model groups were only bound without acupuncture.Observe the general situation of mice in each group, calculate the disease activity index(DAI),observe the morphological changes of colon tissue by HE staining.Use transcriptome sequencing technology to sequence and biologically analyze the colon tissue of mice in each group, and compare and study whether the two acupoints have differences in acupoint efficacy from the perspective of diseased tissue. At the same time, take the heart tissues of each group for comparative analysis, and discuss whether the effects of the two acupoints are different from the perspective of non-diseased tissue. Results Compared with control group, the DAI score of the model group increased significantly(P<0.001),and HE staining results showed significant pathological damage;after acupuncture at Zusanli or Neiguan, the DAI scores of UC mice were reduced considerably(P<0.05),and the pathological damage was significantly improved. The results of transcriptome sequencing and bioinformatics analysis showed that the effects of acupuncture at Zusanli or Neiguan on the colon of diseased tissue involved multiple genes, and 825 differential genes with similar changes(co-DEGs)were mainly enriched in white blood cells and chemokine pathways;there were 168 differentially expressed genes(ST36 only-DEGs)affected only by Zusanli, which were enriched to gastric acid secretion and other pathways;there were 325 differentially expressed genes(PC6 only-DEGs)only affected by Neiguan, which were enriched in cytokine-cytokine receptor interaction and other pathways. There were 95 co-DEGs in the heart of non-diseased tissue, which were mainly enriched in positive regulation of defense response and response to interleukin-1;there were 28 ST36 only-DEGs;there were114 PC6 only-DEGs,which were enriched in cardiovascular diseases and other pathways. Conclusion Acupuncture of"Zusanli"and"Neiguan"could improve UC mice's symptoms and pathological colon changes. However, through transcriptomic analysis,it was found that the two acupoints had differences in gene transcription effects on the colon of diseased tissue and the heart of non-diseased tissue, suggesting that acupoints with different distribution positions had some similarities in macro efficacy.Still, there were some differences at a micro level.

RESUMEN

Single-cell multimodal omics(scMulti-omics)technology is a cutting-edge biotechnique for identifying cell type specificity and exploring gene regulation of cellular processes.Based on the combined application of two or more single-cell omics, thirteen scMulti-omics patterns of multi-dimensional and multi-scale were formed. In this paper,we proposed specific paths and strategies for constructing the database for scMulti-omics technology, preprocessing scMulti-omics data, and developing a prediction model.Combining scMulti-omics technology with acupuncture is a beneficial exploration for the modernization and advancement of inter-disciplinary Chinese medicine. The establishment of a prediction model of how acupuncture therapy regulates diseases using scMulti-omics data can infer the heterogeneous landscape of cell-level,which is anticipated to profoundly elucidate the multi-dimensional cell regulation mechanism of acupuncture in the treatment of diseases.

RESUMEN

Apoptosis refers to the programmed death process of cells modulated by genes. It plays an important role in promoting the evolution of organisms, regulating the development of multiple systems, and maintaining the stability of internal environment. The major apoptosis pathways include mitochondrial pathways, death receptor pathways, and endoplasmic reticulum pathways. With the rapid development of the scientific research on acupuncture, it’s revealed that cell apoptosis should be closely related to the action of acupuncture. So far, the major apoptosis factors involved in relevant researches are P53, Bcl-2 family, Cyt-C, Caspase, Fas/Fasl, TNFR1/TNF-?, and FADD-Caspase8, and PERK, etc. Via influencing these apoptosis factors or genes, acupuncture can produce various effects in improving tissue ischemia, protecting neural cells, reducing inflammation, and correcting endocrine disorders. This article summarized and reviewed the apoptosis pathways and key factors related to the action of acupuncture, to provide some beneficial references for future studies.

RESUMEN

This study was carried out to defect the effects of acupuncture and moxibustion therapy on the proliferation and differentiation of neural stem cells in the olfactory bulb of senescence-accelerated mouse P8. Immunofluorescence double staining methods(BrdU/Nestin, BrdU/MAP-2 and BrdU/beta-tubulin III) were used. The results show that both acupuncture and moxibustion could improve proliferation of neural stem cells, but only moxibustion could induce differentiation of neural stem cells into neuron.

RESUMEN

Metabonomic approaches are believed to be one of the powerful methods for disease diagnosis and assessment of patients' conditions. However, many publications have been providing the evidences of the biases caused by many factors such as diet, physical exercises, environment and individual differences in the clinical researches. For the purpose of ensuring the comparability of metabonomic 1H NMR data from different studies, it is necessary to control those nonexperimental factors in the study design and the data analysis.

RESUMEN

To investigate the growth and osteogenesis characteristics of cultured canine mesenchymal stem cells (cMSCs) under osteogenic induction. We found the cMSCs were isolated from adult canine using density gradient separation method. The cMSCs attachment formed soon after seeding and grew into colonies with the appearance of fibroblastic cells. The osteogenic induction compound of Dexamethasone (Dex), beta-sodium glycerphosphate (beta-GP), ascorbic acid (AA) was added to passaged cMSCs and the proliferation and osteogenic differentiation of them was studied. The morphology of cells was observed by light micrograph and transmission electron microscope. The proliferation and growth characteristics of cMSCs were observed during primary and passage cultures through MTT. The differentiation were assayed by alkaline phophatase (ALP) and osteocalcin (OCN). We found the cMSCs have an active proliferative ability in primary and passage culture, and cMSCs under osteogenic induction have the typical characteristic of a secretory cell; the osteogenic induction compound may induce cMSCs to differentiate to osteoblasts. There are higher expression of ALP and OCN in passage 3 cMSCs under osteogenic induction than that of the osteoblasts osteogenic induction condition. Our research suggest the cMSCs in our culture system are mainly undifferentiated osteoprogenitors and can differentiate to osteoblast under osteogenic induction.

RESUMEN

There are three key factors in tissue engineering: seeding cells, scaffold and their interaction. Although mesenchymal stem cells (MSCs) are potential seeding cells, the problem of what phase MSCs should be used is not yet solved. On the other hand, degradable porous scaffolds which have good mechanics and good biocompatibility are preferred. To choose the optimum seeding cells and the suitable ratio of beta-TCP/PLLA porous scaffold, we observed the phenotype of the male SD rat's osteoblastic MSCs and detected the amount of alkaline phosphatase, osteocalcin and type I collagen secreted by the osteoblastic rMSCs in different phase. About 10, 14 and 20 days after induction, the induced cells came into proliferative phase, matrix synthesis phase and mineralization phase, respectively. Then we chose the suitable cells and seeded them on beta-TCP/PLLA composite scaffolds with different ratios (beta-TCP/PLLA = 1:1; beta-TCP/PLLA = 1:2; and beta-TCP/PLLA = 2:1). Fluorescence microscope, scanning electron microscope and MTT assay were used to observe and to detect the biocompatibility of the scaffolds. The results indicated that all of these materials have biocompatibility to some extent. Cells can grow well on all of the scaffolds. However, scaffold beta-TCP/PLLA = 2:1 seems to be a more suitable tissue engineering scaffold on account of its minimal influence on cell growth and differentiation.

RESUMEN

To establish and observe rabbit hyperlipemia and atherosclerosis model, we combined the method of high-lipid diet and immuoreactive injure. We divided 45 New Zealand rabbit into control group and high-lipid group. According to the time (4, 8, 12 weeks) of established model, the control group and high-lipid group were divided into 3 groups respectively. The blood-lipid, the hemodynamics parameter and vascular intima were determined and observed. The results showed: (1) After feeding 4, 8, 12 weeks, TC and LDL-C of the high-lipid group in the serum increased obviously. After feeding 8 week, TG of the high-lipid group began to increase obviously. HDL-C of the high-lipid group was higher than control group, but with a descendent trend. (2) Blood viscosity of the high-lipid group increased obviously under the 0.512 S(-1) and 5.96 S(-1) at 12 week. Blood flow increased obviously at 8 and 12 week. SBP increased evidently at 8 and 12 week. Alteration of the plasma viscosity and vascular diameter were not obvious. (3) By observing with optical microscope the intima of the control group is smooth. The intima of the high-lipid group has a light incrassation at 4 week. The intima of the high-lipid group has a obvious incrassation at 8 and 12 week. By observing with through transmission electron microscopy (TEM), the lipid vacuole is under the endothelium cell. (4) By adopting the immunohistochemistry, the foam cell that derived from smooth muscle cell were determined. We concluded that the blood lipid can have a prefigurative role in atherosclerosis; blood flow and blood pressure and blood viscosity increase at low shear rate in the course of the atherosclerosis; vascular intima is incrassate and the composition of the AS plaque is variational continually.
