RESUMEN

ObjectiveTo establish a UHPLC-MS/MS quantitative method for the determination of glucuronic acid, tartaric acid, glycolic acid, malic acid, lactic acid, citric acid, DL-2-hydroxybutyric acid sodium, mandelic acid, benzilic acid, hydroxycaprylic acid, lactobionic acid, gluconic acid and N-acetylneuraminic acid in cosmetics. MethodsSamples were prepared by ultrasonic extraction, cleansed by precipitating reagent and followed by high-speed centrifugation of the extraction solution. The supernatant was filtered by 0.22 μm Millipore filter. The continued filtrate was taken for analysis. A reversed phase column, Poroshell 120 EC-C18 (2.7 μm, 4.6 mm×1 000 mm) was used with 0.1% formic acid buffer and acetonitrile as the mobile phase under the condition of gradient elution. The analytes were detected with electrospray ionization source in negative ion mode (ESI-) and multiple reactions monitoring (MRM), and quantified by external standard curve. ResultsThe method showed a good linearity of glucuronic acid, tartaric acid, malic acid, DL-2-hydroxybutyric acid sodium, benzilic acid, hydroxycaprylic acid and N-acetylneuraminic acid within the concentration range of 50.0‒2 000.0 μg·L-1 (r>0.995). The method showed a good linearity of glycolic acid, lactic acid, citric acid and mandelic acid within the concentration range of 100.0‒5 000.0 μg·L-1 (r>0.995). The method showed a good linearity of lactobionic acid and gluconic acid within the concentration range of 50.0‒5 000.0 μg·L-1 (r>0.995). The recoveries were in the range of 92.3%‒114.1%; the relative standard deviations (RSD) were in the range of 0.9%‒6.0% (n=3). The detection limits of glucuronic acid, tartaric acid, malic acid, citric acid, DL-2-hydroxybutyric acid sodium, mandelic acid, benzilic acid, hydroxycaprylic acid, lactobionic acid, gluconic acid and N-acetylneuraminic acid were 0.003% while the detection limits of glycolic acid, lactic acid and mandelic acid were 0.006%. In 10 batches of commercially available cosmetics, eight batches showed positive result. ConclusionThe UHPLC-MS/MS method is efficient, sensitive and accurate and is applicable to the determination of 13 α-hydroxy acidic components in cosmetics.

Purpose@#This study aims to elucidate the longitudinal alterations in frailty and health-related quality of life experienced by elderly patients undergoing surgical treatment for esophageal cancer. Additionally, it seeks to ascertain the impact of preoperative frailty on postoperative health-related quality of life over time. @*Methods@#131 patients were included in the prospective study. Patients' frailty and health-related quality-of-life were assessed utilizing the Tilburg and European Organization for Research and Treatment of Cancer Quality of Life Questionnaire Core 30 at preoperative, 1 week, 1 month, and 3 months, postoperatively. Statistical analyses were performed using generalized estimating equations, repeated-measures analysis of variance, and linear mixed models (LMMs). @*Results@#Out of 131 patients, 28.2% had frailty before surgery, and the prevalence of frailty consistently higher after surgery compared with baseline (67.9%, 51.9%, and 39.7%). There was no significant change in frailty scores in preoperative frail patients within 3 months following surgery (p = .496, p < .999, p < .999); whereas in preoperative non-frail patients, the frailty scores increased at 1 week (p < .001) and then decreased at 1 month (p = .014), followed by no change at 3 months. In addition, preoperative frail patients had significantly worse global quality-of-life (β = −4.24 (−8.31; −.18), p = .041), physical functioning (β = −9.87 (−14.59; −5.16), p < .001), role functioning (β = −10.04 (−15.76; −4.33), p = .001), and social functioning (β = −8.58 (−15.49; −1.68), p = .015), compared with non-frail patients. @*Conclusions@#A significant proportion of participants exhibited a high prevalence of preoperative frailty. These patients, who were preoperatively frail, exhibited a marked reduction in health-related quality-of-life, a more gradual recovery across various functional domains, and an increased symptom burden during the follow-up period. Therefore, it is crucial to meticulously identify and closely monitor patients with preoperative frailty for any changes in their postoperative physiology, role, and social functioning.

Objective:To perform acceptance test and performance assessment for Siemens Biograph Vision 600 positron-emission tomography/computed tomography(PET/CT)according to the national health industry standard WS 817-2023.Methods:Spatial resolution,sensitivity,scatter fraction,count loss and random coincidence,correction accuracy of count loss and random coincidence,time-of-flight(TOF)resolution of the PET component within the PET/CT system were tested through the measurement program(NU2-2018)of National Electrical Manufacturers Association(NEMA),which was installed inside of the equipment,in accordance with the requirement of national health industry standard WS 817-2023.The PET/CT registration accuracy was measured through Gantry_offset acquisition program that was built into the equipment.Results:The transversely and axially spatial resolutions of Biograph Vision 600 PET/CT were respectively 3.69 mm and 4.10 mm at 1 cm away from the center of visual field,and were respectively 4.26 mm and 4.89 mm at 10 cm away from the center of visual field,and were respectively 4.68 mm and 4.89 mm at 20 cm away from the center of visual field.The sensitivity of 10 cm away from center and radial of visual field were respectively 16.12 kcps/MBq and 16.00 kcps/MBq.The peak value of noise equivalent count rate(NECR)was 281.60 kcps,and the corresponding radioactivity concentration of peak value was 30.69 kBq/ml.The NECR peak value,scatter fraction and maximum value of the error of relative count rate were respectively 38.17% and 4.0%.The TOF resolution was 209.87 ps when the radioactivity concentration was 5.3 kBq/mL.The registration accuracy values of Biograph Vision 600 PET/CT were 0.347 mm,-0.226 mm and 3.659 mm at the directions of x,y and z axis.Conclusion:It is feasible to perform the acceptance test according to the WS 817-2023 standard through uses the NEMA NU2-2018 standard measurement program that is installed inside of the equipment.The performance indicators can meet requirement of standard as the current national standard GB/T 18988.1-2013 and the health industry standard WS 817-2023 that will being implemented in the test of Biograph Vision 600 PET/CT,which can pass acceptance.

RESUMEN

Radiotherapy can cause functional and morphological changes in the brain tissues of patients with primary or metastatic malignant brain tumors, leading to radiation-induced brain injury. However, the pathogenesis of radiation-induced brain injury has not yet been unanimously determined, and its research advances and treatment protocols are yet to be elucidated and improved. In this study, we explore the pathogenesis of radiation-induced brain injury from the perspective of vascular injury, inflammatory reactions, neuronal dysfunction, glial cell injury, and gut microbiota and reviewed the advances in research on its treatment and prevention. The purpose is to provide a reference and theoretical basis for the research and clinical diagnosis and treatment of radiation-induced brain injury.

ObjectiveTo reveal the differences of the related pathogenicity gene mutations between sebaceous adenocarcinoma (SC) of scalp and sebaceous adenoma (SA) of scalp on whole exome level. MethodsWhole exome sequencing was performed on a SC sample and a SA sample by Illumina Hiseq 2500 platform. Suspicious single nucleotide variation sites were selected for mutation conservation and functional analysis. SciClone was used to track subclone evolution and clonal map information was obtained for each tumor sample. The high-frequency significant gene mutations in the tumor sample were screened by MutSigCV software, and compared with the known driver genes. ResultsTwo driver genes TFDP1 and ACVR1B harboring mutations in scalp SC compared to SA were found. ConclusionsThe finding of mutation in driver genes TFDP1 and ACVR1B should be confirmed in a large cohort, which might reveal the mechanism of scalp SC development and find a therapeutic target for SC.

Objective:To investigate the effects of down-regulation of FABP5 (fatty acid binding protein 5) on radiation damage of skin cells, and explore underlying mechanism.Methods:A lentiviral vector with down-regulated FABP5 was constructed to infect human immortalized keratinocytes (HaCaT) cells, and the transfection efficiency was examined. The HaCaT cells were divided into blank control group, FABP5 down-regulation group (FABP5), radiation group (IR), and FABP5 down-regulation combined with radiation group (FABP5+ IR). After 6 MV X-ray radiation, cell proliferation viability was measured by CCK-8 assay, cell migration was detected by scratch assay, apoptosis was analyzed by flow cytometry, radiosensitivity was evaluated by cloning formation assay, and the cellular protein expressions of PARP1, γ-H2AX, AKT and p-AKT were detected by Western blot.Results:FABP5 was successfully knocked-down in both RNA level ( t=25.14, P<0.05) and protein level ( t=20.06, P<0.05). The down-regulation of FABP5 decreased the abilities of cells proliferation ( t=3.55, 5.88, 3.18, P<0.05) and migration ( t=15.44, P<0.05), but increased cell resistance to irradiation with a radiosensitization ratio of 0.782. The apoptosis rate of FABP5+ IR group was significantly lower than IR group (22.05±6.71)% vs. (9.82±1.45)%, t=3.08, P<0.05. The protein levels of PARP1 and γ-H2AX in FABP5+ IR group were also lower than those in the IR group 0.04±0.04, 0.11±0.06, 0.26±0.11, 0.22±0.07, 0.21±0.10, 0.52±0.22, 0.57±0.06, 0.43±0.02( t=2.83, 3.07, 4.50, 5.33, P<0.05), while the protein level of p-Akt in FABP5+ IR group was higher than that in IR group ( t=-16.24—3.02, P<0.05). Conclusions:Down-regulation of FABP5 inhibited cell proliferation and migration, increased radioresistance, and reduced radiation-induced apoptosis and DNA damage of skin cells probably through PI3K/AKT signaling pathway.

ObjectiveTo study the possible correlation between serum osteoprotegerin (OPG)/soluble receptor activator of the nuclear factor κB ligand (sRANKL) levels and the left ventricular diastolic dysfunction (LADD) in patients with type 2 diabetes mellitus (T2DM). MethodsTotally 68 T2DM patients and 37 healthy controls were selected. Serum OPG and sRANKL were determined by solid-phase enzyme-linked immunosorbent assay (ELISA). The left ventricular diastolic function of T2DM patients was measured by transthoracic echocardiography, where E/A < 1 were regarded as LVDD. T2DM patients were further divided into two subgroups according to E/A ratio (E/A≥1.0 and E/A<1). Spearman correlation analysis, logistic regression and ROC curves were used to assess the possible correlation between serum OPG/sRANKL and LADD in T2DM patients. ResultsCompared with the healthy controls, serum OPG level in T2DM patients was higher with statistically significant difference (P <0.01), while serum sRANKL level was lower without statistically significant difference (P =0.32). T2DM patients with E/A<1 had significantly higher OPG level and lower sRANKL level than those with E/A≥1(P <0.01) in subgroup analysis. Spearman correlation analysis showed serum OPG level was negatively correlated with E/A ratio, while sRANKL was positively related with E/A ratio. In single factor logistic regression analyses, serum OPG [OR (95% CI)=1.068 (1.031, 1.106), P<0.001] and sRANKL [OR (95% CI)=0.976 (0.959, 0.992), P=0.003] were significant correlation with LVDD in T2DM patients. ROC curve analysis showed that the sensitivity and specificity of combined OPG and sRANKL in diagnosing T2DM patients LADD were 78.13% and 88.3%, respectively (area under the curve: 0.857; 95% CI=(0.768, 0.946); P<0.001). ConclusionsThe elevated OPG and decreased sRANKL levels may be associated with LADD in T2DM patients.

Autophagy often occurs after cells are attacked by oxidative stress, where damaged structures are phagocytic and degraded into nutrients, thereby reducing oxidative damage, promoting the survival of cancer cells and reducing the therapeutic effect of photodynamic therapy (PDT). However, excessive activation of autophagy can promote cell apoptosis. In this paper, the photosensitizer pyropheophorbide-a (Ppa) was used to produce a large amount of reactive oxygen species (ROS) to achieve the effect of killing cancer cells. At the same time, icaritin (Ica), an autophagy inducer, was used to over-activate autophagy, which transformed the protection of cancer cells into the promotion of cancer cell apoptosis, so as to improve the effect of photodynamic therapy. In this study, the interaction force between Ica and Ppa was exploited to successfully construct a self-assembled nanomedicine IP with good stability and high drug load. The synthesis method is simple, through using the drug itself as a carrier, and the loading capacity (LA) of Ica and Ppa can be increased to 83.53% and 16.45% without introducing potential biosafety risks of nanocarriers. Compared with free Ppa, self-assembled nanomedicine IP showed superior performance in cellular uptake and reactive oxygen species production. In addition, the self-assembled nanomedicine IP can reverse the protective autophagy induced by PDT by activating the autophagy of tumor cells, and facilitate apoptosis and antitumor coordination, which significantly improves the antitumor activity of PDT.

Cancer is still one of the major diseases threatening human life and health. At present, how to achieve precise diagnosis and treatment of tumors is the biggest challenge in cancer treatment. Prodrugs use the tumor specificity of targeting molecules to deliver anticancer drugs to tumor sites, which can effectively improve drug bioavailability, therapeutic efficacy and safety, and are currently a hot spot in the research and development of anticancer drugs. The targeting molecules of prodrugs mainly include nucleic acid aptamers, polymers, antibodies, polypeptides, etc. Among them, polypeptides have the advantages of good biocompatibility, controllable degradation performance, high in vivo responsiveness, and simple and easy preparation methods, and are widely used. It is used to construct peptide-drug conjugates (PDC) prodrugs to achieve targeted therapy of tumors. In recent years, with the development of phage peptide library technology and peptide standard solid-phase synthesis technology, more and more targeted peptides have been discovered and effectively synthesized and modified, providing strong support for the development of PDC. This review briefly introduces the types and functions of functional peptides and linkers in PDC, and discusses the application of PDC in chemotherapy, immunotherapy and photodynamic therapy in tumor targeted diagnosis and treatment, and finally summarizes the difficulties faced by PDC drug development.

@#<b>Objective</b> To investigate the outcomes and prognostic factors of differentiated thyroid cancer (DTC) with bone metastasis. <b>Methods</b> A retrospective study was conducted on 108 DTC patients with bone metastasis who were treated in the Cancer Hospital of Chinese Academy of Medical Sciences. Kaplan-Meier survival curves were generated. Log-rank test and Cox proportional hazards model were used to screen the prognostic factors. The correlation between treatment and prognosis was analyzed. <b>Results</b> The median overall survival was 70 months. The 5-, 10-, 15-, and 20-year overall survival rates were 54.4%, 24.3%, 9.8%, and 4.3%, respectively. Univariate analysis showed improved prognosis in patients with single bone metastasis, without skeletal-relatedevents (SREs), and without cervical lymph node metastasis (<i>P </i>= 0.003-0.019). Patients who received combined treatments (<i>P</i> < 0.001) or <sup>131</sup>I treatment alone (<i>P</i> = 0.109) showed better prognosis than those without <sup>131</sup>I treatment. Multivariate analysis identified single bone metastases, SREs, and treatmentas independent prognostic factors. <b>Conclusion</b> In DTC patients with bone metastasis, good prognosis is significantly associated with single bone metastases, absence of SREs, and <sup>131</sup>I therapy in combination with other therapeutic approaches.

Objective This study aims to construct and identify the chimeric antigen receptor NK92 (CAR-NK92) cells targeting NKG2D ligand (NKG2DL) (secreting IL-15Ra-IL-15) and verify the killing activity of NKG2D CAR-NK92 cells against multiple myeloma cells. Methods The extracellular segment of NKG2D was employed to connect 4-1BB and CD3Z, as well as IL-15Ra-IL-15 sequence to obtain a CAR expression framework. The lentivirus was packaged and transduced into NK92 cells to obtain NKG2D CAR-NK92 cells. The proliferation of NKG2D CAR-NK92 cells was detected by CCK-8 assay, IL-15Ra secretion was detected by ELISA and killing efficiency was detected by lactate dehydrogenase (LDH) assay. The molecular markers of NKp30, NKp44, NKp46, the ratio of apoptotic cell population, CD107a, and the secretion level of granzyme B and perforin were detected using flow cytometry. In addition, the cytotoxic mechanism of NKG2D CAR-NK92 cells on the tumor was verified by measuring the degranulation ability. Moreover, after NKG2D antibody inhibited effector cells and histamine inhibited tumor cells, LDH assay was utilized to detect the effect on cell-killing efficiency. Finally, the multiple myeloma tumor xenograft model was constructed to verify its anti-tumor activity in vivo. Results Lentiviral transduction significantly increased NKG2D expression in NK92 cells. Compared with NK92 cells, the proliferation ability of NKG2D CAR-NK92 cells was weaker. The early apoptotic cell population of NKG2D CAR-NK92 cells was less, and NKG2D CAR-NK92 cells had stronger cytotoxicity to multiple myeloma cells. Additionally, IL-15Ra secretion could be detected in its culture supernatant. NKp44 protein expression in NKG2D CAR-NK92 cells was clearly increased, demonstrating an enhanced activation level. Inhibition test revealed that the cytotoxicity of CAR-NK92 cells to MHC-I chain-related protein A (MICA) and MICB-positive tumor cells was more dependent on the interaction between NKG2D CAR and NKG2DL. After stimulating NKG2D CAR-NK92 cells with tumor cells, granzyme B and perforin expression increased, and NK cells obviously upregulated CD107α. Furthermore, multiple myeloma tumor xenograft model revealed that the tumors of mice treated with NKG2D CAR-NK92 cells were significantly reduced, and the cell therapy did not sensibly affect the weight of the mice. Conclusion A type of CAR-NK92 cell targeting NKG2DL (secreting IL-15Ra-IL-15) is successfully constructed, indicating the effective killing of multiple myeloid cells.

Objective To determine bimatoprost, tafluprost ethyl amide, latanoprost, travoprost and tafluprost in eyelash enhancing cosmetics by establishing a LC-MS/MS method. Methods The samples were extracted with a 50% acetonitrile water solution. A salt mixture(4 g NaCl, 1 g MgSO4) was added to the solution to induce phase separation. After centrifugation and filtration, the analysis of five prostaglandin analogs was performed with an Agilent Poroshell 120 PFP-C18 (2.7 μm, 2.1 mm×100 mm) column, using 0.02% formic acid containing 5 mmol·L-1 Acetic acid amine and acetonitrile by gradient elution at a flow rate of 0.5 mL·min-1. The analytes were detected with electrospray ionization source in positive ion mode (ESI+) and multiple reaction monitoring (MRM), and quantified by external standard curve. Results The results showed that it had a good linearity in the range of locatable ambit of concentration with correlation coefficients (r) larger than 0.999. The detection limit of five prostaglandin analogs (LOD) was 0.000 2‒1.5 μg·g-1. The spiked recoveries were 93.2% to 103.5% with a relative standard deviation (RSD) of 1.2% to 3.4%. Conclusion The method is simple, rapid and highly sensitive. It is suitable for the determination of five prostaglandin analogs in eyelash enhancing cosmetics.

RESUMEN

Follicular lymphoma is an indolent malignant tumor originating from lymph nodes and lymphoid tissues, which may affect the patients' quality of survival due to the recurrence and progression. In recent years, with the deepening understand of the molecular biology and signaling pathways, many new targeted drugs for follicular lymphoma have been discovered, such as monoclonal antibodies, checkpoint inhibitors, epigenetic regulation related targeted therapies and signaling pathway inhibitors. In this review, the new progress of immunotherapy for follicular lymphoma is summarized briefly.

ObjectiveTo establish a high⁃performance liquid chromatography (HPLC) quantitative method for the determination of epicatechin (EC), (-)⁃epicatechin gallate (ECG), (-)⁃epigallocatechin (EGC), (-)⁃epigallocatechin gallate (EGCG), (-)⁃gallocatechin (GC), (-)⁃gallocatechin gallate (GCG), (-)⁃catechin gallate (CG), cianidanol (CD) and gallic acid (GA) in cosmetics. MethodsSamples were prepared by ultrasonic extraction and followed by high-speed centrifugation of the extraction solution. The supernatant was filtered by 0.45 μm Millipore filter. The continued filtrate was taken for analysis. A reversed phase column, Kromasil 100-5 C18 (5 μm, 4.6 mm×250 mm) was used with 0.05% trifluoroacetic acid buffer and methanol as mobile phase under the condition of gradient elution. Diode array detection (DAD) method was used for the determination. Qualitative and quantitative determination was conducted in 10 batches of commercially available cosmetics. ResultsThe relative standard deviations (RSD) were in the range of 0.11%-6.30% (n=3); the recoveries were in the range of 84.4%-114.7%. The method showed a good linearity within the concentration range of 0.49-105.39 mg·L-1 (r>0.995). The detection limit was 5 μg·g-1. In 10 batches of commercially available cosmetics, three batches showed positive result, which was consistent with the UV spectrum of the standard. ConclusionThis method is efficient, sensitive and accurate. It is applicable to the determination of EC, ECG, EGC, EGCG, GC, GCG, CG, CD and GA in cosmetics.

Objective: To explore the effect of down-regulation of retinol binding protein 2 (RBP2) expression on the biological characteristics of ovarian cancer cells and its mechanism. Methods: Knockdown of RBP2 and cisplatin (DDP)-resistant ovarian cancer cell line SKOV3/DDP-RBP2i was established, the negative control group and blank control group were also set. Cell counting kit 8 (CCK-8) was used to detect the cell proliferation ability, flow cytometry was used to detect cell apoptosis, scratch test and Transwell invasion test were used to detect cell migration and invasion ability, real-time fluorescent quantitative polymerase chain reaction (RT-qPCR) and western blot were used to detect the expressions of molecular markers related to epithelial-mesenchymal transition (EMT). The effect of RBP2 on the growth of ovarian cancer was verified through experiment of transplanted tumors in nude mice, and the relationships between RBP2 expression and tumor metastasis and patient prognosis were analyzed using the clinical data of ovarian cancer in TCGA database. Results: After down-regulating the expression of RBP2, the proliferation ability of SKOV3/DDP cell was significantly reduced. On the fifth day, the proliferation activities of SKOV3/DDP-RBP2i group, negative control group and blank control group were (56.67±4.16)%, (84.67±3.51) and (87.00±4.00)% respectively, with statistically significant difference (P<0.001). The apoptosis rate of SKOV3/DDP-RBP2i group was (14.19±1.50)%, higher than (8.77±0.75)% of the negative control group and (7.48±0.52)% of the blank control group (P<0.001). The number of invasive cells of SKOV3/DDP-RBP2i group was (55.20±2.39), lower than (82.60±5.18) and (80.80±7.26) of the negative control group and the blank control group, respectively (P<0.001). The scratch healing rate of SKOV3/DDP-RBP2i group was (28.47±2.72)%, lower than (50.58±4.06)% and (48.92±4.63)% of the negative control group and the blank control group, respectively (P<0.001). The mRNA and protein expressions of E-cadherin in the SKOV3/DDP-RBP2i group were higher than those in the negative control group (P=0.015, P<0.001) and the blank control group (P=0.006, P<0.001). The mRNA and protein expression of N-cadherin in SKOV3/DDP-RBP2i group were lower than those in the negative control group (P=0.012, P<0.001) and the blank control group (P=0.005, P<0.001). The mRNA and protein expressions of vimentin in SKOV3/DDP-RBP2i group were also lower than those in the negative control group (P=0.016, P=0.001) and the blank control group (P=0.011, P=0.001). Five weeks after the cells inoculated into the nude mice, the tumor volume of SKOV3/DDP-RBP2i group, negative control group and blank control group were statistically significant different. The tumor volume of SKOV3/DDP-RBP2i group was smaller than those of negative control group and blank control group (P=0.001). Bioinformatics analysis showed that the expression of RBP2 in patients with metastatic ovarian cancer was higher than that without metastasis (P=0.043), and the median overall survival of ovarian cancer patients with high RBP2 expression was 41 months, shorter than 69 months of low RBP2 expression patients (P<0.001). Conclusion: Downregulation of the expression of RBP2 in SKOV3/DDP cells can inhibit cell migration and invasion, and the mechanism may be related to the inhibition of EMT.

RESUMEN

Objective To observe the expression levels of fork head box protein N2 (FOXN2) and fork head box protein F2 (FOXF2) in normal brain tissue, low-grade glioma and high-grade glioma, and to explore the relationship between the two and the grade of glioma, and then deduce their roles in the occurrence and development of glioma, to look for possibilities target of drug therapy. Methods From January 2016 to December 2020, 36 glioma specimens were collected from the Department of Pathology, Yijishan Hospital, Wannan Medical College. The expressions of F0XN2 and F0XF2 were detected by immunohistochemistry and immunofluorescence with the double-blind method, and analyzed by SPSS18. 0 software, with a statistical significance of P<0. 05. Then Western blotting and Real-time PCR experiments were carried out on fresh normal brain tissues, low-grade glioma tissues, high-grade glioma tissues during neurosurgery in Yijishan Hospital of Wannan Medical College from January 2020 to January 2021 (normal brain tissues were all trauma patients). Results The results of immunohistochemistry and immunofluorescence showed that the higher grade of glioma, the lower expressions of FOXN2 and FOXF2 would be (P<0. 05). Western blotting showed that the expressions of FOXN2 and FOXF2 in normal brain tissue and low-grade glioma were higher than those in high-grade glioma (P<0. 05). The result of Real-time PCR showed that the expressions of FOXN2 and FOXF2 in normal brain tissue were higher than those in high-grade glioma (P<0. 01). Conclusion FOXN2 and FOXF2 are expressed in normal brain tissues, and their expression is low in glioma in a grade-dependent manner, suggesting that FOXN2 and FOXF2 are related to the grade and poor prognosis of glioma. Enhancing the expression of F0XN2 and F0XF2 ma)' provide a new idea for target therapy of glioma.

ObjectiveAn HPLC method was established for the determination of azelaic acid and potassium azeloycinate diglycinate in cosmetics. MethodsThe samples were extracted with 60 mmol·L-1 sodium hydroxide water solution-methyl alcohol. After centrifugation and filtration, the analysis of azelaic acid and potassium azeloycinate diglycinate was performed with a SVEA C8(250 mm×4.6 mm, 5 μm) column, using 15 mmol‧L-1 potassium dihydrogen phosphate solution (pH=3.0) and acetonitrile for gradient elution at a flow rate of 1.0 mL·min-1.The analytes were detected with UV detector, and quantified by external standard curve. ResultsThe results showed a good linearity in the range of 5‒1 000 μg‧mL-1 with correlation coefficients (r) larger than 0.999. The detection limit of azelaic acid and potassium azeloycinate diglycinate (LOD) was 0.020% and 0.015%, respectively. The spiked recoveries were 87.66% to 108.96% with the relative standard deviation (RSD) of 0.6% to 3.3%. ConclusionThe method is simple, rapid and highly sensitive. It is suitable for the determination of azelaic acid and potassium azeloycinate diglycinate in cosmetics.

RESUMEN

Objective To investigate the influence of PET/CT imaging conditions (acquisition time, bed overlap, reconstruction matrix, iteration times, filter kernel size, and attenuation correction) on the spatial resolution of images. Methods Two PET/CT devices, GE Discovery Elite and GE Discovery ST-16, were used to scan the elliptical column resolution model in one and two beds (list mode, acquisition time of 6 min). Images were reconstructed under the commonly used clinical reconstruction conditions (Elite: VPFX-S algorithm, ST-16: VUE Point HD algorithm) at 1-6 min/bed, different iteration times of 2-10 times, different filter kernel sizes of 2.0-10.0 mm (Elite), and different reconstruction matrices, with attenuation correction or not. The spatial resolution of reconstructed PET images was represented by the full width at half maximum (FWHM) of the line spread function. Results Under the clinical acquisition conditions, when the acquisition time was 1 min, 2 min, 3 min, 4 min, 5 min, and 6 min, the FWHMElite of spatial resolution at the center of field of view was (4.06 ± 0.08) mm, (4.05 ± 0.20) mm, (4.01 ± 0.01) mm, (4.05 ± 0.07) mm, (4.05 ± 0.03) mm, and (4.08 ± 0.06) mm, and the FWHMST-16 was (5.76 ± 0.12) mm, (5.72 ± 0.11) mm, (5.74 ± 0.09) mm, (5.78 ± 0.05) mm, (5.75 ± 0.09) mm, and (5.77 ± 0.07) mm. When the phantom was located in the center of one bed and the overlap of two beds, the line FWHMElite at the center was (4.04 ± 0.01) mm and (4.04 ± 0.01) mm, and the FWHMST-16 was (5.39 ± 0.19) mm and (5.38 ± 0.07) mm, respectively. The FWHMElite at the center was (4.07 ± 0.18) mm, (4.25 ± 0.10) mm, and (4.73 ± 0.08) mm at the matrices of 256 × 256, 192 × 192, and 128 × 128, respectively. The FWHMElite at the center was (4.65 ± 0.43) mm, (4.77 ± 0.27) mm, (4.02 ± 0.01) mm, (4.11 ± 0.04) mm, and (9.94 ± 0.01) mm at the filter kernel sizes of 2.0 mm-10.0 mm (interval of 2.0 mm), respectively. The FWHMElite at the center was (4.17 ± 0.27) mm, (4.27 ± 0.21) mm, (4.11 ± 0.05) mm, (4.18 ± 0.04) mm, and (4.12 ± 0.06) mm at 2-10 iterations (interval of 2 times), respectively. The FWHMElite at the center was (4.14 ± 0.01) mm and (4.18 ± 0.08) mm with and without attenuation correction, respectively. At the same acquisition time and bed, the spatial resolution of Elite images was improved by about 40.57% compared with that of ST-16 images. Conclusion The spatial resolution of images obtained at the matrix of 256 × 256 is higher than that of images obtained at the matrices of 192 × 192 and 128 × 128 in the same model. Elite images have the best spatial resolution at the reconstruction filter kernel size of 6.0 mm. Under the same imaging conditions, Elite images show significantly better spatial resolution compared with ST-16 images. Acquisition time, overlap of beds, iteration times, and attenuation correction have no significant effect on the spatial resolution of PET images.

Objective To study the radioactive concentration of 131I in the air of workplaces where sodium iodide [131I] oral solution was administrated for patients with differentiated thyroid cancer (DTC) in the Department of Nuclear Medicine, and to estimate the internal radiation dose to the staff. Methods Workplaces of radioiodine 131I therapy for DTC patients in the Department of Nuclear Medicine of a hospital were investigated. Air samples in 131I administration areas and treatment wards were collected respectively and were measured by low-background gamma-ray spectrometry to calculate the activity concentration of 131I in the air and to further estimate the internal radiation dose to staffs. Results The activity concentration in the 131I administration area within the first 3 h of administration was 3~187 Bq/m3. During administration and within the first 3 h of administration, the staff exposed in the administration area for 5~30 min received an internal radiation dose of 0.08~0.50 μSv and 0.00~0.04 μSv, respectively. The highest activity concentration of 131I in the air of the ward was measured on the day of administration, reaching 3091 Bq/m3. After patients were discharged, the activity concentration in the ward gradually decreased to 10~242 Bq/m3 within 48 h. Within 48 h after patients were discharged, the staff exposed in the ward for 5~30 min received an internal radiation dose of 0.01~14.11 μSv. Conclusion A high activity concentration of 131I in the air was recorded during administration for DTC patients in radioiodine 131I therapy, and thus we recommend remote instructed administration or administration through a shielded window. We also recommend that non-treatment related personnel except medical staffs should not enter the ward during patients’ hospitalization at which the activity concentration of 131I in the ward was the highest. After patients were discharged, a delayed entry into the ward is recommended to reduce the internal radiation dose.

OBJECTIVE: To analyze the distribution characteristics of surveillance results of key occupational diseases in Nanning City.METHODS: The occupational health monitoring data of key occupational diseases of Nanning City from 2015 to 2019 were collected using judgment sampling method and analyzed by descriptive epidemiology method. RESULTS: A total of 38 cases of key occupational diseases were reported in Nanning City during the past 5 years.The main diseases were occupational pneumoconiosis and occupational noise-induced deafness(ONID). The rate of occupational health examination was 40.06%, showing an increasing trend with the increase of years(P<0.01). The detection rate of occupational pneumoconiosis was higher than that of ONID(0.19% vs 0.01%, P<0.01). However, the abnormal detection rate of occupational health special examination in noise-exposed workers was higher than that in dust-exposed workers(10.98% vs 0.35%, P<0.01). The detection rate of pneumoconiosis-like changes in dust-exposed workers was the highest in private enterprises(P<0.01). The detection rate of binaural high frequency average hearing threshold ≥40 dB was highest in small and micro enterprises and private enterprises(all P<0.01). The detection rate of blood lead level >400 μg/L was 24.75%, and 98.80% of the abnormal workers were concentrated in medium-sized foreign-funded enterprises. The detection rates of abnormal leukocyte, neutrophil and platelet counts in benzene-exposed workers were 1.17%, 3.21% and 0.26% respectively. CONCLUSION: Among the key occupational disease risk factors in Nanning City, the number of workers exposed to dust and noise is relatively high, which results in serious consequences and harm. But the hazards of lead and benzene cannot be ignored. Emphasis should be placed on strengthening the supervision and management of key occupational diseases in small and medium-sized micro-enterprises and private enterprises.
