Abstract In swine and bovines, leptospirosis prevention and control is carried out via vaccination of susceptible animals using bacterins. However, the efficiency of leptospirosis vaccines has been questioned. This work aimed to investigate the potency of five leptospirosis vaccines sold commercially in Brazil, challenging the animals with one autochthonous strain of Leptospira, Canicola serovar, denoted LO4, isolated from swine. The standard protocol was followed, and renal carriers of Leptospira were identified among the surviving animals by culture and PCR. Of the five vaccines tested, only two proved effective. None of the surviving animals was positive by culture; however, one animal was positive by PCR. Three of the five vaccines sold commercially in Brazil for the immunization of swine or bovines failed the test of the efficacy to protect the vaccinated animals following challenge with an autochthonous Leptospira strain, Canicola serovar. The two vaccines provided protection against the renal carrier state in the surviving animals. The criteria used to produce leptospirosis bacterins sold commercially in Brazil must be reviewed. The industry should support researches on leptospiral vaccinology to improve the quality of the present vaccines and discover new immunogenic strains, because it is known that vaccination is one of the most important tools to increase the reproduction rates in livestock.

Leptospira interrogans serovar Canicola is one of the most important pathogenic serovars for the maintenance of urban leptospirosis. Even though it is considered highly adapted to dogs, serovar Canicola infection has already been described in other animals and even a few human cases. Here, we present the genomic characterisation of two Brazilian L. interrogans serovar Canicola strains isolated from slaughtered sows (L0-3 and L0-4) and their comparison with human strain Fiocruz LV133. It was observed that the porcine serovar Canicola strains present the genetic machinery to cause human infection and, therefore, represent a higher risk to public health. Both human and porcine serovar Canicola isolates also presented sequences with high identity to the Chinese serovar Canicola published plasmids pGui1 and pGui2. The plasmids identification in the Brazilian and Chinese serovar Canicola strains suggest that extra-chromosomal elements are one more feature of this serovar that was previously unnoticed.

Leptospira inadai is classified as a species of the Leptospira intermediate group that has been poorly studied due to its apparent insignificance to human and animal health. Nevertheless, over the last two decades the species has been described in human cases in India and in carrier animals in Ecuador. Here, we present the first identification and genomic characterisation of L. inadai serogroup Lyme isolated from captured rodent in Brazil. Even though the M34/99 strain was not pathogenic for hamsters, it was able to establish renal colonisation. The M34/99 strain presented high similarity with L. inadai serogroup Lyme human reference indicating that animal strain could also infect humans, although it does not represent high risk of severe disease. An extrachromosomal sequence was also identified in M34/99 strain and presented high identity with previously described L. inadai phage LinZ_10, suggesting that phage-like extrachromosomal sequence may be another feature of this understudied species.

Leptospira kirschneri is one of the pathogenic species of the Leptospira genus. Human and animal infection from L. kirschneri gained further attention over the last few decades. Here we present the isolation and characterisation of Brazilian L. kirschneri serogroup Pomona serovar Mozdok strain M36/05 and the comparative genomic analysis with Brazilian human strain 61H. The M36/05 strain caused pulmonary hemorrhagic lesions in the hamster model, showing high virulence. The studied genomes presented high symmetrical identity and the in silico multilocus sequence typing analysis resulted in a new allelic profile (ST101) that so far has only been associated with the Brazilian L. kirschneri serogroup Pomona serovar Mozdok strains. Considering the environmental conditions and high genomic similarity observed between strains, we suggest the existence of a Brazilian L. kirschneri serogroup Pomona serovar Mozdok lineage that could represent a high public health risk; further studies are necessary to confirm the lineage significance and distribution.

We investigated the existence of cross-protection between two anti-leptospirosis monovalent experimental bacterins produced with two strains of Leptospira serogroup Pomona: Fromm strain of serovar Kennewicky, isolated from pigs in the United States, and strain GR6 of serovar Pomona isolated from pigs in Brazil. Both were added of aluminum hydroxide as an adjuvant. Experimental bacterins were tested with the hamster potency test in order to assess protection provided against the disease and against the establishment of kidney infection. Controls were polyvalent commercial vaccine produced with Leptospira strains isolated outside Brazil, which included a representative of Pomona serovar, or Sorensen solution added of aluminum hydroxide adjuvant. The challenge was performed with cross-strains of serogroup Pomona tested in accordance with international standards established for the potency test. After 21 days of the challenge, survivors were killed to evaluate the condition of Leptospira renal carrier. Experimental bacterins protected hamsters against homologous and heterologous strains, demonstrating the existence of cross-protection. The commercial vaccine protected the hamsters challenged with both strains, but there was a high proportion of animals diagnosed as renal carriers when the challenge was performed with strain GR6, isolated from pigs in Brazil.

O objetivo do presente trabalho foi investigar a conveniência do emprego de estirpes de leptospiras autóctones isoladas no Brasil, na coleção de antígenos da microtécnica de soroaglutinação microscópica (SAM) aplicada a leptospirose. Foram amostradas por conveniência 109 propriedades e 9820 bovinos, fêmeas em idade reprodutiva, distribuídos em 85 municípios, dos Estados de Goiás, Mato Grosso, Mato Grosso do Sul, Minas Gerais, Paraná, Rio Grande do Sul, Santa Catarina e São Paulo. Dos 9820 animais examinados, 5806 (59,12%) foram reagentes na SAM para pelo menos um sorovar com a coleção de 23 sorovares de referência. Com a coleção de antígenos de referência e dez estirpes autóctones houve 6400 (65,17%) reagentes, com diferença significativa entre as proporções (p=0,001). Os sorovares mais prováveis identificados com a coleção de antígenos de referência foram Hardjo (43,03%), Shermani (20 %), Wolffi (9,96%), Grippothyphosa (5,42%) e Pomona (4,28%). Com a coleção ampliada por dez estirpes isoladas no Brasil, os sorovares mais prováveis foram Hardjo (31,00%), Guaricura-M4/84 (22,50%), Shermani (15,43%), Wolffi (4,76%), Grippothyphosa (3,71%) e Autumnalis (3,24%). O sorovar Guaricura, estirpe M4/84, isolada de bovinos e búfalos no Estado de São Paulo, despontou como um dos três sorovares mais freqüentes nos Estados de Goiás, Mato Grosso, Mato Grosso do Sul, Minas Gerais e São Paulo. A introdução de estirpes autóctones na coleção de antígenos da SAM propiciou a confirmação do diagnóstico de leptospirose em 594 animais (6,00%) classificados como não reagentes pela coleção de referência (p=0,001).


The aim of this study was to investigate the adequacy of the use of autochthonous strains of leptospires isolated in Brazil, added to antigen collection of the microscopic agglutination test (MAT) applied to the diagnosis of bovine leptospirosis. By means of non-probability sampling, 109 farms and 9,820 cattle, females at reproductive age were chosen from 85 municipalities in the states of Goiás, Mato Grosso, Mato Grosso do Sul, Minas Gerais, Paraná, Rio Grande do Sul, Santa Catarina and São Paulo. Among the 9,820 examined animals, 5,806 (59.12%) were reactants at MAT for at least one serovar using the 23 reference serovars. Employing the collection of reference serovars and the ten autochthonous strains, 6,400 (65.24%) reactants and significant difference (p=0.001) was found. The most probable serovars identified by the collection of reference antigens were Hardjo (43.03%), Shermani (20%), Wolfi (9.96%), Grippothyphosa (5.42%) and Pomona (4.28%). With the collection amplified with the ten strains isolated in Brazil, the most probable serovars were Hardjo (31%), Guaricura-M4/84 (22.50%), Shermani (15.43%), Wolffi (4.76%), Grippothyphosa (3.71%) and Autumnalis (3.24%). The serovar Guaricura, strain M4/84, isolated from bovines and buffaloes in the State of São Paulo, was ranked as one of the three most probable serovars in the states of Goiás, Mato Grosso, Mato Grosso do Sul, Minas Gerais and São Paulo. The addition of autochthonous strains to the MAT antigen collection provided the confirmation of the diagnosis of leptospirosis in 594 cattle (6%) which have been classified as non-reactants by the reference collection (p=0.001).

Bovine semen experimentally contaminated with Leptospira santarosai serovar Guaricura was submitted tothe modified EMJH medium with 5-fluorouracil (300mg/L) and nalidixic acid (20mg/L), named as ôselectivemediumõ and using the serial dilution technique, in order to evaluate the percentage of recovery of the addedmicroorganism. The selective EMJH medium was found with higher percentage of recovery of leptospirasand minor losses of samples due to contamination with opportunistic microorganisms than the non-selectiveEMJH medium: 151/376 (40.0%) of positive growth; and 38/376 (10.0%) contamination and 58/376 (15%) and129/376 (34.0%), respectively. These results were statistically significant (p<0. 0001; Fisher). Differences werefound when the frequencies of positive leptospires recovery have been compared in the serial dilution technique (10-1 to 10-4) between the selective and non-selective media at different dilution factors. At 1/10th dilution the percentages found were (0%, 0/80) and (38%, 30/80), at 1/100th dilution, (3%, 2/80) and (49%, 39/ 80) and at 1/1,000th dilution, (25%, 20/80) and (50%, 40/80), respectively. The percentage of recovery of leptospires was found to be directly proportional to the dilution used. The methodology of the serial dilution technique (setting at least three dilutions) and the use of selective EMJH medium have been found to be efficient for the isolation of leptospires from the bovine semen samples.


Sêmen bovino experimentalmente contaminado com a estirpe Leptospira santarosai Sorovar Guaricura foi utilizado para verificar a porcentagem de recuperação de leptospiras, utilizandoo meio de EMJH com 5-fluorouracil (300 mg/L) e ácido nalidíxico (20 mg/L), denominado ômeio seletivoõ, associado à técnica das diluições seriadas. O meio seletivo EMJH foi encontrado comporcentagem de recuperação mais elevada e com menos perda de amostras devido à contaminação com microorganismos oportunistas quando comparado com o meio EMJH não seletivo, que foram encontrados, respectivamente, com 151/376 (40.0%) de crescimento positivo e 38/376 (10.0%) de contaminação, 58/376 (15%) e 129/376 (34.0%). Estes resultados foram estatíticamente significantes (p<0, 0001; Fisher). Diferenças foram encontradas quando as freqüências de leptospiras recuperadas foram comparadas com a técnica de diluição seriada (10-1 a 10-4) e entre os meios seletivo e não-seletivo, em diferentes níveis de diluição. À diluição de 1/10, as porcentagens encontradas foram (0%, 0/80) e (38%, 30/80); à diluição 1/100, (3%, 2/80) e (49%, 39/80); e à diluição 1/1000, (25%, 20/80) e (50%, 40/80), respectivamente. A porcentagem de recuperação de leptospiras foi diretamente proporcional às diluições empregadas. A metodologia das diluições seriadas (estabelecendo pelo menos três diluições) e o uso de meio EMJH seletivo mostraram ser eficientes no isolamento de leptospiras presentes nas amostras de sêmen bovino.

With the aim of isolating Leptospira spp., blood serum, kidney, liver and genital tract of 137 female swine (40 sows and 97 gilts) and also urine samples from 22 sows were collected in a slaughterhouse in the State of São Paulo, from April 2003 to August 2004. Four isolates were obtained from animals that presented microagglutination test (MAT) titers > 100 for the serovar Pomona and one was obtained from an animal negative by MAT in which Leptospira was isolated from the liver and reproductive tract. The presence of leptospiral DNA was investigated by PCR, and positive results were found in kidneys of 11 females, liver of two, genital tract of two and urine of one of them. Nephrosis, interstitial multifocal nephritis, moderate to severe changing, hyalines cylinders and hemorrhagic focuses, hepatic and uterine horns congestion were histological lesions observed in higher frequency in animals positive for leptospira. The silver impregnation (Warthin Starry) confirmed the presence of spirochetes in renal tubules of four females with positive leptospira cultures from kidneys. The serogroup of the five isolates was identified as Pomona by cross agglutination with reference polyclonal antibodies. Molecular characterization of the isolates was carried out by variable-number tandem-repeats analysis. All the isolates revealed a pattern distinct from the L. interrogans Pomona type strain, but identical to a previously identified pattern from strains isolated in Argentina belonging to serovar Pomona.


Amostras de soro sanguíneo, rim, fígado e trato genital de 137 fêmeas suínas (40 matrizes e 97 marrãs) e de urina de 22 matrizes foram colhidas em abatedouro no Estado de São Paulo, no período de abril de 2003 a agosto de 2004 tendo como objetivo o isolamento de Leptospira spp. Quatro estirpes foram isoladas de animais que apresentaram títulos, no teste de soroaglutinação microscópica (SAM) > 100, para o sorovar Pomona e de um animal, não reagente na SAM, em que houve isolamento de leptospiras do fígado e aparelho reprodutor. A presença do DNA de leptospira foi investigada pela técnica da PCR e foram observados resultados positivos nos rins de 11 fêmeas, no fígado de duas, no aparelho reprodutor de duas e na urina de uma delas. Nefrose, nefrite intersticial multifocal variando de moderada a severa, cilindros hialinos e focos hemorrágicos, congestão hepática e de cornos uterinos foram lesões histológicas evidenciadas com freqüência mais alta em animais positivos para leptospira. A impregnação argêntica (Warthin Starry) confirmou a presença de espiroquetas nos túbulos renais das quatro fêmeas onde houve cultura positiva para leptospiras dos rins. O sorogrupo dos cinco isolados foi identificado como Pomona pela técnica de aglutinação cruzada com anticorpos policlonais de referência. A caracterização molecular dos isolados foi realizada pela análise do número variável de repetições em tandem (VNTR). Os mesmos revelaram um padrão distinto da estirpe padrão de L. interrogans sorovar Pomona, porém idêntico a um padrão previamente identificado em estirpes isoladas na Argentina, pertencentes ao sorovar Pomona.

Inactivation of leptospires in pools of semen from three Holstein Friesian bulls, collected in an artificial vagina, was investigated. Spermatic concentration was adjusted in egg yolk citrate extender, submitted to the following treatments: A (control; without antibiotics); B (penicillin, 1,000 UI/mL - streptomycin, 1,000 icg/mL); C (amoxicillin, 1,000 icg/mL); D (ceptiofur sodium, 1,000 icg/mL); E (amoxicillin 1,000 icg/mL - ceptiofur sodium 1,000 icg/mL). Leptospires (2.0 x 10(6) leptospires/mL) were added into the diluted semen. Recovery of leptospires was obtained in modified EMJH semi-solid medium with and without antibiotics. The antibiotics in the concentrations used did not affect means of percentage of progressive motility and individual progressive motility of spermatozoids. Penicillin-streptomycin presented the best results in leptospire inactivation (97.1 percent). Amoxicillin, ceptiofur sodium and their combination at the concentrations studied presented poor results: 59.29 percent; 32.5 percent and 60.36 percent of inactivation, being less effective in leptospire inactivation than penicillin-streptomycin.
