BACKGROUND:Intervertebral disc degeneration is the basis of spinal degenerative diseases;however, there is no effective treatment. OBJECTIVE:To investigate whether sinomenine can inhibit interleukin-1β-induced apoptosis in nucleus pulposus cells and its molecular mechanism. METHODS:Rat nucleus pulposus cells were cultured in vitro by trypsin combined with type II collagenase digestion, and the cell growth curve was plotted. An appropriate sinomenine concentration was determined using the cell counting kit-8 kit.Nucleus pulposus cells were divided into control group, sinomenine group,interleukin-1β group,sinomenine+interleukin-1β group, zinc protoporphyrin group, zinc protoporphyrin+sinomenine group, zinc protoporphyrin+interleukin-1β group, and sinomenine+zinc protoporphyrin+interleukin-1β group.Proliferative activity, reactive oxygen species content, apoptosis rate, and heme oxygenase-1 expression in nucleus pulposus cells were detected. RESULTS AND CONCLUSION:The rat nucleus pulposus cells cultured in vitro were polygonal, triangular, and short wedge-shaped,and the cell growth showed an"S"curve. The cells grew slowly in the first 3 days of culture, rapidly in 4-6 days, and slowly again in 7-8 days. The cells then entered the"platform stage"where the number of cells no longer increased. The proliferative activity of myeloid cells showed no significant changes when the concentration of sinomenine was≤80 μmol/L(P>0.05).Interleukin-1β significantly reduced the proliferative activity of nucleus pulposus cells, increased the content of reactive oxygen species and led to apoptosis(P<0.01).Sinomenine intervention not only promoted heme oxygenase-1 expression(P<0.05)but also inhibited interleukin-1β-induced decrease in proliferative activity and increase in reactive oxygen species content and apoptosis rate in nucleus pulposus cells(P<0.05).These effects could be reversed by zinc protoporphyrin(P<0.01).

BACKGROUND:Intervertebral disc degeneration is caused by damage and degeneration of the nucleus pulposus and annulus fibrosus tissues inside the intervertebral disc, resulting in structural and functional changes of the intervertebral disc. However, there is yet no effective drug treatment for intervertebral disc degeneration. OBJECTIVE:To investigate the inhibitory effect of syringin on intervertebral disc degeneration. METHODS:A total of 10 male Sprague-Dawley rats were selected, and the coccygeal intervertebral disc(Co4/Co5)of each rat was set as model group,Co5/Co6 intervertebral disc as syringin group, and Co6/Co7 intervertebral disc as control group. The control group did not receive any treatment. In the model group and syringin group,a miniature puncture needle was used to puncture the annulus fibrosus to establish an intervertebral disc degeneration model.Immediately after modeling,2.5 μL of normal saline and syringin solution(5 μmol/L)were given in the model and syringin groups, respectively.Four weeks after injection, the samples were taken. The degree of intervertebral disc degeneration in rats was observed by hematoxylin-eosin and safranine O-fast green staining. The expressions of type Ⅱ collagen, aggrecan and matrix metalloproteinases 3 and 13 in intervertebral disc tissue were analyzed by immunohistochemical staining. RESULTS AND CONCLUSION:Hematoxylin-eosin staining showed that in the model group, the height of intervertebral disc decreased, the cartilage endplate became thinner and cracked, the fibrous ring structure was disordered and cracked, and the nucleus pulposus disappeared;in the syringin group, the height of intervertebral disc was normal or slightly lower than that in the control group, the degree of cartilage endplate degeneration was lighter than that in the model group, the fiber circle permutation was relatively regular with no cracks, and the nucleus pulposus was partially shrunk.Safranine O-fast green staining showed that in the model group, the cartilage endplate of the intervertebral disc was defective and the calcified layer of cartilage became thinner, showing obvious degeneration. The structure and morphology of intervertebral disc cartilage endplate in the syringin group recovered to some extent.Immunohistochemical staining showed that, compared with the control group, the expressions of type Ⅱ collagen and aggrecan in the intervertebral disc cartilage were decreased in the model group(P<0.000 1),while the expressions of matrix metalloproteinases 3 and 13 increased(P<0.000 1).Compared with the model group, the expressions of type Ⅱ collagen and aggrecan in the intervertebral disc cartilage tissue were increased in the syringin group(P<0.001,P<0.000 1),while the expressions of matrix metalloproteinases 3 and 13 decreased(P<0.001,P<0.000 1).These results showed that syringin could improve the structure and function of intervertebral disc by inhibiting the expression of matrix metalloproteinases 3 and 13 and increasing the expression of type Ⅱ collagen and aggrecan, thus preventing and slowing down the procession of intervertebral disc degeneration.

RÉSUMÉ

Defined as a common gynecologic en-docrine disease, endometriosis is characterized by the presence of endometrium outside of the uterus cavity, affecting about 10%to 15%of reproductive-aged women worldwide. Moreover,Asian women tend to have a nine-fold increased risk of endome-triosis compared with their white counterparts in Europe and the United States.Endometriosis not only causes chronic pelvic pain, dysmenorrhea, painful sex and other pain symptoms, but also leads to infertility, pushing the patients into a state of fatigue, high sensitivity and anxiety, thus reduc-ing their quality of life.As the aging of Chinese pop-ulation intensifies and the women's childbearing age delays,it is highly urgent to find solutions to those gynecological endocrine diseases such as en-dometriosis,which hamper women's fertility. In re-cent years, studies have shown that patients with endometriosis have abnormal serum lipid metabo-lism and an increased risk of cardiovascular diseas-es.At the same time,as a type of cholesterol-lower-ing drug, statins can inhibit the progression of en-dometriosis at the cellular and animal levels. Also,a few clinical studies have exhibited that statins ex-ert a positive effect on the treatment of endometri-osis.Therefore,this article aims to review the re-searches regarding the relationship between endo-metriosis and the cardiovascular disease,as well as the research progress of statin therapy on endome-triosis based on the level of cells, animals and hu-man beings.

Objective:To understand the pathogen distributions of community-acquired pneumonia (CAP) in children, and to provide evidence for clinical diagnosis and treatment. Methods:The hospitalized children with CAP in Shanghai General Hospital, Shanghai Jiao Tong University School of Medicine from January to December 2022 were selected as the research subjects. They were divided into infant group (28 d to less than one year), toddler group (one year to less than three years), preschool age group (three years to less than six years), and school age group (not less than six years) by age. According to the onset season, they were divided into spring group (February to April), summer group (May to July), autumn group (August to October), and winter group (January, November to December). Deep airway sputum samples were collected from all patients for bacterial culture identification. Respiratory viruses (influenza A virus (IVA), influenza B virus (IVB), respiratory syncytial virus (RSV), adenovirus, parainfluenza virus type 1 (PIV1), parainfluenza virus type 2 (PIV2), parainfluenza virus type 3 (PIV3)) were detected using direct immunofluorescence assay. Mycoplasma pneumoniae (MP) DNA was detected using fluorescent quantitative polymerase chain reaction, and particle agglutination was used to detect serum MP antibodies. Statistical analysis was performed using the chi-square test. Results:Among the 397 cases of CAP in children, pathogens were detected in 269 cases, with a positivity rate of 67.8%. A total of 309 pathogens were identified, including 204 strains of MP (66.0%), 60 strains of bacteria (19.4%), 42 strains of viruses (13.6%), and three strains of fungi (1.0%). Staphylococcus aureus (19 strains), Haemophilus influenzae (15 strains) and Streptococcus pneumoniae (five strains) were the predominant bacteria, while RSV (19 strains) and PIV3 (nine strains) were the main viruses. The distribution rates of MP, bacteria, and viruses showed statistically significant differences among different age groups ( χ2=99.82, 24.71 and 17.40, respectively, all P<0.05). MP infection was mainly observed in the preschool age group and school age group, and bacterial infection predominantly occurred in the infant group, and viral infection was most common in the toddler group. Among virus infected patients, RSV was detected in the toddler group and the preschool age group, while three cases of PIV3 cases were found in children over five years old. The distribution differences of MP, bacterial and viral infections between different seasons were statistically significant ( χ2=141.65, 20.44 and 31.87, respectively, all P<0.001), with a higher prevalence in winter. RSV infections demonstrated a clear seasonal trend, with 16 cases of RSV infections occurring in winter and spring. Conclusions:MP is the most frequently detected pathogen in children with CAP. Bacterial infection is the most common pathogen in infants with CAP. RSV is the most common viral pathogen, with infections concentrated in the toddler group and the preschool age group, and prevalence in winter and spring. Attention should be paid to PIV3 pneumonia in children over five years old. Rational drug use should be based on the pathogen spectrum characteristics of different seasons and age groups before selecting empirical treatment combinations.

Background Cold environments reduce work efficiency and increase the risk of frostbite. Objective To investigate the effects on cold protection and manual performance of rescue gloves in cold environments, and to preliminarily verify the feasibility of related quantitative characterization methods. Methods An experiment was conducted in a large and a small artificial climate chamber. 12 male right-handed university students were recruited. The experiment was divided into two parts, with 0-10 min as the baseline phase and 10-50 min as the test phase. Subjects sat quietly in a 16 ℃ large chamber, and hands were exposed to a small chamber at 0 ℃ either barely (control group) or with rescue gloves (experimental group). Skin temperatures of the five fingers (thumb, index finger, middle finger, ring finger, and little finger), as well as the dorsal and palm of the hands were measured. In addition, grip strength, manual dexterity, maximum finger flexion, and tactile sensitivity of the hands were measured as the indicators of manual performance. Results In the control group, cold-induced vasodilation occurred when the skin temperature of the five fingers were lowered to about 10 ℃. The maximum rate of change in skin temperature of the control group was in the following order: thumb>index finger>middle finger>little finger>ring finger>dorsal>palm, 165.7 ℃·h−1 in the thumb, 80.0 and 89.9 ℃·h−1 in the palm and dorsal respectively. In addition, the minimum skin temperature of the thumb was 6.6 ℃. Compared with the control group: the rate of change of skin temperature in the experimentalgroup decreased by about 50%; the grip strength decreased by 15% after wearing the rescue gloves (P<0.05); no differences in manual dexterity, maximum finger flexion, and tactile sensitivity were statistically significant (P>0.05). Conclusion This study proved that the rescue gloves improved the thermal comfort of hands while better balancing the effect of cold exposure on manual performance. The quantitative characterization methods can evaluate the protective performance and manual performance of the rescue gloves, which provides a reliable theoretical foundation and reference basis for further experiments.

METHODS Human cervical cancer cisplatin-resistant cells HeLa/DDP were divided into control group, cisplatin group (10 μmol/L cisplatin), cisplatin+ligustilide low-, medium- and high-concentration groups (10 μmol/L cisplatin+25, 50, 100 μmol/L ligustilide). The proliferation, apoptosis, migration and invasion of HeLa/DDP cells were all detected in each group. The mRNA expressions of YAP and transcriptional coactivator with PDZ binding motif (TAZ) as well as the protein expressions of YAP, TAZ, matrix metalloproteinase 2 (MMP2), Ki67, cleaved-caspase-3 and caspase-3 were determined in HeLa/DDP cells. RESULTS Compared with control group, the inhibitory rate, apoptotic rate and cleaved- caspase-3/caspase-3 of cisplatin group were increased significantly; scratch healing rate, the number of invasive cells, the mRNA expressions of YAP and TAZ, and the protein expressions of YAP, TAZ, MMP2 and Ki67 were decreased significantly in cisplatin group (P<0.05). Compared with cisplatin group, the inhibitory rate of cell proliferation, apoptotic rate and cleaved-caspase-3/ caspase-3 were further increased in cisplatin+ligustilide low-, medium- and high-concentration groups, while scratch healing rate, the number of invasive cells, the mRNA expressions of YAP and TAZ, and the protein expressions of YAP, TAZ, MMP2 and Ki67 were further decreased, in a dose-dependent manner (P<0.05). CONCLUSIONS Ligustilide can increase the sensitivity of drug-resistant cervical cancer cells to cisplatin by inhibiting Hippo-YAP signaling pathway.

RÉSUMÉ

Currently, the management of domestic laboratory developed test (LDT) is still in the exploration stage, and new plans and ideas need to be investigated that are suitable for our national development. By analyzing encountered issues, combining the actual needs of the government, healthcare reform, and patients, this article proposes that "promoting healthy development of LDT projects and focusing on the organic combination of patients′ interest protection and innovation support" is an important direction for LDT management, around which the new idea for LDT project management is proposed, that is, LDT management model targeting product registration. The core of this management model is to target product registration and to ensure low service fees for patients and continuous real-time data monitoring. Benefit and risk analysis demonstrate that this management model can to a larger extent balance the realistic demands of patients, medical institutions, in vitro diagnostics companies, and government management departments, which benefits promoting the perfection and development of LDT projects in our country.

RÉSUMÉ

【Objective】 To analyze the association between preschool children′s emotional regulation ability and parents′ participation in accompanying picture book reading, in order to provide theoretical reference for improving children′s emotional regulation abilility. 【Methods】 From September to November 2022, preschool children aged 3 - 6 years from Guangfeng County, Yukan County, Wannian County, Yanshan County and Wuyuan County were selected by random cluster stratified sampling method. Mann-Whitney U test or Kruskal-Wallis H test were used to analyze the correlation of different levels of children′s emotional control ability with different demographic characteristics as well as parents′ participation in children′s picture book reading. Multivariate Logistic regression model was used to analyze the correlation between parents′ involvement in picture book reading and children′s emotional control ability. 【Results】 Finally 848 valid questionnaires were collected. The average age of children was (4.33±0.95)years old, with 432 boys and 416 girls. Multivariate Logistic regression analysis showed that the temperament control ability(>P75) of preschool children was positively correlated with mother being the main accompanying caregiver (OR=4.17, 95%CI: 2.13 - 8.17), and negatively correlated with father being the main accompanying caregiver (OR=0.38, 95%CI: 0.21 - 0.70) or both parents being the main accompanying caregivers (OR=0.14, 95%CI: 0.07 - 0.28)(P<0.05). The social confidence (father as the main caregiver OR=3.53, 95%CI:1.62 - 7.66; mother as the main caregiver OR=3.52, 95%CI: 1.66 - 7.45) and anxiety control abilities(father as the main caregiver OR=4.32, 95%CI: 1.85 - 10.09; mother as the main caregiver OR=3.39, 95%CI: 1.49 - 7.73) of preschool children were positively correlated with the type of accompanying caregiver(P<0.05). Additionally, the anxiety control ability (>P75) of preschool children was negatively correlated with father′s accompanying frequency (sometimes OR=0.09, 95%CI: 0.03 - 0.28; often OR=0.19, 95%CI: 0.06 - 0.60) and mother′s accompanying duration (<1h/d OR=0.33, 95%CI: 0.12 - 0.91) (P<0.05). 【Conclusions】 The frequency and duration of accompanying reading by father or mother are correlated with the emotional regulation ability of preschool children. Parents can moderately participate in picture book reading to promote the development of preschool children′s emotional control ability.
