ObjectiveTo investigate the impact of varying concentrations of baicalein on the proliferation and biological responses of MC3T3-E1 cells, as well as the antibacterial efficacy of baicalein against prevalent oral bacteria, and to elucidate the underlying mechanisms. MethodsMC3T3-E1 cells were exposed to different concentrations of baicalein (0, 6, 12, 18, and 24 μmol/L) and cell viability was determined by using the CCK-8 assay. Alkaline phosphatase (ALP) activity of MC3T3-E1 cells following osteogenic induction was assessed. RT-PCR was used to examine the expression of RunX2, BMP2, and Osterix. After 24 hours of treatment, the antibacterial potential of baicalein against Escherichia coli, Staphylococcus Aureus and Streptococcus Sanguis was evaluated by using the K-B paper disk method. ResultsBaicalein exhibited a modest reduction in proliferation of MC3T3-E1 cells but without affecting their sustained proliferation. Baicalein at a concentration of 18 μmol/L enhanced ALP activity of MC3T3-E1 cells, upregulated BMP2 and Osterix expression, downregulated RunX2 expression, significantly inhibited the proliferation of Staphylococcus Aureus and Streptococcus Sanguis (P < 0.05). ConclusionsBaicalein at an optimal concentration (18 μmol/L) demonstrated a promotional effect on the osteogenic differentiation of MC3T3-E1 cells and effectively suppressed the proliferation of common oral bacteria, including Staphylococcus Aureus and Streptococcus Sanguis.

【Objective】 This study endeavors to introduce the statistical process control (SPC) method to analyze the quality control index concerning red blood cells in additive solution with leukocytes reduced, with the aspiration to advance the effective utilization of blood quality control data, thereby providing empirical foundations for the continual enhancement of blood quality. 【Methods】 Between 2020 and 2022, test data pertaining to the quality control index of red blood cells in additive solution with leukocytes reduced were amassed from six blood stations in Chongqing area. Utilizing Minitab software, the SPC analysis was carried out, p-control charts were delineated, the non-conformance rates of each quality control index along with their 95% confidence intervals were computed, as well as the Process Capability Index (Z value). 【Results】 In accordance with the Whole Blood and Blood Components Quality Requirements, the appraisal of the quality control indexes for red blood cells in additive solution with leukocytes reduced manifested a conformity rate of 100% for appearance, end-of-storage hemolysis rate and sterility test. Nonetheless, the conformity rates for volume, hemoglobin, hematocrit and residual leukocytes did not attain 100%, albeit all were ≥75%. Through the employment of binomial distribution-based p-control charts, the controlled state of the production process was discerned. Although the overarching conformity rate satisfied the national standard stipulations, it was discerned that there were out-of-control points concerning volume, hemoglobin, hematocrit, and residual leukocytes across different institutions, exhibiting palpable trends. The non-conformance rates of all quality control indexes were less than 25%, yet at a 95% confidence level, the residual leukocyte counts from institutions B, C, E, and F did not adhere to the stipulations (exceeding 25%). By architecting the ability evaluation index Z value for count data process capability analysis, it was unveiled that the volume of institution E, the hematocrit of institutions B, C, and F, and the residual leukocytes Z values of all six blood collection and supply institutions were below 2, hinting at avenues for amelioration. 【Conclusion】 The SPC method anchored in binomial distribution exhibits substantial application merit in blood component quality management, facilitating real-time surveillance of blood collection, preparation, and storage procedures.

Objective: To explore the key deubiquitinating enzymes that maintain the stemness of liver cancer stem cells and provide new ideas for targeted liver cancer therapy. Methods: The high-throughput CRISPR screening technology was used to screen the deubiquitinating enzymes that maintain the stemness of liver cancer stem cells. RT-qPCR and Western blot were used to analyze gene expression levels. Stemness of liver cancer cells was detected by spheroid-formation and soft agar colony formation assays. Tumor growth in nude mice was detected by subcutaneous tumor-bearing experiments. Bioinformatics and clinical samples were examined for the clinical significance of target genes. Results: MINDY1 was highly expressed in liver cancer stem cells. The expression of stem markers, the self-renewal ability of cells, and the growth of transplanted tumors were significantly reduced and inhibited after knocking out MINDY1, and its mechanism of action may be related to the regulation of the Wnt signaling pathway. The expression level of MINDY1 was higher in liver cancer tissues than that in adjacent tumors, which was closely related to tumor progression, and its high expression was an independent risk factor for a poor prognosis of liver cancer. Conclusion: The deubiquitinating enzyme MINDY1 promotes stemness in liver cancer cells and is one of the independent predictors of poor prognosis in liver cancer.

ObjectiveTo explore the effect of Gelsemium elegans combined with Mussaenda pubescens on efflux transporter breast cancer resistance protein (BCRP) and cytochrome P450 3A11 (CYP3A11) and their attenuation mechanism, and to investigate whether the nuclear receptors were involved in such regulation by intervening it with nuclear receptor activators. MethodC57BL/6 mice were divided into the blank group, G. elegans (GE, 0.25 g·kg-1)group, GE + M. pubescens (MP) (0.25 g·kg-1+10 g·kg-1) group, GE + pregnane X receptor (PXR) activator (rifampicin)(GE + Rif,0.25 g·kg-1+50 mg·kg-1) group, GE + MP + Rif (0.25 g·kg-1+10 g·kg-1+50 mg·kg-1) group, GE + constitutive androstane receptor (CAR) activator (1,4-Bis [2-(3,5-Dichloropyridyloxy)] benzene, TCPOBOP)(GE + TCP, 0.25 g·kg-1+0.5 mg·kg-1) group, and GE + MP + TCP (0.25 g·kg-1+10 g·kg-1+0.5 mg·kg-1) group. The medication lasted for 14 successive days. One hour after the last administration, the mice were sacrificed by cervical dislocation and the liver tissue was harvested. The left liver tissue was stained with hematoxylin- eosin (HE) for observing the pathological changes. The right liver tissue was used for BCRP and CYP3A11 mRNA and protein expression detection by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot. ResultThe survival rates of mice in the GE + Rif group, GE group, and GE + MP group were 25% (the lowest), 40%, and 80%, respectively, and no death was observed in the other groups. Compared with the obvious lesions in the liver cells of the GE group, the pathological changes in liver cells of the GE + MP group were alleviated, while those in the GE + Rif group were worsened. Compared with the GE + Rif group, the GE + MP + Rif group exhibited relieved pathological changes in liver cells. Both the GE + TCP group and the GE + MP + TCP group showed mild liver lesions. The comparison with the GE + MP group revealed that the pathological changes in the GE + MP + TCP group were slightly relieved. Compared with the blank group, the expression of BCRP protein and mRNA in GE group were significantly decreased (P<0.05,P<0.01).The expression of CYP3A11 protein in GE group were significantly decreased (P<0.01). Compared with the GE group, the GE + MP group displayed remarkably up-regulated BCRP protein and mRNA expression (P<0.05,P<0.01) and CYP3A11 protein expression (P<0.05), but slightly up-regulated CYP3A11 mRNA expression. Compared with the GE group, the GE + Rif group exhibited down-regulated BCRP protein expression (P < 0.05). The protein and mRNA expression levels of BCRP were lower in the GE + MP + Rif group than in the GE + MP group (P<0.05,P<0.01). The PXR activator rifampicin regulated BCRP before and after the combination of G. elegans with M. pubescens. The CYP3A11 protein and mRNA expression levels in the GE + TCP group were higher than those in the GE group (P<0.05,P<0.01). Compared with the GE + MP group, the GE + MP + TCP group showed up-regulated CYP3A11 protein and mRNA expression (P<0.05,P<0.01). CAR activator TCPOBOP also had a regulatory effect on CYP3A11 before and after the compatibility of G. elegans with M. pubescens. ConclusionThe attenuated toxin after the combination of G. elegans with M. pubescens is closely related to the efflux transporter BCRP and the drug-metabolizing enzyme CYP3A11.

Objective:To verify the clinical value of the good outcome following attempted resuscitation (GO-FAR) score in predicting the neurological status of patients with in-hospital cardiac arrest (IHCA) in the Chinese population. Methods:The clinical data of patients with IHCA who were admitted to the Zigong Fourth People's Hospital from January 1 to December 31, 2020 were retrospectively analyzed. Used Glasgow-Pittsburgh cerebral performance category (CPC) score 1 point as the end point, the subjects were divided into 4 groups according to the score: ≤ 0 group, 1-8 group, 9-20 group and ≥ 21 group. Taken the group which GO-FAR score ≤ 0 as the reference group, the odds ratio ( OR) of the other three groups compared with this group was calculated. The receiver operator characteristic curve (ROC curve) was performed to evaluate the predictive value of the GO-FAR score in favorable neurological outcome. A calibration curve was drawn for the Hosmer-Lemeshow test to analyze the degree of calibration of the GO-FAR score for predicting good neurological outcome. Results:A total of 230 IHCA patients were enrolled in the study, including 130 males, aged 74 (65, 81) years old, and 23 case (10.0%) had good neurological prognosis. There were statistically significant differences in GO-FAR-related variables, including age, a normal neurological function on admitted, acute stroke, metastatic cancer, septicemia, medical noncardiac admission, hepatic insufficiency, hypotension, renal insufficiency or dialysis, respiratory insufficiency, pneumonia, etc (all P < 0.05). Taken the GO-FAR score ≤ 0 group as the reference group, the OR values of good neurological prognosis in the GO-FAR score 1-8 group were 0.54 [95% confidence interval (95% CI) was 0.17-1.53, P = 0.250], 9-20 group were 0.17 (95% CI was 0.02-0.67, P = 0.009) and ≥ 21 group were 0.25 (95% CI was 0.05-0.85, P = 0.025). The area under the ROC curve (AUC) of the GO-FAR score for predicting favorable neurological outcome in IHCA patients was 0.653 (95% CI was 0.529-0.777, P = 0.015) and there was no significant difference in Hosmer-Lemeshow test ( P = 0.311). All these suggested that there was no significant difference between the predicted value and the actual value. Conclusions:GO-FAR score can be applied to predict neurological prognosis of IHCA patients in Chinese population. It can help clinicians to predict the prognosis of cardio-pulmonary resuscitation (CPR) and propose critical recommendations in treatment for these patients or their families.

Aim To explore the role of the silent information regulator (silent information regulation 1, SIRTl)-mediated apoptotic pathway in the protection of sevoflurane postconditioning in hippocampal neuronal injury of mice induced by hemorrhagic shock resuscitation (HSR). Methods A mouse model of HSR was established. The male C57BL/6J mice were randomly divided into; sham operation group (Sham group), HSR group (Shock group), sevoflurane treatment group (Sevo group), sevoflurane combined with SIRTl specific inhibitor treatment group (EX527 + Sevo group) and EX527 treatment group (EX527 group). The volume of cerebral infarction was detected by TTC staining method. The changes of hippocampal nerve cells in each group of mice were detected by TUNEL staining method. The learning and memory abilities were detected by Morris water maze test. The expression of SIRTl and apoptosis-related protein levels were delected by Western blot analysis. Results The latency of the model mice reaching the platform in Morris water maze test was prolonged, while the movement distance in the target quadrant was similarly reduced. Besides, the cerebral infarction volume remarkably increased. The number of TUNEL staining positive cells increased, and the expression of SIRTl and Bel-2 decreased while the pro-apoptosis protein Bax, and cleaved-caspase 3 expression level increased; sevoflurane treatment improved nerve injury in HSR. After combined treatment with sevoflurane and SIRTl inhibitor EX527, the protective effect of sevoflurane was attenuated on nerve injury in HSR. Conclusion Sevoflurane may play a protective role against hippocampal neuronal injury caused by HSR mediated by SIRTl apoptosis-related pathway.

Résumé

OBJECTIVE@#To investigate the efficacy, survival and adverse effects of non-transplanted multiple myeloma (MM) patients treated with bortezomib maintenance.@*METHODS@#A total of 25 newly diagnosed/relapsed non-transplanted MM patients treated in West District of Beijing Chaoyang Hospital from June 2004 to November 2015 were analyzed retrospectively. All patients received PD regimen (bortezomib and dexamethasone), including bortezomib at a dose of 1.3 mg/m@*RESULTS@#Till November 1, 2017, 5 patients achieved stringent complete response (sCR), 8 patients achieved complete response (CR), 7 patients achieved very good partial response (VGPR), 4 patients achieved partial reponse (PR), while 1 patient achieved stable disease (SD). After maintenance therapy, 21 patients maintained the efficacy above PR, of which 1 patient was improved from CR to sCR; 4 patients adjusted chemotherapy after disease progressed. Median maintenance therapy was 9 cycles (range from 6 to 31), and the median maintenance time was 27 months (range from 18 to 97). Median follow-up time was 73 months (range from 25 to 171). Median progress-free survival (PFS) time was 30 months (range from 9 to 105) and overall survival (OS) time was 57 months (range from 27 to 160). Till November 1, 2019, 3-year survival rate was 84% (21/25), and 5-year survival rate was 72% (13/18). The most common adverse events were transient leukopenia, thrombocytopenia and peripheral neuropathy, which the patients could tolerate after the prevention and treatment.@*CONCLUSION@#Bortezomib-based maintenance therapy for non-transplanted MM patients can be an option in consideration of its safety and efficacy.

Résumé

OBJECTIVE@#To explore the regulation effect of myeloid leukemia No.1 Chinese herb medicine prescription combined with chemotherapy on Th17 cells in bone marrow fluid of AML patients, so as to provide guidance for improving AML treatment effect and patients' long-term survival.@*METHODS@#Seventy patients with AML who were hospitalized in Department of Hematology, Wuwei People's Hospital from April 2017 to August 2019 were selected and enrolled in AML group, 25 healthy volunteers were selected and enrolled in control group; then according to therapeutic regimen, AML patients were divided into 2 groups: combined therapy group (myeloid leukemia NO.1 Chinese herb medicine prescription combined with chemotherapy) and non-combined therapy group (chemotherapy alone). Flow cytometry was used to detect the ratio of CD3@*RESULTS@#The ratio of CD3@*CONCLUSION@#Th17 cells expression in bone marrow of newly diagnoses and relapsed AML patients significantly increase, and decrease significantly after treatment. Myeloid leukemia No.1 Chinese herb prescription combined with chemotherapy can significantly increase the CR rate and reduce the RL rate for AML.

Résumé

OBJECTIVE@#To detect the level of vascular endothelial growth factor (VEGF) in bone marrow of patients with non-M3 acute leukemia (AL), and estimate its relationship with prognosis.@*METHODS@#From January 2016 to December 2019, 114 patients with AL in department of Hematology, Wuwei People's Hospital were selected as study group, and 25 healthy volunteers were enrolled as control group. The concentration of VEGF in bone marrow was detected by ELISA. The patients were divided into high and low concentration group according to the level of VEGF. The overall survival (OS) and event-free survival (EFS) were compared among different groups.@*RESULTS@#The level of VEGF in patients with AL was significantly higher than that in the control group. The median OS and EFS in the low concentration group was 34.5 and 32 months, respectively, while, in the high concentration group was 30 and 26 months, respectively. The differences between the two groups were statistically significant (P=0.010). There were significant differences in OS rate (P=0.035) and EFS rate (P=0.026) between low and high concentration group. Multivariate analysis showed that high VEGF concentration was an independent risk factor affecting OS (HR=2.619, 95%CI 1.070-6.406, P=0.035) and EFS (HR=2.221, 95%CI 1.074-4.552, P=0.031) in AL patients.@*CONCLUSION@#VEGF highly expresses in the bone marrow of patients with AL at initial diagnosis and relapse, and shows adverse effects on the prognosis.
