Hypopharyngeal squamous cell carcinoma has a high degree of malignancy, hidden location, atypical early symptoms, most patients have reached advanced stage at the time of treatment. Surgical treatment will cause patients to lose laryngeal function and poor postoperative quality of life. The combined application of non-operative therapies such as radiotherapy and chemotherapy can improve the larynx preservation rate of patients and obtain a survival rate similar to that of surgical treatment. Targeted therapy has achieved better results in the treatment of hypopharyngeal squamous cell carcinoma, and immunotherapy has also made great progress in the treatment of head and neck squamous cell carcinoma, which brings new hope for patients with hypopharyngeal squamous cell carcinoma.

3D printing refers to the approach in manufacturing the three-dimensional physical objects by adding materials layer by layer.It features the advantages of quick styling, high flexibility and cost saving.The medical industry is extensively regarded as one of the most important application sectors for 3D printing.In recent years, due to the development of technical and the enhanced support from the nation in the 3D related research, the application of 3D printing technology in the ophthalmology has been increasing.This review made the overview analysis on the 3D printing technology application and possible prospects in the aspects of medical education, foundation research, surgical assistance and individualized treatment of ophthalmology, made a brief introduction on the 3D bioprinting technology attracting extensive attention recently.

OBJECTIVES: The aim of this study was to investigate the feasibility and safety of percutaneous ¹²⁵I seed permanent implantation for advanced hypopharyngeal carcinoma from toxicity, tumor response, and short-term outcome. METHODS: ¹²⁵I seeds implant procedures were performed under computed tomography for 34 patients with advanced hypopharyngeal carcinoma. We observed the local control rate, overall survival, and acute or late toxicity rate. RESULTS: In the 34 patients (stage III, n=6; stage IV, n=28), the sites of origin were pyriform sinus (n=29) and postcricoid area (n=5). All patients also received one to four cycles of chemotherapy after seed implantation. The post-plan showed that the actuarial D90 of ¹²⁵I seeds ranged from 90 to 158 Gy (median, 127 Gy). The mean follow-up was 12.3 months (range, 3.4 to 43.2 months). The local control was 2.1–31.0 months with a median of 17.7 months (95% confidence interval [CI], 13.4 to 22.0 months). The 1-, 2-, and 3-year local controls were 65.3%, 28.6%, and 9.5% respectively. Twelve patients (35%) died of local recurrence, fourteen patients (41%) died of distant metastases, and three patients (9%) died of recurrence and metastases at the same time. Five patients (15%) still survived to follow-up. At the time of analysis, the median survival time was 12.5 months (95% CI, 9.5 to 15.4 months). The 1-, 2-, and 3-year overall survival rates were 55.2%, 20.3%, and 10.9%, respectively. Five patients (15%) experienced grade 3 toxic events and nine patients (26%) have experienced grade 2 toxic events. CONCLUSION: This review shows relatively low toxicity for interstitial ¹²⁵I seed implantation in the patients with advanced stage hypopharyngeal cancer. The high local control results suggest that ¹²⁵I seed brachytherapy implant as a salvage or palliative treatment for advanced hypopharyngeal carcinoma merit further investigation.

ABSTRACT:Objective To explore the effects of virus interleukin‐10 (vIL‐10 ) on different expressions of MHC‐I antigen processing “the operon” .Methods We collected nasopharyngeal carcinoma cells (CNE‐1 and CNE‐2) treated by vIL‐10 at different time points ,and detected the changes of MHC‐I antigen processing “the operons” (TAP‐1 ,TAP‐2 ,LMP‐2 ,LMP‐7 and HLA‐I) by RT‐PCR and Western blot .Results ① mRNA level :There was no difference in the expression of TAP‐1 in CNE‐1 and CNE‐2 cells at various time points .The expressions of TAP‐2 and LMP‐2 in CNE‐1 and CNE‐2 did not change at 1 ,4 ,6 ,12 h ,but downregulated and even disappeared at 24 h .The expression of LMP‐7 in CNE‐1 decreased 4 h after vIL‐10 was added ,and that in CNE‐2 decreased at 6 h .The expression of HLA‐I in CNE‐1 and CNE‐2 showed significant decrease at 24 h .② Protein expression :The expression of TAP‐1 in CNE‐1 and CNE‐2 showed significant decrease at 24 h .The expression of TAP‐2 in CNE‐1 and CNE‐2 was gradually downregulated at different time points .The expressions of LMP‐2 and LMP‐7 in CNE‐2 were gradually downregulated at different periods ,while that in CNE‐1 was only decreased at 12 h .The expression of HLA‐I in CNE‐1 and CNE‐2 was gradually downregulated ,but there was no significant difference at each period in CNE‐1 ,while the expression of HLA‐I in CNE‐2 at 24 h was significantly downregulated .Conclusion vIL‐10 can inhibit the expression of MHC‐I antigen processing “the operon” in NPC in the time‐dependent manner .

Tumor immune evasion is a hallmark of cancer.There are many mechanisms of tumor escape,which mainly include the direct immune escape of tumor and tumor microenvironment mediated immune escape.Recent studies have shown that the development of nasopharyngeal carcinoma and its tumor cells are closely related to the immune escape.The direct immune escape of tumor which is associated with nasopharyngeal carcinoma including the expression decreased or absent of the major histocompatibility complex-Ⅰ on the surface of tumor cell,the lack of costimulatory molecules of tumor and the Fas/FasL system mediated immune escape.And the tumor microenvironment mediated immune escape which is associated with nasopharyngeal carcinoma including the immunosuppressive molecule and the immunosuppressive cells of tumor.Researches on the mechanisms of tumor immune evasion can provide new target for treatment and prevention of nasopharyngeal carcinoma.

Th22,a new subset of helper T cells,which is char-acterized by the secretion of interleukin-22(IL-22),could infil-trate to the epidermis in individuals with inflammatory skin disor-ders.This article introduces the action of Th22 and IL-22 in in-flammatory skin diseases,including psoriasis,atopic dermatitis, systemic lupus erythematosus,systemic sclerosis,aiming at re-vealing the role of Th22 and IL-22 in these diseases,which would not only provide some novel targets of drugs for inflamma-tory skin diseases,but also promote the researches on the pre-vention and treatment of these diseases.

OBJECTIVE@#To study the combined transfection of the siRNA target for Hif-lα and Survivin gene to human NPC CNE-2 cell and its effects on the proliferation and cycle of this cell.@*METHOD@#Combined transfection of the siRNA target for Hif-lα and Survivin gene to human NPC CNE-2 cell, these plasmids were respectively transfected into the same cells. Cell proliferation was detected with MTT assay. The inhibitory effects on target genes were evaluated with RT-PCR and Western Blot at the levels of mRNA and protein, respectively.@*RESULT@#MTT showed that CNE-2 cell proliferation in multi-gene plasmid group was more significantly inhibited than a single gene. The expression of mRNA and protein of two different genes were both decreased in HS group, and the interference effect of multiple genes was better than the single-gene(P<0.01).@*CONCLUSION@#HS group could restrain cell proliferation and interference the mRNA and protein expression in nasopharyngeal carcinoma CNE-2 cell, which was better than the other groups.

OBJECTIVE@#Through analyzing the 13 cases with primary hyperparathyroidism (PHPT) retrospectively, We detected the reasons of the misdiagnosis and the strategy of diagnosis and treatment for the disease.@*METHOD@#Thirteen cases of clinical materials on PHPT were collected from 2002 to 2012. Initial symptom, laboratory examination and imaging findings were analyzed. Thirteen cases of patients with PHPT all were performed surgery. The period of postoperative follow up was from 3 to 24 months, averaged for 12.2 months.@*RESULT@#Surgeries on 13 patients were successful, including 12 cases of parathyroid adenoma and 1 case of parathyroid hyperplasia. After the operation, blood calcium went back to normal, and the symptoms of bone and urinary tract were relieved.@*CONCLUSION@#Initial symptoms of PHPT are predominantly manifested as bone and kidney types. Elevated PTH/blood calcium levels, B-ultrasound/CT and ECT localization are the important diagnosis basis of this disease. Surgical excision of the lesion parathyroid tissue is the most effective treatment.

Objective To construct and identity pSIREN-HIF-1α/shRNA expression vector in order to make foundation of gene therapy for further exploration of RNA interference to nasopharyngeal darcinoma. Methods According to HIF-1αcDNA gene sequence in the gene bank (NM_001530/NM_181054), a pair of 60 nt oligonucleotides each containing the sites of restriction endonuclease at both ends,were designed and synthesized by Reynolds design principles. Oligonucleotides were annealed and ligated with linedrized RNAi-Ready pSIREN-RetroQ-ZsGreen.Transfected into JM109, the recombinants were finally sequenced and identified by 1%agarose gel electrophoresis. Results The size of the target gene fragment amplified by PCR was 470 bp and in accordance with the expected result.pSIREN-HIF-1α was successfully constructed and identitfied by 1%agarose gel electrophoresis.Sequence analysis of inserted fragment revealed the same sequence as synthesized shRNA Oligonucleotides. Conclusion pSIREN-HIF-1α /shRNA expression vector has been successfully constructed, and can make the foundation of research using liposome packaging transfectiing nasopharyngeal darcinoma cell for the next step .

OBJECTIVE@#To investigate the role of VEGF in angiogenesis of nasopharyngeal carcinoma tissue.@*METHOD@#Serum and tissue VEGF were detected by the quantitative enzyme-linked immunosorbent assay (ELISA) method both in 62 nasopharyngeal carcinoma without therapy and 20 nasopharyngitis. The expression of microvessel density in tissue of nasopharyngeal carcinoma and nasopharyngitis were detected by immunohistochemical method.@*RESULT@#(1) There was no significant relationship compared VEGF and MVD in nasopharyngeal carcinoma patient, gender and ages (P>0.05), while the expression of VEGF and MVD in later stage (III+IV) were significant higher than that in earlier stage (I+I) (P0.05).@*CONCLUSION@#VEGF could induce angiogenesis in nasopharyngeal carcinoma tissue, and play an important role in progression in nasopharyngeal carcinoma, VEGF could be an important marker for monitoring prognosis of disease.

OBJECTIVE@#To observe the immunity changes of patients after CIK cells being transfused back, and then to discuss the effects of CIK cells on curing laryngeal cancers.@*METHOD@#Forty eight laryngeal cancer patients with low immune function were collected. The immunity index in the peripheral blood of patients before/after radiotherapy and after CIK cells therapy were measured and compared with normal one.@*RESULT@#After radiotherapy, the percentage of CD3+, CD4+ cells declined, the percentage of CD8+ cells increased; the rate of CD4+ /CD8+ declined and the rate of Th1/Th2 reversed. There were no significant difference between the immunity indexes before and after radiotherapy (P 0.05), while the percentage of NK cells changed obviously (P < 0.05).@*CONCLUSION@#Radiotherapy can restrain the immune function of the patients with laryngeal cancers. CIK therapy is safe and might improve the recent immune function of the patients.

Objective To explore the immune function of the whole body and the tumor site of human nasopharyngeal carcinoma (NPC) patients as well as its correlation with CCR4. Methods The ratios of CD4~+CD25~+T cells and CCR4+cells to lymphocytes were measured by flow cytometry in tissues (25 cases) and peripheral blood (35 cases) from nasopharyngeal carcinoma patients, and then statistical analysis was made. Results The ratios of CD4~+CD25~+T cells and CCR4+ cells in tissue and peripheral blood of NPC were higher than those in the control group (P<0.05). In NPC the ratios of the two cells in tissue were higher than in peripheral blood respectively (P<0.05), but there was no such difference between tissue and peripheral blood in the control group (P>0.05). The ratio of CD4~+CD25~+T cells in tissue at stage Ⅲ+Ⅳ was higher than that at stage Ⅰ+Ⅱ of NPC (P<0.05), but there was no such difference between the two stages in peripheral blood in NPC. There was a positive correlation between CD4~+CD25~+T cells and CCR4+ cells in tissue and peripheral blood of NPC (P<0.05). Conclusion NPC patients have different immunosuppression, and there is even more severe immunosuppression in the tumor site. The ratio of CD4~+CD25~+T cells is correlated with the stage of NPC. Therefore, as NPC progresses to later stages, the percentage of CD4~+CD25~+T cells is increased, which is correlated with poor prognosis. CCR4 plays an important role in reactant of CD4~+CD25~+T cells to tumor sites, and is resistant to immunosurveillance.

OBJECTIVE@#To examine the expression of HIF-1alpha and CD34 in nasopharyngeal carcinoma and nasopharyngitis tissues, evaluate the role of HIF-1alpha and microvessel density (MVD) in nasopharyngeal carcinoma, probe the relationship between HIF-1alpha and MVD.@*METHOD@#The expression of HIF-1alpha was detected in 58 cases with nasopharyngeal carcinoma and 20 samples of nasopharyngitis tissues by immunohistochemical staining SABC methods and SP methods. MVD in tumor was counted by immunostaining with CD34 and analyzed with clinicopathologic parameters.@*RESULT@#The positive rate of HIF-1alpha was 79.31% and 10.00% respectively in nasopharyngeal carcinoma and nasopharyngitis tissues, there was significant difference between them (P<0.01). MVD was 39.74+/-7.41 and 18.43+/-3.24 respectively in nasopharyngeal carcinoma and nasopharyngitis tissues, there was significant difference between them (P<0.01). The expression of HIF-1alpha and MVD were significantly higher in nasopharyngeal carcinoma and in lymph node metastasis group than in no-metastasis group. In different clinical stage, the expression of HIF-1alpha (P<0.05) and MVD (P<0.01) was different. The expression of HIF-1alpha was positively correlated with MVD (P<0.01).@*CONCLUSION@#HIF-1alpha may be involved in carcinogenesis, invasion and metastasis of NPC. Visualization plays a key role in invasion and metastasis of NPC. The expression of HIF-1alpha was positively correlated with tumor visualization in NPC. HIF-1alpha is likely to be a potential target of anticancer therapy.

Objective To investigate the antiproliferative effect of suramin on trabeculectomy in rabit.Design Experimental study. Participants 32 New Zealand albino rabbits.Methods 32 albino rabbits were randomly divided into four groups.Both eyes of rabbits were performed trabaculectomy and placed a piece of cotton with 0.3 mg/ml mitomycin C(MMC)for 2 minutes in standard control group,220 mg/ml suramin for 2 minutes in experimental groupⅠ,250 mg/ml suramin for 2 minutes in experimental groupⅡ.The examination of intraocular pressure was performed before surgery.The filtration bleb and intraocular pressure were observed at 3,7,15, 30 days after surgery.Some conjunctiral specimen was observed with histochemicall(HE staining)and immunohistochemicall(PCNA staining)methods.Main Outcome Measures Intraocular pressure(IOP),filtering bleb appearance,histopathological staining.Results The percentage of the functional blebs in empty control group and experimental groupⅠwas lower than in standard control group,and experimental groupⅡwas higher than in the other 3 groups at 30 days after surgery.Immunohistochemical staining showed the number difference of PCNA positive cell nuclear was no significant between experiment groupⅠand standard control group,but less than empty control group,and experiment groupⅡwas the least(P
