Based on near infrared spectroscopy and high performance liquid chromatography, this paper established the regression relationship between near infrared spectroscopy and index component content of Huoxiang Zhengqi oral liquid, so as to realize the rapid detection of index component content based on near infrared spectroscopy. Magnolol, honokiol and hesperidin were used as the quality indexes of Huoxiang Zhengqi oral liquid. After using the first derivative and normalization pretreatment method, characteristic variables were screened by CARS, and the correction model was finally established by partial least-squares regression (PLSR) method. The method accuracy was evaluated with the external validation, and the prediction results were tested for significance. The results indicated that when the near infrared spectrum was scanned through the bottle, the model's correlation coefficients of prediction (Rp) were higher than 0.99, the root mean square errors of the prediction models (RMSEP) were all less than 0.008 4, and the relative standard errors of prediction set (RSEP) were all less than 2.83%. There was no significant difference in the predicted results between these two kinds of model. The models established in the non-destructive way have good performance and high prediction accuracy. The rapid and nondestructive way has application value in the quality control of Huoxiang Zhengqi oral liquid.

Objective: To explore the active compounds of Huoxiang Zhengqi Oral Liquid for the treatment of coronavirus desease 2019 (COVID-19) by network pharmacology and molecular docking. Methods: The chemical constituents and action targets of Atractylodes chinensis, Citrus reticulata, Magnolia officinalis, Angelica dahurica, Poria cocos, Areca catechu, Pinellia ternata, Glycyrrhiza uralensis, Pogostemon cablin and Perilla frutescens were retrieved from TCMSP. Uniprot database was used to search the corresponding genes of targets, then Cyoscape 3.7.2 software was used to construct the network of medicinal materials-compound-target (gene) for visualization; GO function enrichment analysis and KEGG pathway enrichment analysis were performed through DAVID to predict its mechanism of action, and histograms and bubble maps were plotted by Prism software and Omicshare database for visualization. Results: The network of medicinal materials-compound-target contained 10 medicinal materials, 123 compounds and 257 corresponding target genes, and the key target genes involved PTGS2, HSP90AB1, AR, CAMSAP2, PPARG, NOS2, etc. GO functional enrichment analysis resulted in 278 GO entries (P < 0.05), including 178 biological processes (BP) entries and 36 cellular component (CC) entries, and 64 molecular function (MF) entries. KEGG pathway enrichment analysis revealed that there were 119 (P < 0.05) signaling pathways involving Hepatitis B, small cell lung cancer, non-small cell lung cancer, bladder cancer, prostate cancer and T cell receptor pathways. The results of molecular docking showed that the core compounds such as quercetin, isorhamnetin, irisolidone, kaempferol, wogonin, and baicalein were similar in affinity with the COVID-19 recommended medicine. Among them, quercetin, isorhamnetin and irisolidone had the strongest affinity. Conclusion: The compounds in Huoxiang Zhengqi Oral Liquid can combine with angiotensin converting enzyme II (ACE2) binding to PTGS2, HSP90AB1, AR, CAMSAP2 and other targets to regulate multiple signaling pathways, thus exerting a preventive or therapeutic effect on COVID-19.

The aim of this paper was to investigate the effect of Huoxiang Zhengqi Oral Liquid on intestinal barrier functions in rats with dampness obstructing spleen-stomach syndrome and primarily explore the mechanism. The rat model of dampness obstructing spleen-stomach syndrome was established, and then the modeled rats were randomly divided into the model control group, Huoxiang Zhengqi Oral Liquid high and low dose groups, and natural recovery group according to gender and body weight, with 10 rats in each group. Another 10 rats were taken as blank control group. After each group received the corresponding treatment for 7 days, rat serum was isolated. D-lactic acid content was detected by the MTT method, and diamine oxidase(DAO) activity was detected by the rate method. Colon tissues of the rats were isolated to detect Na~+-K~+-ATPase activity and Ca~(2+)-Mg~(2+)-ATPase activity by phosphate determination method, glutathione peroxidase(GSH-Px) activity was detected by spectrophotometry, catalase(CAT) activity was detected by ammonium molybdate, superoxide dismutase(SOD) activity was detected by hydroxylamine, the expression of occludin protein and ZO-1 protein was detected by immunofluorescence, and the expression levels of occludin protein and ZO-1 protein were detected by Western blot. RESULTS:: showed that low dose Huoxiang Zhengqi Oral Liquid could improve the body weight, diet, stool and urine state of rats with dampness obstructing spleen-stomach syndrome obviously. The D-lactic acid content and the DAO activity in the serum of rats with dampness obstructing spleen-stomach syndrome were reduced obviously. The activities of Na~+-K~+-ATPase, Ca~(2+)-Mg~(2+)-ATPase, GSH-Px, CAT and SOD in rat colon tissues were increased obviously. The occludin proteins and ZO-1 protein expression levels in rat colon tissues were raised obviously. The differences in the above indexes between Huoxiang Zhengqi Oral Liquid group and the model control group were statistically significant(P<0.05). Huoxiang Zhengqi Oral Liquid could effectively restore the intestinal barrier function in rats with dampness obstructing spleen-stomach syndrome and its mechanism may be related to the repair of intestinal mechanical barrier function.

To scientifically evaluate the intervention effect of Chinese medicine preventive administration(combined use of Huo-xiang Zhengqi Oral Liquid and Jinhao Jiere Granules) on community population in the case of coronavirus disease 2019(COVID-19), a large cohort, prospective, randomized, and parallel-controlled clinical study was conducted. Total 22 065 subjects were included and randomly divided into 2 groups. The non-intervention group was given health guidance only, while the traditional Chinese medicine(TCM) intervention group was given two coordinated TCM in addition to health guidance. The medical instructions were as follows. Huoxiang Zhengqi Oral Liquid: oral before meals, 10 mL/time, 2 times/day, a course of 5 days. Jinhao Jiere Granules: dissolve in boiling water and take after meals, 8 g/time, 2 times/day, a course of 5 days, followed up for 14 days, respectively. The study found that with the intake of medication, the incidence rate of TCM intervention group was basically maintained at a low and continuous stable level(0.01%-0.02%), while the non-intervention group showed an overall trend of continuous growth(0.02%-0.18%) from 3 to 14 days. No suspected or confirmed COVID-19 case occurred in either group. There were 2 cases of colds in the TCM intervention group and 26 cases in the non-intervention group. The incidence of colds in the TCM intervention group was significantly lower(P<0.05) than that in the non-intervention group. In the population of 16-60 years old, the incidence rate of non-intervention and intervention groups were 0.01% and 0.25%, respectively. The difference of colds incidence between the two groups was statistically significant(P<0.05). In the population older than 60 years old, they were 0.04% and 0.21%, respectively. The incidence of colds in the non-intervention group was higher than that in the intervention group, but not reaching statistical difference. The protection rate of TCM for the whole population was 91.8%, especially for the population of age 16-60(95.0%). It was suggested that TCM intervention(combined use of Huoxiang Zhengqi Oral Liquid and Jinhao Jiere Granules) could effectively protect community residents against respiratory diseases, such as colds, which was worthy of promotion in the community. In addition, in terms of safety, the incidence of adverse events and adverse reactions in the TCM intervention group was relatively low, which was basically consistent with the drug instructions.

OBJECTIVE: To establish a method for simultaneous determination of nine components in Huoxiang zhengqi oral liquid, and to improve and perfect the quality standard of Huoxiang zhengqi oral liquid. METHODS: The contents of nine components in 10 batches of Huoxiang zhengqi oral liquid were determined by HPLC, such as licorice coumarin, isorlicin, liquiritinapioside, narirutin, liquiritin, saponins, hesperidin, magnolol and honokiol. The determination was performed on Kromasil Eternity XT-5-C18 column with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution (gradient elution) at the flow rate of 1 mL/min. The detection wavelength was set at 220 nm, and column temperature was 25 ℃. The sample size was 10 μL. RESULTS: The linear range of licorice coumarin, isorlicin, liquiritinapioside, narirutin, liquiritin, saponins, hesperidin, magnolol and honokiolin were 0.000 5-0.007 5, 0.000 8-0.025 0, 0.006 1-0.976 0, 0.001 6-0.250 0, 0.007 8-0.025 0, 0.000 4- 0.062 7, 0.008 6-0.276 0, 0.010 0-0.500 0, 0.010 0-0.500 0 mg/mL (r=0.999 2-1.000 0). The detection limits were 0.001 3, 0.000 1, 0.004 7, 0.005 0, 0.012 0, 0.001 3, 0.007 8, 0.007 7 0, 0.005 8 μg/mL, and the quantitative limits were 0.013 0, 0.000 8, 0.047 0, 0.050 0, 0.120 0, 0.013 0, 0.078 0, 0.070 0, 0.058 0 μg/mL, respectively; RSD of precision, stability and repeatability tests were less than 3.0% (n=6). Average recovery rates were 98.67%, 101.85%, 98.97%, 103.05%, 100.00%, 97.78%, 97.91%, 100.13%, 101.95%; RSDs were 1.14%, 2.18%, 0.40%, 0.17%, 1.38%, 0.85%, 1.38%, 0.10%, 1.35% (n=6). CONCLUSIONS: The established method is accurate and reliable, which can provide reference for the establishment of the overall quality control evaluation system and the improvement of quality standard for Huoxiang zhengqi oral liquid.

Objective To classify the essence of dampness obstructing spleen-stomach syndrome (DOSS) in rats and the effect of Huoxiang Zhengqi Oral Liquid (HZOL) on DOSS. Methods Comprehensive physical methods were applied to establish the rat DOSS model. Body weight, body length, tail length, abdominal girth index, and locomotor activity were observed. Moreover, a proton nuclear magnetic resonance (1H-NMR) based metabonomic approach was developed to profile DOSS-related metabolic perturbations in rat serum and feces and to investigate the intervention effect of HZOL in rats. Results Compared with the healthy control group, rats with DOSS showed weight loss, slow growth of body length, increased abdominal girth index, and decreased locomotor activity along with metabolic perturbations in serum and feces. The disturbed metabolic state associated with DOSS in rat serum and feces could be restored by HZOL. Meanwhile, six and eight potential biomarkers related to DOSS in rat serum and feces, respectively, were reversed by administration of HZOL. Conclusion The current study demonstrated that HZOL exerted protective effect against DOSS in rats, which was probably mediated by amelioration of disturbance in energy and amino acid metabolism and gut microbiota.

Objective To establish a UPLC method for detecting content of imperatorin and isoimperatorin in huoxiang zhengqi oral liquid. Methods ACQUITY UPLC BEH C18 (2. 1 mmí50 mm,1. 7 μm) was used and the mobile phase was methanol and water by gradient elution mode. The column temperature was 30 ℃ ,the flow rate was 0. 3 mL·min-1 and the detection wavelength was 248 nm. Results The linear range of imperatorin was 1. 305-13. 050 μg·mL-1 and the regression equation was as follow Y =13 633X+3 976 (r=0. 999 9). The linear range of isoimperatorin was 0. 596-5. 960 μg·mL-1 and the regression equation was Y=10 661X+1 073 (r=0. 999 9). The average recovery was 99. 25% (RSD=0. 74% ) and 98. 94%(RSD = 0. 63% ),respectively. Conclusion The method is accurate, rapid and reliable, and can be used to determine imperatorin and isoimperatorin in huoxiang zhengqi oral liquid.
