SUMMARY: The vomeronasal organ (VNO) is located in the anteroinferior part of the nose and the accessory olfactory organ in mammals which is responsible of sense of smell. This study aims to compare the macro and microanatomical structure of the VNO between sheep and dogs. In the current study, we used ten adult slaughtered sheep and ten adult synchronized dogs with different sexes ages 1-2 years. The head of both animals were preserved in 10 % formalin for one week. This study shows in both animals, the VNO occupies the same position in the cavity of the vomer bone and the same relationship in the cranial part of the nasal cavity. Furthermore, the VNO is divided into three parts based on shape that are the rostral, central, and caudal part. The results show the VNO in sheep has a (U) shape and is opened dorsolaterally. It has a small and narrow cavity. It is long 6 cm long, and it has different diameters on its course. In comparison, the vomeronasal organ in dogs is very developed and has a (J) shape. It has a large and long cavity and ends at the fourth molar. Its length is about 10 cm, and it has one diameter on its course. The VNO receives the blood supply from the sphenopalatine and caudal palatine arteries. The present study shows main differences between sheep and dogs VNO in which the structure of vomeronasal bone between the sheep and dog is completely different. The finding will illustrate fundamental differences and provide specific structural differences between the two species.

Objective:To establish the animal model of cervicofacial venous malformations(VMs)by surgical reconstruction of exter-nal jugular vein in sheep. Methods:The external jugular veins of 5 sheep were dissected, and the position, course, branch and exter-nal diameter were observed and measured. The models of VMs with draining and returning veins were constructed by suturing or constric-ting the proximal part of main trunk and ligating or constricting the distal part of the jugular or branch veins. The animal model was eval-uated by Doppler ultrasound, gross observation and histological observation at the 4th week after surgery. Results:The external jugular veins of sheep is in the lateral side of bilateral neck, and the main trunk is formed by the maxillary vein and lingual facial vein. The ex-ternal diameter ranges from 6 to 12 mm,with an average external diameter of 9.3 mm.Immediately after the external jugular vein was sutured and narrowed at the proximal part of the main vein, the distal part of the vein branch was ligated or narrowed, the blood flow speed slowed down and the veins in the model area bulged.4 weeks after surgery, gross observation showed that most veins narrowed and thrombosis was formed in part of the venous lumen. The central region of some specimens was dilated, and the peripheral collateral veins were dilated in some models.Doppler ultrasonography showed that the lumens of most veins were dilated and the returning veins and the inflow veins were narrowed.Colored blood flow was seen in the lumen.Histological observation showed that the structure of vein endothelium and wall was close to the normal vein, and the vein vessel wall of some specimens was thickened. Conclusion:The VMs model estab-lished by external jugular vein of sheep basically meets the re-quirements and is expected to be used in the therapeutic meth-odology research of cervicofacial VMs.

Abstract This study assessed the anthelminthic feed additive albendazole effect on the main indicators of the quality of sheep meat obtained from infected and deworming animals. A total of 20 heads of Akzhaik meat and wool sheep (i.e. 2 years of age, of different sex) were used in a 20-day experiment and 2 groups of 10 heads were formed (n = 4). The sheep were fed with the basic diet (i.e. control, group CON), without adding anything, and the second group was fed the basic diet orally with the addition of a dose of 1.2 g / head (an experimental group). At the end of the experiment, a control slaughter was carried out from each group in order to study the biochemical composition of meat and its quality. The anthelmintic feed additive albendazole did not have a significant effect on the indicators (organoleptic and physicochemical) of meat quality (P> 0.04). However, the ratio of fat and ash in the meat of the control group has differences in indicators and is reduced by 24.81% and 0.03%, respectively. The drug had a significant effect on the biological value of meat in the experimental group, where there is a higher content of essential amino acids (P = 0.06), nonessential (P = 0.05) concentrations in comparison with the CON groups. The results obtained show that the meat of the experimental groups of sheep, when using the anthelminthic feed additive albendazole, did not have a significant effect on organoleptic parameters, but significantly positively influenced the metabolism, live weight gain and biological value of meat.

Naturally occurring mutations in morphogenetic protein 15 (BMP15) are associated with decreased ovulation rate (OR), litter size (LS), and sterility. It is of a great interest to elucidate BMP15 gene in Cholistani sheep breed to uplift socio-economic status and the knowledge of Cholistani sheep breeding in Southern Punjab, Pakistan. In our study, a total of 50 infertile Cholistani sheep aged between 2-6 years and having no blood relation were screened for BMP15 mutations. For this purpose, a high-quality DNA was extracted from the blood of sheep followed by primer designing, Polymerase Chain Reaction (PCR) amplification, DNA sequencing, and in silico analyses. Out of total 50 samples, 9 samples including case 1 (T3), case 2 (T8), case 3 (T17), case 4 (T22), case 5 (T25), case 6 (T33), case 7 (T40), case 8 (T44), and case 9 (T47) were found positive for a variety of already reported and novel BMP15 mutations. Further in silico analyses of the observed mutations have shown the functional impact of these mutations on different characteristics (molecular weight, theoretical PI, estimated half-life, instability index, sub-cellular localization, and 3D confirmation) of the encoded proteins, possibly altering the normal functionality. In a nutshell, findings of this study have confirmed the possible essential role of the BMP15 mutations in the infertility of the Cholistani sheep.

Urinary tract infections (UTIs) are one of the most commonly occurring infections in outpatient settings and hospital admissions globally. No single medium is capable of supporting the growth and identification of all uropathogens. Recently, in order to improve isolation and presumptive identification in urine samples, chromogenic agar media are being used more and more frequently as versatile primary culture tools. Direct susceptibility testing by disc diffusion on clinical samples offers a rapid and inexpensive means of obtaining information to guide antimicrobial therapy. The objective was to evaluate and compare the effectiveness of conventional culture systems and chromogenic agar medium for the isolation, identification, and direct testing of uropathogen's antibiotic susceptibility. The study comprised 155 clinically suspected UTI patients. Uncentrifuged urine samples underwent wet mount analysis and centrifuged samples for Gram staining before the culture. All urine samples that were received in the lab underwent Direct Susceptibility Testing (DST) right away. A total of 155 urine samples were cultured on all three media (HiChrome UTI Agar, Sheep Blood Agar, and MacConkey Agar). 58 (37.32%) showed significant bacterial growth, while 97 (62.58%) showed no growth. A single organism was isolated in 54 (36% of the samples), and two (1.42%) samples yielded mixed growths of two species. We conclude that the Hichrome agar medium can be a desirable, simple primary isolation and identification medium that significantly lessens the daily workload associated with urine culture in microbiology labs.

Abstract Background: Assessment of animal growth based on live weight (LW) in traditional sheep production systems is limited by the high cost of purchase and maintenance of livestock scales. Objective: To develop and evaluate equations for LW prediction using heart girth (HG) in growing Pelibuey sheep. Methods: A dataset (n=415) of clinically healthy male Pelibuey sheep from two months to one year of age, with an average LW of 25.96 ± 10.25 kg and HG of 68.31 ± 10.53 cm, were used. Three equations were evaluated: LW (kg) = −37.70 + 0.93 × HG (Eq. 1); LW (kg) = −1.74 + 0.19 × HG + 0.008 × HG2 (Eq. 2); and LW (kg) = 0.003 × HG2.68 (Eq. 3). Results: The correlation coefficient between LW and HG was r = 0.94 (p<0.001). The three equations showed a high concordance correlation coefficient (CCCs≥0.97). However, the random error was the main component of the mean square partition of the prediction error (≥82.78%) only for Eqs. 1 and 2. The test for parameter identity (intercept=0; slope=1) was accepted only for Eq. 2 (p>0.05). On the other hand, for Eqs. 1 and 3 the intercept was different from zero and the slope was different from one (p<0.05). Conclusion: The second-degree equation accurately and precisely estimated body weight of growing Pelibuey sheep using the HG as a sole predictor variable.

Abstract The purpose of this paper is to present the results of a characterization of the consumer of sheep meat in the center of the province of Santa Fe, Argentina. The information was obtained through semi-structured online surveys. The results indicate that consumers have a good perception of sheep meat (quality, flavor, forms and moments of consumption), they buy it directly from the field (informal slaughter) and show interest in the traceability of the product. They prefer animals raiced in the outdoor grazing, they demand healthy meat, a brand or a quality mark, and they are willing to pay more for quality attributes. The identified aspects are key when defining the business model or the strategies to be used to achieve competitive and sustainable sheep meat production.

In this study, a single base editing system was used to edit the FecB and GDF9 gene to achieve a targeted site mutation from A to G and from C to T in Ouler Tibetan sheep fibroblasts, and to test its editing efficiency. Firstly, we designed and synthesized sgRNA sequences targeting FecB and GDF9 genes of Ouler Tibetan sheep, followed by connection to epi-ABEmax and epi-BE4max plasmids to construct vectors and electrotransfer into Ouler Tibetan sheep fibroblasts. Finally, Sanger sequencing was performed to identify the target point mutation of FecB and GDF9 genes positive cells. T-A cloning was used to estimate the editing efficiency of the single base editing system. We obtained gRNA targeting FecB and GDF9 genes and constructed the vector aiming at mutating single base of FecB and GDF9 genes in Ouler Tibetan sheep. The editing efficiency for the target site of FecB gene was 39.13%, whereas the editing efficiency for the target sites (G260, G721 and G1184) of GDF9 gene were 10.52%, 26.67% and 8.00%, respectively. Achieving single base mutation in FecB and GDF9 genes may facilitate improving the reproduction traits of Ouler Tibetan sheep with multifetal lambs.

Due to the failure of multiple translational researches, Stroke Therapy Academic Industry Roundtable (STAIR) recommends the use of large animal models of focal cerebral ischemia for preclinical researches. Especially, stroke treatment has currently entered a new era of vascular recanalization. Large animals commonly used in acute ischemic stroke models include dogs, swine, sheep, and non-human primates, which can be used to simulate various aspects of cerebral ischemia and reperfusion (vascular recanalization) in patients. Although large animals have significant advantages due to their proximity to humans in anatomy and physiology, there are also issues with anatomical and physiological specificity and ethical limitations. This article summarizes the large animal ischemic stroke models prepared by craniotomy and endovascular intervention, hoping to help researchers select the most appropriate large animal ischemic stroke model, and then promote the development of stroke translational research.

This study aimed to explore the expression changes of VASA gene in sheep testis development and to construct VASA gene knock-in vector to prepare for the study on the differentiation of sheep germ cells in vitro. The testicular tissues of 3-month-old (3M) and 9-month-old (9M) sheep which represent immature and mature stages, respectively, were collected. The differential expression of VASA gene was analyzed by quantitative real-time PCR (qPCR) and Western blotting, and the location of VASA gene was detected by immunohistochemistry. The sgRNA targeting the VASA gene was designed and homologous recombination vectors were constructed by PCR. Subsequently, plasmids were transferred into sheep ear fibroblasts. The VASA gene was activated in combination with CRISPR/dCas9 technology to further verify the efficiency of the vector. The results showed that the expression level of VASA gene increased significantly with the development of sheep testis (P < 0.01), and was mainly located in spermatocytes and round spermatids. The knock-in vector of VASA gene was constructed by CRISPR/Cas9 system, and the Cas9-gRNA vector and pEGFP-PGK puro-VASA vector were transfected into ear fibroblasts. After CRISPR/dCas9 system was activated, ear fibroblasts successfully expressed VASA gene. The results suggest that VASA gene plays a potential function in sheep testicular development and spermatogenesis, and the VASA gene knock-in vector can be constructed in vitro through the CRISPR/Cas9 system. Our results provided effective research tools for further research of germ cell development and differentiation.

The creole hair sheep (CHS) developed adaptive advantages to the tropical climate, which allow them to have resistance to diseases, high fertility and prolificacy. Currently, there is little research on the productivity of CHS in systems associated with the cultivation of sugarcane, which makes it necessary to carry out research to promote its attributes. The purpose of this research was to analyze the productivity and body development of a crossing of CHS, using production parameters and growth curves.180 animals of single and multiple births were used; the follow weight was recorded: at birth (WB), at weaning (WW); at 210 days (W210), and furthermore, daily gains of pre-weaning (WDPRE) and post-weaning (WPPOS) were recorded. The data were analyzed using descriptive statistics with a GLM with the effects sex (S), type of birth (TB), number of births (NB) and period of birth (PB). The non-linear models Gompertz, Logistic, Brody, Richards and Von Bertalanffy were used in 1455 records of weights at different ages to perform growth curves. All analyzes were performed with the SAS statistical package. The R2, AIC and BIC coefficients obtained by the Gauss-Newton method were considered to select the best fit model. The males presented higher averages in the variables WB, WW, W210) and WPOS. The TB affected all the variables; The WB and the WPPOS were affected by the NB and the PB affected the WB, the WDPRE, the W210 and the WPPOS. The best fit model for the growth curves was of Richards.

We aimed to compare the in silico effect of florfenicol at doses of 20 and 30 mg/Kg in sheep by intravenous (IV) and intramuscular (IM) routes, using PK/PD modeling. We performed a Monte Carlo simulation based on plasma concentration data from a previously published study. We calculated the area under the curve (AUC) and the efficacy rates of florfenicol to bacteriostatic, bactericidal, and bacteriological eradication effects. The dose of 20 mg/Kg IV demonstrated 100, 93, and 0% eradication effects for MICs of 0.5, 1, and above, respectively. The bacteriostatic effect was 99 and 90% for MIC of 4 and 2 µg/ml, while the bactericide was 14% for MIC of 2 µg/ml. The 30 mg/Kg IV dose showed 100% eradication for MIC of 1 µg/mL and 100% bactericidal effect for MIC of 2 µg/mL. There is a 100% of bacteriostatic effect at MIC of 4 µg/ml. Doses of 20 and 30 mg/Kg IM showed 100% eradication for MIC up to 1 µg/mL and 0% for MIC above. The bacteriostatic effect was maintained at 100% for a MIC of 4 µg/mL at both doses. This study shows the bacteriological eradication effect of florfenicol at doses of 20 and 30 mg/Kg, IV, and IM. Therefore, we recommend an in vivo efficacy study with a dose of 30mg/Kg IM in sheep infected with F. necrophorum with MIC greater than two µg/mL.

Background: Litter size at birth (LSB) is one of the most important economic traits in sheep and could be used in genetic improvement schemes for meat production. LSB is inherently a categorical trait and should be analysed with threshold models. Objective: Bayesian threshold models were used to analyze sheep LSB to estimate genetic parameters. Methods: Data was based on 7,901 LSB records from 14,968 dams and 682 sires collected from 1986 to 2012 at Makouie Sheep Breeding Station in Iran. Means of posterior distributions (MPDs) of LSB's genetic parameters were estimated, and the best-fitted models were selected using the deviance information criterion. Results: In the repeated measurement analysis, the estimated direct and maternal heritabilities, and permanent environmental effect (±SE), according to the best-fitted model (model 5), were 0.01 (0.010), 0.02 (0.014), and 0.01 (0.011), respectively. In the univariate analysis, the best estimates of direct and maternal heritabilities were 0.12 (0.064) and 0.08 (0.045), respectively. An increasing trend for direct and maternal heritabilities was observed in parity 2 (0.15 (0.082) and 0.25 (0.083), respectively). In the bivariate analysis, the best estimates of direct and maternal heritabilities for LSB were 0.03 (0.027) and 0.22 (0.041), respectively. The direct and maternal genetic correlations among parities were 0.25 (0.054) and 0.12 (0.021), respectively. Conclusions: The results showed a considerable influence of environmental factors on LSB in each parity of sheep; also, statistically different genetic parameters (p<0.05) were obtained from one parity to another, indicating the different and large influences of genetic and environmental factors for each parity.

Background: The main transmission route of Chlamydia abortus is by ingesting the microorganism that has been eliminated in vaginal secretions, placental membranes or abortions that contaminate the environment and, possibly, through milk and colostrum. Elimination through vaginal secretions is well documented. However, there are no reports about isolation and identification of C. abortus in the colostrum or milk of infected sheep, so it is important to determine whether or not C. abortus may be present in these secretions, which are the only food of lambs. Objective: To detect C. abortus in colostrum, milk, and vaginal secretions of sheep with a history of reproductive disorders. Methods: Colostrum, milk, and vaginal exudates were collected from 66 sheep. The samples were inoculated in mouse fibroblast cell cultures and the presence of C. abortus determined by direct immunofluorescence. Results: 19 out of 66 colostrum samples (28.7%), 14 out of 66 milk samples (21.2%) and 17 out of 66 vaginal swabs (25.7%) were positive for C. abortus. The 50 samples positive for isolation and detected by immunofluorescence, together with 42 negative samples were subjected to qPCR to amplify a fragment of the ompA gene from C. abortus. Thirty-eight of the 92 samples processed by this technique were positive for C. abortus. Conclusion: The results demonstrated the presence of C. abortus in a high proportion in colostrum, milk and vaginal secretions of infected sheep. To the best of our knowledge, this is the first field study confirming the presence of C. abortus in colostrum, which shows that excretion of Chlamydia by lactogenesis could occur in the first hours after birth.

ABSTRACT Objective. To explore the feasibility of developing a sheep model of neurocysticercosis (NCC) by intracranial infection with T. solium oncospheres. Materials and methods. We carried out an experimental infection model of NCC in sheep. Approximately 10 T. solium oncospheres previously cultured for 30 days were inoculated intracranially into ten sheep. The oncospheres, in 0.1 mL of physiological saline, were injected into the parietal lobe through an 18-gauge needle. Results. After three months, granulomas were found in two sheep. In a third sheep we identified a 5 mm diameter cyst in the right lateral ventricle and histological evaluation confirmed that the cyst corresponded to a T. solium larva. Immunohistochemistry with monoclonal antibodies directed against membrane components and excretory/secretory antigens of the T. solium cyst was also used to confirm the etiology of the found granulomas. One of them showed reactivity to the monoclonal antibodies used, thus confirming that it was a cysticercus. Conclusion. This experiment is the proof of concept that it is possible to infect sheep with cysticercosis by intracranial inoculation.
