RESUMO

Objective To establish a simultaneous detection approach for 34 emerging contaminants(ECs)in tap water by liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).Human health risk assessment was performed according to the detection results from 43 tap water samples.Methods Tap water samples were concentrated and extracted by solid phase extraction,and then blown to near dry by nitrogen at 40℃.The sample extracts were dissolved in methanol-water solution(95:5,VN)to 0.5 mL for analyzing.Agilent Jet Stream Electrospray Ionization(AJS ESI)and the multiple reaction monitoring(MRM)mode were performed for MS to acquire the data of 34 ECs.A database including precursor ion,product ion and retention times was established accordingly.Results The average linear correlation coefficients(r)of 34 kinds of ECs was 0.995 9.The limits of detection were 0.01~0.60 ng/L and the recoveries were between 60.7%and 119.8%.The intra-group precisions were between 0.05%~9.89%and the intra-day precisions were between 0.20%~14.40%for the spiked samples.The method was applied to analyze 43 tap water samples and a total of 15 ECs were detected.According to the results,the detection rate of caffeine was the highest(84%),and the concentration range was ND~74.42 ng/L.Among all the ECs detected,1,2,3-benzotriazole had the highest concentration(ND~361.15 ng/L),where detection rate was 44%.Humans may be exposed to these ECs by drinking the tap water.The human health risk assessments of 12 kinds of ECs were carried out,however,the estimated risk was negligible(risk quotient<0.01).Conclusion The method is simple,highly sensitive and selective,and could meet the detection needs of ECs at trace level in tap water.There was no human health risk posed for ECs identified in 43 tap water samples analyzed by this method.

ObjectiveTo explore the possible mechanism of Chuanxiong (Rhizoma Chuanxiong) & Tianma (Rhizoma Gastrodiae) herbal pair in treating migraines based on AMP-activated protein kinase (AMPK)/transient receptor potential A1 channel (TRPA1) pathway. MethodsForty-eight healthy male SD rats were randomly divided into control group, model group, and Chuanxiong Tianma medication group, with 16 rats in each group. The control group and model group were given 10 ml/kg of normal saline by gavage, while the Chuanxiong Tianma medication group was given 0.675 g/kg of Chuanxiong Tianma herbal pair by gavage, once daily for 8 consecutive days in both groups. Migraime model was performed before the last administration, with subcutaneous injection of 10 ml/kg of normal saline in the control group, and subcutaneous injection of 10 ml/kg of nitroglycerin in the model group and Chuanxiong Tianma medication group. The Von Frey filament was used to measure the periorbital mechanical pain threshold of rats. The enzyme-linked immunosorbent assay (ELISA) was used to determine the levels of calcitonin gene-related peptide (CGRP) in rat serum and cerebrospinal fluid. The nitric oxide (NO) assay kit was used to determine the NO level in serum and cerebrospinal fluid. RT-PCR was usedto detect the mRNA expression levels of immediate-early genes in the trigeminal ganglion of rats (c-Fos), CGRP, transient receptor potential V1 channel (TRPV1), AMPK alpha subunit (PRKAA), and TRPA1. Immunofluorescence was used to detect the number of c-Fos-positive cells in the trigeminal cervical complex (TCC) and the protein expression levels of phosphorylated AMPK (pAMPK) and TRPA1 in the trigeminal ganglion. ResultsCompared to those in the control group, the mechanical stimulation threshold and pAMPK protein expression in the model group decreased, while the levels of CGRP and NO in serum, c-Fos, CGRP, TRPV1 and TRPA1 mRNA levels in the trigeminal ganglion, TRPA1 protein expression, and the number of c-Fos-positive cells in the TCC significantly increased (P<0.05). Compared to those in the model group, the mechanical stimulation threshold and pAMPK protein expression in the Chuanxiong Tianma medication group significantly increased, while the levels of CGRP and NO in serum, c-Fos, CGRP, TRPV1 and TRPA1 mRNA levels in the trigeminal ganglion, TRPA1 protein expression, and the number of c-Fos-positive cells in the TCC significantly decreased (P<0.05). ConclusionChuanxiong Tianma herbal pair may improve migraine symptoms by regulating the AMPK/TRPA1 pathway in the trigeminal ganglion and increasing the mechanical pain threshold.

Objective:To explore the significance of next-generation sequencing to detect gene mutations for diagnosis of patients with unexplained pancytopenia.Methods:The next-generation sequencing was used to detect 32 common hematological tumor gene mutations in 113 patients with unexplained pancytopenia in Huazhong University of Science and Technology Union Shenzhen Hospital from February 2016 to February 2019.Results:The next-generation sequencing results showed that of the 32 genes tested in 113 patients, 69 mutations occurred in 25 genes. Among them, 105 patients (92.9%) carried at least 1 gene mutation, and the median number of mutations was 2 (1-5). Fifty-one patients were diagnosed with myelodysplastic syndrome (MDS), and the related mutant genes were detected; 62 patients were diagnosed with atypical aplastic anemia or cytopenia of unknown significance. The total effective rate of treatment for 113 patients was 65.5% (74/113).Conclusion:The next-generation sequencing helps to diagnose patients with unexplained pancytopenia, and provides targeted strategies for finding new treatments and prolonging the overall survival of patients.

Objective To provide useful reference for the pre-operative design and operation by 3D reconstruction and simulation techniques in rhinoplasty.Methods From October 2016 to March 2017,the laser scanning and 3D reconstruction were performed on 25 patients before operations.The facial morphology and the simulated effect were ontained.All patients were female and they were 18 to 39 years old.The average age was 26 years old.The outline and form were normal.We discussed with patients to confirm the ideal nasal 3D simulated effect.In the process of operation,we were guided by the data of soft measures and 3D printing model.Results Among 25 patients who werefollowed up postoperatively.We got intuitive,accurate 3D reconstruction graphs and simulated surgery procedures in all cases.Being applied to actual surgery procedure,23 patients were satisfactory with postoperational results.Two patient's nasal tip dropped.We were satisfied with the results after repair.Conclusions 3D reconstruction and simulation techniques improve surgical precision and operation planning selection,which is a good aided method in rhinoplasty.

We investigated the cellulase-assisted extraction and anti-ultraviolet activity of water-soluble polysaccharides from the root of Flammulina velutipes on Caenorhabditis elegans. A Box-Behnken design experiment with three factors and three levels, including enzymolysis temperature, microwave time, and microwave power, was designed on the basis of the results of single-factor experiments. For improving the polysaccharide yield of F. velutipes root, the following optimal extraction conditions were used: 52.67 degree C enzymolysis temperature, 80s microwave time, and 144 W microwave power. Under optimal conditions, the actual measured value of the yield was 2.01% [w/w] and the predicted value was 2.06% [w/w]. One fraction [FRP-2] was isolated and purified, and its characteristics were analyzed. The average mean molecular weight of FRP-2 was measured to be 2.60×105 Da, and its monosaccharide composition is mainly glucose. The sugar units are present both in the alpha-configuration and beta-configuration. Moreover, FRP-2 exhibited certain antiultraviolet activity to C. elegans when the polysaccharide concentration ranged between 0.05mg/mL and 0.20mg/mL

Objective To establish the in vitro biofilm model of Laribacter hongkongensis(LH),to analyze the type Ⅰ integron related genes carried by LH and to investigate the mechanism of LH resistance to quinolones.Methods The biofilm forming abilities of LH clinical isolates were determined by Giemsa staining qualitative method and by crystal violet staining semi-quantitative method.The sensitivity of LH to norfloxacin,ofloxacin,levofloxacin,ciprofloxacin and lomefloxacin in both planktonic and biofilm conditions were dectermined by broth microdilution susceptibility tests.Type I integron related genes carried in 18 LH strains resistant to quinolone were detected by PCR amplification method.Results The detection results by Giemsa staining demonstrated that 36 strains in 55 LH clinical isolates formed visible biofilm,and the biofilm formation rate was 65.4%(36/55).In the biofilm forming ability detected by crystal violet staining semi-quantitative method,OD560≤0.15 was in 8 strains of LH,0.150.20 in 7 strains respectively.The levels of minimal biofilm inhibitory concentration in norfloxacin,ofloxacin,levofloxacin,ciprofloxacin and lomefloxacin to LH were respectively higher than the minimal inhibitory concentration(MIC) in the corresponding floating bacteria(P<0.05).Among 55 strains of LH,18 strains were resistant to quinolones,the resistance rate was 32.7%,and the type I integron in 18 strains of LH carried the drug resistant genes,these drug resistant genes played the drug resistance role in corresponding bacterial strains.Conclusion Drug resistance gene formation and widespread of LH may be associated with type I integron.

Objective To investigate the effect of different dose of atorvastatin on endothelium dependent vasodilatation function in hypertensives without hyperlipemia in attempt to verify the hypothesis of "beyond antihyperlipimia" effect of statins.Methods Fifty-five hypertensives without hyperlipemia were randomly to receive atorvastatin(10 mg/d,n=25)or(20 mg/d,n=30).Twenty-five normotensives were enrolled in control group.Serum cholesterol were determined.Flow-mediated dilation(FMD)and endothelium-independent dilatation(EID)were measured with high-resolution ultrosonography before and after 4 weeks atorvastatin.Results Compared with control group,FMD were significant decreased in hypertensives without hyperlipemia.FMD were improved after atorvastatin for 4 weeks(atorvastatin 10 mg group:7.5%?2.7% vs 11.5%?3.1%,P

Objective To evaluate the effect of superficial temporal fascia flap for the reconstruction of temporal depression. Methods According to the degree of temporal depression, we applied folded superficial temporal fascia to reconstruct the temporal depression, combined with forehead and orbital subperiosteal facelift. Results Postoperative follow-up for 1~2 years showed that the method used in subperiosteal facelift combined with temporal depression, maintained a satisfactory long-term full contour. And it improved the facial contour.Conclusions Application of superficial temporal fascia flap in reconstructing the temporal depression combined with facelift for one-stage is a good method. It can get the purposes of tightening the skin and improving the facial contour together. Good results are achieved.
