RESUMO

Objective To observe the expression level of PRMT5 in gastric cancer cells, and explore the effect of knocking down the expression level of PRMT5 on the biological behavior of gastric cancer cells. Methods Western blot was used to detect the expression of PRMT5 protein in gastric cancer cell lines MGC803, SGC7901, MKN45 and human gastric epithelial cells GES-1. siRNA1 and siRNA2 plasmids were transfected to knock down the expression of PRMT5. Cell proliferation and apoptosis assay, transwell assay were used to detect cell proliferation, apoptosis and invasion abilities, respectively. The protein expression levels of β-catenin, cyclin D1 and Bax were detected. Results Compared with GES-1 cells, PRMT5 protein expression levels increased in MGC803, SGC7901 and MKN45 cells (P < 0.001). After knocking down the expression of PRMT5, the cell proliferation and invasion abilities were weakened, the apoptosis rate increased (P < 0.05), the expression of β-catenin and Cyclin D1 protein decreased, and the expression of Bax protein increased (P < 0.001). Conclusion The expression level of PRMT5 is increased in gastric cancer cells. Knockdown of PRMT5 expression level could inhibit cell proliferation, invasion and promote cell apoptosis via reducing Wnt/β-catenin signaling pathway.

RESUMO

OBJECTIVE To study the role of Enterococcus in peritoneal cavity infection and the infection -associated factors, by which the prophylactic measures are put forward. METHODS The role of Enterococcus in peritoneal cavity infection and the infection-associated factors were discussed with the analysis of the investigated 33 cases with the peritoneal cavity infection by Enterococcus. RESULTS The peritoneal cavity infection was mainly caused by Enterococcus faecalis, for which penicillin and ampicillin were the first choice of empirical therapy. For regime of the combination use of antibiotics might be feasible for the infection caused by the speeics with unshowed high-level resistance to aminoglycosides. Many related risk factors on peritoneal infection caused by Enterococcus were included such as the colonization of the drug resistant strain of Enterococcus promoted by cephalosporins Ⅲ and the abuse of large amount of antibiotics as a selective pressure on the resistant strain. CONCLUSIONS Much attention should be paid on the role of Enterococcus in peritoneal infection clinically. The effective therapy is associated with the eradication of the pathogen. and the better control measures against infection.

RESUMO

OBJECTIVE To investigate the significance of CNS in clinical infections.METHODS A total of 114 CNS strains isolated from our hospital were identified by conventional procedures and the icaD gene was amplified by PCR.RESULTS Of all CNS strains,the highest isolated rate was S.epidermidis(41.2%).CNS isolated from deep venous catheters,wound secretions and blood had a higher rate of carrying ica operon,accounted for 44.4%,42.1% and 36.8%,respectively,whereas 24.0% in respiratory secretions and 14.1% in urine.Among the ica operon positive CNS strains,the percentages of S.epidermidis and S.haemolyticus were 42.6% and 19.0%.CONCLUSIONS There is a wide range of CNS species carrying ica operon,especially in S.epidermidis.CNS isolated from different specimens might have different significances.It should be cautious to assess the results of isolated CNS from respiratory and urine specimens.The CNS isolates from blood specimens might be contaminated.The PCR method for the ica operon is simple and easy.
