RESUMO
Objective To explore the effect of hydrogen sulfide on inflammatory factors and energy metabolism of mitochondria after limbs reperfusion injury in rats. Methods Sixty rats were divided into three groups:sham operation group, control group(ischemia-reperfusion injury + saline group),and experimental group(ischemia-reperfusion injury + HS group).Wistar rat models of limb ischemia-reperfusion injury were established.Skeletal muscle samples were collected to determine the levels of necrosis decomposition products [including myoglobin(MB),lipoprotein complex(LPC)and lipid peroxide(LPO)];blood samples were collected to determine the levels of interleukin(IL)-1,IL-6 and tumor necrosis factor-α(TNF-α);mitochondria were extracted for mitochondrial transmembrane potential measurement and ATP content detection.Statistical analysis was made on the test results. Results After ischemia reperfusion injury, the levels of MB,LPO,and LPC in skeletal muscle, liver, lung and renal tissues of the control group were significantly increased(MB:P =0.003,P =0.001,P =0.001,P =0.001;LPO:P =0.001,P =0.001,P =0.001,P =0.002;LPC:P =0.000,P =0.002,P =0.002,P =0.003),and hydrogen sulfide treatment during ischemia reperfusion significantly inhibited the production of MB,LPO,and LPC(MB:P =0.021,P =0.036,P =0.005;LPO:P =0.003,P =0.008,P =0.010,P =0.015;LPC:P =0.002,P =0.026,P =0.007,P =0.006).Ischemia/reperfusion of lower extremity in rats resulted in increased levels of IL-1,IL-6,and TNF-α in the serum of rats, and the levels of IL-1,IL-6,and TNF-increased over time, with statistically significant differences in IL-1,IL-6,and TNF-α among groups at 3 h(IL-1:P =0.019,P =0.011,P =0.009,$P_{12_{h}}$=0.008,and P =0.002;IL-6:P =0.026,P =0.009,P =0.002, $P_{12_{h}}$=0.002,P =0.003;TNF-α:P =0.002,P =0.002,P =0.005,$P_{12_{h}}$=0.002,P =0.003).The levels of IL-1,IL-6,and TNF-α in serum were significantly inhibited during ischemia reperfusion(IL-1:P =0.035,P =0.039,P =0.012,$P_{12_{h}}$=0.005,P =0.006;IL-6:P =0.042,P =0.025,P =0.023,$P_{12_{h}}$=0.006,P =0.005;TNF-α:P =0.005,P =0.003,P =0.022,$P_{12_{h}}$=0.005,P =0.005),and such inhibitory effects became even more obvious over time. After limb ischemia and reperfusion in the control group, the mitochondrial transmembrane potential of skeletal muscle cells significantly decreased compared with that of the sham group(t=6.698;P=0.001).After hydrogen sulfide treatment, the mitochondrial membrane potential energy of the experimental group was significantly higher than that of the control group(t=7.507,P = 0.000).The ATP level in the mitochondria of ischemia reperfusion rats in the control group was significantly lower than that in the sham group(t=7.526,P = 0.000).The content of mitochondrial ATP in the experimental group was significantly higher than that in the control group after hydrogen sulfide treatment(t=8.604,P = 0.000). Conclusions Hydrogen sulfide can alleviate the injury of skeletal muscle and distal organs after limb ischemia-reperfusion and reduce local inflammatory reaction. In addition,it is valuable in alleviating mitochondrial transmembrane potential and energy metabolism disorders during reperfusion injury.

Objective:To investigate the mediating roles of perceived social support and relative deprivation in the relationships between life events and Internet addiction among Chinese college students. Methods:Totally 413 college students (200 males and 213 females, aged 16-21 years),completed the Relative Deprivation Questionnaire,Adolescent Self-Rating Life Events Check List,Perceived Social Support Scale and Chinese Internet Addiction Scale Revised to measure the level of relative deprivation, life events, perceived social support and Internet addiction. The Bollen-Stine of structural equation modeling and Bootstrap were used to test the mediating effect of perceived social support and relative deprivation on the relationship between life events and Internet addiction. Results:The scores of PSSS were negatively correlated with the scores of ASLEC,RDQ,and CIAS-R (r=0.11-0.36,Ps <0.05).The scores of ASLEC,RDQ,and CIAS-R were positively correlated with each other (r=0.14-0.23,Ps < 0.01).Path analyses showed that the ASLEC scores were not associated with the CIAS-R scores directly (β =0.11,P >0.05),but indirectly through the mediating roles of the scores of PSSS and RDQ and the chain mediating effect of them among Chinese college students. The structural equation model could be accepted by Bollen-Stine Bootstrap (P <0.01).The confidence interval from Bootstrap output of the mediating effects of the scores of PSSS and RDQ and the chain mediating effect of them were (0.010,0.103),(0.002,0.032) and (0.005,0.061) respectively. Conclusion:It suggests that life events could not directly affect Internet addiction, but indirectly lead to Internet addiction through reducing perceived social support and increasing relative deprivation.
RESUMO

OBJECTIVEThis study investigated the history and gene mutations of a family with X-linked thrombocytopenia, in order to understand the clinical characteristic and molecular pathogenesis of the disease.

METHODSA three-generation X-linked thrombocytopenia family with 13 family members was investigated using PCR-DNA direct sequencing method to screen the exons of WASP gene for mutation analysis.

RESULTSThe WASP gene sequencing of the proband revealed a missense mutation in exon 2 (G291A), resulting in a change of amino acid 86 from arginine to histidine. The patient's mother was the carrier of the heterozygosis mutation in X-chromosome.

CONCLUSIONSWASP mutations may be attributed to the molecular mechanism of X-linked thrombocytopenia. G291A is one of the mutations of WASP.

OBJECTIVETo investigate the effect of flying burthen on blood lipid level in civil aviation flight personnel.

METHODSThe level of total cholesterol (TC), triglyceride (TG) and high density lipoprotein cholesterol (HDL-C) were determined by kits. The relationship between the blood lipid levels and the flight personnel's flying hours, body weight index, smoking, drinking and sport were investigated.

RESULTSThe average annual flying hours of hyperlipidemia group [(561.14 +/- 234.90) h] was significantly higher than that of non-hyperlipidemia group [(500.62 +/- 243.65) h] (P < 0.05). The level of TG of the average annual flying hours more 800 h [(1.61 +/- 0.10) mmol/L] were significantly higher than that of less than 400 h group [(1.31 +/- 0.65) mmol/L]. The morbidity rate of hyperlipidemia among the age 30 to 39 years old flight personnel was the highest.

CONCLUSIONThe average annual flying hours is one of primary factors affecting blood lipid in flight personnel.
