RESUMO

Objective To search the differenially expressed proteins in the serum of patients with unruptured intracranial aneurysm.Methods Eight patients with unruptured intracranial aneurysm,admitted to our hospital from July 2007 to September 2009,and 8 healthy adults(controls)were recruited in our study; two-dimensional differential in-gel electrophoresis(2D DIGE)and matrix-assisted laser desorption/ionization-time of flight mass spectrometry(MALDI-TOF-MS)were employed to analyze and identify the differentially expressed proteins in the serum of these patients and controls.Results As compared with the control group,patients with unruptured intracranial aneurysm had 29 differentially expressed proteins those enjoyed more than 1.5 folds differences; protein at spot 579was expressed as low as 1.81 folds in the serum of patients with unruptured intracranial aneurysm as compared with that in controls(P=0.008); the protein was identified as afamin using MALDI-TOF-MS.Conclusion The low expression of afamin in patients with unruptured intracranial aneurysm probably contributes to the formation and expansion of intracranial aneurysm.

RESUMO

Objective To evaluate the feasibility and advantages ofmidfrontai keyhole approach for treatment of hypertensive ventricular hemorrhage cast. Methods Thirty patients with hypertensive ventricular hemorrhage cast were randomized into 2 groups to receive ventricular drainage (group A) or microsurgical hematoma elimination through midfrontal keyhole approach (group B). The clinical outcomes of the patients were assessed according to the Glasgow Outcome Scale (GOS) after 3-6 months of follow-up. Results IngroupA, 6 patients had GOS grade l, 6 had grade 2-3, and 3 had grade 4-5 outcomes. In group B, 1 patient had GOS grade 1, 2 had grade 2-3, and 12 had grade 4-5 outcomes. The patients in group B had significantly better outcomes than those in group A. Conclusion Microsurgical hematoma elimination through the midfrontal keyhole approach produces better therapeutic effect than ventrieular drainage in the treatment of hypertensive ventricular hemorrhage cast.

Objective To identify G protein signal transduction pathway related genes in peripheral blood mononuclear cells and aneurysm tissues from intracranial aneurysm patients using oligo-microarrays. Methods Blood samples from 50 intracranial aneurysms and 10 healthy human donors were obtained after informed consent. Peripheral blood mononuclear cells (PBMCs) were isolated from blood using Ficoll method. And aneurysm tissue samples from 6 intracraniai aneurysms and 3 healthy human intracranial arteries were obtained. The cDNA were retrotranscribed from the extracted RNA and the biotin-labeled cRNA derived from the transcription of cDNA were fragmented as probes. Then, the probes were hybridized with Affymetrix Human Genome UI33A Array. GeneArmy Scanner was used to screen the signals of hybridization and Microarray Suite software 5.0 was applied to analyze the expression of genes. Results There were 325 genes differently expressed in PBMCs from intracranial aneurysm patients compared to healthy human. And 571 genes were differently expressed above 2-fold level in aneurysmal tissue from intracranial aneurysm patients compared to healthy human intracranial arteries. Function analysis was confirmed from upregulated genes and downregulated genes, and among these genes, there were several genes invovled in G protein signal transduction pathway. There were 3 genes directionally expressed (RA831 upregulated and ARHGAP8, CENTG2 downregulated) both in PBMCs and aneurysm tissues from intracranial aneurysm patients. Conclusions The methodologies of genechip will provide a new powerful approach to help identify the molecular mechanisms of intracranial aneurysm. G protein signal transduction pathway may be the possible mechanism for the pathogenesis of intracranial aneurysms in Chinese.

RESUMO

<p><b>OBJECTIVE</b>To study the effects of Dihydromyricetin (DMY) on antilipid-peroxidation.</p><p><b>METHOD</b>The antilipid-peroxidation of DMY on heart, liver, brain tissue homogenate and mitochondria was measured by the determination of malondiadehyde (MDA) induced by Fe2+ -Vit C, Fe2+ -H2O2, Fe-Cys with TBA spectrometric method.</p><p><b>RESULT</b>DMY could inhibit the lipid peroxidation of homogenate and mitochondria. The inhibition exhibited concentration-dependent manner.</p><p><b>CONCLUSION</b>DMY has good antilipid-peroxidation effect, which is worth studing further.</p>
