RESUMO

Triterpenoids have been described in Andrographis paniculata. Oleanolic acid exhibits high biological activity and is widely used in the clinic, and β-sitosterol not only has good biological activity but also plays an important physiological role in plants. However, analysis of the biosynthetic pathway of triterpenoids in Andrographis paniculata has not been reported. Here, we provide the first report of the isolation and identification of nine 2, 3-oxidosqualene cyclases (ApOSC3 to ApOSC11) from A. paniculata. The results showed that ApOSC4 represented a monofunctional synthase that could convert 2, 3-oxidosqualene to β-amyrin. ApOSC5 as a bifunctional 2, 3-oxidosqualene cyclases, could transfer 2, 3-oxidosqualene to β-amyrin and α-amyrin. ApOSC6 to ApOSC8 composed the multifunctional 2, 3-oxidosqualene cyclases that could convert 2, 3-oxidosqualene to β-amyrin, α-amyrin and one or two undetermined triterpenoids. This study provides a better understanding of the biosynthetic pathway of triterpenoids in A. paniculata, and the discovery of multifunctional 2, 3-oxidosqualene cyclases ApOSC5 to ApOSC8 of the facilitates knowledge of the compounds diversity in A. paniculata.

RESUMO

Ent-labdane diterpenoid lactones, the principal constituents of Andrographis paniculata, have a wide range of pharmacological effects, such as antibacterial, anti-viral, anti-inflammatory, antipyretic, andanti-cancer effect, etc. In this review the biotransformations of four principal ent-labdane diterpenoid lactones of A. paniculata are discussed from the following aspects: substrates and types of reaction, transformation systems, and structure activity relationships. Moreover, the currently-existing problems and the prospects are also described to provide reference for further studying the biotransformation of ent-labdane diterpenoid lactones of A. paniculata.

RESUMO

Andrographis paniculata (Acanthaceae) has been used to treat hepatoprotective, antidiarrhoeal, antiinflammatory, antimalarial, and antihypertensive. The aim of the study was to investigate the antifungal activity of different part of extracts of A. paniculata. The antifungal activity of extracts of A. paniculata was evaluated by Agar well diffusion method against five selected fungal species. Stem extracts of A. paniculata showed high antifungal activity against A.oryzae, Penicillum sp and C.albicans. The root extracts showed high antifungal activity against A. niger, A. flavus, C.albicans, Penicillum sp and A.oryzae and also leaf extracts showed high antifungal activity against Penicillum sp and A. flavus but did not show antifungal activity against C.albicans, A. niger, A.oryzae. The results obtained in the present study suggest that A. paniculata plant can be used in treating various diseases caused by the test organisms.
