Objective: To observe the inhibitory effects of curcumin on hyphal development and biofilm formation of five kinds of non-Candida albicans. Methods: Serial 2-fold dilution assay was used to determine the MICs of curcumin to C. tropicalis, C. glabrata, C. parapsilokis, C. krusei, C. guilliermondii; XTT assay was used to determine the SMIC50 of curcumin to the five non-C. albicans. Inverted microscope and scanning electron microscope (SEM) were applied to inspectting the morphological change of non-C. albicans treated by curcumin, The dilution method was applied to inspecting the hyphae around the colonies. Results: The MICs of curcumin to C. tropicalis, C. glabrata, C. parapsilokis, C. Krusei, C. guilliermondii were 64, 128, 256, 256, and 128 μg/mL, and the SMIC50 were 512, 512, >512, >512, and 512 μg/mL, respectively. We found that curcumin could inhibit the hyphal and biofilm formation of the five kinds of non-C. albicans by observation with inverted microscope and SEM, and curcumin could inhibit the hyphal development around the colonies by observation with optical microscope. Conclusion: Curcumin could inhibit the hyphal development and biofilm formation of the five kinds of non-C. albicans.

The use of hemicellulosic hydrolysates in bioprocesses requires supplementation as to ensure the best fermentative performance of microorganisms. However, in light of conflicting data in the literature, it is necessary to establish an inexpensive and applicable medium for the development of bioprocesses. This paper evaluates the fermentative performance of Scheffersomyces (Pichia) stipitis and Candida guilliermondii growth in sugarcane bagasse hemicellulosic hydrolysate supplemented with different nitrogen sources including rice bran extract, an important by-product of agroindustry and source of vitamins and amino acids. Experiments were carried out with hydrolysate supplemented with rice bran extract and (NH4)2SO4; peptone and yeast extract; (NH4)2SO4, peptone and yeast extract and non-supplemented hydrolysate as a control. S. stipitis produced only ethanol, while C. guilliermondii produced xylitol as the main product and ethanol as by-product. Maximum ethanol production by S. stipitis was observed when sugarcane bagasse hemicellulosic hydrolysate was supplemented with (NH4)2SO4, peptone and yeast extract. Differently, the maximum xylitol formation by C. guilliermondii was obtained by employing hydrolysate supplemented with (NH4)2SO4 and rice bran extract. Together, these findings indicate that: a) for both yeasts (NH4)2SO4 was required as an inorganic nitrogen source to supplement sugarcane bagasse hydrolysate; b) for S. stipitis, sugarcane hemicellulosic hydrolysate must be supplemented with peptone and yeast extract as organic nitrogen source; and: c) for C. guilliermondii, it must be supplemented with rice bran extract. The present study designed a fermentation medium employing hemicellulosic hydrolysate and provides a basis for studies about value-added products as ethanol and xylitol from lignocellulosic materials.

Las alteraciones de frutos de tomate poscosecha por causas microbiológicas conllevan a su deterioro fisiológico produciendo su descomposición. En este estudio, Candida guilliermondii fue examinada en ensayos in vitro e in vivo para determinar su eficacia en el control biológico contra Rhizopus stolonifer en tomates (Lycopersicon esculentum). El efecto antagónico se llevo a cabo usando biomasa de la levadura y sus metabolitos los cuales se identificados en un cromatógrafo gases-masas. Los ensayos in vitro mostraron que la única fase con respuesta antagónica al patógeno fue la fase acuosa sin filtrar. En los ensayos in vivo, la biomasa de Candida guilliermondii presento el mayor porcentaje de protección contra el patógeno (87 %) en comparación con la fase acuosa sin filtrar y la fase orgánica sin concentrar que presentaron el 77 y 80 % de protección respectivamente. Estos resultados son de gran importancia para evitar las perdidas económicas además de garantizar un tiempo de vida mas largo del tomate asegurando una mejor distribución del producto y evitando sus perdidas en estos tiempos de crisis alimentaria en especial en países en desarrollo.


Post-harvest losses in tomato crops can be accredited to a number of causes, which include biological causes that produce physiological deterioration; infection caused by diseases and pests that cause rotting. In this study, we assayed, in vitro and in vivo, the yeast Candida guilliermondii to determine its effectiveness as a biocontrol to reduce the rotting produced by Rhizopus stolonifer on tomatoes (Lycopersicon esculentum). The antagonistic effect was tested using a yeast biomass suspension; gas chromatography and a mass selective detector were used to identify its metabolites. According to the in vitro test, the only phase with an antagonistic response against Rhi%opus stolonifer was the unfiltered water phase. In the in vivo tests, the biomass of Candida guilliermondii presented the highest percentage of protection of the tomato (87 %) compared with the unfiltered aqueous phase and the unconcentrated organic phase, which reached only 77 and 80 % respectively. These results are essential to avoid economic losses caused by fungi and to increase the shelf life of tomatoes improving the distribution of the product and preventing postharvest losses in this time of food crises, especially in developing countries.


As perdas pós-colheita de frutos de tomate são creditadas em várias fontes, incluindo causas biológicas que levam ao deterioro fisiológico e infecção por doenças e pragas que podem levar à sua decomposição. Neste estudo, a levedura Candidaguilliermondii foi examinada in vitro e in vivo para determinar a sua eficácia no controlo biológico de Rhizopus stolonifer em tomates (Lycopersicon esculentum). O efeito antagonista realizou-se utilizando biomassa de levedura e os seus metabolitos, os quais foram identificados por cromatografia gasosa acoplada a um detector selectivo de massa. Os ensaios in vitro mostraram que a única fase com resposta antagonista a Rhizopus stolonifer foi a fase aquosa não filtrada. Nos ensaios in vivo, a biomassa Candida guilliermondii teve a maior percentagem de protecção contra o patógeno (87%) em comparação com a fase aquosa não filtrada e a fase orgânica não concentrada apresentando 77 e 80 % de protecção, respectivamente. Estes resultados são muito importantes para evitar perdas económicas causadas por fungos, além de garantir ao tomate uma vida útil mais longa, o que assegura uma melhor distribuição do produto e evita perdas pós-colheita neste momento de crise alimentar, especialmente nos países em desenvolvimento.

RESUMO

A preliminary study on xylitol production by Candida guilliermondii in sorghum straw hemicellulosic hydrolysate was performed. Hydrolysate had high xylose content and inhibitors concentrations did not exceed the commonly found values in other hemicellulosic hydrolysates. The highest xylitol yield (0.44 g/g) and productivity (0.19 g/Lh) were verified after 72 hours.

La levadura Candida guilliermondii es objeto de estudio debido a su capacidad de producir xilitol aprovechando compuestos hemicelulósicos ricos en xilosa, dado esto, la cepa Candida guilliermondii aislada del fruto del corozo chiquito (Bactris guineensis) fue usada en este estudio con el fin de evaluar su capacidad para producir xilitol sobre un sustrato hidrolizado de cascarilla de arroz. El objetivo de este trabajo fue determinar los parámetros fermentativos como producción de xilitol, productividad volumétrica (Qp) y rendimiento de sustrato en producto (Yp/s) durante la fermentación con la cepa nativa Candida guilliermondii. Se emplearon 200 ml de medio de cultivo hidrolizado de cascarilla de arroz, el cual contenía una concentración de xilosa de 27,5 g/L. La fermentación se llevó a cabo bajo las siguientes condiciones: temperatura 30 ºC, pH del medio 5,8, agitación 120 rpm e inóculo adaptado de 3 g/L. Los resultados mostraron que después de 120 horas de fermentación se obtuvieron 2,6 g/L de xilitol con productividad volumétrica (Qp) de 0,02 g/L-h y rendimiento de sustrato en producto (Yp/s) de 0,13 g/g. De esta manera, la cepa nativa Candida guilliermondii, aislada del fruto de Corozo chiquito (Bactris guineensis), produjo xilitol bajo condiciones específicas de fermentación.


The yeast Candida guilliermondii has been studied due to its ability to produce xylitol in xylose-rich hemicellulosic compounds, Candida guilliermondii strain isolated from the fruit of Corozo chiquito (Bactris guineensis) was used in this study to assess their ability to xylitol production on these substrates. The aim of this study was to determine the fermentation parameters such as xylitol production, volumetric productivity (Qp) and yield of xylitol production (Yp/s) during fermentation with the native strain Candida guilliermondii. Was used 200 ml of culture medium rice husk hydrolysate, which contained a xylose concentration of 27.5 g/L. The fermentation was carried out under the following conditions: temperature 30 ºC, pH of 5.8, agitation 120 rpm and adapted inoculum of 3 g/L. The results showed that after 120 hours of fermentation 2.6 g / L of xylitol was achieved with volumetric productivity (Qp) 0.02 g/L-h and 0.13 g/g yield of xylitol production (Yp/s). The native strain Candida guilliermondii, isolated from the fruit of Corozo chiquito (Bactris guineensis) produced xylitol fermentation under specific conditions.

RESUMO

Candida guilliermondii is found in sea water, animal feces, buttermilk, and beer and has been isolated from human infections, mostly of cutaneous origin. It usually causes skin and soft tissue infections and has decreased susceptibility to fluconazole. Systemic infections occur mostly in immunocompromised patients. A 38-year-old female was admitted with a 4-day fever. Her medical and family histories were unremarkable, except for obesity. She had been taking diet pills for 3 months and had undergone injection therapy into her abdomen for lipolysis for 1 month. She did not respond to empirical antibiotics. A Candida species was isolated from blood cultures and identified as C. guilliermondii based on partial LSU rRNA gene sequence analyses. She was treated with amphotericin B, and recovered completely.

Wheat straw hemicellulosic hydrolysate was used for xylitol bioproduction. The use of a xylose-containing medium to grow the inoculum did not favor the production of xylitol in the hydrolysate, which was submitted to a previous detoxification treatment with 2.5 percent activated charcoal for optimized removal of inhibitory compounds.


Hidrolisado hemicelulósico de palha de trigo foi utilizado para a bioprodução de xilitol. O uso de meio contendo xilose para crescer o inóculo não favoreceu a produção de xilitol no hidrolisado, que foi submetido a um tratamento prévio de destoxificação com 2.5 por cento de carvão ativo para remoção otimizada de compostos inibitórios.

Candida is the most common etiologic agent causing endogenous endophthalmitis resulting due to hematogenous spread from a remote primary focus. Risk factors for the infection include intravenous drug use, hyperalimentation, surgery, malignancy, diabetes, neutropenia, and the use of broad- spectrum antibiotics and immunosuppressive agents, especially corticosteroids. The outcome of candida endophthalmitis is disappointing. One main problem in the management of this infection is that early diagnosis is difficult. Thus, treatment may be delayed and this which often leads to a poor outcome. Candida endophthalmitis, particularly candida guilliermondii endophthalmitis, is extremely rare, although it is becoming more common as the number of chronically debilitated patients and the use of invasive procedures increase. It is an ophthalmologic emergency and commonly takes a tragic course. Therefore, early suspicion and aggressive management are imperative to prevent visual loss. The authors report a case of candida endophthalmitis caused by Candida guilliermondii in a 65-year-old man with mixed gastrointestinal stromal tumor(GIST) and stomach adenocarcinoma.

Yeasts of the genus Candida are of clinical importance and also have many industrial applications, mainly in the food industry. The yeast Candida guilliermondii FTI 20037 has been extensively studied in order to establish a biotechnological process for the production of xylitol. The goal of this study was to verify the taxonomic classification of this strain based on the analysis of rDNA sequences and the xyl1 gene. DNA fragments from these sequences were amplified by PCR and BLAST analysis revealed strong identity with the corresponding sequences from Candida tropicalis. Based on these results, we propose that C. guilliermondii FTI 20037 must be reclassified as C. tropicalis.


As leveduras do gênero Candida possuem tanto importância clínica como diversas aplicações industriais, principalmente na indústria de alimentos. A levedura Candida guilliermondii FTI 20037 tem sido exaustivamente estudada pois pretende-se utilizá-la no estabelecimento de um processo biotecnológico para a produção de xilitol. O objetivo deste trabalho foi verificar a classificação taxonômica desta levedura por análise de sequências do rDNA e do gene xyl1. Fragmentos correspondentes a estas regiões foram amplificados por PCR e a análise destas sequências por BLAST revelou alta identidade com sequências correspondentes de Candida tropicalis. Estes resultados nos levam a propor que C. guilliermondii FTI 20037 deva ser reclassificada como C. tropicalis.

Candida guilliermondii FTI 20037 was grown on a mixed substrate comprising glucose and xylose. Inocula were grown using xylose or glucose as carbon source. Results showed that xylose utilization was delayed until glucose was utilized. Inoculum prepared on glucose showed a lag phase in xylose consumption. Cell mass production was higher when glucose was utilized during fermentation.
