En el beneficiado del cacao se producen una gran cantidad de residuos, las mazorcas generalmente son desechadas dentro de los mismos cultivos y genera problemáticas como la proliferación de insectos y microorganismos patógenos. De estos desechos las cáscaras son las de mayor relevancia. La Escuela de Ingeniería Química de ITCA-FEPADE evaluó añadirle valor a este subproducto, incorporando desechos de cáscara de cacao en la formulación de un sustrato para el cultivo de plántulas de hortalizas. El objetivo de esta investigación fue obtener un sustrato orgánico que sirva de soporte material y nutritivo a partir de cáscaras de cacao criollo. La biomasa vegetal se caracterizó teniendo en cuenta parámetros como el porcentaje de humedad, pH, porcentaje de cenizas, contenido de potasio, nitrógeno y fósforo. Se ejecutaron pruebas comparativas de formulación del sustrato, siembra, cultivo y crecimiento de las plántulas de hortalizas de tomate y pepino, obteniendo como resultado la fórmula óptima de un sustrato y abono orgánico y el mejor medio de desarrollo. De los resultados obtenidos se concluye que, a partir de un adecuado procesamiento y aprovechamiento de las cáscaras de cacao, se puede transformar este desecho en un producto biomaterial alternativo que genera una opción de bioprospección agroindustrial. Los valores obtenidos en la caracterización fisicoquímica de las cáscaras de las mazorcas, dependen de condiciones como el tipo de suelo, variables agrometeorológicas, calidad de agua, abono y especie de la planta de cacao. El escenario de siembra condiciona el desarrollo óptimo de las plántulas; los factores como requerimiento de agua, distribución de nutrientes y estabilidad de las plántulas, se ven afectados por la relación del espacio de germinación. Como resultado de la caracterización fisicoquímica, se obtuvo: pH de 5.7, cenizas 18.83%, humedad 73.56%, celulosa 21.39%, lignina 39.81%, nitrógeno total 0.02%, fósforo total 0.02% y ausencia de potasio.
In the cacao beneficiation, a large amount of waste is produced, generally, the cacao pods are discarded within the same crops and generate problems such as the proliferation of insects and pathogenic microorganisms. The shell are the most relevant this waste. In Escuela de Ingeniería Química of ITCA-FEPADE the incorporation of cacao shell waste in the formulation of a substrate for growing vegetables was evaluated because the giving benefit to these by-products. The objective of this research was to obtain a substrate that serves as material and nutritional support for vegetable seedlings, from creole cacao shells. The vegetal biomass was characterized taking into account parameters such as moisture percentage, pH, ash percentage, potassium, nitrogen and phosphorus content. Subsequently, comparative tests of formulation, planting and growth of vegetable tomato and cucumber seedlings were carry out, obtaining as result, the optimal formula for the substrate and the best development environment. It concludes that, through an adequate processing and use of the cacao shell, it is possible to transform a waste into an alternative biomaterial product that generates an agro industrial bioprospecting option. The values obtained in the physicochemical characterization of the shells of cacao pods depend on conditions such as soil type, agrometeorological variables, water quality, fertilizer and cacao plant species. The planting environment determines the optimal development of the seedlings, factors such as water requirement, nutrient distribution and seedling stability seems affected because relation of the germination space. The results of the characterization were a pH of 5.7, ashes 18.83%, humidity 73.56%, cellulose 21.39%, lignin 39.81%, total nitrogen 0.02%, total phosphorus 0.02%, and total phosphorus 0.02% and an absence of potassium.

Endophytic microorganisms live inside the plants without causing any damage to their hosts. In the agricultural field, these endophytes might be a strategy of biological control for phytopathogens. We aimed to isolate endophytic fungifrom yellowpassion fruit (Passiflora edulis) leaves, evaluating its biocontrol capacity by in vitroantagonism against phytopathogen Colletotrichum sp. CNPU378. We also carried out greenhouse experiments in bean seedlings. A high colonization frequency was obtained (89%), and the molecular identification based on DNA sequencing attested Colletotrichumas the most frequent genus and minor occurrence of Curvulariaendophytes. The endophytes tested showed different types of competitive interactions in in vitro antagonism inhibition rate ranging from 28.8 to 48.8%. There were 10 promising antagonists tested for their antagonist activity of crude extracts of secondary metabolites, in which strain PE-36 (20.8%) stood out among the other strains evaluated. In the greenhouse assay, plants inoculated only with endophyte Colletotrichumsp. PE-36 was symptomless and suggest that the endophyte strengthened the growth promotion in common bean plants, especially in the root length and number of leaves when compared to control plantsand other treatments. Despite many fungiof Colletotrichumgenus being described as causative agents of anthracnose, in this study, the plant sampled was colonized predominantly by Colletotrichumendophytes living in asymptomatic relationship. By the way,we come across a Colletotrichumsp. endophyte able to antagonize a Colletotrichumsp. pathogen

Since drug-resistant nematodes became a common problem in sheep and goat industries, alternative methods using natural products have emerged as a viable and sustainable anthelmintic treatment option. Here, the in vitro effect of essential oil extracted from Lippia gracilis Schauer was assessed on the hatching process of nematodes recovered from naturally infected goats. Essential oil at concentrations of 0.08% (0.008 µL/mL), 0.12% (0.012 µL/mL), and 0.16% (0.016 µL/mL) was able to induce an average inhibition of 74.7, 84 and 93%, respectively. The effective concentration required to inhibit egg hatching in 50% of eggs (EC50) was 0.03452%. Therefore, essential oil of L. gracilis showed promisor in vitro anthelmintic results against egg-hatching of goat gastrointestinal nematodes.(AU)
Como os nematoides resistentes a drogas se tornaram um problema comum nas indústrias de ovinos e caprinos, métodos alternativos que utilizam produtos naturais surgiram como uma opção de tratamento anti-helmíntico viável e sustentável. Aqui, o efeito in vitro do óleo essencial extraído de Lippia gracilis Schauer foi avaliado no processo de eclosão de nematoides recuperados de caprinos naturalmente infectadas. O óleo essencial nas concentrações de 0,08% (0,008 µL/mL), 0,12% (0,012 µL/mL), e 0,16% (0,016 µL/mL)foi capaz de induzir uma inibição média de 74,7, 84 e 93%, respectivamente. A concentração efetiva necessária para inibir a eclosão de ovos em 50% dos ovos (CE50) foi de 0,03452%. Portanto, o óleo essencial de L. gracilis apresentou resultados anti-helmínticos in vitro promissores contra a eclosão de nematódeos gastrintestinais de caprinos.(AU)

Introducción: La bioprospección de metabolitos de interés antropogénico emplea métodos de recolección de microorganismos en ecosistemas extremófilos o endémicos. La microbiota aislada en estos lugares puede o no incluir microorganismos patógenos. Es imprescindible un enfoque interdisciplinario que permita abordar la búsqueda de las especies de interés mientras se preserva la buena salud de los investigadores. Objetivo: Identificar molecular, bioquímica y morfológicamente microorganismos patógenos humanos en cepas celulolíticas de importancia industrial almacenadas en el banco de cepas del Laboratorio de Investigación de la Facultad de Ingeniería Química de la Universidad Central del Ecuador, procedentes del Yasuní, la Antártida y Balzapamba. Métodos: Se realizó un estudio de bioprospección de bacterias celulolíticas empleando técnicas de microbiología ambiental. Se evaluaron las características morfológicas mediante tinciones, como por ejemplo Gram. Además, se realizaron pruebas bioquímicas y antibiogramas para bacterias Gram-negativas y Gram-positivas. Las pruebas moleculares utilizaron extracción de ADN bacteriano para la secuenciación Sanger del gen 16S. Resultados: Se encontraron las especies Klebsiella pneumoniae (Y2 y Y3r) y Nocardia asteroides (Y1 y Y3p) en las muestras de material lignocelulósico recolectadas en Yasuní, mientras que las especies aisladas en la Antártida y en Balzapamba corresponden a Bacillus subtillis. Conclusiones: Se identificaron cepas pertenecientes a diferentes géneros bacterianos. Las bacterias del género Klebsiella, en las muestras colectadas en Yasuní, podrían tener un potencial patógeno. Eso se puede corroborar con técnicas de genotipificación. Por lo tanto, puede existir riesgo para los seres humanos que realizan bioprospección en ese ecosistema y se deben tomar medidas de bioseguridad.
Abstract Background: The bioprospection of metabolites of anthropogenic interests employs methods of collecting microorganisms in extremophile or endemic ecosystems. The microbiota isolated in these places may or may not include pathogenic microorganisms. Therefore, an interdisciplinary approach is essential to address the search of the species of interest while the good health of the researchers is preserved. Objective: To identify in molecular, biochemical and morphologically ways some human pathogenic microorganisms in cellulolytic strains of industrial importance stored in the strain bank of the Research Laboratory of the Faculty of Chemical Engineering at the Central University of Ecuador, from Yasuní, Antarctic and Balzapamba. Methods: IA bioprospecting study of cellulolytic bacteria was performed using environmental microbiology techniques. Morphological characteristics were assessed by Gram staining. In addition, biochemical tests and antibiograms were performed for Gram-negative and Gram-positive bacteria. The molecular tests used extraction of bacterial ADN for 16S gene Sanger sequencing. Results: The species Klebsiella pneumoniae (Y2 and Y3r) and Nocardia asteroides (Y1 and Y3p) were found in samples of lignocellulosic material collected in Yasuni, while the isolated species in Antarctica and Balzapamba correspond to Bacillus subtillis. Conclusions: Strains belonging to different bacterial genera were identified. The bacteria of the genus Klebsiella from the samples collected in Yasuní could have a potential pathogen. This can be corroborated with genotyping techniques. Therefore, there could be a risk to humans who perform bioprospecting in that ecosystem and biosecurity measures should be taken.

Plant species have been used for therapeutic purposes since ancient times and are still in use today since these products represent a source of raw material for the production of phytotherapeutic formulations. Screening and investigation of plants with pharmacological potential require the evaluation of characteristics related to their action, efficacy and safety in different steps. Among these steps, pre- clinical trials are used to evaluate the properties of the test product in in vitro experiments, such as cytotoxicity assays. Within this context, this study consists of a bibliometric analysis of some in vitro cytotoxicity and toxicity assays in erythrocytes used during bioprospecting of medicinal plants. The results demonstrated the wide application of erythrocytes to evaluate the biological effects of medicinal plant extracts. The methods were found to be valid and effective for the preliminary investigation of the in vitro cytotoxicity and toxicity of plant products.
El uso de especies vegetales para fines terapeÌuticos es una praÌctica histoÌrica y todaviÌa bastante actual, ya que estos productos pueden representar una fuente de materia prima para la produccioÌn de formulaciones fitoteraÌpicas. En investigacioÌn de plantas con potencial farmacoloÌgico requiere la evaluacioÌn de su accioÌn, eficacia y seguridad, a traveÌs de diferentes etapas. Entre estas, en los ensayos precliÌnicos se evaluÌan las propiedades del producto-prueba en experimentos in vitro, tales como ensayos de citotoxicidad, entre otros. En este aspecto, el presente estudio consiste en un anaÌlisis bibliomeÌtrico acerca de algunas pruebas de citotoxicidad y toxicidad in vitro en eritrocitos realizados en los ensayos de bioprospeccioÌn de plantas medicinales. Los resultados evidencian la amplia utilizacioÌn de eritrocitos para la evaluacioÌn de los efectos bioloÌgicos de extractos de plantas medicinales, apuntaÌndolos como meÌtodos vaÌlidos y eficaces para la investigacioÌn preliminar de la citotoxicidad y toxicidad in vitro de productos vegetales.

The skin secretion from toads of the Bufonidae family has great potential in the search for new active compounds to be used as drug candidates in treating some diseases, among them cancer. In this context, this study aimed to evaluate the cytotoxic and antimutagenic activity of the parotoid gland secretion extracts of Rhinella marina and Rhaebo guttatus, as well as biochemically analyze transaminases and serum creatinine for liver and renal damage, respectively. Cytotoxicity was performed by the colorimetric method based on MTT (3- [4, 5-dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium bromide) with different concentrations of the extracts in Walker or splenic tumor cell cultures from rats and mice. The micronucleus test was performed with male Swiss mice treated orally with the extracts for 15 days, and then intraperitoneally with N-ethyl-N-nitrosurea (50 mg kg-1). Micronucleated polychromatic erythrocytes (MNPCE) were evaluated in bone marrow. The extracts showed cytotoxic activity in the evaluated cells. There was a significant reduction in the frequency of MNPCE (R. marina = 56% and R. guttatus = 75%, p < 0.001), indicating antimutagenic potential of the extracts. The groups treated only with extract showed an increase in MNPCE frequency, evidencing mutagenic potential. Biochemical analyzes showed no significant difference between treatments. Thus, under our experimental conditions, the extracts of R. marina and R. guttatus skin secretions presented chemopreventive potential for cancer. (AU)
A secreção cutânea de anuros da família Bufonidae tem grande potencial na busca de novos compostos ativos para utilização como fármacos candidatos no tratamento de algumas doenças, entre elas o câncer. Neste contexto, este estudo teve como objetivo avaliar a atividade citotóxica e antimutagênica dos extratos da secreção da glândula parótida de Rhinella marina e Rhaebo guttatus, bem como a análise bioquímica de transaminases e creatinina séricas, para avaliar dano hepático e renal, respectivamente. A avaliação de citotoxicidade foi realizada pelo método colorimétrico baseado no MTT (3-[4, 5-dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium bromide), com diferentes concentrações dos extratos em culturas de células do Tumor de Walker ou células esplênicas de rato e camundongo. O teste do micronúcleo foi realizado com camundongos Swiss machos que receberam tratamento oral com os extratos durante 15 dias, seguido de tratamento intraperitoneal com N-etil-N-nitrosuréia (50 mg kg-1). A frequência de eritrócitos policromáticos micronucleados (PCEMN) foi determinada em medula óssea. Os extratos apresentaram ação citotóxica nas células avaliadas. Houve uma redução significativa na frequência de PCEMN (R. marina = 56% e R. guttatus = 75%, p < 0,001), observando-se um potencial antimutagênico dos extratos. Os grupos tratados somente com os extratos apresentaram um aumento na frequência de PCEMNs, evidenciando um potencial mutagênico. As análises bioquímicas não apresentaram diferença significativa entre os tratamentos. Assim, nas condições experimentais testadas, as secreções cutâneas de R. marina e R. guttatus apresentaram potencial quimiopreventivo para câncer.(AU)

RESUMO A formação de biofilmes e produção de hifas por Candida albicans são importantes fatores de virulência, principalmente, para a aderência à mucosa e invasão tecidual. A busca por metabólitos secundários produzidos por S. mutans é de suma importância, pois poderá fornecer novas estratégias terapêuticas no combate às infecções por Candida, possibilitando o desenvolvimento de medicamentos capazes de inibir os mecanismos de patogenicidade das espécies desse gênero. Desse modo, o objetivo desse estudo foi obter o extrato bruto e frações a partir do sobrenadante da cultura de 4 h de S. mutans (UA159) e avaliar seus efeitos sobre os mecanismos de patogenicidade de C. albicans (ATCC18804) por meio de estudos in vitro. Após o cultivo de S. mutans, o extrato bruto foi obtido via partição líquido-líquido com acetato de etila (3x) e posteriormente fracionado em coluna de sílica derivatizada C-18 eluída com gradiente MeOH:H2O, fornecendo cinco frações (SM-F1, SM-F2, SM-F3, SM-F4 e SM-F5). A identificação das substâncias contidas no extrato bruto e frações foi realizada utilizando cromatografia gasosa acoplada a espectrometria de massas (CGEM), sendo encontradas as seguintes substâncias: ácido octanóico e uridina no extrato bruto, ácido propanóico, (3R)-3-Methyl-1,4-bis(trimethylsilyl)piperazine-2,5- dione, pirimidina, gulose e oleamida na SM-F1 e ácido nicotínico e triptofano na SM- F2. O extrato bruto e as frações foram submetidos aos ensaios de bioatividade sobre a formação de hifas de C. albicans e analisados por microscopia óptica e microscopia eletrônica de varredura (MEV). O extrato bruto e a fração SM-F2, que resultou em maior inibição das hifas, foram investigados na expressão de genes de C. albicans envolvidos no mecanismo de filamentação (CPH1, EFG1, HWP1, UME6 e YWP1) por PCR quantitativo em tempo real (RT-qPCR). Além disso, o potencial de inibição do extrato bruto, frações SM-F1 e SM-F2 foi avaliado sobre a formação de biofilmes de C. albicans, analisados pela quantificação de células viáveis (UFC/mL) e MEV Os resultados demonstraram inibição significativa na formação de hifas de C. albicans pelo extrato bruto e principalmente, pela SM-F2, com alterações significativas na expressão de todos os genes analisados. O extrato bruto e SM-F2 regularam negativamente a expressão dos genes CPH1, EFG1, HWP1 e UME6, em contrapartida, regularam positivamente a expressão do gene YWP1, envolvido no mecanismo de dispersão das leveduras. Nas análises de UFC/mL, os resultados demonstraram redução nas contagens de C. albicans nos biofilmes formados quando em contato com o extrato bruto, frações SM-F1 e SM-F2 em todas as concentrações testadas, sendo que o extrato bruto reduziu totalmente as células de C. albicans na concentração 15 mg/mL. Os resultados do nosso estudo demonstraram que o extrato bruto e frações de S. mutans apresentaram efeitos inibitórios sobre importantes mecanismos de virulência de C. albicans, fornecendo evidências que S. mutans produz substâncias com ação antifúngica, tornando-o promissor na busca de novos compostos antimicrobianos para prevenção e tratamento das candidoses humanas(AU)
The biofilm formation and hyphae production by Candida albicans are important virulence factors, mainly for mucosal adhesion and tissue invasion. The search for secondary metabolites produced by S. mutans is of great importance, since it may provide new therapeutic strategies against Candida infections, enabling the development of drugs that inhibit the mechanisms of pathogenicity of the species of this genus. Thus, the objective of this study was to obtain the crude extract and fractions from the supernatant of the 4 h culture of S. mutans (UA159) and evaluate their effects on the mechanisms of pathogenicity of C. albicans (ATCC18804) through in vitro studies. After culture of S. mutans, the crude extract was obtained via liquid-liquid partition with ethyl acetate (3x) and then fractionated on a C-18 derivatized silica column eluted with MeOH:H2O gradient, providing five fractions (SM-F1, SM-F2, SM-F3, SM-F4 and SM-F5). The identification of the substances contained in the crude extract and fractions was performed by gas chromatography coupled to mass spectrometry (GC-MS). The following substances were found: octanoic acid and uridine in crude extract, propanoic acid, (3R) -3-methyl -1,4-bis (trimethylsilyl) piperazine-2,5-dione, pyrimidine, gulose and oleamide in SM-F1 and nicotinic acid and tryptophan in SM-F2. The crude extract and fractions were submitted to bioactivity assays on hyphae formation by C. albicans and analyzed by optical microscopy and scanning electron microscopy (SEM). The crude extract and SM-F2 fraction, which resulted in highest inhibition of hyphae, were investigated in the expression of C. albicans genes involved in the filamentation mechanism (CPH1, EFG1, HWP1, UME6 and YWP1) by quantitative real-time PCR (RT-qPCR). In addition, the potential of inhibition of the crude extract, SM-F1 and SM-F2 fractions in biofilms of C. albicans were evaluated by the quantification of viable cells (CFU/mL) and SEM. The data obtained were statistically analyzed by Graph Pad Prism 5.0, with a significance level of 5%. The results demonstrated a significant inhibition on C. albicans hyphae formation by the crude extract and mainly by SMF2, with significant alterations in the expression of all genes analyzed. The crude extract and SM-F2 negatively regulated the expression of the CPH1, EFG1, HWP1 and UME6 genes, in contrast, positively regulated the expression of the YWP1 gene, involved in the mechanism of yeast dispersion. In the CFU/mL analyzes, the results showed a reduction in the counts of C. albicans in the biofilms formed when in contact with the crude extract, SM-F1 and SM-F2 fractions in all tested concentrations and the crude extract totally reduced C. albicans cells at 15 mg/mL concentration. The results of our study demonstrated that the crude extract and fractions of S. mutans presented inhibitory effects on important mechanisms of virulence of C. albicans, providing evidence that S. mutans produces substances with antifungal action, making it promising in the search for new compounds antimicrobials for the prevention and treatment of human candidoses(AU)

Background: Cnidarians produce toxins, which are composed of different polypeptides that induce pharmacological effects of biotechnological interest, such as antitumor, antiophidic and anti-clotting activities. This study aimed to evaluate toxicological activities and potential as antitumor and antiophidic agents contained in total extracts from five cnidarians: Millepora alcicornis, Stichodactyla helianthus, Plexaura homomalla, Bartholomea annulata and Condylactis gigantea (total and body wall). Methods: The cnidarian extracts were evaluated by electrophoresis and for their phospholipase, proteolytic, hemorrhagic, coagulant, fibrinogenolytic, neuromuscular blocking, muscle-damaging, edema-inducing and cytotoxic activities. Results: All cnidarian extracts showed indirect hemolytic activity, but only S. helianthus induced direct hemolysis and neurotoxic effect. However, the hydrolysis of NBD-PC, a PLA2 substrate, was presented only by the C gigantea (body wall) and S. helianthus. The extracts from P. homomalla and S. helianthus induced edema, while only C gigantea and S. helianthus showed intensified myotoxic activity. The proteolytic activity upon casein and fibrinogen was presented mainly by B. annulata extract and all were unable to induce hemorrhage or fibrinogen coagulation. Cnidarian extracts were able to neutralize clotting induced by Bothrops jararacussu snake venom, except M. alcicornis. All cnidarian extracts were able to inhibit hemorrhagic activity induced by Bothrops moojeni venom. Only the C. gigantea (body wall) inhibited thrombin-induced coagulation. All cnidarian extracts showed antitumor effect against Jurkat cells, of which C. gigantea (body wall) and S. helianthus were the most active; however, only C. gigantea (body wall) and M. alcicornis were active against B16F10 cells...

Antibiotic resistance results in higher medical costs, prolonged hospital stays and increased mortality and is rising to dangerously high levels in all parts of the world. Therefore, this study aims to search for new antimicrobial agents through bioprospecting of extracts of endophytic fungi from Bauhinia guianensis, a typical Amazonian plant used in combating infections. Seventeen (17) fungi were isolated and as result the methanolic extract of the fungus Exserohilum rostratum showed good activity against the bacteria tested. The polyketide monocerin was isolated by the chromatographic technique, identified by NMR and MS, showing broad antimicrobial spectrum.
La resistencia a los antibióticos conduce a mayores costos médicos, hospitalizaciones prolongadas e incremento de la mortalidad, y está aumentando a niveles peligrosamente altos en todas partes del mundo. Este estudio tuvo como objetivo la búsqueda de nuevos agentes antimicrobianos a través de la bioprospección de extractos de hongos endófitos de Bauhinia guianensis, una planta amazónica típica, utilizada en la lucha contra problemas infecciosos. Fueron aislados 17 hongos; el extracto metanólico del hongo Exserohilum rostratum mostró buena actividad contra las bacterias probadas. Se aisló monocerina policétido por la técnica de cromatografía; este compuesto fue identificado por RM y EM, y mostró un amplio espectro antimicrobiano.

The use of fungi as a source of enzymes has become widespread in various industrial and commercial areas, and Aspergillus section Nigri has significant potential for producing enzymes. The aim of this study was to isolate Aspergillus section Nigri from plant litter and soil from the Atlantic Forest biome and evaluate it with regards to hydrolytic enzyme production. The trials for producing the enzymes were carried out in Petri dishes, using different culture mediums adapted for microbial growth and with the respective substrates for inducing enzyme production - cellulase (carboxymethyl cellulose), protease (skimmed milk), amylase (soluble starch), pectinase (citrus pectin), and phytase (Pikovskaya medium). Forty-two fungi were isolated, 16.7% derived from the plant litter layer and 83.3% derived from soil at a depth of 0 to 5 cm and 5 to 10 cm. All of the isolated lineages presented amylase, protease, and phytase production, with 90.4% producing cellulase and no lineage producing pectinase. From the results, the significant potential for Atlantic Forest fungi as hydrolytic enzyme producers could be perceived. The enzymatic activity evaluations presented a satisfactory result when compared with the scientific literature.(AU)
A utilização dos fungos como fonte de enzimas vem adquirindo status de destaque nas mais variadas áreas industriais e comerciais, e os Aspergillus membros da seção Nigri possuem significativo potencial para produção de enzimas. Os objetivos deste estudo foram isolar e avaliar Aspergillus da seção Nigri de serrapilheira e solos do bioma Mata Atlântica quanto à produção de enzimas hidrolíticas. Os ensaios para produção das enzimas foram realizados em placas de Petri, utilizando diferentes meios de cultivo, adequados ao crescimento microbiano e com a presença dos respectivos substratos indutores à produção das enzimas - celulases (carboximetilcelulose), proteases (leite desnatado), amilases (amido solúvel), pectinases (pectina cítrica) e fitase (meio Pikovskaya). Foram isolados 42 fungos, sendo desse total 16,7% provenientes da camada de serrapilheira e 83,3% provenientes do solo na profundidade de 0 a 5 cm e 5 a 10 cm. Todas as linhagens isoladas apresentaram produção de amilases, protease e fitase, 90,4% produziram celulase, e nenhuma linhagem produziu pectinase. Com esses resultados, percebeu-se significativo potencial dos fungos da Mata Atlântica como produtores de enzimas hidrolíticas. As avaliações da atividade enzimática apresentaram resultado satisfatório quando comparados à literatura científica.(AU)

Parahancornia amapa is an endemic plant species of Amapá in the Amazon region. This study aimed to evaluate the acute toxicity of methanol extract of the bark of P. amapa. Different concentrations of the extract were added to the brine shrimps (Artemia salina) and 50% lethality concentration (LC50) was assessed. The results of the brine-shrimp assay indicated that the extract was found to be non-toxic. Acute toxicity was studied in Wistar rats (males and females) after single dose (2000 mg kg-1) by oral gavage. Food and water intake, body weight, general behavioral changes and mortality of animals were noted. Blood samples were collected for haematological and serum biochemical parameters measurements. Subsequently, they were euthanized and their organs (heart, kidneys, liver and lungs) subjected to macroscopic and histopathological analysis. There was no mortality or any sign of behavioral change or toxicity observed after oral administration of the extract. Nevertheless, it was observed a significant increase (p<0.05) on water intake of female rats treated with the extract of P. amapa (127.64 mL) when compared to control group (101.93 mL). A significant increase (p<0.05) in platelet count was also observed in both male (288.00x103 mm3) and female (220.83x103 mm3) rats that received the extract when compared to their respective control groups (128.33x103 mm3; 109.50x103 mm3). Histopathological changes were not related to the treatment with extract in any of the analyzed organs. These results suggest that P. amapa extract has a non-toxic effect.
Parahancornia amapa é uma espécie endêmica do Estado do Amapá, na região Amazônica. Este estudo teve como objetivo avaliar a toxicidade aguda do extrato metanólico das cascas do caule de P. amapa. Diferentes concentrações do extrato foram adicionadas aos camarões de salmoura (Artemia salina) e a concentração letal média (CL50) foi avaliada. Os resultados desse ensaio indicaram que o extrato não era tóxico. A toxicidade aguda foi estudada em ratos Wistar (machos e fêmeas) após dose única (2000 mg kg-1) por gavagem. Ingestão de água e comida, peso corporal, alterações comportamentais e mortalidade foram anotados. Amostras de sangue foram coletadas para medições de parâmetros hematológicos e bioquímicos. Os animais foram eutanasiados e seus órgãos (coração, rins, fígado e pulmões) submetidos à análise macroscópica e histopatológica. Não houve mortalidade ou qualquer sinal de mudança de comportamento ou toxicidade observada após a administração oral do extrato. No entanto, observou-se um aumento significativo (p<0,05) sobre o consumo de água de ratas tratadas com o extrato de P. amapa (127,64 mL) quando comparadas ao grupo controle (101,93 mL). Um aumento significativo (p <0,05) na contagem das plaquetas também foi observado tanto em ratos machos (288,00x103 mm3) quanto em fêmeas (220,83x103 mm3, respectivamente) que receberam o extrato quando comparados com seus respectivos grupos controle (128,33x103 mm3; 109,50x103 mm3). Alterações histopatológicas não foram relacionadas ao tratamento com extrato em nenhum dos órgãos analisados. Estes resultados sugerem que o extrato de P. amapa tem um efeito geral não-tóxico.

Aniba riparia (Lauraceae) is an important medicinal plant found in the Amazon region and presents alkaloids of the type alkamide known as riparins. Riparin A is structurally represented as the fundamental core of all Amazon riparins. This work aimed to assess the in vitro antioxidant, antitumor and antileishmanial effects of riparin A. Riparin A presented weak antioxidant capacity by tecniques of DPPH (EC50 of 296.2 μg mL-1) and ABTS+ (EC50 of 450.1 μg mL-1), showed moderate activity against colon carcinoma (HCT-116: IC50 of 21.7 μg mL-1) and leishmanicidal activity on promastigotes of L. amazonensis (IC50 of 307.0 ± 79.6, 193.7 ± 44.3 and 81.8 ± 11.2 μg mL-1, respectively, after 24, 48 and 72 h of incubation). Then, in addition to its structural simplicity, riparin A revealed promising biological activities and remarkable in vitro leishmanicidal action, an important result in epidemiological point of view to control leishmaniasis in Brazil, including in the Amazon region.
Aniba riparia (Lauraceae) é uma importante planta medicinal encontrada na região amazônica que apresenta alcaloides do tipo alcamida e conhecidos como riparinas. Este trabalho teve como objetivo avaliar os efeitos antioxidantes, antitumorais e leishmanicidas in vitro da riparina A. Riparina A apresentou fraca capacidade antioxidante pelas técnicas do DPPH (CE50 de 296,2 μg mL-1) e ABTS+ (CE50 de 450,1 μg mL-1), mostrou moderada atividade contra carcinoma de cólon (HCT-116: CI50 de 21,7 μg mL-1) e atividade leishmanicida sobre formas promastigotas de Leishmania amazonensis (CI50 de 307,0 ± 79,6; 193,7 ± 44,3 e 81,8 ± 11,2 μg mL-1, respectivamente, após 24, 48 e 72 h de incubação). Assim, além de sua simplicidade estrutural, a riparina A revelou atividades biológicas promissoras e significativa ação leishmanicida in vitro, resultado importante diante da relevância epidemiológica para controle da leishmaniose no Brasil, inclusive na região amazônica.

A natureza e diversidade das estruturas químicas com atividade farmacológica que se tem encontrado nos organismos marinhos justificam a busca por novos compostos que são de interesse nas mais diversas áreas de aplicação. As espécies de macroalgas vermelhas, em especial Laurencia spp., merecem destaque pela enorme variedade de terpenos e acetogeninas que produzem, sendo consideradas de grande potencial na produção de novos fármacos. O estudo de seus constituintes pode fornecer importantes subsídios para a quimiotaxonomia, ecologia química, caracterização das espécies e avaliação do potencial biotecnológico. Baseado nisso, Laurencia aldingensis, L. dendroidea e Laurenciella sp. foram selecionadas para o presente estudo para isolamento, caracterização e teste de atividades biológicas dos seus compostos. A técnica do DNA barcoding foi utilizada como ferramenta de diagnóstico para garantir a similaridade entre as amostras de cada espécie, que foram coletadas em época e locais diferentes. Do extrato orgânico de Laurencia aldingensis, nove substâncias foram isoladas, sendo quatro esfingosinas (1-4), três terpenos (5-7) e duas novas substâncias halogenadas (8 e 9). Do extrato orgânico de Laurencia dendroidea formam isolados dois terpenos halogenados conhecidos (10, 11) e, do extrato de Laurenciella sp. três novas substâncias halogenadas alifáticas insaturadas (12-14), assim como um ácido graxo (15) e um esterol (16) conhecidos. Dentre elas, a 8 apresentou atividade citotóxica, mas não se mostrou seletivo, e as substâncias 4 e 11 apresentaram atividade esquistossomicida, bastante promissora. No entanto, nenhum deles apresentou atividade antioxidante. Diante desta investigação, podemos dizer que as informações geradas com os estudos de Laurencia aldingensis, L. dendroidea e Laurenciella sp. expandiram significantemente o conhecimento no que tange a diversidade química no gênero e o potencial biológico-farmacêutico dos mesmos
The nature and diversity of chemical structures with pharmacological activity that have been found in marine organisms justifies the search for new compounds that may have applications in various areas of interest. Species of red seaweeds, especially Laurencia spp., are special because of the unprecedented variety of terpenes and acetogenins they produce that are considered potentially useful for the production of new drugs. Study of their constituents can also provide important insights relating to their chemotaxonomy, chemical ecology, characterization of species and biotechnological potential. On this basis Laurencia aldingensis, L. dendroidea and Laurenciella sp., were selected for study and isolation, characterization, and biological activity assessment of isolatable quantities of their compounds. The technique of DNA barcoding was used as a diagnostic tool to ensure similarity between samples of each species collected at different times and places. From the organic extract of Laurencia aldingensis nine compounds were isolated; four sphingosines (1-4), three terpenes (5-7) and other two new halogenated compound (8, 9). From the organic extract of Laurencia dendroidea two known halogenated terpenes (10, 11) were isolated while from a similar extract of Laurenciella sp., three new halogenated aliphatic compounds (12-14) were isolated together with known fatty acid (15) and sterol (16). Among all isolates, 8 demonstrated unspecific cytotoxic activity and compounds 4 and 11 showed promising schistosomicidal activity. In applied antioxidant assays none of the isolates we noted to have activity. From the overall investigation it is also clear that the information gleaned from the studies of Laurencia aldingensis, L. dendroidea and Laurenciella sp., significantly expanded our knowledge base concerning chemical diversity in the genus Laurencia and their biological-pharmaceutical potential

Background Snakebite is considered a neglected tropical disease by the World Health Organization. In Brazil, about 70% of the envenomation cases are caused by Bothrops snakes. Its venom may provoke hemorrhage, pain, necrosis, hemolysis, renal or cardiac failure and even death in victims. Since commercial antivenom does not efficiently neutralize the local toxic effects of venoms, natural products have been tested in order to provide alternative or complementary treatment to serum therapy. Therefore, the present study aimed to evaluate the ability of the seaweed Plocamium brasiliense and its active derivatives to neutralize hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic activities of B. jararaca venom. Methods Specimens of P. brasiliense were collected in Rio de Janeiro state, Brazil, dried and submitted to oil extraction using four solvents of increasing polarities, n-hexane (HEX), dichloromethane (DCM), ethyl acetate (ETA) and hydroalcoholic solution (HYD). The solvents were evaporated, yielding HEX, DCM, ETA and HYD extracts. Further, all extracts were dissolved in dimethylsulfoxide. In addition, two monoterpenes (8-bromo-3,4,7-trichloro-3,7-dimethyl-1E, 5E-octadiene and 1,8-dibromo-3,4,7-trichloro-3,7-dimethyl-1E, 5E-octadiene) and a cholesterol fraction were isolated from the extract of P. brasiliense prepared in hexane. Algal samples were incubated for 30 minutes with B. jararaca venom, and then tested for lethality; hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic effects. Results Most of the algal extracts inhibited the toxic effects with different potencies. The DCM extract was the most effective, since it inhibited all types of toxic activity. On the other hand, the HYD extract failed to inhibit any effect. Este trabalho teve por objetivo a investigação fitoquímica e propriedades antioxidantes de extratos das folhas de Trigynaea oblongifolia Schltdl (Annonaceae), Ottonia frutescens Trel (Piperaceae), e Bathysa australis (St Hill) Hooz (Rubiaceae), bem como avaliar, in vitro, a letalidade frente ao microcrustáceo Artemia salina Leach. Os extratos foram preparados por maceração em metanol 10% (p/v) por sete dias, à temperatura ambiente. A atividade antioxidante dos extratos foi determinada pela metodologia que utiliza o radical estável DPPH. A toxicidade dos extratos foi avaliada frente ao microcrustáceo A. salina. Os extratos de O. frutescens e B. australis apresentaram as seguintes classes de metabólitos secundários: Alcalóides, Antraquinonas, Cumarinas, Polifenóis (Taninos), Saponinas. Nos extratos de T. oblongifolia, além dos metabólitos citados anteriormente, foi detectada a presença de Flavonóides. A atividade antioxidante, observada em 30 minutos na concentração de 24 µg/mL de extrato, foi de: O. frutescens - 38,3%, T. oblongifolia - 32,3%, e B. australis - 32,1%. A Concentração Letal, CL50, dos extratos em A. salina foi de: O. frutescens - 149,75 ± 1,02 µg/mL, T. oblongifolia - 148,8 ± 1,74 µg/mL, e B. australis - 684 ± 9,04 µg/mL. Neste contexto, destacamos as espécies, nativas da Floresta Atlântica, O. frutescens e T. oblongifolia de grande potencial na bioprospecção de moléculas biologicamente ativas.
This study aimed to carry out phytochemical and antioxidant properties, and assess, in vitro, the lethality of a leaf extract Trigynaea Schltdl oblongifolia Schltdl (Annonaceae), Ottonia frutescens Trel (Piperaceae), Bathysa australis (St Hill) Hooz (Rubiaceae) front of the microcrustaceans Artemia salina Leach. Extracts were prepared by soaking in 10% methanol (w/v) for seven days at room temperature. The antioxidant activity of extracts was determined by the stable radical DPPH method. The toxicity of the extracts was evaluated against the microcrustaceans A. saline. Extracts of O. frutescens and B. australis showed the following classes of secondary metabolites: Alkaloid, Anthraquinones, Coumarins, polyphenols (tannins), Saponin. The extracts from T. oblongifolia, besides the aforementioned metabolites the presence of flavonoids was detected. The antioxidant activity was observed in 30 minutes, the concentration 24 mg/mL of extract was: O. frutescens - 38.3%, T oblongifolia - 32.3%, and B. australis - 32.1%. The lethal concentration, LC50, extracts of A. salina was: O. frutescens - 149.75 ± 1.02 mg/mL, T. oblongifolia - 148.8 ± 1.74 mg/mL, and B. australis - 684 ± 9.04 mg/mL. In this context, we highlight the species, native to the Atlantic Forest, O. frutescens and T. oblongifolia great potential in bioprospecting of biologically active molecules.

In an effort to raise awareness of the major environmental challenges facing the regions coastal and marine ecosystems, and to highlight the potential research and socioeconomic benefits of this program, the authors provide summaries of key lectures and conclusions presented in the international conference on Central American Marine Biodiversity and Genomics, in April of 2012, dedicated to Genomic Archiving and Coastal Marine Biodiversity Exploration, Conservation and Sustainable Development, and describe the main components of the initiative for the benefit of other actors, stakeholders and donors in the field of marine biodiversity.

Background: Cellulases and lipases have broad industrial application, which calls for an urgent exploration of microorganisms from extreme environments as valuable source of commercial enzyme. In this context, the present work describes the bioprospection and identification of deep-sea bacteria that produce cellulases and lipases, as well their optimal temperature of activity. Results: The first step of this study was the screening of cellulolytic and lipolytic deep-sea bacteria from sediment and water column, which was conducted with substrates linked with 4-Methylumbelliferyl. Among the 161 strains evaluated, 40 were cellulolytic, 23 were lipolytic and 5 exhibited both activities. Cellulolytic and lipolytic bacteria are more common in sediment than at the water column. Based on the ability to produce cellulases and lipases three isolates were selected and identified (16S rRNA sequencing) as Bacillus stratosphericus, B. aerophilus and B. pumilus. Lipases of strain B. aerophilus LAMA 582 exhibited activity at a wide temperature range (4º to 37ºC) and include psychrophilic behaviour. Strain Bacillus stratosphericus LAMA 585 can growth in a rich (Luria Bertani) and minimal (Marine Minimal) medium, and does not need an inducer to produce its mesophilic cellulases and lipases. Conclusions: Deep-sea sediments have great potential for bioprospection of cellulase and lipase-producing bacteria. The strains LAMA 582 and LAMA 585 with their special features, exhibit a great potential to application at many biotechnology process.
