ABSTRACT Discharge of coke-oven wastewater to the environment may cause severe contamination to it and also threaten the flora and fauna, including human beings. Hence before dumping it is necessary to treat this dangerous effluent in order to minimize the damage to the environment. Conventional technologies have inherent drawbacks however, biological treatment is an advantageous alternative method. In the present study, bacteria were isolated from the soil collected from the sites contaminated by coke-oven effluent rich in phenol and cyanide. Nucleotides sequence alignment and phylogenetic analysis showed the identity of the selected phenol and cyanide degrading isolates NAUN-16 and NAUN-1B as Pseudomonas putida and Pseudomonas stutzeri, respectively. These two isolates tolerated phenol up to 1800 mg L-1 and cyanide up to 340 mg L-1 concentrations. The isolates were immobilized on activated charcoal, saw dust and fly ash. The effluent was passed through the column packed with immobilized cells with a flow rate of 5 mL min-1. The isolates showed degradation of phenol up to 80.5% and cyanide up to 80.6% and also had the ability to reduce biological oxygen demand, chemical oxygen demand and lower the pH of effluent from alkaline to near neutral. The study suggests the utilization of such potential bacterial strains in treating industrial effluent containing phenol and cyanide, before being thrown in any ecosystem.

Ten bacterial strains that utilize cyanide (CN) as a nitrogen source were isolated from cassava factory wastewater after enrichment in a liquid media containing sodium cyanide (1 mM) and glucose (0.2% w/v). The strains could tolerate and grow in cyanide concentrations of up to 5 mM. Increased cyanide levels in the media caused an extension of lag phase in the bacterial growth indicating that they need some period of acclimatisation. The rate of cyanide removal by the strains depends on the initial cyanide and glucose concentrations. O objetivo do presente estudo foi verificar os efeitos sobre a cinética do cianeto, em suínos, em diferentes fases da vida, usando o tiocianato como biomarcador. Vinte e dois suínos, foram divididos em quatro grupos (60 dias da idade, 95 dias da idade, 80 dias do gestação e 21 dias de lactação), e receberam por via oral, a dose única de 3.0 mg /kg de peso vivo de cianeto do potássio (KCN). As concentrações do tiocianato no sangue foram medidas dentro de 24h. O tempo máximo (Tmax) e constante de eliminação (Kel) foram mais elevados em porcas lactantes (15 hs e 0.045, respectivamente); por ouro lado, a maior concentração do tiocianato (Cmax) foi observada nas fêmeas grávidas (161.8). A meia vida de eliminação (t1/2) e o volume da distribuição (Vd) foram mais elevados nas fêmeas adultas (41, 57 e 1.23, respectivamente). Contudo a área sob a curva (AUC) do tiocianato foi mais elevado nos animais novos (354183,28), e o clearance o mais baixo (0.007) nestes animais. Concluindo, os resultados do presente estudo, evidenciam que o metabolismo do cianeto, varia extremamente, considerando o estado fisiológico dos suínos fêmeas, e que são os animais novos, provavelmente, os mais sensíveis aos efeitos tóxicos, da exposição crônica as baixas doses do cianeto.
The aim of the present study was to determine the effects of the swine, in different periods of life on the toxicokinetics of cyanide using thiocyante as biomaker. Twenty and two swines, was divided into four groups (60 days of age, 95 days of age, sows with 80 days of gestation and lactating swine), were dosed orally with 3,0 mg/kg/ body weigth of potassium cyanide (KCN). Thiocyanate concentrations in blood were measured within 24h. The time of peak concentration (Tmax) and constant of elimination (Kel) were higher in lactating sows (15 hs and 0,045, respectively); on the other hand, the maximum plasma concentration (Cmax) of thiocyanate was observed in pregnant females (161,8). The elimination half life (t1/2) and volume of distribution (Vd) were higher in adult sows (41, 57 and 1,23, respectively). Whereas the clearance and the area under the curve (AUC) of thiocyanate was higher in young animals (354183,28) the clearance was lower (0,007) in these animals. In coclusion , the results of the present study evidence that the metabolism of cyanide varies greatly considering the physiologic state of female swine being the young animals probably more sensitive to the toxic effects of chronic exposure to low doses of cyanide.

Chromobacterium violaceum is a Gram-negative bacterium found in a wide variety of tropical and subtropical ecosystems. The complete genome sequence of C. violaceum ATCC 12472 is now available, and it has considerable biotechnological potential for various applications, such as environmental detoxification, as well as medical and agricultural use. We examined the biotechnological potential of C. violaceum for environmental detoxification. Three operons, comprising the ars operon, involved in arsenic resistance, the cyn operon, involved in cyanate detoxification, and the hcn operon, encoding a cyanase, responsible for biogenic production of cyanide, as well as an open reading frame, encoding an acid dehalogenase, were analyzed in detail. Probable catalytic mechanisms for the enzymes were determined, based on amino acid sequence comparisons and on published structural information for these types of proteins

A clear association between seasonal outbreaks of a paralytic disease called konzo and toxic effects from consumption of insufficiently processed bitter cassava roots has been demonstrated in Bandundu region, Zaire. A community-based survey of 6764 inhabitants identified 110 live and 24 dead konzo-affected persons with a history of isolated non-progressive spastic paraparesis of abrupt onset. The start of these annual outbreaks of konzo in 1974 coincided with the completion of a new tarmac road to the capital, which facilitated the transport of cassava and made it the main cash crop. The extensive cassava sales encouraged the consumption by the peasant families of roots that had not been adequately processed; frequent acute cyanide intoxications resulted when the naturally occurring cyanogens in the roots were eaten. The disease mainly appeared in the dry season when there was high consumption of insufficiently processed cassava and the diet lacked supplementary foods with sulfur-containing amino acids which promote cyanide detoxification. These results, which confirm the earlier findings in East Africa, show that, owing to the high cyanide and low sulfur dietary intake, there is an increased risk of konzo outbreaks in cassava-growing areas during periods of adverse agro-economic changes
