Effect of Yindan Xinnaotong capsules on Bcl-2 and Bax expressions in the hippocampal CA1 region and learning and memory abilities of vascular dementia rats / 中华神经医学杂志

ABSTRACT
Objective To study the impact of Yindan Xinnaotong capsules on learning and memory abilities and Bcl-2 and Bax expressions in the hippocampal CA1 region of rats with vascular dementia (VD).Methods All rats were randomly divided into normal group,sham-operated group,vehicle group and low-,middle-and high-dose Yindan Xinnaotong capsule treatment groups.The animal models of VD in all the groups,excepted the normal group and sham-operated group,were established by two-vesssel-occlusion+sodium nitroprusside method.Yindan Xinnaotong capsule treatment groups were intragastrically given Yindan Xinnaotong capsules by dose of 0.24 g/(kg·d),0.48 g/(kg· d) and 0.96 g/(kg ·d) for 28 days,respectively.The Bcl-2 and Bax expressions were detected by immunohistochemistry staining; the capabilities of learning and memory were tested by Morris water maze.Results Morris water maze test showed that the escape latency of the vehicle group was (94.32±16.53),which was significantly longer than that of the normal group and sham-operated group (P<0.05); the middle dose and high dose Yindan Xinnaotong capsule treatment groups had the escape latency of (74.36±15.67) and (65.73 ±14.84),which were significantly shorter than that of vehicle group (P<0.05).As compared with the normal group and sham-operated groups,number of the number of crossing the platform and target quadrant dwell time in the vehicle group were significantly reduced (P<0.05); in the middle dose and high dose Yindan Xinnaotong capsule treatment group,crossing the platform and the target quadrant dwell time were (2.30 ±0.42) and (79.85 ±5.58),(2.71 ±0.45) and (83.38 ±5.36),which were significantly increased than those in the vehicle group (1.15±0.24) and (66.80±7.20,P<0.05).As compared with those in the normal group and sham-operated group,the Bcl-2 and Bax positive expressions in the other groups were significantly increased (P<0.05),and the changes in the vehicle group were the most obvious (P<0.05); as compared with those in the vehicle group,the Bcl-2 expression were significantly increased and the Bax expression was obviously reduced in groups of middle dose and high dose (P<0.05).Conclusion Yindan Xinnaotong capsules can improve the learning and memory abilities; it can also increase the Bcl-2 expression and decrease the Bax expression.
