RESUMO

A microagglutination test using trypsin-treated and Coomassie blue-stained Trypanosoma cruzi epimastigote antigen was adapted for the diagnosis of Chagas' disease. When incorporated in the test, 2-mercaptoethanol treatment of chagasic sera had no influence on antibody titer. In contrast, titers in sera from patients with visceral leishmaniasis, African trypanosomiasis, and autoimmune disorders, subjected to similar treatment, showed remarkable decline. Accordingly, a lower cut-off point for Chagas' disease serological negativity could be taken resulting in a higher sensitivity (95.6%); the specificity was 94.7%. Similar specificities were obtained with Leishmania donovani chagasi and L. d. donovani antigens applied to homologous visceral leishmaniasis and heterologous Chagas' sera. Of 316 nonchagasic sera, only 3 with leptospirosis and 1 with leprosy showed seropositive titers prior to and after 2-mercaptoethanol treatment.

RESUMO

The fuchsin salts currently available in Latin American market have shown some instability when in solution, according to the classic Ziehl-Neelsen method, resulting in a total precipitation of the salt. The authors indicate a new technique for the preparation of this solution, in order to minimize the action of interfering factors responsible for the precipitation, obtaining thus a greater solubility of the salt, as well as the solution stability. The method effectiveness is reinforced by the utilization of a smaller amount of the salt and the attainment of a larger storage period for the solution.

Os sais de fucsina atualmente encontrados no mercado Latino-americano, têm demonstrado uma certa instabilidade, quando em soluçäo, segundo indicaçöes clcássicas do método de Ziehl-Neelsen, resultando em precipitaçäo total do sal. Os autores indicam nova técnica de preparaçäo desta soluçäo, de forma a minimizar a açäo dos fatores interferenciais responsáveis pela precipitaçäo, obtendo maior solubilidade do sal e estabilidade da soluçäo. A eficiência do método é ainda reforçada pela utilizaçäo de menor quantidade do sal e possibilidade de maior tempo de estocagem da soluçäo

In Spurr-embedded biopsies for ultrastructural examinations, Mycobacterium leprae hardly differed from the surrounding tissues using the staining technique of Richardson. Also, the usual histological staining methods of Ziehl-Neelsen and Fite did not achieve positive results for the determination of M. leprae. Therefore, we applied the methylene blue-borax and basic fuchsin technique for the demonstration of the bacilli in plastic-embedded tissue of 11 patients suffering from Hansen's disease. In every patient the diagnosis was confirmed by histological examination of skin biopsies. Portions of the biopsies of nine patients were then fixed in glutaraldehyde and osmium tetroxide and embedded according to Spurr. In the other two cases, the material was first fixed in 10% Formalin and embedded in paraffin. After cutting 3-micron sections for routine histological examination, the remaining material was prepared adequately for ultrastructural examination. Using the methylene blue-borax and basic fuchsin technique, the semithin sections of plastic-embedded material presented a considerably more differentiated picture than other comparable methods. M. leprae located in foamy cells or in the tissue stained violet. These findings were corroborated in subsequent electron-microscopic examinations. The semithin sections thus prepared allow, through the clear demonstration of the microorganisms, a precise demarcation of the ultrathin area.

Two methods, the carbol fuchsin with acetic acid differentiation and the periodic acid-carbol pararosaniline, were used for demonstrating leprosy bacilli in skin smears. Bacillary smears from 200 long-treated patients with tuberculoid, borderline and lepromatous leprosy were stained with periodic acid-carbol pararosaniline. There were significantly greater BI and MI determinations than with classic carbol fucsin staining. With the former stain bacilli were found in 69 of 96 skin smears in which no bacilli could be seen by the latter stain. It is suggested that under the action of antileprosy drugs some leprosy bacilli may lose their acid-fastness and become chromophobic; chromophobic bacilli can be restored to their staining with periodic acid pretreatment. Leprosy bacilli in their chromophobic form can survive in healing and apparently healed lesions even after prolonged chemotherapy and can be a possible source of relapse.

RESUMO

A modified allochrome staining procedure is presented as being the most reliable and sensitive method for demonstrating mycobacteria in tissue sections. The technic is as follows: Deparaffinize formalin fixed sections, oxidize in 10% periodic acid for 24 hours, differentiate in 1% HCl-70% ethanol, stain in Weigert's iron hematoxylin nuclear stain, and counterstain in picro-methyl glue. Mycobacteria stained brilliant red in contrast with the allochrome-stained background tissues, and apparently otherwise chromophobic bacilli are demonstrated.

RESUMO

Reduced acid-fast staining of leprosy bacilli occurs during the dewaxing of paraffin sections by xylene and alcohols; the older and more decrepit bacilli being especially affected. By the use of non-deparaffinized sections, the leprosy bacilli which could not be stained with the usual carbol fuchsin are strongly stained. Moreover, non-deparafinized sections can be used for the periodic acid-carbol pararosanilin stain or methenamine silver stain for demonstrating mycobacteria.

Acid fast staining of mycobacteria in the form of beadings is obtained by means of a carbolfuchsin solution (Ziehl-Neelsen stain) prepared from pararosaniline or from certain kinds of basic fuchsin. After such acid-fast stains, the intensity of the bacilli's colouring was rather poor and unstable, so that some bacilli lost their acid-fast stain. In contrast, an acid-fast staining of mycobacteria in rod form results by using a carbolfuchsin prepared from rosaniline or from other basic fuchsins included new fuchsin. The spectrophotometric and thin-layer chromatographic data indicate that the main component of those basic fuchsins showing beady staining may be pararosaniline, whereas the main ingredient of basic fuchsin with staining the bacteria in rod form may be its higher homologues. Neither chloride nor acetate of the fuchsin could affect the appearance and number of stained bacilli. The commercially available "basic fuchsin" is either the chloride or acetate of pure pararosaniline or consists of variable mixtures of it with higher homologues. Consequently, only a basic fuchsin which has an absorption maximum at lambda greater than or equal to 552 nm could be employed for the acid-fast stain of mycobacteria in a stable manner. Pararosaniline included some basic fuchsins, composed mainly from pararosaniline, should not be selected for the preparation of the carbolfuchsin formula.
