ABSTRACT

Regulatory T cells (Tregs) play a key role in immune system homeostasis and tolerance to antigens, thereby preventing autoimmunity, and may be partly responsible for the lack of an appropriate immune response against tumor cells. Although not sufficient, a high expression of forkhead box P3 (FOXP3) is necessary for their suppressive function. Recent reports have shown that histones deacetylase inhibitors increased FOXP3 expression in T cells. We therefore decided to investigate in non-Tregs CD4-positive cells, the mechanisms by which an aspecific opening of the chromatin could lead to an increased FOXP3 expression. We focused on binding of potentially activating transcription factors to the promoter region of FOXP3 and on modifications in the five miRs constituting the Tregs signature. Valproate treatment induced binding of Ets-1 and Ets-2 to the FOXP3 promoter and acted positively on its expression, by increasing the acetylation of histone H4 lysines. Valproate treatment also induced the acquisition of the miRs Tregs signature. To elucidate whether the changes in the miRs expression could be due to the increased FOXP3 expression, we transduced these non-Tregs with a FOXP3 lentiviral expression vector, and found no changes in miRs expression. Therefore, the modification in their miRs expression profile is not due to an increased expression of FOXP3 but directly results from histones deacetylase inhibition. Rather, the increased FOXP3 expression results from the additive effects of Ets factors binding and the change in expression level of miR-21 and miR-31. We conclude that valproate treatment of human non-Tregs confers on them a molecular profile similar to that of their regulatory counterpart.

ABSTRACT

This paper investigates the spontaneous decay rate of elliptical plasmonic nanostructures. The refractive index was analyzed using the effective medium theory (EMT). Then, the polarizability, spontaneous radiative, non-radiative decay rate, and electric field enhancement factor were characterized for the targeted elliptical nanostructures at different aspect ratios. All of the optical analyses were analyzed at different distances between the excited fluorescent coupled atom and the plasmonic nanostructure (down to 100 nm). This work is promising in selecting the optimum elliptical nanostructure according to the required decay rates for optical conversion efficiency control in energy harvesting for solar cells and optical sensing applications.

ABSTRACT

The sediment compartment has the ability to trap large amounts of radionuclides and to indicate the radiological impact of pollution. The present work shows the results obtained related to the concentrations of 137Cs and natural radionuclides in sediment in the Burullus Lake, Egypt. The average values of 226Ra, 232Th, and 40K in the bottom sediments collected from the east of the Burullus Lake ranged from 10.3 to 21.8 Bq/kg, from 11.9 to 34.4 Bq/kg, and from 268 to 401 Bq/kg, respectively. The study has shown that 40K concentration is nearly uniform throughout the studied area while 226Ra and 232Th are more concentrated in the northeastern shore. Lake sediments showed contamination with 137Cs (2.7-15.9 Bq/kg). The 137Cs sediment activities indicated higher concentrations in the off-shore sites. Concentrations of all γ -ray emitting radionuclides except 40K in water samples were below the detection limits. The 40K sediment-water distribution coefficients of the near-shore samples were higher than the off-shore samples.

ABSTRACT

Malakoplakia is a rare chronic granulomatous disease that may affect many organs, including the lung, brain, adrenal glands, pancreas, bone, and the genitourinary tract. The gastrointestinal tract is the most common site of involvement outside of the urinary tract. Herein, we present a case of a 65-year-old male who presented with abdominal pain and unintentional weight loss. Physical examination findings were unremarkable, but colonoscopy revealed an isolated large, flat, soft, and pale lesion in the transverse colon. Histopathological examination of the lesion showed expansion of the lamina propria due to numerous lymphocytes, plasma cells, neutrophils, and polygonal cells. Von Kossa stain showed small intracytoplasmic basophilic granular inclusions, and these histiocyte cells showed numerous Michaelis-Gutmann bodies, findings considered as diagnostic features of colonic malakoplakia. This is a rare case of isolated malakoplakia of the transverse colon diagnosed on endoscopic specimens. The majority of reported cases have shown an association between systemic diseases and colorectal adenocarcinoma. In addition, most reported cases of colonic involvement have been in the rectum, sigmoid, and right colon.

ABSTRACT

Research of the effect of salinity on the fate of radionuclides has been focused on seas or estuarine systems while there is almost no information on marine environments with a salinity higher than that of sea water. The hypersaline Bardawil lagoon is a concentration basin, with evaporation exceeding precipitation. This study presents the characteristics of some environmental factors including salinity and their influence on the distribution of natural and artificial radionuclides in different compartments of the lagoon. The concentrations of (238)U, (234)Th, (228)Ra and (137)Cs in sediments show some degree of dependency on the water's salinity. Migration of these radionuclides in the lagoon's sediments must take place from high salinity to low-salinity regions. Cluster analysis revealed the data structure for sediment by separating (137)Cs and (40)K from (232)Th, (226)Ra, and (234)Th and for sand by separating (40)K from the other radioisotopes.

ABSTRACT

The radioactivity levels are poorly studied in non-coastal arid regions. For this reason, 38 locations covering an area of about 350 km(2) in northeast Sinai, Egypt, were investigated by γ-ray spectroscopy. Moderately significant correlations among (238)U, (234)Th, and (226)Ra isotopes and low significant correlations between the concentrations of (238)U-series and (232)Th in sand were obtained. No evidence of correlation was found between the concentrations of radioisotopes and pH, grain size, total organic matter content, bicarbonate or calcium carbonate concentrations of the sand samples. The mean values of soil-to-plant transfer factor were 0.15, 0.18, 1.52 and 0.74 for (226)Ra, (232)Th, (40)K, and (137)Cs, respectively. The range of concentrations of (226)Ra,( 232)Th, and (40)K in water samples collected from five wells were<0.4-0.16,<0.4-0.13, and<0.15-1.62 Bq l(-1), respectively. The mean absorbed dose rate in outdoor air at a height of 1 m above the ground surface for the sand samples was 19.4 nGy h(-1). The Ra(eq) activities of the sands are lower than the recommended maximum value of 370 Bq kg(-1) criterion limit for building materials.

ABSTRACT

Natural radioactivity concentrations due to the coal mining in Gabal El-Maghara, North Sinai, Egypt, were determined using gamma-ray spectroscopy. Coal, water and soil samples were investigated in this study. The (226)Ra, (232)Th and (40)K activity concentrations in coal before extraction were 18.5 +/- 0.5, 29.5 +/- 1.2 and 149.0 +/- 8.4 Bq kg(-1), respectively. These concentrations were reduced to 18-22% after extraction due to the clay removal of the coal ore. The activity contents of the water and soil samples collected from the surrounding area did not show any evidence of enhancement due to the mining activities. Absorbed dose rate and effective dose equivalent in the mine environment were 29.4 nGy h(-1) and 128.0 microSv a(-1), respectively. The measured activity concentrations in the mine environment and the surrounding areas (5 km away from the mine) are similar to that found in other regions in North and South Sinai. Based on the measurements of gamma-ray emitting radionuclides, the mine activity does not lead to any enhancement in the local area nor represents any human risk.
