The present work evaluated whether dietary and pharmacological interference on cholesterol synthesis were capable of inducing alterations in blood and yolk cholesterol levels and the secretion of corticosterone metabolites. Forty-five 40-day-old quails were divided into three experimental groups: vegetal fat diet, 2% beef fat (tallow) diet and vegetal fat diet with simvastatin administration (3.13 mg/kg/day). During all experiments, the animal weights and food consumption were recorded and blood and faecal samples (days 0, 15, 30, 45 and 60), as well as eggs (days 30, 45 and 60), were collected. Analysis of serum and yolk cholesterol was performed and faecal corticosterone levels were measured. No differences were observed on blood cholesterol or faecal corticosterone between all treatments, despite a tendency of increased cholesterol in the group with the animal fat diet. However, quails submitted to an animal fat diet displayed an increase in yolk cholesterol at day 30 of the treatment and the egg yolks of quails treated with simvastatin exhibited a decrease in cholesterol content by the end of the treatment at 60 days. These results improved the knowledge regarding the physiology of quails and offered support to other studies concerning the consequences of the pharmacological treatment and the dietary manipulation of cholesterol levels.

The aim of the study was to evaluate the seasonality of andrological characteristics and hormonal profile of captive maned wolves (Chrysocyon brachyurus, Illiger 1811). Three adult males were evaluated from the Companhia Brasileira de Metalurgia e Mineração Scientific Breeding Center in Araxá, MG, Brazil, over 13 months. Semen was collected 2-3 times weekly and analysed. Scrotal circumference, biometrics and testicular volume were measured. Stool samples were collected 2-3 times weekly to analyse corticosteroid and testosterone metabolite concentrations. A success rate of 100% was achieved in the collection attempts during the breeding season (BS) and 77.8% during the non-breeding season (NBS). The interval to achieve penile erection was 1-5 min in the BS and 6-10 in the NBS (p < 0.001). Of the ejaculates collected, 80.0% contained sperm during BS, while 28.6% did during the NBS. The ejaculate had only one fraction, was odourless, predominantly translucent (72.4%), with a watery appearance, pH 6.7 and osmolarity of 352.8 mOsmol. Seasonal influences were seen in ejaculate volume (1.3 ml vs 0.4 ml), number of spermatozoa per ejaculate (73.9 × 10(6) vs 6.1 × 10(6) ) and percentage of live sperm (82.0% vs 66.1%) between the BS and NBS (p < 0.05), respectively. A high percentage of major sperm defects were observed in both seasons (50.1% in BS; 65.7% in NBS). Testicular volume was larger (p < 0.05; right testicles 13.1 cm(3) in BS vs 4.0 cm(3) in NBS, while left testicles 12.9 cm(3) in BS vs 5.3 cm(3) in NBS) and testicular consistency increased in the BS. No difference was seen in the basal faecal metabolite concentrations of testosterone; however, the corticosteroid concentrations were higher in the BS. Based on these results, it is possible to conclude that the collection of semen is feasible in captive maned wolves without compromising libido, seminal characteristics and reproductive behaviour and that sperm production is influenced by seasonality; however, it appears that there is no seasonal influence on basal testosterone concentrations.

We evaluated the influence of two cooling rates (from 25 to 5 degrees C) on post-thaw function of frozen sperm in ocelots (Leopardus pardalis; n=3 males) and tigrinas (Leopardus tigrinus; n=4 males). Seven normospermic (>70% normal sperm) electroejaculates from each species were diluted with a 4% glycerol freezing medium, divided into two aliquots, and assigned to one of two cooling rates: fast or slow (0.7 or 0.16 degrees C/min, respectively). Sperm motility index (SMI) and percentage of sperm with an intact acrosome were assessed before freezing and after thawing, and the ability of sperm to bind to the zona pellucida of IVM domestic cat oocytes were assessed in a competitive in vitro sperm-binding assay. Regardless of the cooling rate, frozen-thawed sperm from both species exhibited a SMI of 50; approximately 20 and approximately 32% of post-thaw sperm had an intact acrosome in ocelots and tigrinas, respectively (P<0.05). The mean (+/-S.E.M.) number of sperm bound per oocyte was higher for fast-cooled (8.5+/-1.3) than slow-cooled (2.5+/-0.3; P<0.01) ocelot sperm. In contrast, more tigrina sperm bound to domestic cat oocytes when cooled slowly versus quickly (5.8+/-0.9 versus 2.7+/-0.4, P<0.05). In conclusion, cryopreservation decreased sperm function in both species, and the oocyte-binding assay was the most efficient method to detect functional differences in post-thaw sperm.

Captive adult male ocelots (Leopardus pardalis, n = 3), margays (L. wiedii, n = 3) and tigrinas (L. tigrinus, n = 4) in two locations in southern Brazil were studied for 14 consecutive months to evaluate the effect of season on testicular function. Reproductive evaluations, including testicular measurements, electroejaculation and blood collection were conducted monthly. Fecal samples were collected weekly for androgen metabolite analysis to assess testicular steroidogenic activity. Ocelots had the highest number of motile spermatozoa in the ejaculate (114.7+/-15.8 x 10(6); P < 0.05), the highest percentage of morphologically normal spermatozoa (82.4+/-1.2%; P < 0.05) and the highest concentration of fecal androgens (1.71 vs. 0.14 microg/g; P < 0.05). Margays and tigrinas had lower numbers of motile spermatozoa (23.4+/-2.8 x 10(6), 74.2+/-8.9 x 10(6), respectively), lower percentages of morphologically normal spermatozoa (57.4+/-2.8, 59.2+/-3.5%, respectively), and lower fecal androgen concentrations (0.15+/-0.01, 0.23+/-0.01 microg/g, respectively). Serum testosterone concentrations were similar among the three species. Fecal androgen concentrations were not affected by season, with the exception of the ocelot where concentrations were higher (P < 0.05) in the summer. Ejaculates were collected throughout the year; however, peaks in average sperm production were observed during the summer for all species. In summary, this study has identified several species differences in male testicular traits among ocelots, margays and tigrinas. Results of longitudinal reproductive assessments suggest males of each species are capable of breeding throughout the year.

Reproductive endocrine patterns were characterized in female ocelots (Leopardus pardalis; n = 3), tigrinas (Leopardus tigrinus; n = 2), and margays (Leopardus wiedii; n = 2) housed in captivity in southern Brazil. Females were maintained as singletons and exposed to natural fluctuations in photoperiod. Cyclic changes in ovarian steroids were monitored by analyzing estrogen and progestogen metabolites in fecal samples collected five times weekly for 14 to 18 months. Based on intervals between fecal estrogen peaks, mean (+/- SEM) duration of the estrous cycle was 18.4 +/- 1.6 days for the ocelots (range, 7-31 days; n = 75 cycles), 16.7 +/- 1.3 days for the tigrinas (range, 11-27 days; n = 23 cycles), and 17.6 +/- 1.5 days for the margays (range, 11-25 days; n = 32 cycles). Fecal progestogen analyses combined with two laparoscopic observations of the ovaries confirmed that ocelots and tigrinas did not ovulate spontaneously. In contrast, non-mating-induced luteal phases of 40.1 +/- 6.3 days in duration (range, 30-60 days) were observed frequently in both margays. There was no evidence of gonadal seasonality in margays in either follicular or luteal activity. In ocelots, cyclic changes in estrogen excretion were observed during each month of the year; however, only one female cycled continuously. In the other two ocelots, periods of acyclicity of several months' duration were observed. It was not possible to conclude whether tigrinas were aseasonal because estrous cyclicity was observed in only one of two individuals. In the female that cycled, a 3-month period of acyclicity was observed in the late fall/early winter. These data demonstrate similarities among three felid species of the genus Leopardus, including evidence they are polyestrous but experience unexplained periods of ovarian inactivity. Only the margays differed by exhibiting occasional spontaneous, non-mating-induced ovulations. Historically, these species have not bred well in captivity. However, it is hoped that understanding the biological similarities and differences among them could lead to improved management strategies that may one day result in increased reproductive success. It has been hypothesized that oils containing high levels of omega-3 polyunsaturated fatty acids, such as canola and fish oil, could counteract some of the adverse effects induced by phthalates. In the present study, the influence of different oily vehicles on di-butyl phthalate (DBP)-induced testicular toxicity and lipid profile was investigated. Pregnant Wistar rats were treated by oral gavage from gestation days 13 to 20 with DBP (500 mg/kg/day) diluted in three different vehicles: corn, canola or fish oil. Male fetuses were analyzed on gestation day 20. DBP exposure lowered intratesticular testosterone levels and anogenital distance, regardless of the vehicle used. The percentage of seminiferous cords containing multinucleated gonocytes and cord diameter was increased in DBP-exposed groups, compared with vehicle controls, with no difference between the three DBP-exposed groups. Clustering of Leydig cells was seen in all DBP groups. Lipid profile indicated that administration of canola and fish oil can increase the content of omega-3 fatty acids in rat testis. However, content of omega-3 was diminished in DBP-treated groups. Overall, our results indicate that different oily vehicles did not alter fetal rat testicular toxicity induced by a high DBP dose.

Artemisinins combination therapy (ACT) is the first choice therapy for falciparum malaria. Data on the safety of ACTs in pregnancy are limited and controversial and the use is not recommended on the first trimester. To evaluate the effects of isolated and combined artesunate (AS)/mefloquine (MQ) on embryo rats, pregnant rats were treated orally with AS (15 and 40 mg/kg body weight (bwt)/day), MQ (30 and 80 mg/kg bwt/day) and AS/MQ (15/30 and 40/80 mg/kg bwt/day) on days 9-11 post coitum (pc). The dams were euthanized on day 12 pc and gestational and embryos histological parameters were evaluated. Embryolethality and histopathological anomalies were significant when AS was given alone or combined with MQ. Combination of AS and MQ did not enhance their toxicity compared to their separate administrations; on the other side, there was a reduction in the toxic effects of the AS when combined with MQ. Isolated MQ did not induce developmental toxicity.

The phenomenon of teratozoospermia in felids is not fully understood. In this study, we investigated the testicular androgen:estrogen balance in domestic cats and correlated these data with epididymal sperm morphology and the degree of spermatogenic activity. During spring and summer, testes and blood samples were obtained from 37 mixed-breed domestic cats (12 to 48 mo). The epididymal sperm were harvested and evaluated for sperm counts, motility, and morphology. Distal cytoplasmic droplets were not considered a defect, and samples were considered normozoospermic if they contained more than 60% normal sperm (N = 25) or teratozoospermic if they contained less than 45% normal sperm (N = 12). The testicular and serum concentrations of testosterone (T) and 17ß-estradiol (E2) were determined with an enzyme immunoassay. The gonadosomatic index and epididymal sperm numbers and motility did not differ between groups. The percentage of normal sperm was higher in normozoospermic (74.3 ± 2.0, mean ± SEM) than in teratozoospermic samples (43.1 ± 1.4). The most prevalent sperm defects in the teratozoospermic group were abnormal acrosomes (9.7 ± 2.0) and bent midpieces (12.2 ± 2.0) or tails (24.0 ± 2.7) with cytoplasmic droplets. Histomorphometric data were similar between groups, although there was a lower Leydig cell nuclear volume in teratozoospermic samples. Normozoospermic samples contained a higher percentage of haploid cells and had a higher index of total spermatogenic transformation than teratozoospermic samples. Serum concentrations of T (0.5 ± 0.1 vs. 0.8 ± 0.4 ng/mL) and E2 (9.5 ± 1.2 vs. 11.4 ± 2.3 pg/mL) and testicular T concentrations (471.6 ± 65.3 vs. 313.4 ± 57.6 ng/g) were similar between groups. However, compared with normozoospermic samples, teratozoospermic samples had higher testicular E2 concentrations (8.5 ± 3.6 vs. 5.4 ± 0.5 ng/g) and a lower T:E2 ratio (31.8 ± 4.1 vs. 87.2 ± 11.6). There were significant correlations between testicular E2 values and percentages of normal sperm (r = -0.55) as well as those with primary sperm defects (r = 0.58) or abnormal acrosomes (r = 0.64). The T:E2 ratio was also correlated with meiotic index (r = 0.45) and percentage of normal sperm (r = 0.58). In conclusion, a high testicular E2 concentration and a reduced T:E2 ratio were significantly associated with higher ratios of abnormal sperm types, suggesting that the balance between androgens and estrogens is an important endocrine component in the genesis of teratozoospermia in felids.

This study investigated the effects of different doses of 17-beta-estradiol (E(2)) in Rhamdia quelen. Groups of males exposed to different doses of E(2) (0.1 mg kg(-)(1), 1 mg kg(-)(1) and 10 mg kg(-)(1)) were compared with non-exposed male and female fish groups. Among the considered biomarkers, no significant differences were observed for micronuclei test, reduced glutathione concentration and lipid peroxidation. All E(2)-treated individuals had decreased glutathione S-transferase activity. Increased catalase and superoxide dismutase activities, increased vitellogenin expression and decreased metallothionein concentration were observed in males treated with the highest dose. Liver of all test groups showed necrotic areas, but cytoplasm vacuolization was again found only in the individuals exposed to highest dose. E(2) causes deleterious hepatic effects to R. quelen, and vitellogenin expression, catalase and superoxide dismutase activity and metallothionein concentration represent appropriate biomarkers for studying E(2) effects. Additionally, the response of some biomarkers was similar in males exposed to E(2) and unexposed females, and therefore exposure to endocrine disruptors may cause consequences for fish populations.

High temperature is an important factor for reproduction and can induce testicular degeneration. Pentoxifylline is a methylxanthine phosphodiesterase inhibitor with anti - inflammatory and anti - apoptotic properties. Considering the protective propert ies of pentoxifylline and the harmful effects of heat, the present study aimed to use pentoxifylline to reduce the damage induced by heat to the testis. Adult mal e Wistar rats were exposed to testicular heat shock (43ºC for 15 min) , treated with 50 or 100 mg/ k g of pentoxifylline and evaluated at 3, 7, 15, 30 and 60 days after heat shock. Pentoxifylline treatment did not change testicular weight, histomorphometrical parameters or plasma testosterone concentration. However, pentoxifylline inhibited germ cell apoptosis and reduced the severity of pathological lesions at 30 and 60 days after testicular heat shock. In conclusion, pentoxifylline treatment seem ed to inhibit pro - inflammatory and apoptotic mechanisms triggered by testicular heat shock, improving sper matogenesis regeneration.

The intensification of animal production systems presents a potential impact on the welfare of animals. The objective of this work was to assess the welfare of quail Coturnixcoturnix japonicain two maintenance systems: battery cages (BC),and enriched aviary (EA),with saw-dust bedding, sand-bathing area and nests. The experiment procedure involved eight animals per holding area and four repetitions per treatment, an overall of 64 quails. Welfare was assessed through behavioral freedom, sanitary freedom (feather condition and injuries), blood analyses and glicocorticoid metabolites measurement in droppings. Results are presented in the BC order, followed by EA. Water drinking behavior and agonistic behavior were different between treatments (P<0.05). Feather condition was adequate in both treatments, except for the head in BC quails. Blood data were statistically different forred blood cells, hematocrit, hemoglobin, blood proteins, eosinophils, heterophils, lymphocytes and heterophil: lymphocyte ratio. Glicocorticoid metabolites levels were significantly different between maintenance systems. The behavioral and physiological welfare indicators showed higher welfare degree for quails in enriched aviary as compared to battery cages system.


O objetivo deste trabalho foi avaliar o bem-estar de codornas (Coturnix coturnix japonica) em dois sistemas de manutenção: gaiolas industriais em baterias (BC) e aviários enriquecidos (EA) com cama de serragem, caixa de areia e ninhos. O procedimento experimental envolveu oito animais por área experimental equatrorepetiçõesportratamento, totalizando64codornas. O bem-estar animal foi avaliado por meio da liberdade comportamental e da liberdade sanitária -condição das penas e ferimentos -, análises de sangue e mensuração de glicocorticoides nas fezes. Atividade de beber água e comportamento agonístico foram diferentes entre os tratamentos (P<0,05). A condição das penas foi adequada em ambos os tratamentos, exceto para a condição das cabeças em codornas do tratamento BC. Os dados do sangue foram diferentes para eritrócitos, hematócrito, hemoglobina, proteínas sanguíneas, eosinófilos, heterófilos e linfócitos e para a relação heterófilo:linfócito. Os níveis de metabólitos glicocorticoides foram significativamente diferentes entre os sistemas de manutenção. Os indicadores comportamentais e fisiológicos de bem-estar mostraramalto grau de bem-estar para as codornas no EA em relação às no BC.

Foram avaliadas as medidas testiculares e o comportamento sexual durante a colheita de sêmen de seis (6) jumentos da raça Pega, com idades variando entre 3 a 9 anos. Os valores médios obtidos para o comprimento, largura e altura testiculares foram, respectivamente, 10,12 x 7,38 x 7,68 cm para o testículo esquerdo e 10,35 x 6,73 x 7,12 cm para o direito. Näo foi encontrada diferença significante (a=0,05) entre os volumes do testículo esquerdo e direito de um mesmo reprodutor, cujos valores médios foram 201,36 cm cúbicos e 182,34 cm cúbicos, respectivamente. Num total de 68 colheitas, o tempo de reaçäo médio (medido desde que o animal era colocado em presença da fêmea até a ejaculaçäo) foi de 10,98 minutos, variando de dois a 45 min. (coeficiente de variaçäo = 59 por cento). Durante o período de excitaçäo observaram-se reaçöes, consideradas por vários autores como características da espécie, tais como salto sem ereçäo ou com ereçäo apenas parcial do pênis, "flehmen" e movimentos mastigatórios

RESUMEN

Foram estudadas as características físicas e morfológicas do sêmen de seis (06) jumentos da raça Pêga, com idade variando entre 3 e 9 anos. As colheitas de sêmen foram realizadas durante a estaçäo de monta, numa frequência de 2 ejaculaçöes por semana, totalizando setenta (70) amostras. Os valores médios encontrados para os parâmetros espermáticos foram os seguintes: volume total (59,88 mais ou menos 15,54 ml), volume de gel (8,19 mais ou menos 4,07 ml), motilidade espermática total (81,93 mais ou menos 6,27 por cento), motilidade progressiva (72,86 mais ou menos 7,20 por cento), vigor espermático (4,63 mais ou menos 0,49), concentraçäo espermática (444,11 mais ou menos 182,72 x 10 à terceira potência/mm à terceira potência), total de espermatozóides por ejaculado (24,88 mais ou menos 9,59 x 109), total de defeitos espermáticos (15,60 mais ou menos 3,62 por cento), defeitos maiores (8,55 mais ou menos 2,14 por cento) e defeitos menores (7,04 mais ou menos 2,56 por cento). Essas médias näo diferem muito daquelas reportadas para asininos. No entanto, ao serem comparadas com dados relativos a equinos, observa-se a existência de diferenças em alguns aspectos. O volume total do ejaculado de jumentos é inferior, porém com menor incidência e volume de gel, portanto, a porçäo rica em espermatozóides apresenta praticamente o mesmo volume. Entretanto, a concentraçäo espermática e o número total de espermatozóides säo maiores do que em garanhöes. Além disso, pela avaliaçäo da motilidade e vigor espermáticos, constata-se que os jumentos apresentam, de um modo geral, ejaculados de melhor qualidade. Em programas de inseminaçäo artificial estas características indicam a possibilidade de maior número de doses por ejaculado e melhores índices de congelabilidade

Amostras de sêmen e plasma seminal de seis jumentos da raça Pêga foram analisadas para determinaçäo do pH (n=70), osmolalidade (n=69) e níveis de Na (n=49), K (n=51), Ca (n=51) e Mg (n=51). Os valores de pH variaram de 7,0 a 7,8 com uma média de 7,29 mais ou menos 0,18 para o total de ejaculados. A osmolalidade do plasma seminal oscilou entre 273 e 373 mOsm tendo como média global o valor de 302,89 mais ou menos 15,03 mOsm, o que corresponde a um ponto crioscópico de -0,56§C. As concentraçöes média de Na, K, Ca e Mg foram, respectivamente, 238,26 mais ou menos 67,92, 105,20 mais ou menos 32,14, 13,24 mais ou menos 1,62 e 1,54 mais ou menos 0,10 mg para cada 100 ml de plasma seminal. Os resultados obtidos no presente estudo foram confrontados com dados publicados relativos a jumentos e garanhöes, sendo em geral muito próximos. A exceçäo foram os níveis de Mg que se mostraram relativamente baixos nos animais por nós encaminhados. Pela semelhança existente entre as características seminais de asininos e equinos, acredita-se que os métodos utilizados para a congelaçäo de sêmen equino possam, em princípio, ser utilizados para o sêmen de jumentos. A confirmaçäo desta hipótese fica na dependência de estudos complementares
