RESUMEN

Constructing three-dimensional (3D) aligned nanofiber scaffolds is significant for the development of cardiac tissue engineering, which is promising in the field of drug discovery and disease mechanism study. However, the current nanofiber scaffold preparation strategy, which mainly includes manual assembly and hybrid 3D printing, faces the challenge of integrated fabrication of morphology-controllable nanofibers due to its cross-scale structural feature. In this research, a trench-guided electrospinning (ES) strategy was proposed to directly fabricate 3D aligned nanofiber scaffolds with alternative ES and a direct ink writing (DIW) process. The electric field effect of DIW poly(dimethylsiloxane) (PDMS) side walls on guiding whipping ES nanofibers was investigated to construct trench design rules. It was found that the width/height ratio of trenches greatly affected the nanofiber alignment, and the trench width/height ratio of 1.5 provided the nanofiber alignment degree over 60%. As a proof of principle, 3D nanofiber scaffolds with controllable porosity (60-80%) and alignment (30-60%) were fabricated. The effect of the scaffolds was verified by culturing human-induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs), which resulted in the uniform 3D distribution of aligned hiPSC-CMs with ∼1000 µm thickness. Therefore, this printing strategy shows great potential for the efficient engineered tissue construction.

RESUMEN

The severe hazard of chlorinated volatile organic compounds (CVOCs) to human health and the natural environment makes their abatement technology a key topic of global environmental research. Due to the existence of Cl, the byproducts of CVOCs in the catalytic combustion process are complex and toxic, and the possible generation of dioxin becomes a potential risk to the environment. Well-qualified CVOC catalysts should process favorable low-temperature catalytic oxidation ability, excellent selectivity, and good resistance to poisoning, which are governed by the reasonable adjustment of acidity and redox properties. This review overviews the application of different types of multicomponent catalysts, that is, supported noble metal catalysts, transition metal oxide/zeolite catalysts, composite transition metal oxide catalysts, and acid-modified catalysts, for CVOC degradation from the perspective of balance between acidity and redox properties. This review also highlights the synergistic degradation of CVOCs and NOx from the perspective of acidity and redox properties. We expect this work to inspire and guide researchers from both the academic and industrial communities and help pave the way for breakthroughs in fundamental research and industrial applications in this field.

Surface lattice oxygen is crucial to the degradation of volatile organic compounds (VOCs) over transition metal oxides according to the Mars-van Krevelen mechanism. Herein, λ-MnO2 in situ grown on the surface of CoMn spinel was prepared by acid etching of corresponding spinel catalysts (CoMn-Hx-Ty) for VOC oxidation. Experimental and relevant theoretical exploration revealed that acid etching on the CoMn spinel surface could decrease the electron cloud density around the O atom and weaken the adjacent Mn-O bond due to the fracture of the surface Co-O bond, facilitating electron transfer and subsequently the activation of surface lattice oxygen. The obtained CoMn-H1-T1 exhibited an excellent catalytic performance with a 90% acetone conversion at 149 °C, which is 42 °C lower than that of CoMn spinel. Furthermore, the partially maintained spinel structure led to better stability than pure λ-MnO2. In situ diffuse reflectance infrared Fourier transform spectroscopy confirmed a possible degradation pathway where adsorptive acetone converted into formate and acetate species and into CO2, in which the consumption of acetate was identified as the rate-limiting step. This strategy can improve the catalytic performance of metal oxides by activating surface lattice oxygen, to broaden their application in VOC oxidation.

The development of highly efficient and stable monolithic catalysts is essential for the removal of volatile organic compounds (VOCs). Copper foam (CF) is a potential ideal carrier for monolithic catalysts, but its low surface area is not conducive to dispersion of active species, thus reducing the interface interaction with active species. Herein, a vertically oriented Cu(OH)2 nanorod was in situ grown on the CF, which acted as the template and precursor to synthesize CoCu-MOF. The optimized catalyst (12CoCu-R) delivers excellent performance for acetone oxidation with a T90 of 195 °C. Impressively, the catalyst demonstrated satisfactory stability in long-term, cycle, water resistance, and high airspeed tests. Therefore, the present study provides a novel strategy for rationally designing efficient monolithic catalysts for VOC oxidation and other environmental applications.

The extraction, characterization and antioxidant activity of polysaccharides from Choerospondias axillaris leaves were investigated in the present study. Two purified polysaccharide fractions, CALP-1 and CALP-2, were isolated from crude Choerospondias axillaris leaf polysaccharides (CALP) by DEAE-52 cellulose chromatography and Sephadex G-100 column chromatography. The characteristics of CAL-1 and CALP-2 were determined by using High-performance Gel Permeation Chromatography (HPGPC), High-Performance Anion-Exchange Chromatography, HPAEC (HPAEC-PAD) and Fourier transform infrared spectroscopy (FTIR). CALP-1 with molecular weight of 11.20 KDa was comprised of Rhamnose, Arabinose, Galactose, Glucose, Xylose, Mannose and galacturonic acid in a molar ratio of 5.16:2.31:5.50:27.18:1.00:0.76:1.07. CAL-2 with molecular weight of 8.03 KDa consisted of Rhamnose, Arabinose, Galactose, Glucose, and galacturonic acid at a ratio of 1.38:3.63:18.84:8.28:1.45. FTIR revealed that CALP-1 and CALP-2 were acidic polysaccharides. The antioxidant activity of crude CALP, CALP-1 and CALP-2 was evaluated in vitro. The fraction CALP-2 was demonstrated to be of polysaccharide nature containing a large percentage of Galactose but no Xylose and Mannose. The antioxidant activity assays showed that CALP-1 and CALP-2 exhibited antioxidant and scavenging activities on hydroxyl and DPPH radicals in vitro. Compared with pure polysaccharide, crude CALP exhibited stronger anti-oxidant activities. These results will provide a better understanding of Choerospondias axillaris leaf polysaccharide and promote the potential applications of Choerospondias axillaris leaf polysaccharide in the pharmacological field and as a natural antioxidant.

The development of catalysts for volatile organic compound (VOC) treatment by catalytic oxidation is of great significance to improve the atmospheric environment. Size-effect and oxygen vacancy engineering are effective strategies for designing high-efficiency heterogeneous catalysts. Herein, we explored the in situ carbon-confinement-oxidation method to synthesize ultrafine MnOx nanoparticles with adequately exposed defects. They exhibited an outstanding catalytic performance with a T90 of 167 °C for acetone oxidation, which is 73 °C lower than that of bulk MnOx (240 °C). This excellent catalytic activity was primarily ascribed to their high surface area, rich oxygen vacancies, abundant active oxygen species, and good reducibility at low temperatures. Importantly, the synthesized ultrafine MnOx exhibited impressive stability in long-term, cycling and water-resistance tests. Moreover, the possible mechanism for acetone oxidation over MnOx-NA was revealed. In this work, we not only prepared a promising material for removing VOCs but also provided a new strategy for the rational design of ultrafine nanoparticles with abundant defects.

INTRODUCTION: The on-line analysis of active pharmaceutical ingredients (APIs) during the extraction process in herbal medicine is a challenge. Establishing a reliable and robust model is a critical procedure for the industrial application of on-line near-infrared (NIR) technology. OBJECTIVE: To evaluate the advantages of on-line NIR model development using system optimisation strategy, Glycyrrhiza uralensis Fisch was used as a case. The content of liquiritin and glycyrrhizic acid was monitored during pilot scale extraction process of Glycyrrhiza uralensis Fisch in three batches. METHODS: High-performance liquid chromatography (HPLC) was used as reference method for content determination of liquiritin and glycyrrhizic acid. The quantitative models of on-line NIR were developed by system optimisation of processing trajectory. For comparison, the models were simultaneously developed by stepwise optimisation. Moreover, the modelling parameters obtained through system optimisation and stepwise optimisation were reused in three batches. Root mean square error of prediction (RMSEP) and residual predictive deviation (RPD) were used to assess the model quality. RESULTS: The average values of RMSEP and RPD of systematic model for liquiritin in three batches were 0.0361, 4.1525 (first batch), 0.0348, 4.7286 (second batch) and 0.0311, 4.9686 (third batch), respectively. In addition, the modelling parameters of systematic model for glycyrrhizic acid in three batches were same, and the average values of RMSEP and RPD were 0.0665 and 5.2751, respectively. The predictive performance and robustness of systematic models for the three batches were better than the comparison models. CONCLUSION: The work demonstrated that system optimisation quantitative model of on-line NIR could be used to determine the contents of liquiritin and glycyrrhizic acid during Glycyrrhiza uralensis Fisch extraction process.

OBJECTIVE: To conduct a case series analysis on Professor Wang Qi's prescriptions for oligoasthenospermia and a preliminary discussion on the clinical application of the pathogenesis theory of "kidney deficiency with dampness, heat, stasis, toxin and parasites" so as to provide some reference for the diagnosis and treatment of oligoasthenospermia by traditional Chinese medicine (TCM). METHODS: We selected 13 representative cases of oligoasthenospermia treated by Professor Wang Qi, analyzed the basic conditions of the patients, changes of the disease and prescriptions used, and investigated the treatment of oligoasthenospermia in view of Professor Wang Qi's TCM theory on andrology. RESULTS: Excellent therapeutic effect was achieved in 7, indefinite effect in 5 and no effect in 1 of the 13 cases. Totally 38 prescriptions were employed in the treatment, of which Bajitian was used in 30, Tusizi in 27, Gouqizi in 25, Sangshen in 25, Shuizhi in 24 and Danggui in 22. The drugs used in the treatment of 10 or more patients included Bajitian in 12, Sangshen in 11, Shuizhi in 11, Tusizi in 10 and Gouqizi in 10 cases. CONCLUSIONS: Professor Wang Qi's prescriptions are effective for the treatment of oligoasthenospermia and the pathogenesis theory of "kidney deficiency with dampness, heat, stasis, toxin and parasites" is valuable in guiding the clinical diagnosis and treatment in traditional Chinese medicine.

Objective: To analyze the academic thought, medication experience and prescription rules of Academician Wang Qi in the treatment of premature ejaculation (PE) using the TCM inheritance support platform (V2.5). METHODS: We collected and sorted out the medical records on the treatment of PE from Academician Wang Qi's Clinic. We established a database of medical records on the TCM inheritance support platform, analyzed the drugs and prescriptions in the database and explored new prescriptions using "statistical reports" and "data analysis" systems on the platform. RESULTS: A total of 91 effective prescriptions were recorded, involving 148 TCM drugs, with Phellodendron, Amomum Villosum, Polygala Tenuifolia, Tuckahoe, Lodestone, Oyster, Acanthopanax Senticosus, Uncaria, Tribulus, and Keel as the top 10 with the highest frequency of use, which were featured mainly by "warm" and "cold" concerning the four natures, "sweet", "bitter" and "pungent" relating to the five flavors, and acting on "kidney meridian", "liver meridian" and "heart meridian" in terms of the meridian tropisms. In addition, 5 new prescriptions were obtained through unsupervised entropy hierarchical clustering. CONCLUSIONS: In the treatment of PE, Academician Wang Qi employs tranquilizing the mind and consolidating the kidney (An Zhi Gu Shen) as the primary strategy, taking into account the three organs of heart, liver and kidneys, focusing on the phase of calming the mind or regulating the liver or clearing the kidney or controlling fire, and adding or reducing drugs according to different conditions and syndromes, which conforms to his diagnosis and treatment mode of "body differentiation-disease differentiation-syndrome differentiation". The analysis of the potential new prescriptions also accords with Academician Wang Qi's rules of medication, which can provide some ideas for the clinical treatment of and scientific researches on premature ejaculation in the future.

The chemical properties of characteristic components are significant to the manufacturing quality control of big brand traditional Chinese medicine. In this study, the Huangjing Zanyu Capsules were used as the research carrier to determine the content of five characteristic components including icraiin, emodin, schisandrin A, 2,3,5,4'-tetrahydroxystilbene-2-O-ß-D-glucoside, and osthole simultaneously by high-performance liquid chromatography(HPLC). The results showed that the chemical properties of five cha-racteristic components had a good linear relationship(r>0.999 9) within the quantitative range; the relative standard deviations(RSD) was 0.11%-2.0% and 0.25%-2.8% respectively for intra-day and inter-day precision; the RSD of repeatability was 1.8%-2.6%; the RSD of stability within 48 hours was 0.19%-2.8%, and the average recovery rate was 95.52%-100.1%, all meeting the requirements of pharmaceutical quantitative analysis. Additionally, the interval estimation method was used to directly reflect the distribution of samples with abnormal chemical properties of characteristic components, and the results showed ten samples were detected beyound the 95% control line of confidence level. Multivariate statistical process control(MSPC) method was used to monitor the abnormal samples of Huangjing Zanyu Capsules collectively, and the results showed that two samples were beyond the 95% control line of Hotelling's T~2 and three samples beyond the 95% control line of squared prediction error(SPE), indicating consistent quality control of Huangjing Zanyu Capsules. In conclusion, the proposed method is not only accurate and efficient but also a compensation for the traditional single-component quality control method, providing a scientific basis for the quality control in manufacturing process of Huangjing Zanyu Capsules. Furthermore, it could also serve as a reference method for the quality control in manufacturing big brand traditional Chinese medicine.

BACKGROUND: The occurrence of segmental/lobar pattern pneumonia (S/L-PP) in children has recently increased. The pathogens of the disease may change for the misuse of antibiotics and the application of vaccines. Therefore, pathogens positive in hospitalized children with S/L-PP and their association with clinical characteristics may have changed. The aim of this study was to analyze the pathogens positive in hospitalized children with S/L-PP and their association with clinical characteristics. METHOD: The current study analyzed the epidemiological and clinical characteristics of pathogens positive in children with S/L-PP under 14 years old at a single hospital between 1st Jan 2014 and 31st Dec 2018 retrospectively. The pathogens were detected by microbial cultivation, indirect immunofluorescence of the kit (PNEUMOSLIDE IgM), Elisa, and/or real-time PCR in the samples of the patients. RESULTS: A total of 593 children with S/L-PP received treatment at a single hospital during the study period by inclusion criteria. Four hundred fifty-one patients were single positive for one pathogen and 83 patients were positive for at least 2 pathogens. Mycoplasma pneumoniae (M.pneumoniae) (72.34%) was the most commonly detected pathogen, followed by Streptococcus pneumoniae (S.pneumoniae) (8.77%). The prevalence of M.pneumoniae in children with S/L-PP increased with time (p < 0.05). The positive rate of M.pneumoniae increased with ages of patients (p < 0.05). M.pneumoniae was statistically associated with the extrapulmonary manifestations while S.pneumoniae was statistically associated with abnormal white blood cells (WBCs) and C reactive proteins (CRPs) (p < 0.05). CONCLUSION: M.pneumoniae was the most positive pathogen in children with S/L-PP. The positive rate of M.pneumoniae in children with S/L-PP increased with time and the ages of children. M.pneumoniae was associated with extrapulmonary manifestations while S.pneumoniae was associated with abnormal WBCs and CRPs.

Penta-graphene has been intensively studied owing to its superior properties such as being an intrinsic semiconductor and having two dimensional stability. However, the nonmagnetic character makes it difficult for straightforward application in the fields of spintronic or information storage. Here, the deposition effects of Fe-group and Co-group transition metal (TM = Fe, Ru, Os; Co, Rh, Ir) clusters on the penta-graphene have been systemically investigated for their electronic and magnetic properties by using density functional theory (DFT) calculations. We found that the TM deposition stability on penta-graphene is overall greater than that on graphene. Importantly, TM adatoms (adclusters) not only change penta-graphene from being a wide band-gap semiconductor to a narrow band-gap semiconductor, but also introduce large magnetic moments into systems simultaneously. It is worth noting that the Ir5 cluster on penta-graphene is a good candidate for realizing the magnetic half-metallic materials. Our calculated results demonstrate that adatoms can exhibit large out-of-plane magnetic anisotropy energy, e.g., the Os adatom presents the largest value of 113 meV. Therefore, from the application point of view, magnetic functionalization of penta-graphene by TM clusters facilitates its application as a spintronic device or a high-density information storage device.

OBJECTIVE: To analyze the medication rules for oligoasthenozoospermia (OAZ) observed by Wang Qi, an academician, master of Traditional Chinese Medicine (TCM) and initiator of andrology in TCM. METHODS: We collected the outpatient cases of OAZ treated by Wang Qi and established a database of clinical medical records using the TCM Inheritance Auxiliary Platform. Employing the integrated rule-based system for analysis of the software, we modified the mutual information method, complex system entropy clustering analysis and other data mining methods, and summarized the medication rules Wang Qi followed in the treatment of OAZ. RESULTS: A total of 134 prescriptions made by Wang Qi for the treatment of OAZ were collected, involving 110 TCM drugs, which are mainly neutral and warm in nature and taste sweet and mostly act through the liver and kidney meridians. The core formula ingredients of the prescriptions included Morinda officinalis, Cuscuta chinensis, Lycium barbarum, Mulberry, Angelica sinensis, Astragalus mongholicus and Fish Maw, and most frequently Morinda officinalis, Cuscuta chinensis, Lycium barbarum and Mulberry. CONCLUSIONS: Wang Qi holds that kidney deficiency, dampness-heat, blood stasis and toxin are the main pathogenic factors for OAZ. The basic treatment of OAZ is to invigorate the kidney and replenish the essence, and meanwhile activate blood circulation, dissipate stasis and eliminate dampness-heat.

Microwave-assisted extraction was employed to extract polyphenols from the leaf of Magnolia kwangsiensis Figlar & Noot. The yield of polyphenols was 2.44±0.02 % under the optimal conditions of RSM: acetone concentration of 70 %, ratio of solvent to material of 21 mL⋅g-1 and extraction time of 16 min. The antioxidant activities were evaluated in terms of total antioxidant ability, reducing power, DPPH⋅ and ⋅ OH scavenging activity. Results showed the polyphenols presented potential antioxidant activities, especially the stronger scavenging activity on ⋅ OH. In term of ⋅ OH scavenging activity, the IC50 value of NKA-9 purification was 0.335 mg mL-1 , equivalent to 35.23 % of VC . The IC50 values of crude extract and ethyl acetate extract were 0.580 and 0.828 mg mL-1 , equivalent to 60.99 % and 87.07 % of VC . Results indicated that M. kwangsiensis leaf polyphenols present potential antioxidant activities that make it beneficial for human health by preventing or reducing oxidative damage.

OBJECTIVE: To explore the regulatory effect of Huatan Qushi Decoction (HTQSD) on lipid metabolism and semen quality in obese rats with oligoasthenozoospermia (OA) and its underlying mechanisms. METHODS: Ninety-six 8-week-old male Wistar rats were randomly divided into six groups of equal number: blank control, OA model control, Wuzi Yanzong Pills (WZYZP), and high-, medium- and low-dose HTQSD, the blank controls fed with a normal diet and the others with a high-fat diet, all for 8 weeks. Meanwhile, the rats in the blank control and OA model control groups were treated intragastrically with normal saline, those in the WZYZP group with WZYZP at 1.07 g/kg, and those in the high-, medium- and low-dose HTQSD groups with HTQSD at 26.25, 13.125 and 6.5625 g/kg, respectively. The body weights of the animals were obtained at 1, 4 and 8 weeks of modeling and intervention, the levels of serum TG, TC, HDL-C and LDL-C, the liver and testis indexes and semen quality detected at 4 and 8 weeks, and the expression of HO-1 mRNA determined at 8 weeks. RESULTS: At 8 weeks of modeling and intervention, the rats in the model control group, compared with the blank controls, showed significant increases in the body weight (ï¼»523.1 ± 25.54ï¼½ vs ï¼»451.50 ± 27.53ï¼½ g, P < 0.01) and levels of serum TG (ï¼»8.58 ± 0.39ï¼½ vs ï¼»2.18 ± 0.28ï¼½ nmol/L, P < 0.001), TC (ï¼»4.41 ± 1.44ï¼½ vs ï¼»1.68 ± 0.18ï¼½ nmol/L, P < 0.01) and LDL-C (ï¼»2.06 ± 0.17ï¼½ vs ï¼»0.48 ± 0.57ï¼½ nmol/L,P<0.01), but decreases in the level of serum HDL-C (ï¼»27.92 ± 0.40ï¼½ vs ï¼»57.47 ± 1.52ï¼½ nmol/L,P < 0.01), sperm motility (ï¼»11.31 ± 4.87ï¼½% vs ï¼»39.66 ± 2.77ï¼½%, P < 0.01), sperm concentration (ï¼»31.07 ± 10.52ï¼½ vs ï¼»65.37 ± 6.30ï¼½ ×106/ml, P < 0.01) and the expression of HO-1 mRNA (0.16 ± 0.03 vs 1.06 ± 0.20, P < 0.01). The rats in the medium-dose HTQSD group, in comparison with the model controls, exhibited remarkable decreases in the body weight (ï¼»445.13 ± 34.19ï¼½ vs ï¼»523.1 ± 25.54ï¼½ g, P < 0.01) and levels of serum TG (ï¼»2.05 ± 0.27ï¼½ vs ï¼»8.58 ± 0.39ï¼½ nmol/L,P < 0.01), TC (ï¼»1.63 ± 0.21ï¼½ vs ï¼»4.41 ± 1.44ï¼½ nmol/L,P < 0.01) and LDL-C (0.45 ± 0.07) vs ï¼»2.06 ± 0.17ï¼½ nmol/L, P < 0.01), but increases in the level of serum HDL-C (ï¼»48.35 ± 3.63ï¼½ vs ï¼»27.92 ± 0.40ï¼½ nmol/L, P < 0.01), sperm motility (ï¼»32.84 ± 6.22ï¼½% vs ï¼»11.31 ± 4.877ï¼½%, P < 0.01) and sperm concentration (ï¼»46.90 ± 6.39ï¼½ vs ï¼»31.07 ± 10.52ï¼½ ×106/ml, P < 0.05), and the low-dose HTQSD group showed a significantly up-regulated expression of HO-1 mRNA (0.76 ± 0.13 vs 0.16±0.03, P < 0.01). CONCLUSIONS: HTQSD can regulate the blood lipid level and improve semen quality in obese rats with oligoasthenozoospermia by promoting serum anti-oxidation and reducing oxidative stress.

The quality of compound traditional Chinese medicine is the prerequisite and foundation for its stable efficacy. Based on the quality by design( QbD) concept,the controllable extraction times,extraction time and the ratio of water were the critical process parameters( CPPs) in the Tuomin Dingchuan Prescription extraction process. The CQAs corresponding to CPPs were screened from the four potential critical quality attributes( p CQAs),namely the extraction amount of solid matter,the content of amygdalin,the content of cimicifugoside and the content of 5-O-methylvisammioside by orthogonal experiment. The extraction amount of solid matter and the content of amygdalin were determined as CQAs in the extraction process by the variance analysis of Box-Behnken experimental. The optimal extraction process based on the linear model between CQAs and CPPs of the extraction process was immersion in water for 30 minutes,extraction for three times,extraction for 100 minutes each time and 10 times of water volume. The control space was established for the extraction amount of solid matter and the content of amygdalin,and both of them could be controlled simultaneously to achieve the optimization objective. The molding ratio of Tuomin Dingchuan granules was regarded as CQA in forming process. On the basis of the single factor investigation,the ratio of dry extract powder to excipient and the ratio of ethanol were determined as CPPs. The central composite design( CCD) was used to optimize the forming process of Tuomin Dingchuan granules. The results showed that the dextrin was used as the filler; the ratio of dry paste to dextrin was 1 ∶1; and 0. 3 m L·g-1 of 70% ethanol was added as binder. The soft material and granules conformed to the actual production requirements.

BACKGROUND/AIMS: Based on the theory of constitution in Traditional Chinese Medicine (TCM), the Chinese Han population has been classified into nine constitutions. Of these, Yang deficiency constitution mainly exhibit cold intolerance while Yin deficiency constitution mainly exhibit heat intolerance. Some studies have been carried out to explore the modern genetic and biological basis of such constitution classification, but more remains to be done. MicroRNA (miRNA) serves as post-transcriptional regulators of gene expression and may play a role in the classification process. Here, we examined miRNA expression profile of saliva to further improve the comprehensiveness of constitution classification. METHODS: Saliva was collected from Chinese Han individuals with Yang deficiency, Yin deficiency and Balanced constitutions (n=5 each), and miRNA expression profile was determined using the Human miRNA OneArray®v7. Based on 1.5 Fold change, means log2|Ratio|≥0.585 and P-value< 0.05, differentially expressed miRNA was screened. Target genes were predicted using DIANA-TarBasev7.0 and analysis of KEGG pathway was carried out using DIANA-mirPathv.3. RESULTS: We found that 81 and 98 differentially expressed miRNAs were screened in Yang deficiency and Yin deficiency constitution, respectively. Among them, 16 miRNAs were identical and the others were unique. In addition, the target genes that are regulated by the unique miRNAs were significantly enriched in 27 and 20 signaling pathways in Yang deficiency and Yin deficiency constitution, respectively. Thyroid hormone signaling pathway is present in both constitutions. These unique miRNAs that regulated target genes of thyroid hormone signaling pathway may be associated with cold intolerance or heat intolerance. CONCLUSION: The results of our study show that Yang deficiency and Yin deficiency constitutions exhibit systematic differences in miRNA expression profile. Moreover, the distinct characteristics of TCM constitution may be explained, in part, by differentially expressed miRNAs.

OBJECTIVE: To investigate the effect of Bazi Granules on sperm quality in male rats with oligoasthenozoospermia (OAS) induced by multi-glycosides of tripterygium wilfordii (GTW). METHODS: Thirty-six SD male rats were randomly divided into six groups of equal number: normal control, OAS model control, Wuziyanzong Pills (WYP), and low-, medium- and high-dose Bazi. The OAS model was established in the rats except those of the normal control group by intragastrical delivery of GTW at 30 mg/kg/d for 40 days. From the 41st day, the animals of the normal and OAS model control groups were fed with distilled water, those of the WYP group treated by gavage with WYP at 1.02 g/kg/d, and those of the low-, medium- and high-dose Bazi groups intragastically given Bazi Granules 3 (5.27 g/kg), 6 (10.54 g/kg) and 12 (21.08 g/kg) times, respectively, that of the human-equivalent dose. Semen parameters and the contents of superoxide dismutase (SOD) and malondialdehyde (MDA) in the testis tissue were determined after 28 days of treatment. RESULTS: After treatment, the rats of the of the high-, medium- and low-dose Bazi groups, compared with the OAS model controls, showed significant increases in sperm concentration (ï¼»1 050.71 ± 203.71ï¼½, ï¼»1 370.06 ± 166.01ï¼½ and ï¼»1 302.53 ± 476.51ï¼½ vs ï¼»617.01 ± 237.08ï¼½ ×106/ml, P < 0.05), sperm motility (ï¼»0.56 ± 0.24ï¼½%, ï¼»0.73 ± 0.14ï¼½% and ï¼»0.70 ± 0.23ï¼½% vs ï¼»0.07 ± 0.05ï¼½%, P < 0.05), sperm average path velocity (ï¼»85.71 ± 30.35ï¼½, ï¼»83.83 ± 10.31ï¼½ and ï¼»75.06 ± 19.70ï¼½ vs ï¼»43.45 ± 38.74ï¼½ µm/s, P < 0.05), sperm curvilinear velocity (ï¼»101.76 ± 23.28ï¼½, ï¼»119.60 ± 21.22ï¼½ and ï¼»102.11 ± 32.89ï¼½ vs ï¼»53.63 ± 47.91ï¼½ µm/s, P < 0.05), sperm straight line velocity (ï¼»62.75 ± 7.63ï¼½, ï¼»67.80 ± 5.05ï¼½ and ï¼»64.11 ± 12.03ï¼½ vs ï¼»40.18 ± 36.86ï¼½ µm/s, P < 0.05), and the SOD level (ï¼»380.23 ± 75.07ï¼½, ï¼»349.53 ± 97.48ï¼½ and ï¼»415.07 ± 72.01ï¼½ vs ï¼»304.62 ± 27.17ï¼½ U/mg, P < 0.05), but a remarkable decrease in the MDA level (ï¼»0.33 ± 0.16ï¼½, ï¼»0.22 ± 0.05ï¼½ and ï¼»0.34 ± 0.22ï¼½ vs ï¼»0.73 ± 0.20ï¼½ nmol/mg, P < 0.05). CONCLUSIONS: Bazi Granules can significantly improve the sperm quality of OAS rats, which may be related to its abilities of repairing oxidative stress injury and enhancing oxidation resistance.

BACKGROUND: Atherosclerosis is a chronic and complex disease caused by lipid disorder, inflammation, and other factors. It is closely related to cardiovascular diseases, the chief cause of death globally. Peroxisome proliferator-activated receptors (PPARs) are valuable anti-atherosclerosis targets that showcase multiple roles at different pathological stages of atherosclerosis and for cell types at different tissue sites. AIM OF REVIEW: Considering the spatial and temporal characteristics of the pathological evolution of atherosclerosis, the roles and pharmacological and clinical studies of PPARs were summarized systematically and updated under different pathological stages and in different vascular cells of atherosclerosis. Moreover, selective PPAR modulators and PPAR-pan agonists can exert their synergistic effects meanwhile reducing the side effects, thereby providing novel insight into future drug development for precise spatial-temporal therapeutic strategy of anti-atherosclerosis targeting PPARs. KEY SCIENTIFIC: Concepts of Review: Based on the spatial and temporal characteristics of atherosclerosis, we have proposed the importance of stage- and cell type-dependent precision therapy. Initially, PPARs improve endothelial cells' dysfunction by inhibiting inflammation and oxidative stress and then regulate macrophages' lipid metabolism and polarization to improve fatty streak. Finally, PPARs reduce fibrous cap formation by suppressing the proliferation and migration of vascular smooth muscle cells (VSMCs). Therefore, research on the cell type-specific mechanisms of PPARs can provide the foundation for space-time drug treatment. Moreover, pharmacological studies have demonstrated that several drugs or compounds can exert their effects by the activation of PPARs. Selective PPAR modulators (that specifically activate gene subsets of PPARs) can exert tissue and cell-specific effects. Furthermore, the dual- or pan-PPAR agonist could perform a better role in balancing efficacy and side effects. Therefore, research on cells/tissue-specific activation of PPARs and PPAR-pan agonists can provide the basis for precision therapy and drug development of PPARs.

BACKGROUND: The treatment of metabolic system, cardiovascular system, and nervous system diseases remains to be explored. In the internal environment of organisms, the metabolism of substances such as carbohydrates, lipids and proteins (including biohormones and enzymes) exhibit a certain circadian rhythm to maintain the energy supply and material cycle needed for the normal activities of organisms. As a key factor for the health of organisms, the circadian rhythm can be disrupted by pathological conditions, and this disruption accelerates the progression of diseases and results in a vicious cycle. The current treatments targeting the circadian rhythm for the treatment of metabolic system, cardiovascular system, and nervous system diseases have certain limitations, and the identification of safer and more effective circadian rhythm regulators is needed. AIM OF THE REVIEW: To systematically assess the possibility of using the biological clock as a natural product target for disease intervention, this work reviews a range of evidence on the potential effectiveness of natural products targeting the circadian rhythm to protect against diseases of the metabolic system, cardiovascular system, and nervous system. This manuscript focuses on how natural products restore normal function by affecting the amplitude of the expression of circadian factors, sleep/wake cycles and the structure of the gut microbiota. KEY SCIENTIFIC CONCEPTS OF THE REVIEW: This work proposes that the circadian rhythm, which is regulated by the amplitude of the expression of circadian rhythm-related factors and the sleep/wake cycle, is crucial for diseases of the metabolic system, cardiovascular system and nervous system and is a new target for slowing the progression of diseases through the use of natural products. This manuscript provides a reference for the molecular modeling of natural products that target the circadian rhythm and provides a new perspective for the time-targeted action of drugs.
