RESUMEN

Lumpy skin disease (LSD) is a contagious non-zoonotic viral disease of cattle. The disease raises great concern due to the recent rapid spread toward free countries and reoccurrence in countries where control and preventive measures had achieved eradication. Deep nodules involving skin, subcutaneous tissue, and occasionally muscles are localized mostly in the head, neck, perineum, genitalia, udder, and limbs. LSD can cause large economic losses mainly because of the decline in milk production and the decrease in hide value, in addition to the ban of movement of animals and animal products.

Lumpy skin disease (LSD) is a highly infectious disease of cattle caused by a virus of the Poxviridae family, genus Capripoxvirus. The present study was designed to determine the prognostic ability of serum IL-6 in LSD using a binary logistic regression model at baseline sampling. A 17-day cohort study was conducted on a recent outbreak of LSD among cattle in the Lodhran District of Punjab, Pakistan. Infected cattle were divided into two categories based on their clinical status on day 17 as recovered (n = 33) or unrecovered (n = 17). Nodular lesions and scab specimens (n = 50) were used for the isolation of the lumpy skin disease virus and were confirmed by PCR. In recovered animals, hematological results showed marked leukocytosis, eosinophilia, lymphocytosis, neutrophilia, and monocytopenia. However, marked erythrocytosis, leukopenia, and thrombocytopenia were observed in the unrecovered animals at the final sampling point of the study. Serum levels of total protein, albumin, and glucose were significantly higher in the recovered animals. Meanwhile, aspartate aminotransferase, alkaline phosphatase, lactate dehydrogenase, creatinine phosphokinase, total bilirubin, and direct bilirubin were found considerably higher in the unrecovered group. Receiver-operating characteristic curve analysis for serum IL-6 at baseline predicts the extended clinical conditions at the cut-off value of 85.16 pg/mL (55% specificity, 94% sensitivity, area under the curve 0.8039, respectively). In conclusion, the disease-induced hematological and biochemical alterations were significantly ameliorated in the recovered animals. In addition, the study revealed that serum IL-6 can be used as a valid marker for predicting the clinical worsening of LSD in cattle.

Mycoplasma mycoides subsp. mycoides (Mmm) is the causative agent of contagious bovine pleuropneumonia (CBPP). Lumpy skin disease (LSD) is a viral disease of cattle caused by lumpy skin disease virus (LSDV). LSD and CBPP are both transboundary diseases spreading in the same areas of Africa and Asia. A combination vaccine to control CBPP and LSD offers significant value to small-scale livestock keepers as a single administration. Access to a bivalent vaccine may improve vaccination rates for both pathogens. In the present study, we evaluated the LSDV/CBPP live combined vaccine by testing the generation of virus neutralizing antibodies, immunogenicity, and safety on target species. In-vitro assessment of the Mycoplasma effect on LSDV growth in cell culture was evaluated by infectious virus titration and qPCR during 3 serial passages, whereas in-vivo interference was assessed through the antibody response to vaccination. This combined Mmm/LSDV vaccine could be used to protect cattle against both diseases with a single vaccination in the endemic countries. There were no adverse reactions detected in this study and inoculated cattle produced high levels of specific antibodies starting from day 7 post-vaccination, suggesting that this combination vaccine is both safe and effective.

BACKGROUND: New Lumpy Skin Disease (LSD) outbreaks are currently circulating in Chaharmahal and Bakhtiari province, (western Iran, in central Zagros Mountains). OBJECTIVES: The aim of this study was to model the risk probability of LSD in this area. METHOD: Data were collected from veterinary organizations between 2012 and 2016. During this period, 290 outbreaks were registered. The herds were grazing and zero grazing. The average size of herds was 2958. We analyzed the potential for an outbreak of LSD in this area basing on the grid maps with the resolution of 1 km. In this study, 22 environmental variables (19 bioclimatic and 3 topography variables) were used to explore the environmental influences on LSD outbreak by maximum entropy ecological niche modeling (MaxEnt). RESULTS: The results showed that mainly the central, northern, northeast and southern parts of Charmahal and Bakhtiari were the most very high risk areas for LSD. The MaxEnt model performed, with an area under the receiver operating characteristic curve (AUC) of 0.977 and 0.972 for training and test data, respectively. It showed high accuracy for predicting the prevalence of LSD (AUC close to 1). The precipitation of coldest season, isothermality and mean temperature of wettest season made the greatest contribution to the model (68.1%). Evaluating the importance of environmental variables, which were derived from the jackknife test, stated the precipitation of the wettest period and the coldest season and isothermality as the bioclimatic variables in explaining LSD prevalence compared to the other variables. CONCLUSION: The MaxEnt model could be applied to predict the LSD risk probability of occurrence in central Zagros Mountains of Iran based on the current prevalence data of the disease. Also, the model confirmed that coexistence of weather conditions including defined humidity and temperature is necessary for the disease occurrence.

In this comparative study, we examine the safety of the sheeppox (SPP) and goatpox (GTP) vaccines and the protective response of these vaccines in cattle against a virulent lumpy skin disease (LSD) field strain. The vaccine safety was tested in rabbits, mice and cattle using ten times recommended dose. In the safety trial, none of the vaccinated animals showed any deviation from physiological norms or fever, inappetence or local/ generalized skin reactions. In the challenge trial, both SPP and GTP vaccine groups developed virus-neutralizing antibodies with an average titre of 2.1 log2 at 21 days post-vaccination. No significant difference in seroconversion was found in cattle vaccinated with SPP and GTP vaccines (P ≥ 0.05). When challenged with a virulent LSD field strain, one animal vaccinated with the SPP Niskhi vaccine strain showed typical LSD skin lesions at the injection sites of different dilutions of the challenge virus. All animals vaccinated with GTP G20-LKV vaccine strain showed full protection. After infection with the challenge virus, unvaccinated fully susceptible control cattle showed characteristic clinical signs of LSD. The average protective index for SPP and GTP vaccine groups was 5.3 ± 1.42 and 5.9 ± 0.00, respectively.
