ABSTRACT

In current study, the enhancement effect of magnetite on anaerobic digestion was evaluated at increased organic loading rate (OLR) from 1.6 to 25.6 kg COD·m-3·d-1. The supplement of magnetite enhanced the methane yield by 7-483% accompanied with faster VFAs conversion. Microbial analysis suggested the varied enhancing effect achieved at different OLRs was attributed to different syntrophic interactions triggered by magnetite. More specially, an electroactive syntropy was established between Trichococcus with Methanobacterium at OLR lower than 6.4 kg COD·m-3·d-1, while with the OLR increase, more acid fermentative bacteria (Propionimicrobium, Syner-01) were enriched and further enhanced methanogenesis in a syntrophic way with Methanosaeta. Overall, the incorporation of magnetite was a promising approach to achieve efficient anaerobic digestion, OLR was also critical factor affecting the methanogenesis and should be carefully regulated in future application.

ABSTRACT

The coronavirus disease 2019 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) threatens global public health. The development of a vaccine is urgently needed for the prevention and control of COVID-19. Here, we report the pilot-scale production of an inactivated SARS-CoV-2 vaccine candidate (BBIBP-CorV) that induces high levels of neutralizing antibodies titers in mice, rats, guinea pigs, rabbits, and nonhuman primates (cynomolgus monkeys and rhesus macaques) to provide protection against SARS-CoV-2. Two-dose immunizations using 2 µg/dose of BBIBP-CorV provided highly efficient protection against SARS-CoV-2 intratracheal challenge in rhesus macaques, without detectable antibody-dependent enhancement of infection. In addition, BBIBP-CorV exhibits efficient productivity and good genetic stability for vaccine manufacture. These results support the further evaluation of BBIBP-CorV in a clinical trial.

ABSTRACT

Background: In treating androgenetic alopecia, 5% minoxidil is a commonly used topical drug. By using electrodynamic microneedle at the same time may increase absorption of minoxidil and further stimulate hair growth.Objective: A 24-week, randomized, evaluator blinded, comparative study was performed to evaluate the efficacy of treating Chinese male androgenetic alopecia using microneedle combined with 5% minoxidil topical solution. Methods: Randomized subjects received topical 5% minoxidil (group 1, n = 20), local electrodynamic microneedle treatments (group 2, n = 20), or local electrodynamic microneedle treatments plus topical 5% minoxidil (group 3, n = 20). A total of 12 microneedle treatments were performed every 2 weeks with 2ml 5% minoxidil delivery in group three during each microneedle treatment. Patient receiving topical 5% minoxidil applied 1 ml of the solution twice daily over the course of the study. A total of 60 Chinese male subjects with Norwood-Hamilton type III-VI androgenetic alopecia were treated.Results: The mean improvement in total hair density from baseline to 24 weeks was 18.8/cm2 in group 1, 23.4/cm2 in group 2, and 38.3/cm2 in group 3. The hair growth in the three groups was significantly different (P = 0.002), but there were no significant differences in toxicity found between the three groups.Conclusions: Treatment with microneedle plus topical 5% minoxidil was associated with the best hair growth.

ABSTRACT

Small dams are widely constructed in urban rivers as landscape engineering practice, which increasingly cause eutrophication problems. Phosphorus retention in dammed rivers is a critical factor driving eutrophication, but it is little known in urban landscape river systems controlled by small dams. In this study, we investigated the retention of different phosphorus species along an urban landscape river with 30 rubber dams. We found that 42.5% (7.69 metric tons/yr) of the total phosphorus (TP) was trapped within dams, of which total particulate phosphorus (TPP) retention load accounted for 81.5%. From first river segment BBF-4# to the segments further downstream, the TP retention rate sharply decreased from 47.6% to -8.3%-9.2%, and phosphorus was mainly retained in the uppermost segment of the dammed river. The retention rate of dissolved reactive phosphorus (86.3%) was higher than that of TPP (40.3%) because of biological uptake. Further, with a retention rate of -11.3%, the dammed river was a net source of dissolved organic phosphorus. Different hydrological regimes, due to seasonal events and dam management, greatly influenced phosphorus retention within the dammed river, resulting in higher retention loads in the rainy season than in the dry season, and very low retention loads in the snowmelt season, with 1.48, 0.55 and 0.06 t/month, respectively. Our findings imply that management practices should focus on reducing the phosphorus export from the upper watershed and improving the hydrodynamic conditions of the dammed urban landscape river with regard to eutrophication.

ABSTRACT

Eutrophication is one of the greatest threats to global freshwater ecosystems. The phytoplankton responses to nutrient inputs vary in different water bodies, so it is particularly important to determine the nutrient thresholds and synergistic interactions between nutrients in different freshwater ecosystems. Field sampling and bioassay experiments were conducted to determine the thresholds of soluble reactive phosphorus (SRP), nitrate-nitrogen (NO3-N), and ammonium-nitrogen (NH4-N) in Miyun Reservoir. A separate nutrient addition bioassay was designed to assess the synergistic interactions between these nutrients. Chlorophyll a (Chl a) concentrations were used to estimate phytoplankton biomass. The results showed the following: (1) nutrient threshold bioassay indicated that eutrophication thresholds of SRP, NO3-N, and NH4-N should be targeted at below 0.04 mg P L-1, 0.5 mg N L-1, and 0.3 mg N L-1, respectively, to limit the growth of phytoplankton. (2) The stimulatory effect of "NH4-N plus P" on phytoplankton biomass was greater than "NO3-N plus P" at the same N concentration, and "NH4-N plus NO3-N" did not show such associated stimulatory effect as "NH4-N plus P" or "NO3-N plus P". (3) The average concentrations of total phosphorus (TP), NO3-N, and NH4-N in Miyun Reservior were 0.017 mg P L-1, 0.620 mg N L-1, and 0.143 mg N L-1, respectively. The reservoir-wide average Chl a is below 20 µg L-1 on an annual basis. (4) Ammonium was an important factor for the growth of phytoplankton and inputs of both NH4-N and NO3-N should be reduced to control bloom formation. Our findings imply that although P load reduction is important, appropriate reductions of all forms of N in watershed is recommended in the nutrient management strategy for Miyun Reservoir.

ABSTRACT

The continuous and sporadic human transmission of highly pathogenic avian H5N1 and H7N9 influenza viruses illustrates the urgent need for efficacious vaccines. However, all tested vaccines for the H5N1 and H7N9 viruses appear to be poorly immunogenic in mammals. In this study, a series of vaccines was produced using reverse genetic techniques that possess HA and NA genes from the H5N1 virus in the genetic background of the high-yield strain A/PR/8/34 (H1N1). Meanwhile, a group of H7N9 VLP vaccines that contain HA from H7N9 and NA and M1 from A/PR/8/34 (H1N1) was also produced. The HA amino acids of both the H5N1 and H7N9 vaccines differed at residues 226 and 228, both of which are critical for receptor specificity for an avian or mammalian host. Mice received two doses (3µg of HA each) of each vaccine and were challenged with lethal doses of wild type H5N1 or H7N9 viruses. The results showed that a recombinant H5N1 vaccine in which the HA amino acid G228 (avian specificity) was converted to S228 (mammalian specificity) resulted in higher HI titers, a lower viral titer in the lungs, and 100% protection in mice. However, a H7N9 VLP vaccine that contains L226 (mammalian specificity) and G228 (avian specificity) in HA showed better immunogenicity and protection efficacy in mice than VLP containing HA with either L226+S228 or Q226+S228. This observation indicated that specific HA residues could enhance a vaccine's protection efficacy and HA glycoproteins with both avian-type and human-type receptor specificities may produce better pandemic influenza vaccines for humans.

ABSTRACT

OBJECTIVE: To explore the effectiveness of the traditional Chinese herbal medicine Lingzhi (Ganoderma lucidum) preparation in treating simian acquired immune deficiency syndrome (SAIDS). METHODS: Five female adult Chinese rhesus monkeys were inoculated rectally with SIVmac239, and were all diagnosed as SAIDS by laboratory and clinical examinations 17 months later. Of these 5 monkeys, 3 (#393, #374, and #381; treatment group) were orally administered with Ganoderma lucidum (2 spores powder capsules plus 2 spores oil capsules on a daily basis), and the remaining other two monkeys (#348 and #361) served as control and did not receive treatment. RESULTS: Animal #393 (treatment group), #361 (control group) and #348 (control group) died of SAIDS (opportunity infection) 3.5 months, 6 months, and 11 months later, respectively. Two animals (#374 and #381) survived. The necropsy revealed depletion and/or exhaustion of their lymphoid tissue. In the monkey #374, the peripheral CD4(+) T lymphocyte increased by 30% in the 6(th) month compared with the baseline level and then fluctuated. The plasma viral load gradually fell and reached about 1 log(10) in the treatment group, but remained stable in the control group. As shown by pathological examinations, the lymph node and spleen of monkeys #374 (treatment group) and #381 (treatment group) showed rehabilitation and reconstruction in the lymphatic tissue, thymus, nerve tissue of gyrus hippocampi, pituitary gland, pineal body, thyroid gland, adrenal gland, and ovary. In the control group, however, animals experienced depletion of lymph nodes, atrophy of spleen, disappearance of thymus, and other disorders in endocrine organs. CONCLUSION: Ganoderma lucidum preparation may have certain protective effect on the immune system, nervous system, and endocrine system of monkeys with SAIDS.

ABSTRACT

In this article, polyphosphate-accumulating organisms (PAOs) from a lab-scale continuous-flow two-sludge system was isolated and identified, the different phosphorus accumulation characteristics of the isolates under anoxic and aerobic conditions were investigated. Two kinds of PAOs were both found in the anoxic zones of the two-sludge system, one of them utilized only oxygen as electron acceptor, and the other one utilized either nitrate or oxygen as electron acceptor. Of the total eight isolates, five isolates were capable of utilizing both nitrate and oxygen as electron acceptors to uptake phosphorus to some extent. And three of the five isolates showed good phosphorus accumulative capacities both under anoxic or aerobic conditions, two identified as Alcaligenes and one identified as Pseudomonas. Streptococcus was observed weak anoxic phosphorus accumulation because of its weak denitrification capacity, but it showed good phosphorus accumulation capacity under aerobic conditions. One isolates identified as Enterobacteriaceae was proved to be a special species of PAOs, which could only uptake small amounts of phosphorus under anoxic conditions, although its denitrification capacity and aerobic phosphorus accumulation capacity were excellent.

ABSTRACT

The ability of simultaneous biological phosphorus and nitrogen removal was investigated in a lab-scale continuous-flow two-sludge system. Alternating anaerobic and anoxic conditions were combined with contact oxidation stage for treating raw municipal wastewater. Long-term experiments showed that the contradiction of competing for the organic substrate between denitrifying bacteria and PAOs (phosphorus accumulating organisms) in traditional phosphorus and nitrogen removal system has been resolved. The system can adapt to low influent COD/TN ratio (C/N). Furthermore the SRT (sludge retention time) of nitrifying sludge and denitrifying phosphorus removal sludge can be controlled at optimal conditions respectively. The removal efficiency of COD, TP, TN, and NH4-N was 81.78%, 92.51%,75.75%, and 84.47% respectively. It was also found that the appropriate influent C/N should be controlled at the range of 3.8-6, while the optimal C/N to the system ranged between 4-5, and the BFR (bypass sludge flow rate) should be controlled at 0.35 around.
