ABSTRACT

ObjectiveTo investigate the therapeutic effect of Xuebijing injection (XBJ) on sodium taurocholate (Na-Tc)-induced severe acute pancreatitis (SAP) in rats. MethodForty rats were randomly assigned into 5 groups: sham operation group, SAP model group, and low-, medium-, and high-dose (4, 8, 12 mL·kg·d-1, respectively) XBJ groups. SAP model was established by retrograde injection of Na-Tc (1 mL·kg-1) into the biliary and pancreatic ducts. XBJ was injected intraperitoneally 3 days before and 0.5 h after modeling. The ascitic fluid volume and the pancreas weight-to-body weight ratio were measured. The pathological changes of pancreatic tissue were observed via hematoxylin-eosin (HE) staining. The protein levels of formyl peptide receptor 1 (FPR1) and nucleotide-binding oligomerization domain-like receptor 3 (NLRP3) in pancreatic tissue were detected by immunohistochemistry. Western blot was employed to determine the expression levels of NADH-ubiquinone oxidoreductase chains 1-6 (MT-ND1, MT-ND2, MT-ND3, MT-ND4, MT-ND5, and MT-ND6) in rat plasma. ResultCompared with sham operation group, the SAP model group showcased increased ascitic fluid volume and pancreas weight-to-body weight ratio (P<0.05), serious lesions in pancreatic tissue, increased total pathological score (P<0.05), and up-regulated protein levels of FPR1 and NLRP3 in pancreatic tissue (P<0.05). The model group had lower MT-ND2 level (P<0.05) and higher MT-ND1, MT-ND3, and MT-ND6 levels in plasma (P<0.05) than the sham operation group, while MT-ND4 and MT-ND5 had no significant differences between the two groups. Compared with SAP model group, the XBJ treatment decreased ascitic fluid volume and pancreas weight-to-body weight ratio (P<0.01), ameliorated pancreatic lesions, and down-regulated the protein levels of FPR1 and NLRP3 in pancreatic tissue (P<0.01). The treatments, especially high-dose XBJ (P<0.01), down-regulated the expression of MT-ND1 (P<0.01), MT-ND3 (P<0.01), MT-ND6 (P<0.01), and MT-ND4 and did not change that of MT-ND5. ConclusionXBJ may antagonize partial mitochondrial N-formyl peptides and excessive inflammatory response mediated by FPR1/NLRP3 to treat SAP in rats.

ABSTRACT

Psammosilene tunicoides is one of the main ingredients of the "Yunnan Baiyao". P. tunicoides is an endangered species included in the secondary protection list in China Plant Red Data Book as well as the endemic species in Southwest China. Its natural resources could not meet the needs of pharmaceutical production. Construction of core collection of P. tunicoides will lay the foundation for germplasm improvement and molecular breeding. The sequence variation of the key enzymes gene locus (β-AS) were carried out to survey the population structure and population history of the species. Among the 11 populations across its geographical range, 36 haplotypes were identified. The levels of haplotype diversity (Hd=0.905) were high, while the levels of population differentiation (GST=0.280) were low. Analysisof molecular variance (AMOVA) indicated that a significantly greater proportion of total genetic variationpartitioned among populations thanwithin populations (values of 77.43% and 22.57%, respectively). These results in combination with the star-like phylogenetic network analysis indicate that Hap1 as an ancestral haplotypewas shared in four populations, Hap2, Hap4, Hap15 and Hap16 are occurred in two populations, the remains as private haplotype only distributed in single population. The strategy of core collection was constructed in order to maximumpreserve genetic diversity of P. tunicoides.

ABSTRACT

Nanotechnology has broad prospects and great potential in improving the druggability of active ingredients of Chinese materia medica (CMM). This article systematically summarized the methods and advantages of nanotechnology in improving oral absorption, changing the distribution in the body, increasing the targeting ability, and improving transdermal and mucous membrane absorption of active ingredients of CMM. In addition, we analyzed the problems in the development of improving the druggability of active ingredient of CMM by nanotechnology, put forward the strategies for solving these problems, and discussed the research prospects.

In this review, the ability of borneol and menthol as the common penetration enhancers in traditional Chinese medicines was summarized, and the effect of borneol and menthol on the ability of nanopreparations across the main physiological barriers of the human body promoting drug absorption or increasing the distribution of drugs was also reviewed, which aims to provide reference for promoting the treatment effect of nanopreparations through the combination of traditional Chinese medicine and modern preparation technology.

ABSTRACT

[Objective] To explore the mechanism of acupuncture at Zusanli point(ST36) in the treatment of functional gastrointestinal disorders(FGIDs). [Methods] A total of 40 Wistar rats were randomized into normal(n=10),model(n=10), Zusanli point(ST36),non-acupuncture point(n=10). FGIDs model was established by exerting the factors of cold-constraint stimulation and hunger disorder synthesis . And the content of rats serum and stomach tissue Gastrin (GAS) and Motilin(MTL) was observed by radioimmuno-assay. [ Results] Compared with the normal group, the content of rats serum and stomach tissue GAS and MTL of the model group was significantly reduced(P<0.01);Compared with the model group, the content of rats serum and stomach tissue GAS and MTL of ST36 group was significantly ascended(P<0.01);Compared with the non-acupuncture point group, the content of rats serum and stomach tissue GAS and MTL of ST36 group had statistically significant differences(P<0.01). [Conclusion] Acupuncture at ST36 had obvious improvement in FGIDs, its mechanism was closely related to GAS and MTL.

ABSTRACT

Gitelman's syndrome (GS) is an autosomal recessive hereditary tubulopathy.It is caused by sodium reabsorption in the distal convoluted tubule of the renal unit,which is not yet fully cured.It is characterized by great heterogeneity with clinical manifestations.Some patients with no symptoms,were confirmed by hypokalemia with physical examination.The others can be manifested as fatigue,nocturia,polydipsia and hypotension.Less literature is reported about GS combined with osteoarthrosis.In this report,the author reported a patient of GS with osteoarthrosis characterized by hypokalemia,hypomagnesemia,metabolic alkalosis,lower blood pressure,and swelling and pain in left knee joint.The symptoms were improved after treatment with potassium and magnesium supplementation

ABSTRACT

Objective To learn current status of psychological capital and health-promoting lifestyle among duty nurses,and to analyze their relationship.Methods A total of 527 duty nurses from tertiary referral hospital affiliated to Zhejiang Chinese Medical University were selected and investigated using rating scale with Nurses' Psychological Capital Questionnaire,health-promoting lifestyle questionnaire and a self-designed questionnaire about demographic information.We process the result by pearson correlation analysis and multiple linear regression analysis.Results Overall respondents showed high psychological capital scores(79.66 ± 14.07),and the four dimension scores rated in sequence were self-efficiency(67.81%),resilience(66.47%),optimism(66.17%),hope(65.00%).Overall score in health-promoting lifestyle was (60.38 ± 17.71).The five dimension scores rated in sequence were occupational safety (68.14%),self-actualization (62.77%),development of interpersonal support (60.99%),balanced diet (58.70%),sports health care(34.32%).The result of pearson correlation analysis showed positive correlation between psychological capital and health-promoting lifestyle among duty nurses (P < 0.05).Self-realization and occupational safety has important influence on psychological capital.Conclusions Health-promoting lifestyle was an important influencing factor of psychological capital.Effective strategies should be implemented to combine elevation of psychological capital and health-promoting lifestyle,to promote better physical and mental health and to obtain holistic health care quality.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the anti-inflammatory and analgesia mechanism of electroacupuncture (EA) device of point injection (PI) on rats of inflammatory pain.</p><p><b>METHODS</b>48 Sprague Dawley (SD) rats were randomly assigned into a control group, a model group, an EA+PI group, an EA device of PI (EAPI) group, an EA group and a PI group, eight rats in each one. The rats in the control group were subcutaneously injected with 50 μL of liquid paraffin oil solvent into the dorsum of left hindpaw, while rats in the remaining groups were treated with 50 μL of complete freund's adjuvant (CFA) at identical location to induce the model of inflammatory pain. After model establishment, the rats in the EA+PI group, EAPI group, EA group and PI group were treated with EA+PI,EA device of PI, EA and PI, respectively, once every other day (the 2nd day, 4th day and 6th day). Each treatment was given for 30 min. The mechanical withdrawal threshold, thermal withdrawal threshold and foot swelling before and 1 d to 6 d after model establishment were observed; the western blotting method was applied to measure IL-1β expression in inflammatory tissue of skin.</p><p><b>RESULTS</b>After model establishment, compared with the control group, the mechanical withdrawal threshold and thermal withdrawal threshold were reduced (all<0.05) and the foot swelling was increased in the rest groups (all<0.05). After treatment, the mechanical withdrawal threshold and thermal withdrawal threshold in the EAPI group were significantly increased compared with those in the EA+PI group, EA group and PI group (all<0.05), but the foot swelling was reduced (all<0.05). The IL-1β expression in the model group was higher than that in the control group (<0.05); after treatment, the IL-1β expression in the EAPI group was lower than that in the model group, EA group and PI group (all<0.05), but no significantly different from that in the EA+PI group (>0.05).</p><p><b>CONCLUSIONS</b>The efficacy of EA device of PI on inflammatory pain is superior to EA combined with PI, EA alone and PI alone, which is suitable for further popularization and application.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the efficacy and safety of Pai-Neng-Da Capsule (panaxadiol saponins component, PND), a new Chinese patent medicine, on patients with chronic aplastic anemia (CAA) and to explore the optimal therapeutic regimen for CAA.</p><p><b>METHOD</b>A total of 36 patients with CAA were enrolled and divided into three groups: the AP group (20 cases, andriol 120 mg/day + PND 240 mg/day), the ACP group (13 cases, andriol 120 mg/day + cyclosporine 3-6 mg kd(-1) day(-1) + PND 240 mg/day), and the PND group (3 cases, PND 240 mg/day). All patients were treated and followed up for 6 months. Peripheral blood counts, renal and hepatic function and Chinese medical (CM) symptoms of patients were assessed and all indices were gathered at the beginning and end of the study.</p><p><b>RESULT</b>In the AP group, no significant hematologic difference was observed at the end of 6-month treatment comparing with the beginning. In the ACP group, the blood counts were maintained at the same level after the 6-month treatment. In the PND group, trilineage hematologic improvement was displayed at the end of 6-month treatment comparing with the beginning. No significant difference was showed in renal and hepatic function in all patients. All patients' clinical symptom improved according to CM symptom score. The effective rates were 95%, 73% and 100%, respectively.</p><p><b>CONCLUSION</b>PND improved the efficacy and decreased side effects by cutting down the dosage of andriol, and it could also improve patients' clinical symptom and quality of life. PND were effective and safe in the treatment of CAA, it could be used alone or in combination with pharmacological agents such as andriol and cyclosporine.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the relation between Pi deficiency syndrome (PDS) and the configuration and functions of extensor digitorum longus (EDL)and soleus (SOL).</p><p><b>METHODS</b>Totally 36 ICR mice were randomly divided into 3 groups according to weight matching principle, the control group, the exhausted group, and the rhubarb group, 12 in each group. Two PDS models were established by either purgation with rhubarb diarrhea (as Group A) or exhausted swimming plus sleep deprivation (as Group B).The cross sectional area (CSA) of type I and II fibers of extensor digitorum longus (EDL) and soleus (SOL), relative proportions of type I and II fibers were measured by m-ATPase histochemical method. The isotonic contraction and the maximum tetanus contraction of EDL and SOL were detected by PowerLab system.</p><p><b>RESULTS</b>Compared with the control group, the body weight, body temperature, and the general health condition of PDS model rats obviously decreased; the spleen index and the thymus index were also lower; the maximal isotonic contraction and the maximum tetanus contraction obviously decreased; the cross section areas of EDL and SOL were reduced with loosely arranged cells. In EDL, the proportion of type I fibers was added and the proportion of type II fibers was lowered. In SOL, there was no change in the proportion of type I and type II fibers.</p><p><b>CONCLUSIONS</b>EDL and SOL were obviously atrophied in the two PDS model mice. The type I fibers of SOL was more significantly atrophied in Group B.</p>

ABSTRACT

Objective:To determine residual sulfite in traditional Chinese medicinal materials or pieces processed by sulfur fumi-gation respectively by iodine titration, acid-base titration and ion chromatography and compare the results. Methods:The three meth-ods were used to determine four kinds of Chinese herbal medicines including Codonopsis radix, Dioscoreae rhizoma, Achyranthis bident-atae Radix and Atractulodis Macrocephalae Rhizoma, the recovery tests were also performed and the results were analyzed and compared to summarize the characteristics and quality control requirements of each method. Results:Iodine titration and acid-base titration had the advantages of simple operation process and low cost. However, there were many interference factors in the two methods, and due to different principles, they were suitable for the determination of different varieties of herbal medicines. Ion chromatography method had the advantages of high sensitivity and strong specificity, while the cost was high. Conclusion: It is suggested that proper methods should be chosen for the determination of sulfur dioxide residues according to actual situations.

BACKGROUND: Intraoperative Cell Salvage (ICS), hereby referred to 'mechanical red cell salvage', has been widely used in adult elective major surgeries to reduce requirement for homologous red blood cell transfusion and its associated complications. However, amount of free haemoglobin (fHb) from ICS has been shown related to incidence of renal failure. fHb is the most important indicator of quality control of cell salvaged blood, thus monitoring the fHb concentration is imperative to minimise renal injury. However, currently there has been lacking quick biochemical markers to monitor the levels of fHb during ICS. The aim of this study was to screen quick biochemical markers for evaluating the amount of fHb during use of intraoperative cell salvage. METHODS: Twenty patients undergoing elective cardiovascular surgery were enrolled. Blood was collected and processed using a Fresenius continuous auto-transfusion system device. The concentration of fHb, albumin (Alb), and calcium (Ca) in three washing modes were measured, and their clearance rates were calculated. The correlations among the clearances and concentrations of fHb, albumin, and calcium were analysed. RESULTS: In three washing modes, concentrations of albumin and calcium are significantly associated with amount of fHb:fHb(g/L) = 0.111 Alb(g/L) -0.108, R = 0.638, p = 0.000; fHb(g/L) = 1.721 Ca(mmol/L) +0.091, R = 0.514, p = 0.000. Furthermore, the clearance rates of albumin and calcium significantly predict clearance of fHb, CR(fHb) = 0.310 CR(ALB) + 0.686, R = 0.753, p = 0.000, CR(fHb) = 0.073 CR(Ca) + 0.913, R = 0.497, p = 0.000. CONCLUSIONS: In clinic practice, clearance rates of albumin, or calcium can be used to evaluate the quality of salvaged blood, fHb. Bed-side measurement of calcium could offer a more feasible means for clinicians to undertake a real-time assessment of fHb.

ABSTRACT

This study is to investigate the modulation of Kudiezi (KDZ) injection on differential protein expression in cerebral cortex of rats with cerebral ischemic stroke and heat toxin syndrome established by intraperitoneal injection of carrageenan and middle cerebral artery occlusion (MCAO) methods. According to random number table rats were divided into three groups: drug group, model group and sham group. The tripheye tetrazolium chloride (TTC) staining and HE staining were used to observe brain tissue injury of rats. After therapeutic intervention with above drug for seventy-two hours, the level of differential protein expression was analyzed by two-dimensional gel electrophoresis (2-DE). The results show that there are differential protein expressions between cerebral ischemic stroke and heat toxin syndrome rats and sham rats. Furthermore, as a Chinese medicine injection with effect of clearing heat, resolving toxin and dredging collaterals, KDZ injection can decrease alleviate morphological changes of cerebral ischemia, regulate the levels of some differential proteins expression.

ABSTRACT

Although traditional Chinese medicines (TCMs) play important role in drug discovery and human health, the actual value of TCMs has not been fully recognized worldwide due to its complex components and uncontrollable quality. For the modernization and globalization of TCMs, it is important to establish selective, sensitive and feasible analytical methods for determination and quantification of bioactive components of TCMs in body fluids primarily due to the low concentration, the complex nature of the biological matrices, and multi-components and their metabolites present in biological fluids. The present review summarizes the current extraction techniques, chromatographic separation and spectroscopic (especially mass spectrometric) analysis methods and new trends on the analysis of bioactive components and metabolites of TCMs in biological fluids. In addition, the importance of establishment of pharmacokinetics and bioavailability profiles and simultaneous determination of multi-active components in TCMs is discussed to provide proper examples of analytical methods for pharmacological and clinical studies of TCMs.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the possible angiogenesis mechanism of Huazhuo Jiedu Hewei Recipe (HJHR) in preventing and treating precancerous lesions of gastric cancer (PLGC).</p><p><b>METHODS</b>Totally 66 Wistar rats were randomly divided into 6 groups, i.e., the normal control group, the model group, the retinoic acid (RA) group, the high dose HJHR group, the middle dose HJHR group, the low dose HJHR group, 11 in each group. PLGC model was duplicated by inserting a spring with Helicobacter. Corresponding medicines were administered to rats in each medicated group once daily by gastrogavage, 2 mL each time for 12 successive weeks. The effect of HJHR on hypoxia induced factor (HIF-1alpha) and vascular endothelial growth factor (VEGF) of PLGC in chronic atrophic gastritis (CAG) rats' gastric mucosa was observed by immunohistochemical assay and Western blot method.</p><p><b>RESULTS</b>Compared with the normal control group, the expression of VEGF and HIF-1alpha increased in the model group (P < 0.05). Compared with the model group, the expression of VEGF and HIF-1alpha decreased in each medicated group (P < 0.05). Besides, they were lower in the high and middle dose HJHR groups than in the RA group and the low dose HJHR group (P < 0. 05). There was no statistical difference between the low dose HJHR group and the RA group (P > 0.05).</p><p><b>CONCLUSION</b>HJHR could prevent and treat PLGC of CAG rats possibly through decreasing the expression of HIF-1alpha and VEGF in a dose-dependent manner.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the therapeutic effect of acupuncture-moxibustion and Tuina massage for treatment of sub-health.</p><p><b>METHODS</b>A systematic evaluation on randomized controlled trial, quasi-randomized control trial or controlled clinical trial of treatment of sub-health with acupuncture-moxibustion and Tuina was carried out according to the assessment methods recommended by the Cochrane Collaboration.</p><p><b>RESULTS</b>There are totally 9 articles and 937 subjects met the inclusion criteria and are included into the assessment system. The average quality of methodologies adopted is not so high, and with many limitations. Meta-analysis or descriptive analysis on the data collected: (1) General effect: Tuina massage therapy is better than auricular-point-pressing therapy; acupuncture is better than self health care; acupoint injection is better than intramuscular injection, Tuina is similar to Chinese herb. (2) Quality of life and pain score: Tuina is better than blank control. (3) Individual effect: the effect of Tuina massage on lassitude is better than acupuncture. (4) Effect on sleep quality index: Tuina massage is better than acupuncture.</p><p><b>CONCLUSION</b>There are not enough evidences to approve that the effect of acupuncture-moxibustion and Tuina massage on sub-health is better than that of other therapies. Therefore, more high quality randomized controlled trials with strict and scientific designation are necessary for obtaining more and better evidences.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the clinical effect and safety of Tuina for treatment of somatic pain of sub-health.</p><p><b>METHODS</b>A randomized, double-blind and blank parallel controlled trial was done. The experiment group was treated with Tuina and the control group lied down for rest, 45 minutes each time, twice each week for three weeks.</p><p><b>RESULTS</b>Tuina treatment could improve more on sensory, affective, evaluation, pain rating index and extant pain intensity of the pain index, and score of subjective sensation of life quality and health status together with physiology and psychology field of life quality.</p><p><b>CONCLUSION</b>Massage is an effective therapy for treatment of somatic pain of sub-health without adverse reactions and it should be generalized to application.</p>

<p><b>OBJECTIVE</b>To study the effects of Ligusticum and its two components (Sodium Ferulate and Ligustrazine as main efficacy components in Ligusticum for invigorating blood circulation) on muscle atrophy in a hind limb unloaded rat model.</p><p><b>METHODS</b>The tail-suspended rats were subjected to a 14-days disuse, immunohistochemistry and hemorheology were used to study the effects of medicines on soleus muscle.</p><p><b>RESULTS</b>Compared with HLU+ W: (1) The CSA of soleus type I fibers in HLU + SfH and HLU+ TmpH increased by 37.3% and 39.4% respectively (P < 0.05). (2) Expression level of MHC II were inhibited in all treatment groups (P < 0.01). (3) Expression of MHC II in nuclear bag 2 fiber were altered from positive to negative. (4) The blood viscosity in low shear rates decreased obviously (P < 0.01), even near to control.</p><p><b>CONCLUSION</b>Ligusticum and its two main efficacy components (Sodium Ferulate and Ligustrazine) can prevent soleus atrophy induced by disuse, and Sodium Ferulate and Ligustrazine in high dose showed most efficacy.</p>

ABSTRACT

<p><b>AIM</b>To study the effect of radix-astragali compound(RC) on muscle atrophy in tail-suspended rats. Muscle weight, fiber type distribution, cross-sectional area (CSA), and activity of myosin adenosine triphosphatase (ATPase) in rat soleus muscle were investigated.</p><p><b>METHODS</b>The tail-suspended rats were subjected to a 14 days simulated weightlessness, during which period, RC or saltwater was given via intragastric instillation during tail suspension. The changes of soleus muscle weight were scaled by muscle-to-body weight ratio. The activities of myosin ATPase of muscle fibers were detected by method of Ca(2+) -ATPase.</p><p><b>RESULTS</b>After a 14 days tail suspension it was found: in rats treated with RC, soleus muscle-to-body weight ratio rose by 33.33% (P < 0.01), both CSA of type I and II fiber drastically enhanced by(143.03%, P < 0.01; 83.25%, P < 0.01), the percentage of type I fiber significantly declined compared to the untreated rats.</p><p><b>CONCLUSION</b>RC is able to effectively prevent muscle atrophy caused by tail suspension and restrain the increase in the myosin ATPase activities caused by simulated weightlessness.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the therapeutic effect of acupuncture and moxibustion on takayasu arteritis of brachiocephalic artery type and the mechanism.</p><p><b>METHODS</b>Thirty-one cases were treated by acupuncture and moxibustion at main acupoint Renying (ST 9) and adjuvant acupoints selected according to the symptoms. The therapeutic effect and changes of color Doppler's ultrasonic flow image (CDFI) of relevant arteries in patients before and after treatment were investigated.</p><p><b>RESULTS</b>The clinical total effective rate was 90.3%. Of the 31 cases, 5 cases were clinically cured, 16 cases were markedly effective, 7 cases improved and 3 cases were ineffective; and it was showed that acupuncture and moxibustion could enlarge the inner diameter of the injured blood vessel, increase the volume of blood flow and the elastic index of the injured blood vessel, and improve the abnormal velocity and the abnormal crest form of blood flow.</p><p><b>CONCLUSION</b>Acupuncture and moxibustion has a good therapeutic effect on takayasu arteritis of brachiocephalic artery type; acupuncture and moxibustion can improve the stenotic degree of the injured blood vessel and abnormal hemodynamics of blood flow in the limb and brain.</p>
