ABSTRACT

In order to judge the future development trend of science and technology, plan ahead and lay out the frontier technology fields and directions, China Association of Chinese Medicine(CACM) has launched consultation projects for collecting "major scienti-fic issues and engineering technology difficulties in traditional Chinese medicine(TCM)" for the industry for three consecutive years since 2019. Up to now, 18 projects have been selected as major issues for research, and some experience and achievements have been made. These projects have been applied in important scientific and technological work such as scientific and technological planning and deployment at all levels of national, local, and scientific research institutions, the selection and cultivation of major national scientific and technological projects, and the construction of innovation bases, giving full play to the role of the think tank advisory committee of CACM. This study reviewed the selection of major issues for the first time, systematically combed its application in the national layout of science and technology, and put forward the existing problems and improvement suggestions, aiming to provide new ideas for further improving the selection of major issues and research direction, providing a theoretical basis and decision support for the national scientific and technological layout in the field of TCM, and promoting scientific and technological innovation to facilitate the high quality development of TCM.

ABSTRACT

ObjectiveTo explore the practical value of environment-friendly sample release agent combined with ultrasound in the preparation of pathological tissue sections. MethodsFrom February 2013 to December 2022, 2 518 pathological specimens submitted by Foshan Municipal Hospital of Traditional Chinese Medicine were selected as the study objects. Two samples of the same specimen were randomly divided into two groups: the environment-friendly fast group, in which the pathological tissue sections were made by using the environment-friendly sample release agent combined with ultrasound; and the traditional group, in which formaldehyde, ethanol and xylene were used to make slices in the conventional way. The differences of hematoxylin (HE) staining effect, immunohistochemistry (IHC) staining effect and MDM2 gene detection result of atypical lipomatous tumor/highly differentiated liposarcoma (ALT/WDL) tissue sections between the two groups were compared. Results① The wax of the two groups' pathological tissues was dehydrated well and the tissue hardness was moderate. After HE staining, the sections of the two groups were intact, without cracks and tremor marks, and the contrast between nucleus and cytoplasm was appropriate, with good transparency, uniform staining, and no tissue loss. The excellent rate and score of HE staining in the environmental fast group were higher than those in the traditional group, but the difference was not statistically significant (χ2 = 3.125,P1 = 0.070;t = 0.965,P2 = 0.334). ②After IHC staining of the two groups of sections, the positive location of the cells was accurate, the staining was specific and uniform, the staining intensity was moderate, the staining sensitivity was good, and there was no tissue loss. The excellent rate of IHC staining and the positive rate of IHC staining in the environmental fast group were lower than those in the traditional group, but the difference was not statistically significant (χ12 = 2.769,P1 = 0.092;χ22 = 0.800,P2 = 0.375). ③The background and outline of the two groups of WDL tissue sections were clear, the staining was uniform, the cells were clear and visible, the nuclear boundary was clear, the hybridization signal was clear and bright under the background fluorescence, and there was no miscellaneous signal. The two groups of sections were hybridized successfully, and MDM2 showed positive amplification. The number of cells successfully hybridized in the environment-friendly fast group was lower than that in the traditional group, but the difference was not statistically significant (t = 1.414,P = 0.230). ConclusionsThe tissue treatment method of using environment-friendly sample release agent combined with ultrasound can ensure the detection effect of HE staining, IHC staining and MDM2 gene detection of pathological tissue sections, and is more efficient and environment-friendly, suitable for promotion and use in hospitals at all levels.

ABSTRACT

ObjectiveTo explore the mechanism of Dendrobium huoshanense in the treatment of gastric ulcer (GU) based on network pharmacology and in vivo experiment. MethodThe active components of D. huoshanense were searched from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and literature, and the targets of the components were screened from TCMSP and SwissTargetPrediction. GU-related genes were retrieved from GeneCards, Online Mendelian Inheritance in Man (OMIM), and DisGeNET. Thereby, the common targets of the disease and the medicinal were yielded and the protein-protein interaction (PPI) network was constructed, followed by Gene Ontology (GO) term enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment. According to the predicted results, hematoxylin-eosin (HE) staining, immunohistochemistry, enzyme-linked immunosorbent assay (ELISA), Western blot, and real-time fluorescent quantitative polymerase chain reaction (Real-time PCR) were used to validate the effects of D. huoshanense on acetic acid-induced GU in rats. ResultA total of 63 active components of D. huoshanense and 37 target genes of D. huoshanense for the treatment of GU were screened out. PPI network analysis yielded several possible core anti-GU targets of D. huoshanense. They influenced the development of GU by acting on signaling pathways such as phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt), hypoxia inducible factor-1 (HIF-1), tumor necrosis factor (TNF), and nuclear factor-κB (NF-κB), and various biological processes. The in vivo experiment showed that D. huoshanense significantly reduced the levels of inflammatory factors such as interleukin-1β (IL-1β), interleukin-6 (IL-6), and TNF-α in the serum of model rats (P<0.05, P<0.01), increased gastric blood flow (GBF) at the ulcer margin, raised the expression of epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) at the ulcer margin (P<0.01), significantly down-regulated protein and mRNA expression of PI3K and Akt, and up-regulated protein and mRNA expression of phosphatase and tensin homolog deleted on chromosome ten (PTEN) in the gastric tissues of GU rats (P<0.01). ConclusionThrough regulating EGFR/PI3K/Akt signaling pathway, D. huoshanense can inhibit tissue inflammation, increase gastric microcirculatory blood flow at the ulcer margin, and promote cell proliferation and repair of damaged gastric mucosa.

ABSTRACT

This study mainly introduced the research on Chinese medicine toxicology funded by the National Natural Science Foundation of China(NSFC) in 2012-2021 and analyzed the research content. Furthermore, key research topics and characteristic research projects were discussed, such as the toxicity mechanism, relationship between toxicity and efficacy, toxicity-alleviating mechanisms, and new technology and methods. The review suggested that researchers should gain an in-depth understanding of the "toxicity" of Chinese me-dicine, turned to characteristic research topics, and build a toxicological research paradigm suited to the characteristics of Chinese medicine in project application.

ABSTRACT

Books on Chinese herbal medicines have shown that Dendrobium has the effect of nourishing Yin and reinforcing Yin,usually used for constipation induced by spleen Yin deficiency in clinical application. D. huoshanense,as an independent species among many species of Dendrobium,has no experimental studies about its effects on spleen Yin deficiency-type constipation. The purpose of this experiment was to illustrate the therapeutic effect of D. huoshanense on the constipation of spleen Yin deficiency type in rats,investigate its preliminary mechanism,and compare it with the D. officinale and D. nobile contained in the Chinese Pharmacopoeia to clarify its characteristics. The spleen Yin deficiency model was replicated in 70 rats by the composite factor method,and then the model rats were randomly divided into 7 groups: model group,Liuwei Dihuang Pills group( LWDHP),D. huoshanense high( DHS-H),medium( DHS-M),low( DHS-L) dose groups,D. nobile group( DNS),and D. officinale group( DOS),and another 10 rats were used as normal group( Normal). After 7 continuous days of administration,the fecal water content and intestine propulsion rate of each group were detected. HE staining was used to observe the pathological damage of ileum and colon in each group. Immunohistochemistry and Western blot were used to detect aquaporin 3( AQP3) expressions,while the expression levels of the somatostatin( SS) and motilin( MTL) in the ileum of each group were detected by enzyme-linked immunosorbent assay. The results showed that as compared with the model group,the rats in each drug-administered group had increased number of fecal pellets,increased fecal water content,and the increased intestinal propulsion rate( P<0. 01),while the pathological damage of the ileum and colon was significantly reduced; the expression of AQP3 protein was significantly decreased( P<0. 01); the level of MTL was significantly increased and the level of SS was decreased( P<0. 01). All DHS groups showed a good dose-effect relationship,and the same dose treatment effect was equivalent to that of DOS,but it was superior to DNS. Therefore,DHS has a significant therapeutic effect on constipation of spleen Yin deficiency type,and its mechanism may be related to intestinal motility and water-liquid metabolism,with a good therapeutic effect.

ABSTRACT

OBJECTIVE: To observe early intervention effects of Modified Shuyu Pill (MSP) on vascular cognitive impairment no dementia (VCIND). METHODS: Totally 100 patients VCIND were randomly assigned to the treatment group (43 cases) and the control group (33 cases). On the basis of the treatment targeting risk factors of blood vessels, patients in the treatment group were treated by MSP, while those in the control group were treated by donepezil hydrochloride. The therapeutic course was 16 weeks. The neuropsychological scales [mini-mental state examination (MMSE) and Montreal cognitive assessment (MOCA) score] and Chinese medicine dementia syndromes scales were observed before and after treatment. RESULTS: The MMSE and MOCA score of the two groups increased when compared with the same group before treatment (P < 0.01). But there was no statistical difference in MMSE or MOCA score after treatment between the two groups (P > 0.05). The Chinese medicine dementia syndromes scales significantly decreased in the treatment group when compared with before treatment (P < 0.01). But there was no statistical difference in Chinese medicine dementia syndromes scales in the control group between before and after treatment (P > 0.05). There was statistical difference in Chinese medicine dementia syndromes scales after treatment between the two groups (P < 0.01). CONCLUSION: MSP could effectively intervene the progress of VCIND.

ABSTRACT

<p><b>OBJECTIVE</b>To study the protective effect of medicated serum prepared with Taohong Siwu Tang on hydrogen peroxide-injured human umbilical vein endothelial cells (HUVECs).</p><p><b>METHOD</b>Sprague Dawley rats were orally administered with 10 mL x kg(-1) extracts from Taohong Siwu Tang (1.75 g crude drug), twice a day for three days, in order to prepare medicated serum of Taohong Siwu Tang. The effect of medicated serum pre-treated with Taohong Siwu Tang (with concentrations of 5%, 10%, 15%) on reduction of H2O2-induced cell activity was detected MTT. Cell morphological changes were observed under microscope. The effect of medicated serum prepared with Taohong Siwu Tang on the apoptosis and antioxidant capacity of HUVECs was detected with the content of malondialdehyde (MDA) and dehydrogenase (LDH), the activity of superoxide dismutase (SOD) and AO/EB staining. The expression of Caspase-3 was determined by western blot.</p><p><b>RESULT</b>Compared with the blank serum control group, cell were significantly less active after being damaged by H2O2 (400 micromol x L(-1)). Medicated serum could significantly improve the SOD activity, reduce the levels of LDH and MDA, and inhibite the expression of Caspase-3. AO/EB staining and inverted microscope also showed that medicated serum prepared with Taohong Siwu Tang could reduce cell apoptosis induced by H2O2.</p><p><b>CONCLUSION</b>Medicated serum prepared with Taohong Siwu Tang has significant protective effect on HUVECs injured by H2O2.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the anti-platelet activation effect and partial mechanisms of Taohong Siwu decoction (TSD).</p><p><b>METHOD</b>The effect to venous thrombosis model and pulmonary thromboembolism model induced by vein injecting ADP and Adr was observed. The platelet adhesion rate was analyzed by using spinning glass bottle, and the platelet aggregation rate induced by ADP, Adr was analyzed by using turbidimetry. The acute blood stasis rat model was established to analyze the content of plasm TXB2 and PGI2 by RIA, and the content of VWF, GMP-14 by ELISA.</p><p><b>RESULT</b>TSD could effectively reduce platelet the adhesion rate of normal rat, inhibit the platelet aggregation of normal rat induced by ADP, Adr. It significantly reduced the plasma TXB2 VWF, and GMP-140 level of blood stasis rats. It also had significant tendency to increase 6-keto-PGF1alpha level.</p><p><b>CONCLUSION</b>TSD possessed obvious activity of inhibiting platelet activation. The mechanism related with the restraining of platelet adhesion, platelet aggregation and platelet releasion.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To seek for new active components from Caesalpinia sappan.</p><p><b>METHOD</b>The chemical constituents were isolated and identified by means of chromatographic and spectroscopic technologies methods.</p><p><b>RESULT</b>Nine compounds were isolated, and their structures were identified as 3, 8-dihydroxy4, 10-dimethoxy-7-oxo-[2] benzopyrano[4,3-b] benzopyran (1), 3-deoxysappanchalcone (2), sappanchalcone (3), 3-deoxysappanone B (4), rhamnetin (5), protosappanin C (6), 3, 7-dihydroxy-chroman-4-one (7), dimethyl adipate (8), daucosterin (9).</p><p><b>CONCLUSION</b>Compound 1 was a new compound, and compounds 7, 8 were isolated from this plant for the first time.</p>

ABSTRACT

Objective:To observe the therapeutic effects of treating traumatic gonarthromeningitis with acupuncture combined Chinese herbs.Methods:Seventy Patients were randomly divided into two groups,with 35 cases in each group.The treatment group was treated with Chinese herbal medicine,acupuncture and cupping therapy.The control group was treated with Diclofenac Sodium Tablets,Corydalis Analgesic Tablets and Vitamin C Tablets.Results:The total effective rate of the treatment group was higher than that of the control group.There was a statistically significant difference between the two groups(P<0.05).Conclusion:The combined therapy with acupuncture and Chinese herbal medicine has a remarkable therapeutic efrect on treating traumatic gonarthromeningitis.
