ABSTRACT

OBJECTIVE: To observe the effect of "Xingnao Kaiqiao" needling on the recovery of general anesthesia in mid-aged and elderly patients undergoing painless gastrointestinal endoscopy. METHODS: A total of 140 patients were randomly divided into control and treatment groups, with 70 cases in each group. After termination of the endoscopy, patients of the treatment group received acupuncture stimulation of Neiguan (PC 6), Shuigou (GV 26) and Sanyinjiao (SP 6) with sterilized disposable filiform needles which were manipulated with twirling-reducing method for 3 min in PC 6, with lifting-thrusting reinforcing method in SP 6, and with bird-peck needling in GV 26. The patients of the control group were not given any treatment. Before, the immediate time of termination of the endoscope examination, resuscitation and 10 min after the endoscope examination, the mini-mental state examination (MMSE) scores were obtained to assess the patients' recognition function. The Bi-spectral Index (BIS) values were recorded to assess the patients' depth of sedation and anesthesia, and the Observer's Assessment of Alertness/Sedation Scale (OAA/S) was used to evaluate the resuscitation/sedation level. RESULTS: Compared with the control group, the MMSE score of the treatment group was significantly higher at the 10th min after endoscope examination (P<0.05),suggesting an improvement of the cognitive function after acupuncture intervention. The duration from general anesthetic state (BIS values=40-60 points) to mild sedation (BIS values=80-90 points) was significantly shorter in the treatment group than in the control group (P<0.05). No significant differences were found between the two groups in the OAA/S score at the 10th min after endoscope (P>0.05). CONCLUSIONS: "Xingnao Kaiqiao" needling stimulation has a significant effect on the recovery of middle-aged and elderly patients undergoing painless gastroscopy during general anesthesia.

ABSTRACT

OBJECTIVE: To research the effects of soothing liver and invigorating spleen recipes on expression of TLR4 mRNA and protein expression in hepatic tissue of rats with non-alcoholic steatohepatitis and its mechanism. METHODS: 72 male SD rats were randomly divided into 8 groups: normal group, model group, high-dose soothing liver group (receiving gavage of Chaihu Shugan Powder 9. 6 g/kg), low-dose soothing liver group (receiving gavage of Chaihu Shugan Powder 3.2 g/kg), high-dose invigorating spleen group (receiving gavage of Shen Ling Baizhu Powder 30 g/kg), low-dose invigorating spleen group (receiving gavage of Shen Ling Baizhu Powder 10 g/kg), high-dose integrated Group (receiving gavage of Chaihu Shugan Powder and Shen Ling Baizhu Powder combination recipes 39.6 g/kg), low-dose integrated Group (receiving gavage of Chaihu Shugan Powder and Shen Ling Baizhu Powder combination recipes 13.2 g/kg), 9 rats were in each group. Used high fat diet (10 mL/kg) to establish experiment model of NASH rat. At the end of the sixteenth weeks, the levels of serum lipids, liver lipids and serum aminotransferase were measured by automatic biochemical analyzer; Liver pathology was analyzed by HE and Oil red O staining; TLR4 mRNA was assayed by real-time fluorescent quantitative polymerase chain reaction (RT Q-PCR); TLR4 protein was detected by Western blot. RESULTS: Compared with normal group, the levels of TC, LDL-C in the serum, TC,TG as well as the expression of TLR4 mRNA and protein in the hepatic tissue were dramatically increased in model group (P < 0.01). Compared with the model group, the levels of serum lipids, liver lipids, the expression of TLR4 mRNA and protein in the hepatic tissue were decreased in each treatment group (P < 0.05 or P < 0.01). CONCLUSION: Soothing liver and invigorating spleen recipes can inhibit hepatic TLR4 expression, that may be one of their therapeutic mechanisms. There is much difference between high-does and low-does treatment groups in various testing items, which shows that there is does-effect relationship in intervention NASH.

ABSTRACT

OBJECTIVE: To study on the effect and mechanism of curcumin on inhibiting injury induced by free radical in pulmonary fibrosis. METHOD: One hundred and forty-four male SD rats were randomly divided into 6 groups (24 rats in each group). Rats in the model control group, positive medicine group, and high, moderate and low curcumin groups were injected with a single dose of bleomycin by trachea, and rats in sham-model control group with same volume normal saline. One day after the injection, curcumin solution of different dosages (200,100,50 mg x kg(-1) x d(-1)) was respectively given to rats in the high, moderate and low curcumin group by daily gastrogavage, while equal volume of normal saline was given to those in the sham-model control group and model control group, and an equal volume of prednisone (0.56 mg x kg(-1) x d(-1)) was saline was given to those in positive medicine control group. On the 7, 14, 28 days, the contents of GSH-Px, SOD, MDA and iNOS in pulmonary tissues of different groups were measured. RESULT: Curcumin can raise the content of SOD and GSH-Px and lessen the level of MDA and iNOS. CONCLUSION: Curcumin can regulate the level of free radical in the body of rats with pulmonary fibrosis and lessen the oxidative injury of pulmonary tissues caused by free radical, in the body of rats with pulmonary fibrosis. The mechanisms of curcumin on idiopathic pulmonary fibrosis lie in adjusting the level of free radical and inhibiting the injury of lung tissue induced by free radical.
