ABSTRACT

The Brazilian Cerrado biome consists of a great variety of endemic species with several bioactive compounds, and Anadenanthera peregrina (L.) Speg is a promising species. In this study, we aimed to perform phytochemical characterization and evaluate the antioxidant and antibacterial activities against Staphylococcus aureus and Escherichia coli of the hydroethanolic extract of A. peregrina stem bark. The barks were collected in the Botanical Garden of Goiânia, Brazil. The hydroethanolic extract was obtained by percolation and subjected to physicochemical screening, total phenolic content estimation, high-performance liquid chromatography (HPLC) fingerprinting, and antioxidant (IC50 values were calculated for the 2,2-diphenyl-1-picrylhydrazyl assay - DPPH) and antibacterial activity determination. The pH of the extract was 5.21 and density was 0.956 g/cm3. The phytochemical screening indicated the presence of cardiac glycosides, organic acids, reducing sugars, hemolytic saponins, phenols, coumarins, condensed tannins, flavonoids, catechins, depsides, and depsidones derived from benzoquinones. The extract showed intense hemolytic activity. The total phenolic content was 6.40 g GAE 100 g-¹. The HPLC fingerprinting analysis revealed the presence of gallic acid, catechin, and epicatechin. We confirmed the antioxidant activity of the extract. Furthermore, the extract did not inhibit the growth of E. coli colonies at any volume tested, but there were halos around S. aureus colonies at all three volumes tested. These results contribute to a better understanding of the chemical composition of A. peregrina stem bark and further support the medicinal applications of this species.

Objetivou-se avaliar o consumo de forragem e o desempenho de ovinos mantidos em pastagem de capim-aruana, submetidos a porcentagens crescentes de proteína bruta (PB) no suplemento, na época seca. Vinte borregos da raça Santa Inês foram utilizados em delineamento inteiramente ao acaso, com cinco tratamentos e quatro repetições. Os suplementos foram fornecidos em 1,0% do peso corporal, nas porcentagens de 0, 15, 20, 25 e 30%. O aumento de proteína bruta influenciou o consumo total de matéria seca (kg/dia) e a porcentagem do peso vivo, com valores máximos estimados de 1.296g (3,2% de MS) com 21,48 e 21,89% de PB no suplemento, respectivamente. O consumo de forragem máximo, estimado de 893g/dia, ocorreu com a PB de 21,5%. O aumento de PB nos suplementos resultou em efeito quadrático sobre o ganho médio diário, com valor máximo de 104g/dia com a PB de 23% no suplemento. Recomenda-se o uso de suplementos múltiplos com 21 a 23% de PB fornecidos na proporção de 1% do peso corporal (PC) para ovinos mantidos em pastos de capim-aruana na época seca...


The aim of this study was to evaluate the forage intake and grazing sheep performance keep on Aruana grass subjected to increasing crude protein (CP) levels in the supplement on dry season. Twenty Santa Ines male lambs were used, with initial body weight of 31.80kg by a completely randomized design with five treatments and four replications. The supplements were provided daily at 1% of body weight, with protein levels of 0, 15, 20, 25 and 30%. The increase of the crude protein levels promoted a squarely effect on dry matter intake (kg/day and % of BW), with maximum estimated values of 1296g and 3.2% of DM in CP levels of 21.48 and 21.89, respectively. The maximum forage intake estimated of 893g/day occurred in CP level de 21.51%. The increased of crude protein level in supplements increased squarely the average daily gain, with a maximum of 104g/day, for the 23% crude protein in the supplement. Thus, the use of the multiple supplements supplied in 1% of body weight with CP levels ranged 21 a 23% is indicated for sheep grazing Aruana grass on dry season...

ABSTRACT

This study evaluated the effect of feed restriction and goat genotype on meat quality. Three genotypes (Brazilian native breed Canindé; Brazilian native breed Moxotó; and F1 Boer crossbred animals obtained by crossing Boer bucks with local breed does) and three different feeding regimens (ad libitum fed, AL; restricted fed at 75% of the ad libitum, R.75; or restricted fed at 50% of the average ad libitum intake, R.50) were used. There was no difference (P>0.05) in chemical composition, total and soluble collagen, and shear force of the Longissimus lumborum muscle among genotypes. However, AL had greater amounts of soluble collagen and crude protein in the muscle (P<0.05) than R.75. No difference (P>0.05) was observed for the myofibrillar fragmentation index. The goat genotype presented few differences in their fatty acid profiles. However, goats fed ad libitum had a more favorable fatty acid profile for human health with greater concentrations of oleic acid, unsaturated fatty acids, and conjugated linoleic acid.

ABSTRACT

ETHNOPHARMACOLOGICAL RELEVANCE: Cenostigma macrophyllum Tul. var. acuminata Teles Freire (Leguminosae-Caesalpinioideae), popularly known in Brazil as "caneleiro", is widely used in folk medicine against gastrointestinal diseases. In previous studies, the ethanol extract of leaves from Cenostigma macrophyllum Tul. var. acuminata Teles Freire had shown antinociceptive, anti-inflammatory, antibacterial, antioxidant and antiulcerogenic activities. AIM OF THE STUDY: The aim of this study was to assess the gastroprotective effect of the hydroalcoholic fraction of leaves of Cenostigma macrophyllum Tul. var. acuminata Teles Freire (Cm-FHA), as well as to elucidate the possible underlying mechanisms of action. MATERIALS AND METHODS: Mice were used for the evaluation of the acute toxicity, and mice and rats to study the gastroprotective activity. The potential gastroprotective of Cm-FHA was assessed on different gastric ulcer models in rodents, such as absolute ethanol, HCl/ethanol, ischemia-reperfusion, cold restraint stress and indomethacin. The participation of prostaglandins, NO-synthase pathway and ATP-sensitive potassium channels (KATP) in gastroprotective activity of Cm-FHA were evaluated after treatment with a cyclooxygenase inhibitor (indomethacin), a NO-synthase inhibitor (L-NAME) and a KATP channel blocker (glibenclamide 5mg/kg), respectively. Likewise, the catalase activity was determinated in order to assess the possible participation of antioxidant mechanisms. RESULTS: No signs of acute toxicity was observed after oral acute administration of Cm-FHA, considering the analyzed parameters. Likewise, Cm-FHA promoted a protective effect against gastric ulcers induced by absolute ethanol (lesion inhibition by 40% at both 100 and 200mg/kg), HCl/ethanol (lesion inhibition by 50 or 48% at 100 or 200mg/kg, respectively), ischemia-reperfusion (lesion inhibition by 49 or 90% at 100 or 200mg/kg, respectively) and cold restraint stress (lesion inhibition by 63 or 76% at 100 or 200mg/kg, respectively), as well as a increase of catalase activity was observed. Otherwise, Cm-FHA was not able to protect gastric mucosa against indomethacin-induced lesions. Nitric oxide release, the of KATP channels opening and antioxidant activity are the possibly involved in the Cm-FHA-induced gastroprotective activity. CONCLUSION: This study corroborates the folk medicine use of Cenostigma macrophyllum for treatment of gastric ulcers, as well as reinforces this species as a valuable source of promising natural drugs with gastroprotective activity.

ETHNOPHARMACOLOGICAL RELEVANCE: Remirea maritima Aubl., popularly known as "capim-da-praia", is popularly employed in the treatment of diarrhea, kidney disease, fever, and for analgesic and anti-inflammatory purposes through the preparation of teas. Few studies have focused on the chemical composition and its biological properties. AIM OF THE STUDY: This work evaluated the antinocipetive, anti-inflammatory and antioxidant activities of the aqueous extract from Remirea maritima Aubl. as well as the isolation and identification of the chemical compounds. MATERIALS AND METHODS: Compounds were isolated from aqueous extract of Remirea maritima through preparative HPLC and the structures were identified by means of NMR and MS analysis. The tests for antinociceptive, anti-inflammatory, and antioxidant activities, along with motor coordination test (Rota rod), were performed over the aqueous extract. RESULTS: The phytochemical investigation of aqueous extract of Remirea maritima resulted in the isolation of three flavone glycosides. The structures of these compounds were determined by means of MS and 1D and 2D NMR data as vitexin-2″-O-ß-D-glucopyranoside, isovitexin-2″-O-ß-D-glucopyranoside, and luteolin-7-O-glucuronide. Acute pretreatment with aqueous extract (100, 200 or 400mg/kg, i.p.) caused a significant decrease (p<0.001) in the number of abdominal writhes. In the formalin test, higher doses significantly inhibited the late (inflammatory pain) phase of formalin-induced licking (p<0.05 or 0.001). In the hot plate test, there was no significant difference in nociceptive behavior, discarding the possible central effect of the aqueous extract. In the rota rod test, it was verified that the aqueous extract in all concentration evaluated does not alter the motor coordination of mice, such antinociceptive results were unlikely to be caused by motor abnormality. In the peritonitis test, induced by carrageenan, the treatment with aqueous extract produced a significant reduction in leukocyte migration in all concentration evaluated. Additionally, a significant reduction of lipoperoxidation (TBARS test) and in nitric oxide formation (.NO Scavenging assay) was observed in antioxidant activity assay. CONCLUSION: The biological and phytochemical investigations of the aqueous extract of Remirea maritima resulted in the identification of three flavone glycosides that have been described here for the first time in Remirea and effective analgesic activity in various pain models, probably mediated via the inhibition of peripheral mediators which could be related to its strong antioxidant effect observed in vitro.

ABSTRACT

A regulação do metabolismo lipídico com drogas ou alimentos naturais é um alvo importante para diminuir o risco de doenças cardiovasculares. O objetivo deste estudo foi verificar o efeito da lecitina de soja na dislipidemia e na hipertrofia ventricular de camundongos hipercolesterolêmicos. Utilizaram-se quatro grupos de camundongos LDLr-/- com 3 meses de idade que receberam a seguintes dietas: Grupo S- ração padrão; Grupo S+Lec- ração padrão e lecitina de soja; Grupo HL- ração hiperlipídica; e Grupo HL+Lec- ração hiperlipídica e lecitina de soja. Após 15 dias, o sangue foi coletado para análise sérica dos lipídeos e da proteína C reativa. O ventrículo esquerdo foi separado, a proporção peso ventricular (mg) pelo peso do animal (g) foi calculada e, em seguida, processado histologicamente. Cortes histológicos foram corados com hematoxilina/eosina e picrosírius red para avaliar alterações morfológicas e morfométricas ventriculares. A lecitina de soja apresentou efeito antidislipidêmico e aumentou os níveis séricos de HDL nos camundongos do grupo S+Lec. Entretanto, nos camundongos do grupo HL+lec, a lecitina de soja não preveniu a dislipidemia, apenas aumentou o nível sérico do HDL. Este efeito nestes animais influenciou no processo inflamatório cardiovascular, reduzindo o nível sérico de proteína C reativa; e prevenindo a hipertrofia ventricular esquerda. A utilização da lecitina de soja representa um tratamento e/ou uma prevenção alternativa de baixo custo para as dislipidemias não associadas com dieta hiperlipídica. Contudo, a lecitina de soja aumenta os níveis séricos do HDL prevenindo o desenvolvimento da HVE mesmo em dislipidemias associadas com dieta hiperlipídica.

The aim of this study was to investigate the effects of soy lecithin on left ventricular hypertrophy (LVH) and dyslipidemia in hypercholesterolemic mice. We used four experimental groups of LDLr-/- mice (aged 3 months), which received the following diets: Group S: standard diet, Group S+Lec: standard diet and soy lecithin; Group HL: hyperlipidic diet and Group HL+Lec: hyperlipidic diet and soy lecithin. After 15 days on these diets, blood was collected for analysis of serum lipids and C-reactive protein. The left ventricle was dissected out and weighed and the ratio of its weight to the body weight of the animal was calculated, after which it was processed histologically. Sections were stained with hematoxylin-eosin and picrosirius red, to assess morphological and morphometric changes in the ventricle. In Group S+Lec, the soy lecithin had an antidyslipidemic effect and enhanced the serum levels of HDL. However, in the mice in group HL+Lec, soy lecithin did not prevent dyslipidemia, only increasing the serum level of HDL. These effects in these animals influenced the cardiovascular inflammatory process, reducing the level of serum C-reactive protein and preventing LVH. Soy lecithin could thus be used as a treatment or a low-cost alternative preventative measure against dyslipidemia associated with a non-fat diet. However, soy lecithin increases the serum level of HDL, reducing the risk of LVH even in dyslipidemia associated with a high-fat diet.

Com o objetivo de avaliar a produtividade e qualidade do funcho (Foeniculum vulgare Mill.) produzido nas condições edafoclimáticas de Sergipe, foi conduzido cultivo orgânico de funcho em Frei Paulo-SE. Em junho de 2008 e de 2009, início do período de chuvas, foram instalados plantios por meio de mudas. Adicionalmente, em dezembro de 2010 foi realizada colheita de frutos a partir de plantas obtidas após poda drástica e rebrota. Foi realizada colheita de frutos e determinação da produtividade de dezembro a janeiro de 2008, 2009 e em dezembro de 2010. Verificou-se produtividade de 720 Kg.ha-1, no primeiro plantio e de 349 Kg.ha-1 no plantio de 2009. As plantas obtidas por rebrota em 2010 resultaram em produtividade de 589 Kg.ha-1. Sugere-se que a maior produtividade verificada no primeiro ano seja decorrente da maior precipitação média e conseqüente maior disponibilidade de água. A produtividade verificada nas plantas que rebrotaram em 2010 foi também superior a verificada em 2009, o que pode ser explicado tanto em função da maior precipitação, quanto em função do provável maior desenvolvimento das raízes e maior número de ramos após poda, devido à quebra da dominância apical. Esses dados sugerem importância da disponibilidade hídrica na determinação da produtividade na região e explicam a variação na produtividade verificada. A massa seca dos frutos (5, 29 g para 1000 frutos) e o alto teor de óleo essencial verificado (3,2 por cento), assim como a composição do óleo, revelam a alta qualidade dos frutos produzidos na região.


Aiming to evaluate the productivity and quality of fennel fruits produced in the countryside of Sergipe, with no irrigation, they were evaluated organic crops in Frei Paulo-SE. They were installed crops in 2008 and 2009 from seeds, on beginning rainy time. Seedlings obtained from seeds were transplanted in June 2008 and 2009. Besides, after the last harvest of the plants, grown in 2009, it was done a drastic pruning on plants, on May, 2010. The plants were held on the area until their fruits harvest, on december and January of 2008, 2009 and December 2010. The fennel fruits yield was 720 Kg ha -1 in the first crop (2008) and 349 Kg ha -1 at following crop (2009), while the fruits yield obtained from pruned plants was 589 Kg.ha-1. It is suggested that the higher yield at the first year has been due to the higher average rainfall and its greater availability of water. The observed yield in sprouted plants was higher (at 2010) than fennel yield of 2009. It can be explained as a function of precipitation, likely as a function of greater root development and increased number of branches and umbels after pruning and breaking of apical. These results reveal water availability importance to fennel yield. The dried mass of fruits (5.29 g to 1000 fruits), high content of essential oil in fruits (3.2 percent) and the essential oil composition reveal good quality of fennel fruits of Sergipe.

ABSTRACT

We evaluated the effects of a substrate in the biosynthesis of nitric oxide (NO)-l-arginine (LARG)-on hepatic lesions caused by ischemia/reperfusion (I/R) injury in rabbit livers. Rabbits were pretreated with LARG (150 mg/kg IV) or saline solution 0.9% (SS) before the hepatic I/R procedure. The effects of LARG on hepatic injury were evaluated before and after I/R. The warm hepatic I/R procedure produced profound acute liver injury, as indicated by elevated values of serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactic dehydrogenase (LDH), as well as a high apoptotic cell count. All changes were attenuated by treatment with LARG before the hepatic I/R procedure. These results suggested that LARG produced protective effects on hepatic I/R lesions. This protective effect of LARG was probably associated with blocking generation of superoxide anions during the hepatic I/R procedure.

ABSTRACT

Pseudomonas aeruginosa LBI isolated from petroleum-contaminated soil produced rhamnolipids (RL(LBI)) when cultivated on soapstock as the sole carbon source. HPLC-MS analysis of the purified culture supernatant identified 6 RL homologues (%): R(2) C(10) C(10) 28.9; R(2) C(10) C(12:1) 23.0; R(1) C(10) C(10) 23.4; R(2) C(10) C(12) 11.3; R(2) C(10) C(12) 7.9; R(2) C(10) C(12) 5.5. To assess the potential antimicrobial activity of the new rhamnolipid product, RL(LBI), its physicochemical properties were studied. RL(LBI) had a surface tension of 24 mN m(-1) and an interfacial tension of 1.31 mN m(-1); the cmc was 120 mg l(-1). RL(LBI) produced stable emulsions with hydrocarbons and vegetable oils. This product showed good antimicrobial behaviour against bacteria: MIC for Bacillus subtilis, Staphylococcus aureus and Proteus vulgaris was 8 mg l(-1), for Streptococcus faecalis 4 mg l(-1), and for Pseudomonas aeruginosa 32 mg l(-1). RL(LBI) was active against phytopathogenic fungal species, MIC values of 32 mg l(-1) being found against Penicillium, Alternaria, Gliocadium virens and Chaetonium globosum. Due to its physicochemical properties and antimicrobial behaviour, RL(LBI) could be used in bioremediation treatment and in the food, cosmetic and pharmaceutical industries.

ABSTRACT

The relative effectiveness of daily supplementation of iron deficiency during pregnancy using 15 mg/day of iron from iron-bis-glycinate chelate (71 pregnant women), or 40 mg iron from ferrous sulfate (74 pregnant women) was evaluated by measuring hemoglobin, transferrin saturation and serum ferritin, at the beginning of the study (< 20 weeks of pregnancy) and at 20-30 weeks and 30-40 weeks thereafter. Ingestion for 13 weeks or more was considered adequate. Seventy three percent of the Ferrochel consuming group and 35% of the ferrous sulfate consuming group were considered to have taken the treatment adequately. The decrease in levels of all the measured parameters was significantly less pronounced in the group that consumed Ferrochel in spite of the lower treatment dose. Iron depletion was found in 30.8% of the women treated with Ferrochel and in 54.5% of the women than consumed ferrous sulfate. Of the factors responsible for non compliance taste was reported in 29.8% of the ferrous sulfate consumers and none in the groups that consumed Ferrochel. It is concluded that daily supplementation with Ferrochel was significantly more effective, in spite of the lower dose, than supplementation with ferrous sulfate.

ABSTRACT

The authors present the angiographic aspects of four patients having an arteriovenous transhepatic transtumoral fistula (two arterioportal and two arteriovenous). The angiographic results of the occlusion with cyanoacrylate (Hystoacryl) of the right hepatic artery in one and by the infusion of Lipiodol in two are also shown. The physiopathological characteristics of each kind of fistula are discussed and the consequences of the passage of the quimioembolizing mixture through the tumor, regarding intrahepatic abscess formation, irregular clinical results and pulmonary complications are commented. These complications make the diagnosis of a transtumoral transhepatic arteriovenous fistula, previously to the chemoembolic treatment important, even though being frequently difficult, because of the reduced caliber of the feeding artery and eventual low transfistular bloodflow. The efficient occlusion of the arterioportal fistula with Histoacryl was favourably compared to the infusion of Lipiodol, which was unable to occlude the arteriovenous fistula. The contraindication to perform chemoembolic treatment of hepatic tumors, when an intrahepatic transtumoral arteriovenous fistula is present and the embolization of the fistular feeding artery is stressed. Ultrasonography using color Doppler and sono-enhancing contrast is appointed as the ideal non-invasive means of making the diagnosis of an hepatic transtumoral fistula and makes it eligible to be the first test in the algorithm for the appraisal of hepatic tumors.

ABSTRACT

The effect of feeding rice and bean diets in both hepatic and plasmatic activity of gamma-glutamyltranspeptidase-(GGT-EC 2.3.2.2) activity was evaluated in growing-rats (Experiment A) and mature rats (Experiment B). During 28 days, the animals were fed with isocaloric-diets composed by tree levels of rice, bean or rice-and-bean protein. Similarly with the aproteic group, a significant increase on both the hepatic and plasmatic GGT activity were showed with the lowest levels of protein, when compared with 25% casein control group. This rise was more effective in growing-rats fed on legume-based diets (as bean or rice-and-bean diets), making evident a differential effect of age and an exacerbated effect of the protein restriction with the lowest sulfur amino acids disposal. These alterations suggest a metabolic adaptation of GGT to both the inadequate protein and sulfur-amino acid levels, thus supporting the hypothesis that the Glutathione levels may be reduced by these legume-based diets.

ABSTRACT

The frequency of anemia, iron deficiency and iron body stores was assessed in 155 pregnant teenagers of low socioeconomic status in a prenatal care unit of a beneficent hospital in São Paulo, Brazil. By the criterion of the World Health Organization (Hb < 12 g/dL) 14.2% of the pregnant adolescents had anemia. The iron deficiency diagnostic by saturation of transferrin < 16% and zinc protoporphyrin concentration > 60 (mol/mol heme were 45.8 and 42.6%, respectively. The iron body store (serum ferritin < 12 micrograms/L) was depleted for 48.4% of adolescents. It is concluded that the iron nutritional status of these adolescents were characteristics of the pregravidic inadequate iron store. Despite low percentage of the anemia, the high frequency of iron deficiency and depleted iron stores suggest a practical procedure to detect iron deficiency and the use of iron supplementation in teenagers.

ABSTRACT

Gamma-glutamyltranspeptidase (GGT-EC 2.3.2.2) activity and glutathione (GSH) content were measured in livers of female weanling Wistar rats (N = 5-18), submitted to rice-and-bean diets (13 and 6% w/w protein), both supplemented or not with DL-methionine (0.5 and 0.23 g/100 g dry diet, respectively). After 28 days, the rats on the rice-and-bean diets showed significantly higher levels (four times higher) of liver GGT activity and a concomitant 50% lower concentration of liver GSH in comparison with control groups feeding on casein. The addition of DL-methionine to rice-and-bean diets significantly increased the liver GSH content, which reached levels 50% higher than those found in animals on casein diets. The increase in GSH was accompanied by a decrease in liver GGT activity, which did not reach levels as low as those observed in the control groups. No significant correlation could be established between GGT and GSH changes under the present experimental conditions. Linear correlation analysis only revealed that in animals submitted to unsupplemented rice-and-bean diets GSH concentration was positively associated (P < 0.05) with weight gain, food intake and food efficiency. GGT, however, was negatively correlated (P < 0.05) with food intake only, and exclusively for supplemented rice-and-bean diets. The high levels of GGT activity observed in the present study for rats receiving a rice-and-bean mixture could be a result of the poor quality of these diets associated with their deficiency in sulfur amino acids. The results also suggest that diet supplementation with methionine could be important in the reduction of the deleterious effects of GSH depletion by restoring the intracellular concentration of this tripeptide.

ABSTRACT

A total of 1752 strains of osmophilic yeasts were isolated from honey and pollens. Forty-three strains of osmophilic yeasts produced polyols, among which 6 strains produced erythritol in good yields. On the other hand, 52 osmophilic yeasts converted sucrose to fructooligosaccharides, among which 8 strains produced both extra and intracellular beta-fructofuranosidase, which converted sucrose to fructooligosaccharides. This investigation concluded that osmophilic yeasts converted sucrose not only to polyols, but also to fructooligosaccharides in good yields.

ABSTRACT

To promote therapeutic educational activities for nursing personnel in order to decrease stress, to improve interpersonal relations and the search for self-knowledge are the objectives of the courses promoted by the Departament of Nursing of FCM and by The Continued Education Nursing Service of the University Hospital of UNICAMP. Respiration, relaxation, body sensibilization and awareness, and theater interpretation techniques were taught in the following courses: The Hospital and Human Relations, Dance and Creativity, Yoga and Mental Relaxation. The estrategy employed was "group experience", with the participation of nurse's aides, nurses technicians, practical nurses, and registered nurses during working hours in 15 to 20 meetings per course. "Individual statements" written by the participants were used as a research tool, and the method employed was content analysis. The evaluation demonstrated that the "experience" facilitated relations among the members of the work team, opened the space for effective communication, favored self-knowledge, and helped with the problem solving. The analysis demonstrated the importance of the continuity of alternative courses to help employes improve their relations with themselves, with others and with their work, and to properly value their health/learning.
