ABSTRACT

Andrographis paniculata is an important medicinal plant in the Lingnan region of China, which has the functions of clearing heat, removing toxins, and resisting bacteria and inflammation. The TCP gene family is a class of transcription factors that regulate plant growth, development, and stress response. In order to analysis the role of the TCP gene family under abiotic stress in A. paniculata, this study identified the TCP gene family of A. paniculata at the genome-wide level and analyzed its expression pattern in response to abiotic stress. The results showed that the A. paniculata TCP gene family had 23 members, with length of amino acid ranging from 136 to 508, the relative molecular mass between 14 854.71 and 55 944.90 kDa, and the isoelectric point between 5.67 and 10.39. All members were located in the nucleus and unevenly distributed on 13 chromosomes. Phylogenetic analysis classified them into three subfamilies: PCF, CIN and CYC/TB1. Gene structure and conserved motif analysis showed that most members of the TCP gene family contained motif 1, motif 2, motif 3 in the same order and 1-3 CDS. The analysis of promoter cis-acting elements showed that the transcriptional expression of the TCP gene family in A. paniculata might be induced by light, hormones, and adversity stress. In light of the expression pattern analysis and qRT-PCR verification, the expression of ApTCP4, ApTCP5, ApTCP6, and ApTCP11 involved in response by various abiotic stresses such as drought, high temperature, and MeJA. This study lays the foundation for in-depth exploration of the functions of A. paniculata TCP genes in response to abiotic stress.

ABSTRACT

The plant growth, development, and secondary metabolism are regulated by R2 R3-MYB transcription factors. This study identified the R2 R3-MYB genes in the genome of Andrographis paniculata and analyzed the chromosomal localization, gene structure, and conserved domains, phylogenetic relationship, and promoter cis-acting elements of these R2 R3-MYB genes. Moreover, the gene expression profiles of R2 R3-MYB genes under abiotic stress and hormone treatments were generated by RNA-seq and validated by qRT-PCR. The results showed that A. paniculata contained 73 R2 R3-MYB genes on 21 chromosomes. These members belonged to 34 subfamilies, 19 of which could be classified into the known subfamilies in Arabidopsis thaliana. The 73 R2 R3-MYB members included 36 acidic proteins and 37 basic proteins, with the lengths of 148-887 aa. The domains, motifs, and gene structures of R2 R3-MYBs in A. paniculata were conserved. The promoter regions of these genes contains a variety of cis-acting elements related to the responses to environmental factors and plant hormones including light, ABA, MeJA, and drought. Based on the similarity of functions of R2 R3-MYBs in the same subfamily and the transcription profiles, ApMYB13/21/35/67/73(S22) may regulate drought stress through ABA pathway; ApMYB20(S11) and ApMYB55(S2) may play a role in the response of A. paniculata to high temperature and UV-C stress; ApMYB5(S7) and ApMYB33(S20) may affect the accumulation of andrographolide by regulating the expression of key enzymes in the MEP pathway. This study provides theoretical reference for further research on the functions of R2 R3-MYB genes in A. paniculata and breeding of A. paniculata varieties with high andrographolide content.

ABSTRACT

INTRODUCTION: The aim of this prospective study is to evaluate the effects of combining topical airway anaesthesia with intravenous induction on haemodynamic variables during the induction period in patients undergoing cardiac surgery. METHODS AND ANALYSIS: This randomised, double-blind, controlled, parallel-group, superiority study from 1 March 2021 to 31 December 2021 will include 96 participants scheduled for cardiac surgery. Participants will be screened into three blocks (ASA II, ASA III, ASA IV) according to the American Society of Anesthesiologists (ASA) grade and then randomly allocated into two groups within the block in a 1:1 ratio. Concealment of allocation will be maintained using opaque, sealed envelopes generated by a nurse according to a computer-generated randomisation schedule. In addition to general intravenous anaesthetics, participants will receive supraglottic and subglottic topical anaesthesia. Changes in arterial blood pressure and heart rate in both groups will be recorded by an independent investigator at the start of anaesthesia induction until the skin incision. If vasopressors are used during this period, the frequency, dosage and types of vasopressors will be recorded. The incidence and severity of participants' postoperative hoarseness and sore throat will also be assessed. ETHICS AND DISSEMINATION: This study was approved by the Ethics Committee of Qianfoshan Hospital of Shandong Province (registration number: YXLL-KY-2021(003)). The results will be disseminated through a peer-reviewed publication and in conferences or congresses. TRIAL REGISTRATION NUMBER: NCT04744480.

ABSTRACT

Nine resorcinol derivatives including two new ones, 5-[(8Z,11Z,14Z)-nonadeca-8,11,14-trienyl] resorcinol (1) and 5-[(8Z,11Z,14E)-heptadeca-8,11,14-trienyl] resorcinol (2), were isolated from the leaves of Syzygium samarangense. The new structures were elucidated by means of extensive spectroscopic techniques including interpretation of 1D and 2D NMR spectra. Among them, compounds 3, 4, 6 and 7 exhibited significant α-glucosidase inhibitory activities with IC50 of 3.16, 3.16, 2.34 and 0.99 µM, respectively. This finding provides evidence that resorcinol derivatives with long aliphatic chain function as new promising antidiabetic alternatives.

ABSTRACT

This research used batch soil column experiment to study the effects of irrigating with reclaimed water and tap water on the soil chemical properties and culturable microorganisms. The results indicated that reclaimed water could markedly increase the soil organic material (OM) and total nitrogen (TN) content, but it had no obvious effect on total phosphorus (TP), available phosphorus (AP) and pH value. Reclaimed water irrigation could significantly enhance the amounts of surface soil bacteria and actinomycetes at a depth of 0-20 cm, but it had little effect on the biomass of 20-40 cm and 40-60 cm soil layers. The dominant bacteria in tap water irrigation area was the genus Bacillus whereas that of reclaimed water irrigation area was the genus Acinetobacter. Tap water irrigation area had four endemic genera and reclaimed water irrigation area had six endemic genera. Reclaimed water had no obvious effect on the microbial community Shannon diversity of 0-20 cm soil layer, while it decreased Pielou evenness index, and improved Margalef richness index. Through SPSS 17. 0 correlation analysis between soil microbes quantity and soil chemical properties, it was shown that the soil microbes quantity was positively correlated with OM, TN, TP and AP, but negatively correlated with soil water content (SWC) and pH value. Based on CANOCO 4.5 detrended correspondence analysis (DCA) and redundancy analysis (RDA) between soil microbes species and soil chemical properties, it was shown that AP had the strongest correlation with the microbial community (P = 0.002). TN and TP had larger impact on Streptococcus, Aeromonas and Neisseria. OM and AP had larger impact on Aerococcus, Planococcus and Halobacterium.

ABSTRACT

The complete nucleotide sequence of the sugar beet (Beta vulgaris ssp. vulgaris) chloroplast genome (cpDNA) was determined in this study. The cpDNA was 149,637 bp in length, containing a pair of 24,439 bp inverted repeat regions (IR), which were separated by small and large single copy regions (SSC and LSC) of 17,701 and 83,057 bp, respectively. 53.4% of the sugar beet cpDNA consisted of gene coding regions (protein coding and RNA genes). The gene content and relative positions of 113 individual genes (79 protein encoding genes, 30 tRNA genes, 4 rRNA genes) were almost identical to those of tobacco cpDNA. The overall AT contents of the sugar beet cpDNA were 63.6% and in the LSC, SSC and IR regions were 65.9%, 70.8% and 57.8%, respectively. Fifteen genes contained one intron, while three genes had two introns.

ABSTRACT

In the present paper, the micro-NIR spectrometer with the splitter of linear variable filter was used to develop the recognition models of the West Lake Longjing tea and the ordinary flat tea of the year 2012 and 2013. The NIR spectral data of different years and different storage times were decomposed by PCA algorithm. The PLS-DA models were developed by the representative samples selected by the mathematical characteristics of PCA-scores' distribution in order to analyze the reason for the inadaptability of the models according to mathematical principles and find out the solution for its correction. Being examined by the external validation set, the adaptability of the authenticity identification model was enhanced effectively. The result of this research indicated that, for the West Lake Longjing tea and the ordinary flat tea, the correct recognition rate of the model developed by all different-year samples' NIR spectral data would be enhanced effectively. The model developed by the NIR spectral data of different storage time samples indicated that the physicochemical properties of the ordinary flat tea have changed remarkably after cryopreservation for 3 months, while the physicochemical properties of the West Lake Longjing tea are relatively stable. The model adaptabilities for different years and different storage times were studied according to the mathematical perspective of the principal component characteristics of spectral data. After the authenticity identification model of West Lake Longjing tea was developed, the prediction accuracy was enhanced effectively. This research would provide reference for not only the application of NIR spectroscopy in quality grading and safety of agricultural products, but also the enhancement of the prediction accuracy of the NIR grading models for agricultural products.

ABSTRACT

Fifteen compounds were obtained from the twigs and leaves of Caesalpinia minax. Their structures were identified as apigenin (1), 5,7,3',4'- tetrahydroxy-3-methoxyflavone (2), luteolin-5, 3 '-dimethyl-ether (3), thevetiaflavon (4), apigenin-7-O-beta-D-glucuronide (5), bonducellin (6), 7-hydroxy-3-( 4-hydroxybenzylidene )-chroman-4-one (7), 3-deoxysappanchalcone (8), 5-acetonyl-7-hydroxy-2-methyl chromone (9), 4-(trans)-acetyl-3,6,8-trihydroxy-3-methyl-dihydronaphthalenone (10), 4-(cis)-acetyl-3,6,8-trihydroxy-3-methyl-dihydronaphthalenone (11), vanillic acid (12), omega-hydroxypropioquaiacone (13), syringaresinol (14) and uracil (15). All compounds were isolated from C. minax for the first time. Compounds 1-14 were phenolic compounds and compounds 1-5, 9-13 and 15 were isolated from the genus Caesalpinia for the first time.

ABSTRACT

Five new 1,3-diarylpropan-1-ols, combretol A-E (1- 5), together with one known coumarin (6) and ten known triterpenes (7-16), were isolated from Combretum yunnanense. Their structures were determined by spectroscopic investigation, including ¹H and ¹³C NMR, NOESY, HSQC, HMBC, and HRESIMS analyses. This is the first report on the occurrence of 1,3-diarylpropan-1-ols and coumarin in the Combretum genus including C. yunnanense. Also, to the best of our knowledge, 1,3-diarylpropan-1-ols are rare in nature.

ABSTRACT

OBJECTIVE: To investigate the effect of hyperbaric oxygen(HBO)therapy on mitochondrial free radicals after transient focal cerebral ischemia in rats. METHODS: The male SD rats were randomly assigned into two groups, control and HBO groups. All animals were subjected to 90 min intra-luminal middle cerebral artery occlusion (MCAO) with the regional cerebral blood flow monitored in vivo by laser Doppler flowmetry. HBO treatment was performed in a pressure chamber with 100% O(2)(3 ATM 1 h) 3 h after ischemia. Twenty-four hours after ischemia, mitochondria in the ischemic core and penumbra were isolated and the contents of H(2)O(2), O(2)(*-), MDA, SOD, GSH-PX and GSH in mitochondria were measured respectively. RESULT: After cerebral ischemia-reperfusion, contents of mitochondrial H(2)O(2), O(2)(*-), MDA increased, while the SOD, GSH-PX and GSH in the mitochondria decreased significantly both in the ischemic core and the ischemic penumbra, compared with those in the normal controls(P<0.05). In the ischemic penumbra, HBO therapy increased significantly the content of O(2)(*-)(P<0.05), enhanced the activity of SOD, and decreased the level of MDA (P<0.05). However, HBO therapy did not change the level of MDA, though it also increased the content of O(2)(*-) and the activity of SOD in the ischemic core. HBO therapy had no significant effect on the contents of H(2)O(2), GSH-PX and GSH in the ischemic mitochondria. CONCLUSION: HBO therapy initiated early after acute transient cerebral ischemia in rats can increase the mitochondrial free radicals level, but also increase the activity of the anti-radical enzymes. HBO treatment inhibits the lipid peroxidation damage of mitochondria in the ischemic penumbra, but not in the ischemic core, which indicates that the mitochondrial function plays a role in the reaction of the free radical in the ischemic area after HBO therapy.

ABSTRACT

Improving the efficiency of fertilization is an effective method of enhancing income for farmers, but it depends on measuring the soil nutrients accurately and rapidly. Near infrared reflectance spectroscopy (NIRS) is a fast method to detect the soil nutrients. In order to evaluate the feasibility of using NIRS to determine the soil N and P contents, the soil samples were collected from different LULC (land use and land cover) types in Daxing district, Beijing, and their biochemical parameters were determined by traditional chemical method. Then, the near infrared reflectance spectra of the samples were acquired, and NIRS models were built using partial least square regression (PLS) and Fourier transform technology for the total N and total P. The determination coefficients of cross validation for the total N and total P were 0.862 6 and 0.668 5 respectively. Ten samples were used to test the performance of the models. The coefficients of correlation between the chemically determined value and the NIRS predicted one were 0.969 8(N) and 0.830 7(P) respectively. The root mean standard error of prediction for N and P were 0.009 5(%) and 0.008 6(%), respectively. The ratios of RMSEP to SD(RPD) were 3.78(N) and 1.69(P). The results showed that the NIRS could be used to evaluate the soil N accurately and the soil P roughly.

ABSTRACT

It take long time and high cost to measure sulfate reducing bacteria (SRB) in wastewater of oil field. A rapid quantitative method was developed by combining polymerase chain reaction(PCR) and most probable number (MPN) to measure sulfate reducing bacteria (SRB) in wastewater of oil field. The bacterium solution was directly prepared from wastewater for PCR amplification, which ensured quantitative accuracy. Reaction system and amplification condition were designed using universal primers DSR1F and DSR5R of dissimilatory sulfite reductase in SRB. The result show that the accuracy of this method is two magnitude higher than that of MPN. The whole measuring process take 3 - 4 hours and the reproducibility of this method is extremely stable, being fit to practical process.
