ABSTRACT

Objective: To explore the influence of early psychological intervention on AIDS patients receiving hospital-led case management. Methods: Between December 2022 and May 2023, 100 cases of AIDS patients were gathered at the Fifth Hospital of Shijiazhuang. They were randomly assigned to either a psychological intervention group or a conventional intervention group. Each group consists of 50 individuals affected by AIDS. The conventional intervention group received care through traditional care models. In contrast, the psychological intervention group received immediate psychological support upon being screened positive for HIV antibodies, in addition to the conventional intervention group. The intervention period for both groups lasted 3 months, during which medication adherence, adverse emotional conditions, self-management skills, and quality of life were compared. Results: During the intervention period, the psychological intervention group exhibited higher medication adherence than the conventional intervention group. Post-intervention, the levels of anxiety as assessed by the SAS and the degree of depression as evaluated by the SDS of the conventional intervention group were higher than those of the psychological intervention group. Additionally, the psychological intervention group demonstrated higher scores in 7 dimensions the total score on the self-management ability scale, and a higher score in the WHOQOL-HIV-BREF compared to the conventional intervention group. Conclusion: In the context of hospital-led case management for AIDS, early psychological intervention in patients has been shown to enhance medication adherence, reduce negative emotions, improve self-management skills, and enhance overall quality of life.

ABSTRACT

Background: Intestinal obstruction frequently leads to acute kidney injury (AKI), yet understanding the specific clinical features and influencing factors in these patients remains limited. Objective: This study aims to comprehensively evaluate the clinical features and affecting factors of AKI in patients with intestinal obstruction. Methods: We conducted a systematic search of PubMed, Embase, Cochrane Library, China National Knowledge Infrastructure (CNKI), Wanfang Data Knowledge Service platform, VIP, and China Biology Medicine Literature Database from 2010 to the present. Relevant literature addressing the clinical features or affecting factors of AKI in patients with intestinal obstruction was retrieved. The study group (SG) comprised patients with intestinal obstruction complicated by AKI, while the control group (CG) included patients without AKI after intestinal obstruction. Meta-analysis was performed to investigate the clinical features of both patient groups and funnel plots were employed to assess publication bias. Results: This study included 5 articles with a total of 7583 patients (1472 in the SG and 6111 in the CG). Meta-analysis results indicated a higher prevalence of hypertension (OR=1.80, 95% CI: 1.59-2.04, P < .00001), cardiovascular disease (OR=1.60, 95% CI: 1.06-2.41, P = .03), and diabetes (OR=1.61, 95% CI: 1.35-1.91, P < .00001) in both groups. Additionally, factors such as infection rate (OR=4.03, 95% CI: 2.10-7.73, P < .0001), age (OR=2.90, 95% CI: 1.07-7.90, P = .04), and BMI (OR=1.31, 95% CI: 1.15-1.48, P < .0001) significantly influenced AKI occurrence. No significant difference was observed in the history of pelvic surgery between both groups (OR=0.91, 95% CI: 0.53-1.55, P = .73). Funnel plot analysis suggested minimal publication bias. Conclusions: Age, BMI, hypertension, cardiovascular disease, diabetes mellitus, and infection emerge as primary influencing factors for AKI in patients with intestinal obstruction. Our findings advocate for early interventions to mitigate the global incidence of AKI in this patient population.

ABSTRACT

Xyloglucan endotransglucosylase/hydrolases (XTHs) play a crucial role in plant growth and development. However, their functional response to phytohormone in sugar beet still remains obscure. In this study, we identified 30 putative BvXTH genes in the sugar beet genome. Phylogenetic and evolutionary relationship analysis revealed that they were clustered into three groups and have gone through eight tandem duplication events under purifying selection. Gene structure and motif composition analysis demonstrated that they were highly conserved and all contained one conserved glycoside hydrolase family 16 domain (Glyco_hydro_16) and one xyloglucan endotransglycosylase C-terminus (XET_C) domain. Transcriptional expression analysis exhibited that all BvXTHs were ubiquitously expressed in leaves, root hairs and tuberous roots, and most of them were up-regulated by brassinolide (BR), jasmonic acid (JA), abscisic acid (ABA) and gibberellic acid (GA3). Further mutant complementary experiment demonstrated that expression of BvXTH17 rescued the retarded growth phenotype of xth22, an Arabidopsis knock out mutant of AtXTH22. The findings in our work provide fundamental information on the structure and evolutionary relationship of the XTH family genes in sugar beet, and reveal the potential function of BvXTH17 in plant growth and hormone response.

ABSTRACT

The SEVEN IN Absentia (SINA), a typical member of the RING E3 ligase family, plays a crucial role in plant growth, development and response to abiotic stress. However, its biological functions in oil crops are still unknown. Previously, we reported that overexpression of AtSINA2 in Arabidopsis positively regulated the drought tolerance of transgenic plants. In this work, we demonstrate that ectopic expression of AtSINA2 in soybean improved the shoot growth, grain yield, drought tolerance and seed oil content in transgenic plants. Compared to wild type, transgenic soybean produced greater shoot biomass and grain yield, and showed improved seed oil and drought tolerance. Physiological analyses exhibited that the increased drought tolerance of transgenic plants was accompanied with a higher chlorophyll content, and a lower malondialdehyde accumulation and water loss during drought stress. Further transcriptomic analyses revealed that the expressions of genes related to plant growth, flowering and stress response were up- or down-regulated in transgenic soybean under both normal and drought stress conditions. Our findings imply that AtSINA2 improved both agricultural production and drought tolerance, and it can be used as a candidate gene for the genetic engineering of new soybean cultivars with improved grain yield and drought resistance.

ABSTRACT

Icariin, a flavonoid glycoside isolated from Epimedium brevicornum, exerts a variety of biological activities. However, its effects on depression-induced glucocorticoid resistance in asthma and the underlying mechanisms have not been elucidated. In this study, a murine model of asthma with depression was established by exposure to ovalbumin combined with chronic unpredictable mild stress, and icariin was given orally during ovalbumin challenge and chronic unpredictable mild stress exposure. Depression-like behaviors were assessed by the open field test, forced swim test, and tail suspension test. The characteristic features of allergic asthma, including airway hyperreactivity, histopathology, inflammatory cytokine levels in bronchoalveolar lavage fluid, and immunoglobulin E and corticosterone levels in serum, were examined. Following splenocyte isolation in vitro, the inhibitory effects of corticosterone on the proliferation and cytokine secretion of splenocytes, glucocorticoid receptor DNA-binding activity, and expression of p-glucocorticoid receptor s226, glucocorticoid receptor α, and p-p38 mitogen-activated protein kinase in splenocytes were determined. We found that icariin had limited effects on depression-like behaviors, however, it markedly suppressed airway hyperresponsiveness, inflammatory infiltration in lung tissues, levels of interleukin-4, interleukin-5, and interleukin-6 in bronchoalveolar lavage fluid, and immunoglobulin E in serum. Furthermore, icariin improved the inhibitory effects of corticosterone on lipopolysaccharide-stimulated splenocytes, increased the glucocorticoid receptor expression and glucocorticoid receptor DNA-binding activity, and inhibited the phosphorylation of glucocorticoid receptors S226 and p38 mitogen-activated protein kinase. Taken together, icariin improved glucocorticoid resistance in a murine model of asthma with depression associated with enhancement of glucocorticoid receptor function and glucocorticoid receptor expression, and its effects on the glucocorticoid receptor function were related to decreased phosphorylation of glucocorticoid receptors S226 and p38 mitogen-activated protein kinase.

ABSTRACT

Intratumoral hypoxia, which is in favour of cancer cell proliferation, invasion and metastasis, also inhibits photodynamic therapy (PDT) badly. Herein, second near-infrared (NIR-II) photocatalytic O2 production is established to realize hypoxia relief. MoS2/Co3S4@PEG (MSCs@PEG) nanoflowers (100-150 nm) are prepared via a two-step hydrothermal method. These samples possess high NIR-II harvest and photothermal conversion (39.8 %, 1064 nm) ability. That not only reveals photothermal therapy (PTT) but also lifts the thermal energy of nanomaterials to replenish extra energy, making sure the co-excitation of MoS2 (1.14 eV) and Co3S4 (1.40 eV) by low-energy NIR-II (1064 nm, 1.16 eV) laser. The investigation of band structure further displays the Z-Scheme characterization of MSCs heterostructure. These photo-excited holes/electrons hold great redox ability to form O2 (water splitting) and reactive oxygen species (ROS), simultaneously. In addition, MSC-2@PEG can be served to mimic catalase, peroxidase, and glutathione (GSH) oxidase to further boost oxidative stress. It is noted that heterostructure discovers the greater nanozyme activity, attributing to the lower resistance for charge transfer. Moreover, MSC-2@PEG displays a novel biodegradation ability to induce the elimination via urine and faeces within 14 days. Given the superparamagnetic and photothermal effect, the nanocomposite can be used as magnetic resonance and photothermal imaging (MRI and PTI) contrast. Associated with dual-imaging, intracellular O2 supplementation, and synergistic chemotherapy (CDT)/PTT/PDT, MSC-2@PEG possess great tumor inhibition that also efficiently motivates immune response for anticancer.

ABSTRACT

Electroacupuncture (EA) could enhance neuroregeneration and posttraumatic conditions; however, the underlying regulatory mechanisms remain ambiguous. PDCD6 (programmed cell death 6) is an established proapoptotic regulator which is responsible for motoneuronal death. However, its potential regulatory role in post-spinal cord injury (SCI) regeneration has remained largely unknown. Further investigations are warranted to clarify the involvement of PDCD6 post-SCI recovery and the underlying mechanisms. In our study, based on bioinformatics prediction, we found that miR-34a-3p might be an upstream regulator miRNA for PDCD6, which was subsequently validated through combined utilization of the qRT-PCR, western blot, and dual-luciferase reporter system. Our in vitro results showed that miR-34a-3p might promote the in vitro differentiation of neural stem cell (NSC) through suppressing PDCD6 and regulating other important neural markers such as fibroblast growth factor receptor 1 (FGFR1), MAP1/2 (MAP kinase kinases 1/2), myelin basic protein (MBP), ßIII-tubulin Class III ß-tubulin (ßIII tubulin), and glial fibrillary acidic protein (GFAP). Notably, in the post-SCI rat model, exogenous miR-34a-3p agomir obviously inhibited the expression of PDCD6 at the protein level and promoted neuronal proliferation, motoneurons regeneration, and axonal myelination. The restorations at cellular level might contribute to the improved hindlimbs functions of post-SCI rats, which was manifested by the Basso-Beattie-Bresnahan (BBB) locomotor test. The impact of miR-34a-3p was further promoted by EA treatment in vivo. Conclusively, this paper argues that a miR-34a-3p/PDCD6 axis might be a candidate therapeutic target for treating SCI and that the therapeutic effect of EA is driven through this pathway.

ABSTRACT

The efficacy of phototherapy is dependent on intracellular O2 concentration and NIR harvest. Here, a simple nanoplatform with nanoenzyme mediated phototherapy enhances anticancer capacity. Mn-CoS@carbon (CMS/C) di-shell hollow nanospheres (50 nm) are synthesized successfully through two-step consecutive Kirkendall process. The nanoheterostructure reveals the higher near-infrared (NIR) light absorption and photothermal conversion rate of 66.3% than pure CoS (45.5%), owing to the decreased band gap and multi-reflection of incident light in the hollow structure. And CMS/C reveals the reactive oxygen species (ROS) production and nanoenzyme activities (mimic peroxidase and catalase) that are 6 and 2 times than those of pure CoS. Furthermore, the nanoenzyme exhibits NIR-enhanced abilities to produce more OH and O2 facilitating anticancer. In addition, it also depletes glutathione (mimicking glutathione oxidase), to disturb intracellular redox-homeostasis, boosting the increase of oxidative stress. With grafting bovine serum albumin (BSA) and drug loading, CMS/C@BSA-Dox integrated multi-therapy make the great anticancer effect in vitro and vivo. After that, the nanocomposite could be biodegraded and eliminated via urinary and feces within 14 days. Based on this work, the efficient charge-separation can be designed to reveal high performance nanoenzymes as well as photosensitizers for anticancer.

INTRODUCTION: Currently, the first choice for the clinical treatment of static tremor in Parkinson's disease (PD) is drug therapy, however side effects are common. In recent years, the effects of physical therapy on PD has become a serious research focus. Studies have indicated that aerobic and resistance exercises alleviate PD movement disorders and improve aerobic capacity, but the effects of Qigong on PD static tremor and aerobic capacity remain unknown. METHODS AND ANALYSIS: OBJECTIVE: To observe the effects of Zhan Zhuang Qigong on upper limb static tremor and aerobic capacity in patients with PD, we established a rigorous randomised, parallel-controlled, assignment hidden, evaluator-blinded protocol. METHODS: Seventy-two patients with PD, at the Affiliated Hospital of Shandong University of Traditional Chinese Medicine, were recruited and randomly divided into a control (n=36) and experimental group (n=36). The intervention group received conventional medicine plus Zhan Zhuang Qigong exercises five times a week at 30 min each time, over an 8-week period. The long-term effects of Zhan Zhuang Qigong on PD was investigated after the intervention. Phyphox APP, CRST, CPET, UPDRS(II, III) were used to evaluate tremor, aerobic capacity, and motor function in groups. DISCUSSION: We are investigating the effects of Zhan Zhuang Qigong on upper limb static tremor and aerobic capacity in patients with PD. If positive are identified, they will add a new research direction and evidence for the clinical exploration of exercise therapy for PD. ETHICS AND DISSEMINATION: This study was approved by the Ethics Committee of Shandong University of Traditional Chinese Medicine (Approval Number: 2021-025-KY). The Committee will be informed of any changes to the trial protocol, such as intervention intensity, outcome indicators and data collection. Study results will be presented as a paper at an international conference or in a journal. TRIAL REGISTRATION NUMBER: ChiCTR2100053529.

ABSTRACT

In order to optimize water and fertilizer use in the double-cropping rice in eastern Fujian Province, a field runoff plot experiment was conducted to investigate rice yield, nutrient uptake, and runoff losses of N (nitrogen) and P (phosphorus) in the T0(no chemical fertilization with traditional flooding irrigation), T1(common chemical fertilizer of 273 kg N·hm-2, 59 kg P·hm-2, and 112 kg K·hm-2 combined with traditional flooding irrigation), T2(chemical fertilizer of 240 kg N·hm-2, 52 kg P·hm-2, and 198 kg K·hm-2 combined with traditional flooding irrigation) and T3(chemical fertilizer combined with shallow intermittent irrigation) treatments. Results showed that early rice grain yield in the T1, T2 and T3 treatments significantly increased by 0.7, 1.0, 1.1 times, late rice grain yield significantly increased by 0.9, 1.1, 1.0 times compared to that in the T0 treatment, respectively. The T1, T2 and T3 treatments significantly increased the uptake of N and P in aboveground parts of the plants, especially in grains. The T1, T2 and T3 treatments significantly increased N uptake by 1.1, 1.2, 1.2 times, increased P uptake by 0.9, 1.4, 1.6 times in early-season grains, and significantly increased N uptake by 0.8, 1.0, 1.0 times, increased P uptake by 0.7, 0.9, 0.9 times in late-season grains, compared to T0, respectively. Furthermore, T3 increased agronomic N use efficiency (AEN) and agronomic P use efficiency (AEP) by 71.1% and 69.2% in early rice plants, increased AEN and AEP by 26.4% and 25.0% in late rice plants, whereas T3 decreased total dissolved N (DN) by 16.0% in comparison with T1. Dissolved inorganic N loss in surface runoff occurred mainly in the form of NO3--N (nitrate N) under different water and fertilizer regimes. However, there were no significant differences in AEN and AEP between T2 and T3 treatments. These findings suggested that optimal applications of water and fertilizers (T3) might increase N and P uptake in rice plants, maintain yield, and reduce N loss, especially in the form of NO3--N in surface water from early rice field. In general, this study could provide theoretical support for the optimization of irrigation and fertilization and for the control of N and P non-point source pollution from the double cropping rice paddy fields in eastern Fujian Province.

ABSTRACT

Heavy metal pollution is consistently a critical global issue, and bioremediation is regarded as one of the most promising approaches. In this work, the biosorption characteristics of Cd(II), Pb(II), Cu(II) and Zn(II) from aqueous solutions using three phosphorus-accumulating organisms (PAOs) biomasses, Ochrobactrum cicero (PAB-006), Stenotrophomonas maltophilia (PAB-009), and Pseudomonas putida (PAB-0031), as biosorbents were investigated. Results indicated that the equilibrium biosorption capacities of biosorbents to heavy metal ions were sensitive to the solution pH, and increased with increasing pH values. The experimental data of Cd(II), Pb(II), Cu(II) and Zn(II) biosorption were in good agreement with the Pseudo-second-order, Redlich-Peterson and Temkin models, implying that the biosorption was a hybrid chemical reaction-biosorption process. In addition, the theoretical maximum biosorption capacities of Cd(II), Pb(II), Cu(II) and Zn(II) were calculated to be 67.84, 80.23, 50.56 and 63.07 mg/g for PAB-006, 59.99, 87.71, 39.26 and 64.00 mg/g for PAB-009 and 68.31, 85.43, 38.97 and 62.85 mg/g for PAB-031, respectively (pH = 5.0 ± 0.1, T = 25 °C), according to the parameters of the Langmuir model. Moreover, ionic strength had negligible influences or slight promoting effects, while humic acid exhibited positive effects on the removal of heavy metals. Further, PABs were stable and displayed excellent reusability. Characterization techniques of FTIR and XPS revealed that surface complexation, ion exchange, hydrogen bonding and electrostatic interaction were the main mechanisms involved in the biosorption process. In summary, the biosorbent PABs possessed high biosorption performance with excellent reusability, and which hold the great application prospect in the treatment of heavy metal contaminated water.

ABSTRACT

Danhong injection (DHI) restrains diabetic retinopathy and nephropathy (DR and DN) advancement in diabetic mice. However, the downstream mechanism of its modulation is not fully studied. Diabetic model mice (db/db mice) were intravenously injected with DHI and corresponding virus particles. MiR-30d-5p and JAK1 were detected. The body weight and fasting blood glucose mice were measured every 4 weeks. The renal tissues and serum of mice were collected, and the contents of creatinine and blood urea nitrogen were biochemically analyzed. IL-6, IFN-γ and TNF-α were detected by ELISA, with the pathological conditions of renal tissues in mice by He staining, and the adjustment conditions by TUNEL. Human retinal pigment epithelium (ARPE-19) cells were selected to induce DR model in vitro by high glucose, and exposed to DHI for treatment. The corresponding plasmids were transfected, and miR-30d-5p and JAK1 were detected, with the proliferation ability by plate cloning, apoptosis by flow cytometry, and cell migration ability by Transwell. The angiogenesis ability of cells was assessed by tube formation assay. The targeting relationship between miR-30d-5p and JAK1 was detected. The results manifested that miR-30d-5p was declined in DR and DN, while JAK1 expression was elevated. DHI was able to improve DR and renal injury. DHI could regulate the miR-30d-5p-JAK1 axis in vivo, and miR-30d-5p targeted and regulated JAK1. Upregulation of miR-30d-5p or inhibition of JAK1 could improve DR and renal injury. The results implies that DHI can repress the development of DR and DN by elevating miR-30d-5p and targeting JAK1.

BACKGROUND: In China, depressive disorders have been estimated to be the second leading cause of years lived with disability. However, nationally representative epidemiological data for depressive disorders, in particular use of mental health services by adults with these disorders, are unavailable in China. The present study, part of the China Mental Health Survey, 2012-15, aims to describe the socioeconomic characteristics and the use of mental health services in people with depressive disorders in China. METHODS: The China Mental Health Survey was a cross-sectional epidemiological survey of mental disorders in a multistage clustered-area probability sample of adults of Chinese nationality (≥18 years) from 157 nationwide representative population-based disease surveillance points in 31 provinces across China. Trained investigators interviewed the participants with the Composite International Diagnostic Interview 3.0 to ascertain the presence of lifetime and 12-month depressive disorders according to DSM-IV criteria, including major depressive disorder, dysthymic disorder, and depressive disorder not otherwise specified. Participants with 12-month depressive disorders were asked whether they received any treatment for their emotional problems during the past 12 months and, if so, the specific types of treatment providers. The Sheehan Disability Scale (SDS) was used to assess impairments associated with 12-month depressive symptoms. Data-quality control procedures included logic check by computers, sequential recording check, and phone-call check by the quality controllers, and reinterview check by the psychiatrists. Data were weighted according to the age-sex-residence distribution data from China's 2010 census population survey to adjust for differential probabilities of selection and differential response, as well as to post-stratify the sample to match the population distribution. FINDINGS: 28 140 respondents (12 537 [44·6%] men and 15 603 [55·4%] women) completed the survey between July 22, 2013, and March 5, 2015. Ethnicity data (Han or non-Han) were collected for only a subsample. Prevalence of any depressive disorders was higher in women than men (lifetime prevalence odds ratio [OR] 1·44 [95% CI 1·20-1·72] and 12-month prevalence OR 1·41 [1·12-1·78]), in unemployed people than employed people (lifetime OR 2·38 [95% CI 1·68-3·38] and 12-month OR 2·80 [95% CI 1·88-4·18]), and in people who were separated, widowed, or divorced compared with those who were married or cohabiting (lifetime OR 1·87 [95% CI 1·39-2·51] and 12-month OR 1·85 [95% CI 1·40-2·46]). Overall, 574 (weighted % 75·9%) of 744 people with 12-month depressive disorders had role impairment of any SDS domain: 439 (83·6%) of 534 respondents with major depressive disorder, 207 (79·8%) of 254 respondents with dysthymic disorder, and 122 (59·9%) of 189 respondents with depressive disorder not otherwise specified. Only an estimated 84 (weighted % 9·5%) of 1007 participants with 12-month depressive disorders were treated in any treatment sector: 38 (3·6%) in speciality mental health, 20 (1·5%) in general medical, two (0·3%) in human services, and 21 (2·7%) in complementary and alternative medicine. Only 12 (0·5%) of 1007 participants with depressive disorders were treated adequately. INTERPRETATION: Depressive disorders in China were more prevalent in women than men, unemployed people than employed, and those who were separated, widowed, or divorced than people who were married or cohabiting. Most people with depressive disorders reported social impairment. Treatment rates were very low, and few people received adequate treatment. National programmes are needed to remove barriers to availability, accessibility, and acceptability of care for depression in China. ABSTRACT

Rice cultivation usually involves high water and fertilizer application rates leading to the nonpoint pollution of surface waters with phosphorus (P) and nitrogen (N). Here, a 10-year field experiment was conducted to investigate N and P losses and their impact factors under different irrigation and fertilization regimes. Results indicated that T2 (Chemical fertilizer of 240 kg N ha-1, 52 kg P ha-1, and 198 kg K ha-1 combined with shallow intermittent irrigation) decreased N loss by 48.9% compared with T1 (Chemical fertilizer of 273 kg N ha-1, 59 kg P ha-1, and 112 kg K ha-1 combined with traditional flooding irrigation). The loss ratio (total N loss loading/amount of applied N) of N was 9.24-15.90%, whereas that of P was 1.13-1.31% in all treatments. Nitrate N (NO3--N) loss was the major proportion accounting for 88.30-90.65% of dissolved inorganic N loss through surface runoff. Moreover, the N runoff loss was mainly due to high fertilizer input, soil NO3--N, and ammonium N (NH4+-N) contents. In addition, the N loss was accelerated by Bacteroidetes, Proteobacteria, Planotomycetes, Nitrospirae, Firmicutes bacteria and Ascomycota fungi, but decreased by Chytridiomycota fungi whose contribution to the N transformation process. Furthermore, T2 increased agronomic N use efficiency (AEN) and rice yield by 32.81% and 7.36%, respectively, in comparison with T1. These findings demonstrated that T2 might be an effective approach to ameliorate soil chemical properties, regulate microbial community structure, increase AEN and consequently reduce N losses as well as maintaining rice yields in the present study.

ABSTRACT

In this study, we aim to evaluate the efficacy and safety of Guizhi Decoction associated formulas for the treatment of allergic rhinitis. A total of seven online databases were searched to collect studies published up to Feb 23rd, 2020. Study quality of each included article was evaluated by the Cochrane Collaboration risk of bias tool. Systematic reviews were conducted based on the Cochrane systematic review method by using RevMan 5.3 Software. Among the included trials, Guizhi Decoction associated formulas alone (or plus Western medicine, or acupoint-based therapy) were main therapies in experimental groups. Interventions in control groups include Western medicine, Guizhi Decoction associated formulas alone, Chinese patent medicine, and placebo control. Primary outcomes in this study include recovery rate, accumulative marked effective rate, accumulative effective rate, and recurrence rate. Finally, 23 trials involving 2281 participants were included. Results of systematic reviews show that Guizhi Decoction and associated formulas alone, plus Western medicine or plus acupoint-based therapies, were significantly better compared with using Western medicine alone in terms of efficacy. In addition, the formulas plus nasal TCM fumigation therapy could improve effective rate for AR treatment compared to using the formulas alone. More types and cases of adverse events were reported in the control groups (Western medicine alone), but events of included trials were all mild and did not need specific medical intervention. More RCTs of high quality, and large sample size, with appropriate blinding methods or nonblinded pragmatic trials of Guizhi Decoction and associated formulas for AR are needed.

ABSTRACT

Background: Parkinson's disease (PD) is the second most common neurodegenerative disease worldwide and involves deficiencies in alpha-synuclein (α-Syn) degradation. Effective therapeutic strategies for PD are urgently needed. L-asparaginase (L-ASNase) has been developed for therapeutic applications in many fields because it catalyzes the hydrolysis of asparagine and glutamine in cancer cells, which may also activate autophagy and induce the degradation of accumulated α-Syn. However, the efficacy and related mechanism of L-ASNase in PD remain poorly understood. Methods: We determined the correlation between L-ASNase and autophagic degradation of α-Syn in a cell model of PD. Mitochondrial function and apoptosis were examined in the presence or absence of L-ASNase. Then, we applied GC-MS/MS targeted amino acid metabolomics analysis to validate the amino acid regulation induced by L-ASNase treatment. Glutamine was added to verify whether the neuroprotective effect was induced by deprivation of glutamine. α-Syn-related autophagy and mitochondrial fusion/fission dynamics were detected to explore the mechanism of L-ASNase-based therapy in PD. Results: L-ASNase activated the autophagic degradation of α-Syn in a cell model of PD without cytotoxicity at specific concentrations/times. Under these conditions, L-ASNase showed substantial neuroprotective effects, including improvements in mitochondrial function and decreased apoptosis. Through GC-MS/MS targeted analysis, glutamine metabolism was identified as the target of L-ASNase in PD treatment, and the neuroprotective effect of L-ASNase was reduced after glutamine supplementation. Conclusions: Our study demonstrated for the first time that L-ASNase had a neuroprotective effect on a cell model of PD through a moderate deprivation of glutamine, which induced autophagic activation and mitochondrial fusion. Therefore, we demonstrated that L-ASNase could be a promising and effective drug for PD treatment.

Seed development plays an important role during the life cycle of plants. Linseed flax is an oil crop and the seed is a key organ for fatty acids synthesis and storage. So it is important to understand the molecular mechanism of fatty acid biosynthesis during seed development. In this study, four small RNA libraries from early seeds at 5, 10, 20 and 30 days after flowering (DAF) were constructed and used for high-throughput sequencing to identify microRNAs (miRNAs). A total of 235 miRNAs including 114 known conserved miRNAs and 121 novel miRNAs were identified. The expression patterns of these miRNAs in the four libraries were investigated by bioinformatics and quantitative real-time polymerase chain reaction (qPCR) analysis. It was found that several miRNAs, including Lus-miRNA156a was significantly correlated with seed development process. In order to confirm the actual biological function of Lus-miRNA156a, over-expression vector was constructed and transformed to Arabidopsis. The phenotypes of homozygous transgenic lines showed decreasing of oil content and most of the fatty acid content in seeds as well as late flowering time. The results provided a clue that miRNA156a participating the fatty acid biosynthesis pathway and the detailed molecular mechanism of how it regulates the pathway needs to be further investigated.

ABSTRACT

BACKGROUND: Current management of patients with atrial fibrillation (AF) is limited by low detection of AF, non-adherence to guidelines, and lack of consideration of patients' preferences, thus highlighting the need for a more holistic and integrated approach to AF management. OBJECTIVE: The objective of this study was to determine whether a mobile health (mHealth) technology-supported AF integrated management strategy would reduce AF-related adverse events, compared with usual care. METHODS: This is a cluster randomized trial of patients with AF older than 18 years of age who were enrolled in 40 cities in China. Recruitment began on June 1, 2018 and follow-up ended on August 16, 2019. Patients with AF were randomized to receive usual care, or integrated care based on a mobile AF Application (mAFA) incorporating the ABC (Atrial Fibrillation Better Care) Pathway: A, Avoid stroke; B, Better symptom management; and C, Cardiovascular and other comorbidity risk reduction. The primary composite outcome was a composite of stroke/thromboembolism, all-cause death, and rehospitalization. Rehospitalization alone was a secondary outcome. Cardiovascular events were assessed using Cox proportional hazard modeling after adjusting for baseline risk. RESULTS: There were 1,646 patients allocated to mAFA intervention (mean age, 67.0 years; 38.0% female) with mean follow-up of 262 days, whereas 1,678 patients were allocated to usual care (mean age, 70.0 years; 38.0% female) with mean follow-up of 291 days. Rates of the composite outcome of 'ischemic stroke/systemic thromboembolism, death, and rehospitalization' were lower with the mAFA intervention compared with usual care (1.9% vs. 6.0%; hazard ratio [HR]: 0.39; 95% confidence interval [CI]: 0.22 to 0.67; p < 0.001). Rates of rehospitalization were lower with the mAFA intervention (1.2% vs. 4.5%; HR: 0.32; 95% CI: 0.17 to 0.60; p < 0.001). Subgroup analyses by sex, age, AF type, risk score, and comorbidities demonstrated consistently lower HRs for the composite outcome for patients receiving the mAFA intervention compared with usual care (all p < 0.05). CONCLUSIONS: An integrated care approach to holistic AF care, supported by mHealth technology, reduces the risks of rehospitalization and clinical adverse events. 'Friar' plum (Prunus salicina Lindl.) fruit were transferred to shelf life (25 °C) following different storage periods at low (0 °C) or intermediate (5 °C) temperature. The earliest flesh reddening appeared in plums during shelf life removed after 28 d at 0 °C and 14 d at 5 °C, respectively, in comparison with turning yellow in plums remained at 25 °C immediately after harvest. The flesh reddening developed rapidly thereafter. Anthocyanins, in particular, cyanidin 3-O-glucoside, significantly accumulated in the reddening tissue, and activities of enzymes associated with the phenylpropanoid pathway were considerably activated after the removal. The removal elicited extremely high ethylene production in plums, which might mediate the activation of the anthocyanin biosynthesis in response to cold stress signal. The results provided a potential approach for postharvest regulation of flesh color and accumulation of bioactive substances in plums, which could lead to attractive attributes and health-promoting effects on consumers.

Cistanches Herba is a popular Traditional Chinese Medicine and functional food that is used to treat kidney yang deficiency (KYD) in China. In this study, we investigated the effects of different drying methods for Cistanches Herba on kidney invigoration and yang strengthening. We established a mouse model of KYD by intraperitoneal injection of hydrocortisone for 8 days. We dried slices of Cistanches Herba in the sun, in the shade, in a microwave, or in an oven at 40°C, 60°C, 80°C or 100°C, then prepared and administered extracts to the mice by gastric gavage. We measured and evaluated the echinacoside (ECH) and acteoside (ACT) contents of the extracts, as well as the mice's body weight; testicular, epididymal, hepatic, and renal coefficients; and semen quality. All the Cistanches Herba extracts, obtained using different drying techniques, improved symptoms of KYD diagnosis in mice. Among them, treatments with Cistanches Herba dried in a microwave and dried in an oven at 100°C had the best therapeutic effects. Our results suggested that the higher the total content of ECH and ACT in Cistanches Herba extracts, the better the effects of kidney invigoration and yang strengthening. In addition, shorter drying times at higher temperatures lead to the highest recoveries of active components, and Cistanche dried in the sun at a medium-high temperature can improve sperm quality in mice.
