ABSTRACT

Insomnia is a common sleep disorder around the world, which is harmful to people's health, daily life, and work. The paraventricular thalamus (PVT) plays an essential role in the sleep-wake transition. However, high temporal-spatial resolution microdevice technology is lacking for accurate detection and regulation of deep brain nuclei. The means for analyzing sleep-wake mechanisms and treating sleep disorders are limited. To detect the relationship between the PVT and insomnia, we designed and fabricated a special microelectrode array (MEA) to record electrophysiological signals of the PVT for insomnia and control rats. Platinum nanoparticles (PtNPs) were modified onto an MEA, which caused the impedance to decrease and improved the signal-to-noise ratio. We established the model of insomnia in rats and analyzed and compared the neural signals in detail before and after insomnia. In insomnia, the spike firing rate was increased from 5.48 ± 0.28 spike/s to 7.39 ± 0.65 spike/s, and the power of local field potential (LFP) decreased in the delta frequency band and increased in the beta frequency band. Furthermore, the synchronicity between PVT neurons declined, and burst-like firing was observed. Our study found neurons of the PVT were more activated in the insomnia state than in the control state. It also provided an effective MEA to detect the deep brain signals at the cellular level, which conformed with macroscopical LFP and insomnia symptoms. These results laid the foundation for studying PVT and the sleep-wake mechanism and were also helpful for treating sleep disorders.

ABSTRACT

Acupoint specificity for diseases has consistently been the focus of acupuncture research owing to its excellent prospects for clinical diagnosis and treatment. However, the specificity of cardiovascular and sleep functions in terms of electrical signals at acupoints remains unclear. In this study, five volunteers were recruited and their electrophysiological signals of GV20 (baihui), RN17 (danzhong), PC6 (neiguan), and SP6 (sanyinjiao) and the corresponding sham points, Pittsburgh sleep quality index, blood pressure, and echocardiography were monitored over four periods of 90-day head-down bed rest (HDBR). The results demonstrated that the power and characteristic amplitude of the acupoints were more significant than those of the sham points under normal conditions. And along with the altered physiological condition of the body after HDBR, the differential signal characteristic amplitude (DSCA) and the power of the acupoints were decreased to a larger extent than those of the sham points. In addition, the difference between the power of acupuncture and sham points was also reduced. During the recovery period, except for GV20, the power and DSCA of other acupoints did not return to normal. In terms of DSCA, GV20 is related to human sleep function and other acupoints are related to cardiovascular function. The above results show that the electrophysiological signals of acupoints are disease-specific and more accurately reflect the changes of physiological homeostasis. The research conduces to the development of acupuncture-based disease diagnosis and treatment integrated methods, and the realization of the portable and accurate diagnosis and regulation of diseases in space medicine.

ABSTRACT

BACKGROUND: Sonchus oleraceus is a large and widespread plant in the world. It is edible to humans as a leaf vegetable and is also used as a folklore medicinal herb in the treatment of infections and inflammatory disease, but limited research on its chemical constituents has been done. OBJECTIVE: To isolate and identify the bioactive ingredients from S. oleraceus. METHODS: 20kg of S. oleraceus was extracted twice with 75% alcohol. The concentrated extract was suspended in H2O and partitioned with petroleum ether, dichloromethane, ethyl acetate and n-butanol, respectively. The ethyl acetate phase was subjected to repeated normal chromatography on a silica gel column chromatography and eluted with a gradient of CH2Cl2-MeOH to give 12 crude fractions. Fraction 6 was subjected to ODS silica gel column chromatography, Sephadex LH-20 and HPLC to yield 1 and 2. Cell viability of 1 and 2 on A549, H292 and Caco2 cell lines were assayed by MTT method. Apoptosis analysis and apoptosis related proteins were detected subsequently. RESULTS: Two new sesquiterpenes were isolated from S. oleraceus and identified by NMR spectra and HR-ESIMS. 1 selectively suppressed the viability of A549 and H292 cells with IC50 values of 14.2, and 19.5µM respectively, while possessing no cytotoxicity against Caco2 cells (IC50 > 100µM). 2 did not exhibit cytotoxicity against A549, H292 and Caco2 cells (IC50 > 100µM). 1 significantly decreased the density of live cells and could cause cell apoptosis at 10 and 20µM in a dose-dependent manner. After treatment of 1 for 24h, the level of cleaved caspase-3 was increased accompanied by the reduction in procaspase-3 expression, and the downregulation of Bcl-2 was associated with the enhancement of Bax expression. 1 could lead to the up-regulation of cytochrome c and activation of caspase-9. CONCLUSION: 1 and 2 are new sesquiterpenes from S. oleraceus. 1 could induce apoptosis in A549 and H292 cells through Bax/caspase-9 pathway.

ABSTRACT

Two endophytic strains SS01 and SS02 with the potential for producing steroidal saponins were isolated from the underground stems of Paris polyphylla var. chinensis Franch. The TLC comparison indicated that there are 3 sports with similar R(f) between the metabolites of SS01 and the saponins of Paris polyphylla var. chinensis Franch. And there are 2 sports with similar R(f) between the metabolites of SS02 and the saponins of Paris polyphylla var. chinensis The and that biochemical SS01 Franch. characteristics of morphology, physiological belonged showed to Enterobacteriaceae and SS02 belonged to Bacillus sp.. The 16S rDNA of SS01 and SS02 were PCR and sequenced. The accessions of GenBank are AY842143 and AY842144, respectively. The two 16S rDNA phylogenetic trees were constructed by comparing with the published 16S rDNA sequences of the relative bacteria species. In the first phylogenetic tree SS01 and Cedecea davisae DSM 4568 was the closest relative with 98.9% sequence similarity, and in the second phylogenetic tree SS02 and Paenibacillus daejeonensis was the closest relative with 97.7% sequence similarity. According to the phylogenetic analysis they were identified as Cedecea davisae SS01 and Paenibacillus daejeonensis SS02, respectively.
