ABSTRACT

Objective:To observe the changes of inflammatory damage in the brain of rats after focal cerebral ischemia-reperfusion, and explore the effect of the initiation of IκB kinases β (IKKβ), which is the key protein of activating nuclear factor (NF)-kappa B signaling pathway in inflammatory response, and the mechanism of electroacupuncture inhibiting inflammatory damage. Methods:A total of 240 male Sprague-Dawley rats were randomly divided into sham group, ischemia-reperfusion group, electroacupuncture group, IKKβ silencing group, IKKβ overexpression group and IKKβ overexpression + electroacupuncture group, each group was further divided into six hours, twelve hours, 24 hours, 48 hours and 72 hours subgroups. The right middle cerebral artery occlusion reperfusion model was established by modified thread embolization. The IKKβ gene was intervened by gene silencing and gene overexpression technology. Results:Compared with the model group, the neurological function score increased (P < 0.05), the cerebral infarction volume decreased (P < 0.05), the activation of NF-κB p65 was inhibited, and the content of proinflammatory factors decreased (P < 0.05) in IKKβ silencing group. Compared with IKKβ silencing group, the above results were significantly worse in IKKβ overexpression group (P < 0.05), and microglia in cerebral ischemic cortex were significantly activated. The activation of microglia and activation of IKKβ were significantly inhibited in IKKβ overexpression + electroacupuncture group. Conclusion:IKKβ gene silencing could inhibit the inflammatory response of cerebral ischemic cortex mediated by NF-κB signaling pathway, and over-expression of IKKβ could lead to severe inflammatory damage in ischemic cortex. Electroacupuncture could inhibit the inflammatory damage after focal cerebral ischemia-reperfusion by regulating the activity of IKKβ.

ABSTRACT

Objective To observe the effect of electroacupuncture(EA)at Baihui(GV20)and Siguan(Hegu/LI4 and Taichong/LR3)of affected side on expression of Tax1-binding protein 1(TAX1BP1)in cerebral cortex in focal cerebral ischemia-reperfusion rats,so as to in-vestigate its protective mechanism in inhibiting nuclear factor kappa B(NF-κB)signaling pathway and promoting neurobehavioral recovery. Methods A total of 105 Sprague-Dawley rats were randomly assigned into sham group,model group and EA group.Each group was ran-domly assigned into reperfusion six hours,twelve hours,24 hours,48 hours,72 hours groups after two hours of ischemia.The model was es-tablished by right middle cerebral artery occlusion and reperfusion.EA group received electroacupuncture at Baihui and left Siguan(Hegu and Taichong)acupoints.Neurobehavioral evaluation,TAX1BP1 protein expression,TAX1BP1 positive cell count,zink finger protein A20 expression, and nuclear NF-κB p65 protein expression were tested in each group. Results There was no neurological deficit in the sham group. Compared with the model group, the neurological scores at 48 hours, 72 hours after reperfusion decreased in EA group (P<0.05). Compared with the control group,the TAX1BP1 expression at twelve hours,24 hours and 48 hours after reperfusion increased in the model group(P<0.05),and further increased at twelve hours,24 hours,48 hours and 72 hours after reperfusion in EA group(P<0.05),and peaked at 24 hours after reperfusion.Compared with the sham group,the expression of A20 and NF-κB p65,and the number of TAX1BP1 positive cells increased in the model group(P<0.05),and the expression of A20 and the number of TAX1BP1 positive cells further increased,(P<0.05)and the expression of NF-κB p65 decreased in EA group(P<0.05)at 24 hours after reperfusion.Immunofluorescence labeling indicat-ed that TAX1BP1 protein primarily expressed in the cytoplasm,TAX1BP1 protein and A20 protein co-expressed in the cytoplasm.Immuno-histochemistry showed indicated that NF-κB p65 mainly expressed in the nucleus in the model groupr,and mainly expressed in the cyto-plasm in the EA group.Conclusion Electroacupuncture could significantly inhibit neuronal NF-κB signaling pathway and promote neurobe-havioral recovery in focal cerebral ischemia-reperfusion,which may related with up-regulating TAX1BP1 protein expression.

ABSTRACT

Objective To observe the effect of electroacupuncture (EA) at Baihui (GV20), Dazhui (GV14) and Shenshu (BL23) acu-points on cognitive function and the synapse of neurons in hippocampal CA1 in SAMP8 mice,to explore the mechanism of EA in the treat-ment of Alzheimer's disease(AD).Methods A total of 24 seven-month-old SAMP8 mice were randomly divided into model group(n=12) and EA group (n=12), and the same age SAMR1 mice were as control group (n=12).The EA group accepted EA at Baihui, Dazhui and Shenshu for 30 days.They were assessed with Morris maze test.The expression of synaptophysin(SYN)and postsynaptic density protein 95(PSD95)in hippocampal CA1 region were detected with immunohistochemistry.The morphology and density of synapse in hippocampal CA1 region was observed with transmission electron microscopy.Results Compared with the model group,the latency of Morris maze de-creased in EA group(P<0.05),the time staying in the quadrant of the platform increased(P<0.05),as well as the number passing the origi-nal platform(P<0.05),with the more expression of SYN and PSD95 in hippocampal CA1 region(P<0.001),and more and completed syn-apse.Conclusion EA can improve the learning and memory ability of SAMP8 mice by increasing the expression of SYN and PSD95 to pro-tect the ultrastructure of synapses in hippocampal CA1 region.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect and safety of the complementary use of the extract of Artemisia annua L. (EAA) on treating active rheumatoid arthritis (RA).</p><p><b>METHODS</b>A randomized controlled clinical trial was performed. All the 159 participates with active RA were randomly assigned to the control group (80 cases) and EAA group (79 cases) using concealed random allocation method. In the control group, patients were medicated with leflflunomide and methotrexate for 48 weeks; and patients in the EAA group were administrated with leflflunomide, methotrexate plus EAA (30 g/d). At the time points of 0, 12, 24 and 48 weeks, the clinical outcome measures, including objective pain score, tenderness score, number of painful joints, number of swollen joints, health assessment questionnaire (HAQ) score for quality of life, levels of serum rheumatoid factor (RF), anti-cyclic citrullinated protein antibodies (CCP-Ab), erythrocyte sedimentation rate (ESR), C reactive protein (CRP), visual analogue score for pain (VAS), and the overall effificacy were detected and recorded.</p><p><b>RESULTS</b>The objective pain score, number of painful joints and ESR at 12 weeks, tenderness score and HAQ at 24 weeks, and the tenderness score, number of painfull joints, number of swollen joints, HAQ, CRP, RF and CCP-Ab at 48 weeks were signifificantly improved in the EAA group compared with the control group (P<0.01 or P<0.05). At 24 and 48 weeks, the overall effificacy of the EAA group was signifificantly higher than the control group (P<0.01). There were signifificantly higher withdrawal rate of corticosteroids within 12 weeks post-treatment and lower incidence rate of adverse effects in the EAA group compared with the control group (P<0.01 or P<0.05).</p><p><b>CONCLUSION</b>EAA plus methotrexate and leflflunomide were more effective and safer than the routine use of methotrexate and leflflunomide in the treatment of active RA.</p>

ABSTRACT

ObjectiveBy studying acupuncture at Neiguan (PC6), Shuigou (GV26), and Baihui (GV20) in septic shock patients, to observe the effect of acupuncture on T cell subsets, vital signs, white blood cell (WBC) count, and procalcitonin (PCT) level in septicshock patients.MethodSixty patients were randomized into a treatment group and a control group. The control group was intervened by symptomatic managements including supplementing blood volume, anti-inflammation, nutrition support, and organ protection, and the treatment group was by acupuncture at Neiguan, Shuigou, and Baihui in addition to the intervention given to the control group. The T cell subsets contents were detected before and after intervention, and serum PCT level, APACHEⅡscore, vital signs, and WBC count were determined on day 1, 3, 5, and 7.ResultIn the treatment group, T cell subsets including CD3﹢, CD4﹢, and CD4﹢/CD8﹢increased significantly after intervention (P<0.01,P<0.05), while CD8﹢dropped significantly afterintervention (P<0.05), and the changes in the treatment group were all markedly superior to that in the control group (P<0.05). The decrease of PCT level in the treatment group was more significant than that in the control group after treatment, the PCTlevel dropped significantly on the 5th day in the treatment group compared to that before treatment, and the inter-group difference was statistically significant after the 3rd day (P<0.01); the APACHEⅡscore in the treatment group was different from that in the control group after 3 d treatment, and the score was significantly changed in the treatment group after 5 d treatment (P<0.05); the body temperature, heart rate, inspiration, WBC count all showed decrease or improvement, and the improvements of vitalsigns in the treatment group were more significant than that in the control group (P<0.01,P<0.05).ConclusionAcupuncture at Baihui, Shuigou, and Neiguan can improve the immune function of septic shock patients, and its treatment effect is confirmed.

ABSTRACT

Objective To study the clinical effects of addition treatment of liver-kidney yin deficiency and postmenopausal osteoporosis with Liuwei Dihuang pill.Methods 120 patients suffering from postmenopausal osteopo-rosis who were determined to be subject to liver -kidney Yin deficiency by doctors of traditional Chinese medicine when they received treatment at the outpatient clinic of traditional Chinese medicine department of our hospital in the period from February 2010 to june 2013 were divided into three groups:group A,B and C,with the random number table.For group A:provided the honeyed pills made up by supplemented six -ingredient rehmannia pill,and taken two pills per day continuously for six months;For group B:provided 5mg of zoledronic acid injection for intravenous drip infusion;For group C:provided traditional Chinese medicine honeyed pills and zoledronic acid for treatment at the same time;all of the three groups take Calcium carbonate D3 at the same time;The three groups of cases until the 12th month were observed and followed -up.The clinical efficacy of three groups were compared.Results A,C group 3 months after treatment with estradiol levels compared with before treatment,the difference was statistically significant (t =2.54,P =0.05,t =2.07,P =0.06).A group 3 months after treatment,the VAS scores at 6 months respectively (3.75 ±1.32),(2.06 ±1.53)points before treatment,and the differences were statistically significant(t =10.18, P =0.00,t =9.26,P =0.00).A group 3 months after treatment,the SF -36 scores at 6 months were (90.82 ± 12.38),(105.82 ±9.06)% respectively,compared with that before treatment,the differences were statistically significant(t =8.40,P =0.00,t =5.21,P =0.00).A group 6 months after treatment of lumbar 1 -4 bone density-were (0.85 ±0.07)g/m2 ,and before treatment[(0.76 ±0.07)g/m2 ]comparison,the difference was statistically significant(t =7.56,P =0.00);A group 6 months after treatment of Wards bone density compared with before treatment,the difference was statistically significant(t =8.07,P =0.00).C group 6 months after treatment of lumbar 1 -4 bone density(0.91 ±0.07)g/m2 ,and B Group[(0.88 ±0.05)g/m2 ]comparison,the difference was statistical-ly significant(t =27.19,P =0.00);C group after treatment,the bone mineral density of Wards area for 6 months were (0.62 ±0.04)g/m2 .The difference was statistically significant (t =26.22,P =0.00),compared with the B group. The TCMsyndromes of TCMin the 6 months after treatment in A group were (5.42 ±2.13),and the difference was statistically significant (t =13.29,P =0.00),compared with the B Group (10.75 ±4.08).(t =13.29,P =0.00). Compared with B group,the difference was statistically significant(t =26.22,P =0.00).A group after the treatment of TCMsyndrome score was 6 months(5.42 ±2.13),and B Group[(10.75 ±4.08)],the difference was statistically significant(t =13.29,P =0.00).Conclusion The addition of Liuwei Dihuang pill can improve BMD,alleviate the ostealgia and improve life quality of patients suffering from postmenopausal osteoporosis,and has no obvious adverse effects.It can enhance curative effects through using with diphosphonate.

ABSTRACT

Objective To explore the effect and mechanism of electroacupuncture on eNOS in mobilizing endothe -lial progenitor cells (EPCs) in rat bone marrow and peripheral blood to promote revascularization in focal cerebral ischemi -a/reperfusion rat.Methods A total of 100 healthy male adult Sprague-Dawley (SD) rats were randomly divided into nor-mal group ( N) , model group ( I/R) , electroacupuncture group ( I/RE) and I/RE plus L-NAME ( A specific antagonist of eNOS) group (I/REL), and were further divided into 1 d, 2 d and 7 d subgroups after reperfusion, 10 rats in each group, in addition to the N group .The rats received filament occlusion of the right middle cerebral artery for 1.5 hours followed by reperfusion .“Baihui” ( GV 20)/“Siguan” ( Hegu LI 4/Taichong LR 3) were selected as acupucture points .Flow cytome-ter was used to detect the percentage of EPCs in bone marrow and peripheral blood .The expression of VEGFR2 mRNA was tested by fluorescence quantitative PCR .Immunohistochemical staining was used to detect VEGFR 2 +cells and to stain the CD34 +microvessels.Results Compared with the I/R group, there was a significant up-regulation of the percentage of EPCs in bone marrow and peripheral blood by electroacupuncture (P<0.01,P<0.05), but decreased by the inhibitor of eNOS (P<0.01).Compared with the I/R group, the VEGFR2 +cells, expression of VEGFR2 mRNA and CD34 +mi-crovessels were significantly increased in the I/RE group ( P<0.01 ) , but decreased in the I/REL group ( P<0.01 ) . Conclusions Electroacupuncture can effectively mobilize EPCs to promote the revascularization in focal cerebral ischemia /reperfusion rat .This effect is attenuated by inhibitor of eNOS , suggesting that the activation of eNOS mediated by electroa-cupuncture may be related to mobilizing EPCs to promote the revascularization .

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of bitter-cold herbs easing dampness method (BCHEDM) plus Sanhuang Yilong Decoction (SYD) combined with methotrexate (MTX) on expression levels of interleukin-1 (IL-1), IL-6, and IL-17 in rheumatoid arthritis (RA) patients of accumulated dampness-heat syndrome (ADHS).</p><p><b>METHODS</b>From January 2011 to January 2013 recruited were 90 RA inpatients of ADHS at Department of Integrative Medicine on Rheumatoid Disease, General Hospital of Chengdu Military Region. They were assigned to the treatment group (45 cases) and the control group (45 cases) according to the random digit table produced by SPSS 11.5 Software. Patients in the treatment group were treated by heavy bitter-cold herbs plus SYD combined with MTX, while those in the control group were treated by MTX alone. Expressional levels of IL-1, IL-6, and IL-17 in serum were detected by enzyme linked immunosorbent assay (ELISA) before treatment, at week 2 and 4 after treatment. Erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), and disease activity score in 28 joints (DAS28) were detected as well.</p><p><b>RESULTS</b>After two or four weeks of treatment, ESR, CRP, and DAS28 decreased more in the treatment group than in the control group with statistical difference (P < 0.05, P < 0.01). After four weeks of treatment, IL-1, IL-6, IL-17, ESR, CRP, and DAS28 in the treatment group were all lower than before treatment and those of the control group at corresponding time points with statistical difference (P < 0.01).</p><p><b>CONCLUSION</b>SYD combined MTX could play roles of improving inflammatory indices within 2 weeks, and inhibiting the expression of IL-1, IL-6, and IL-17 within 4 weeks.</p>

ABSTRACT

@# Objective To observe the effect of electroacupuncture at Hegu (LI04) on capillary density and expression of CD34 after focal cerebral ischemia/reperfusion in rats. Methods 90 rats were randomly divided into normal group (n=6), model group (n=42) and electroacupuncture group (n=42). A rat model of focal cerebral ischemia/reperfusion was made by filament occlusion in the model group and the electroacupuncture group. And the electroacupuncture group received electroacupuncture. They were observed 2 h, 12 h, 24 h, 3 d, 7 d, 14 d, 21 d after reperfusion 1 h followed ischemia. HE staining was used to observe and calculate the infarct volume and capillary density, and immunohistology was used to detect the expression of CD34 after neurologic symptoms rating. Results Compared with the model group, the neurologic symptoms score significantly decreased 2 h after reperfusion in the electroacupuncture group (P<0.01), and still decreased 21 d after reperfusion (P<0.05). The infarct volume significantly was smaller in the electroacupuncture group than in the model group (P<0.01). The capillary density was higher 2 h, 24 h, 3 d, 7 d, 14 d after reperfusion in the electroacupuncture group than in the model group (P<0.05). Compared with the model group, the expression of CD34 significantly increased 3 d after reperfusion in the electroacupuncture group (P<0.01),and it reached a peak at 7 d after reperfusion (P<0.01), and still higher at 14 d. Conclusion Electroacupuncture can promote the expression of CD34 and the capillary density, and stimulate the angiogenesis after focal ischemia/reperfusion.

ABSTRACT

Objective To investigate the stromal cell derived factor 1 α (SDF-1 α) and chemokine CXC motif receptor 4 (CXCR4) axis using acupoint electroacupuncture (EA) on the brains of rats after focal cerebral ischemia and reperfusion.Methods Ninety-eight Sprague-Dawley (SD) rats were randomly divided into a control group (8 rats),a model group (50 rats),and an EA group (40 rats).The animal model of focal brain ischemia-reperfusion was made with all the rats in the model group and EA group by using the filament occlusion technique.The model and EA groups were subdivided into 5 subgroups according to the sampling time points on the 1 st,3rd,7th,14th or 21st day after ischemia-reperfusion.The EA was administered bilaterally to the rat analog of the Hegu point (LI 4) in the EA group.The model and control groups received no special treatment.Immunohistochemical methods were employed to detect CXCR4-positive cells in the model and EA groups.The expressions of SDF-1α and CXCR4 mRNA were detected with RT-PCR methods in the 3rd,7th and 14th day subgroups.Results With the prolongation of reperfusion,SDF-1α mRNA expression in the model group had a single peak-like increase in the ischemic area of the cerebral cortex.It had increased significantly by the 3rd day,reached its peak value at the 7th day and then decreased gradually.SDF-1α mRNA expression in the EA group behaved similarly,but SDF-1 mRNA expression was significantly higher in the EA group than in the other two.In the model and EA groups CXCR4 mRNA relative values were higher at the 7th and 14th day than at day 3,and the expression at day 14 was significantly greater than at day 7.CXCR4 mRNA values in the EA group were significantly higher than in the model group at each time point.The expression of CXCR4-positive cells began to increase on the 1 st day in the model group,reached its peak value at the 7th day,then decreased by day 14,but it was still strongly expressed highly at the 21st day.Compared with the model group,the expression in the EA group showed the same pattern,but the number of CXCR4-positive cells in EA group was significantly higher.Conclusion Point EA can activate the SDF-1α and CXCR4 axis to promote angiogenesis after focal cerebral ischemia and reperfusion,at least in rats.

Objective To explore the effect of electroacupuncture on CD 34 +VEGFR2 +endothelial progenitor cell (EPC)-derived vessels and stromal cell-derived factor-1α(SDF-1α)/CXCR4, and study its mechanism of promoting an-giogenesis in hippocampus after focal cerebral ischemia /reperfusion .Methods A total of 180 healthy male adult Sprague Dawley (SD) rats were randomly divided into sham operation (sham) group, model (I/R) group, electroacupuncture (I/RE) group, I/RE plus AMD3100 (A specific antagonist of CXCR4) group (I/REA) and AMD3100 (I/RA) group. The rats received filament occlusion of the right middle cerebral artery for 2 hours followed by reperfusion .Electroacupunc-ture was applied to “Baihui” (GV20)/“Siguan” (Hegu LI 4/Taichong LR 3) acupoints for 30 min, once a day.The mR-NA expression of SDF-1αand CXCR4 were detected by reverse transcription-polymerase chain reaction ( RT-PCR) .Double immunofluorescence was used to stain CD 34 +VEGFR2 +EPC-derived vessels.Results Compared with the sham group, the mRNA expressions of SDF-1αand CXCR4 were significantly upregulated in I/R and I/RE group ( P<0.05 ) , but that in I/RE group was more significantly increased than I/R group(P<0.05).In addition, the mRNA expression of SDF-1αand CXCR4 were highly increased on day 1 in the I/REA group than that of I/RE group, but decreased than that of I/RE group on day 7 after reperfusion (P<0.01).CD34 +VEGFR2 +EPCs-derived vessels were obviously increased on 3d and 7d in the I/RE group compared with that of the I/R group, and significantly decreased on 7d in the I/REA group compared with that of the I/RE group ( P<0.01) .Conclusions Electroacupuncture can effectively promote an-giogenesis through upregulating the expression of SDF-1αand CXCR4 in rat ischemic hippocampus after focal cerebral is-chemia/reperfusion.

ABSTRACT

<p><b>OBJECTIVE</b>To probe the function of interleukin-17 (IL-17) in rheumatoid arthritis (RA) patients of accumulated dampness-heat obstruction in joints syndrome (ADOJS) by detecting levels of IL-17 in serum and the synovial fluid and analyzing its correlation with erythrocyte sedimentation rate (ESR) and C reactive protein (CRP).</p><p><b>METHODS</b>From January 2011 to January 2013, recruited were 90 RA inpatients of ADOJS at Department of Integrative Medical Rheumatism, General Hospital of Chengdu Military Region, of which 28 patients had knee joint effusion. Besides, 30 healthy volunteers who received physical examination at our hospital were recruited as the normal control group, and 30 patients with osteoarthritis (OA) who had knee joint effusion were recruited as the synovial fluid control group. The expression levels of IL-17 in serum and the synovial fluid were detected by enzyme linked immunosorbent assay (ELISA), and contents of ESR and CRP were detected in RA patients. Then correlation analyses were performed between levels of IL-17 and contents of ESR and CRP.</p><p><b>RESULTS</b>Compared with the normal serum control group, the expression levels of IL-17 in serum of RA patients significantly increased (P < 0.05). Compared with the serum of RA patients and the synovial fluid of OA patients, the expression levels of IL-17 in the synovial fluid of RA patients significantly increased (P < 0.05). The expression levels of IL-17 in serum of RA patients were not correlated with ESR or CRP (r = 0.092, -0.082; P > 0.05), and the expressional levels of IL-17 in the synovial fluid of RA patients were not correlated with ESR or CRP (r = 0.113, -0.034; P > 0.05).</p><p><b>CONCLUSIONS</b>IL-17 was the main effector cytokine of Th17 cells. The expressional levels of IL-17 significantly increased in serum and the synovial fluid of RA patients of ADOJS, but with no correlation to ESR or CRP. It indicated that IL-17 participated in the occurrence and development of RA. Concrete mechanisms needed to be further proved in larger samples.</p>

ABSTRACT

To study the impact of five different origin processing methods, namely natural drying, drying in baking shop, drying by microwave heating, drying in drum and drying with sulphur fumigation, on the quality of Lonicerae Japonicae Flos from Donghai cultivation base in Jiangsu Province, with the contents of chlorogenic acid and galuteolin and the similarity in HPLC fingerprints as the evaluation indicators. The results showed that different origin processing methods had significant impact on the content of chlorogenic acid and the similarity in HPLC fingerprints, but with no significant difference on the content of galuteolin. By means of drying by microwave heating and drying in drum, the samples showed higher contents of chlorogenic acid, respectively 3.67% and 3.39%. The similarities of HPLC fingerprints were 0.815 and 0.793, respectively. By means of the drying in baking shop and the drying with sulphur fumigation, the contents of chlorogenic acid in the samples were 2. 87% and 2. 53% , respectively. The similarities of HPLC fingerprints were 0.964 and 0.765, respectively. The lowest content of chlorogenic acid in naturally dried samples was 1.92%. The similarity of HPLC fingerprints was 0.940. According to the findings as well as the internal control standards for Lonicerae Japonicae Flos herbs of Jiangsu Kanion Pharmaceutical Co. , Ltd. , the optimum processing method for Lonicerae Japonicae Flos from Donghai cultivation base was the drying in baking shop. This study provided a theoretical basis for determining the processing method for Lonicerae Japonicae Flos from Donghai cultivation base of Jiangsu Province.

ABSTRACT

Objective To observe the effect of electroacupuncture at Hegu (LI04) on capillary density and expression of CD34 after focal cerebral ischemia/reperfusion in rats. Methods 90 rats were randomly divided into normal group (n=6), model group (n=42) and electroacu-puncture group (n=42). A rat model of focal cerebral ischemia/reperfusion was made by filament occlusion in the model group and the elec-troacupuncture group. And the electroacupuncture group received electroacupuncture. They were observed 2 h, 12 h, 24 h, 3 d, 7 d, 14 d, 21 d after reperfusion 1 h followed ischemia. HE staining was used to observe and calculate the infarct volume and capillary density, and immu-nohistology was used to detect the expression of CD34 after neurologic symptoms rating. Results Compared with the model group, the neuro-logic symptoms score significantly decreased 2 h after reperfusion in the electroacupuncture group (P<0.01), and still decreased 21 d after re-perfusion (P<0.05). The infarct volume significantly was smaller in the electroacupuncture group than in the model group (P<0.01). The cap-illary density was higher 2 h, 24 h, 3 d, 7 d, 14 d after reperfusion in the electroacupuncture group than in the model group (P<0.05). Com-pared with the model group, the expression of CD34 significantly increased 3 d after reperfusion in the electroacupuncture group (P<0.01), and it reached a peak at 7 d after reperfusion (P<0.01), and still higher at 14 d. Conclusion Electroacupuncture can promote the expression of CD34 and the capillary density, and stimulate the angiogenesis after focal ischemia/reperfusion.

ABSTRACT

OBJECTIVE: To study the effects of Compound Ganshao Paste (CGP) on the levels of sex hormones and hepatic transaminases in polycystic ovarian syndrome (PCOS) rat model. METHODS: The PCOS rat model was established using Kafali modeling method. Rats were randomly divided into five groups, i. e., the normal control group, the PCOS model group, the Diane-35 group, the Compound Ganshao Caspule group, and the CGP group. The levels of sex hormones were detected using radioimmunoassay. The levels of hepatic transferases were detected using method of enzyme coupling ratio. RESULTS: Compared with the normal control group, the serum levels of luteinizing hormone (LH), testosterone (T), follicle stimulating hormone (FSH), estradiol (E2), and progesterone (P) decreased, showing statistical difference (P<0.05). Compared with the model group, the serum levels of LH, T, and FSH decreased, while serum E2 and P increased in the three medication groups, showing statistical difference (P<0.05). There was insignificant difference in the serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) among the normal control group, the model group, and the CGP group (P>0.05). Compared with the Diane-35 group and the Compound Ganshao Capsule group, the levels of hepatic transaminases of the CGP group was lower with statistical difference (P<0.05). CONCLUSIONS: CGP could improve the ovarian functions through adjusting the endocrine functions of PCOS model rats, thus stimulating the follicular development and ovulation. CGP did not add the hepatic burden.

ABSTRACT

Objective To investigate the mechanism by which electro-acupuncture (EA) promotes revascularization in the brain after focal cerebral ischemia and reperfusion.Methods The Sprague-Dawley rat model of focal cerebral ischemia was made by filament occlusion. The rats were randomly divided into a control group, a model group, and an EA group. The model and EA groups were each divided into 5 subgroups receiving reperfusion 1, 3,7, 14 or 21 days after ischemia. EA was given at the bilateral Hegn point (LI 4) in the EA group. The expression of stromal cell-derived factor-1α(SDF-1α) mRNA was detected using a RT-PCR in the 3, 7 and 14 day subgroups.The immunohistochemical method was employed to detect the expression of SDF-1α protein. Results Compared with the control group, expression of SDF-1α protein increased significantly in the model and EA groups. Compared with the model group, the expression of SDF-1α mRNA increased significantly in the 3, 7 and 14 day subgroups.SDF-1α protein expression and microvessel count increased slightly but not significantly in the 1d subgroup, but the increases were significant in the 3, 7, 14 and 21 day subgroups.Conclusions EA may promote angiogenesis in an ischemic area of the cortex by increasing the expression of SDF-1αmRNA and its protein after focal cerebral ischemia and reperfusion.

ABSTRACT

<p><b>OBJECTIVE</b>To demonstrate the vasodilatation activity of the coumarin-containing Angelica dahurica var. formosana and to further analyze active components in the herb extracts.</p><p><b>METHODS</b>(1) The vasodilatation effects induced by different extracts (cyclohexane, ethyl acetate, acetone, methanol, 95 % ethanol and water) of Angelica dahurica var. formosana on mouse thoracic aorta pre-contracted with phenylephrine were investigated. (2) The amount of imperatorin and isoimperatorin in each extract was measured by high-performance liquid chromatography. (3) The vasodilatation effects of imperatorin and isoimperatorin on mouse thoracic aorta were compared using the same in vitro method. (4) The vasodilatation mechanism of imperatorin in the mouse thoracic aorta pre-contracted with phenylephrine was studied using the methods of denuded endothelium, NG-nitro-L-arginine methylester (L-NAME, a nitric oxide synthase inhibitor), and propranolol.</p><p><b>RESULTS</b>(1) The cyclohexane and ethyl acetate extracts of Angelica dahurica var. formosana decreased the maximal response of phenylephrine-induced mouse thoracic aorta contraction dose-dependently, with 50% inhibiting concentration (IC(50)) values of 35.3+/-12.4 mg/L and 40.5+/-12.0 mg/L, respectively. The vasodilatation effect of imperatorin and isoimperatorin was dose-dependent. (2) The cyclohexane extract, showing the strongest vasodilatation effect, possessed the highest contents of imperatorin (4.09%) and isoimperatorin (0.27%, w/w). There was a correlation between the vasodilatation activity and the contents of imperatorin and isoimperatorin in the extracts. (3) The vasodilatation effect of imperatorin was about 4-fold stronger than that of isoimperatorin. (4) The vasodilatation effect of imperatorin was signifificantly attenuated to 24.88%+/-4.06% in the denuded endothelium group compared with the intact endothelium group. And 1 mmol/L L-NAME reduced the imperatorin-induced vasorelaxation by 32.18 %+/-11.29 %.</p><p><b>CONCLUSIONS</b>The principal effective component of Angelica dahurica var. Formosana was found to be imperatorin. Imperatorin-induced vasodilatation is at least partially regulated by nitric oxide, and has no correlation to beta-receptor.</p>

ABSTRACT

To explore the mechanism of action of butylphalide (NBP) against the injury following oxygen glucose deprivation/reoxygenation (OGD/R) in rat cortical neurons, neurons of Wistar newborn rats were prepared by filtering through a mesh, centrifugation and trypsogen digestion. A simple, stable and reliable in vitro model of OGD/R of neurons was established. We studied the activation, the nuclear translocation of NF-kappaB p65 and the mRNA expression of iNOS affected by NBP in each group neuron by RT-PCR. NBP is proved to be able to add cellular vigor and decrease LDH release. The mRNA expression of iNOS in neurons after OGD 4 h/R 8 h decreased when treated with NBP. There is statistical difference between each concentration of NBP that it adds cellular vigor, decreases LDH release and expression of iNOS in neurons after OGD 4 h/R 8 h. There is also statistical difference between NBP (100 micromol x L(-1)) and PDTC (100 micromol x L(-1)). It is proved that NBP can protect neurons, block upregulation of iNOS mRNA, and restrain activation of NF-kappaB in neurons.

ABSTRACT

OBJECTIVE: To investigate the clinical efficacy of Tiaomian Huayu Decoction (THD) in treating patients with endometriosis. METHODS: Patients in the treated group were administered with THD, those in the control group were given provera. T-cell sub-population including CD3, CD4, CD8 and CD4/CD8 ratio, and natural killer (NK) cell in peripheral blood in the two groups before and after treatment were determined. RESULTS: Among the 66 patients in the treated group, the treatment results was cured in 10, significantly effective in 22, effective in 25 and ineffective in 9, the total effective rate was 86.4%, while of the 30 in the control group, the corresponding number was 2, 6, 12, 10 and 66.7%, which was significantly different from that in the treated group respectively (P<0.05). The levels of CD4 and CD4/CD8 ratio were higher, CD8 and NK were lower in the treated group than those in the normal group before treatment (P < 0.01), while these indexes were restored to some extent but still did not reach the normal level after treatment. All the above-mentioned indexes in the control group were insignificantly changed after treatment (P >0.05). CONCLUSION: THD has definite effect in treating endometriosis, and it can also regulate the immune function of patients.
