ABSTRACT

Selenium is commonly used as a supplement in the poultry diet and plays an important role in male fertility. However, the effect of selenium nanoparticles (Se-NPs) on exosome secretion associated with spermatozoa in the epididymis is largely unknown. H&E staining, Immunohistochemistry, Immunofluorescence and Western blot were performed to study the effect of Se-NPs on exosomes secretion associated with sperm maturation in epididymis. The results indicated that the Se-NPs showed a significant contribution to sperm concentration by light microscopy. It was observed that there was an increase in the spermatozoa concentration in the epididymis of the treated group as compared to the control group. Furthermore, exosome secretion, the expression of tumor susceptibility gene-101 (TSG-101) and cluster of differentiation (CD-63) proteins was identified by immunochemistry, immunofluorescence assay, and western blotting. After nano-selenium treatment, the exosome markers TSG-101 and CD-63 were strong positive immunoreactivity and immunosignaling in the lumen followed by epithelial lining of the epididymis. However weak positive immunoreactivity and immunosignaling were seen of TSG-101 and CD63 in the control group. In addition, highly significant protein expression of TSG-101 and CD63 in the treated group as compared to the control group was confirmed by western blotting. In conclusion, the above findings provide rich evidence about the Se-NPs play a dynamic role in exosome secretion that might be essential for sperm motility and maturation within epididymis.

ABSTRACT

The porous structure and hydrophilicity of coating shells affect the nutrient controlled-release performance of castor oil-based (CO) coated fertilizers. In order to solve these problems, in this study, the castor oil-based polyurethane (PCU) coating material was modified with liquefied starch polyol (LS) and siloxane, and a new coating material with cross-linked network structure and hydrophobic surface was synthesized, and used it to prepare the coated controlled-release urea (SSPCU). The results demonstrated that the cross-linked network formed by LS and CO improved the density and reduced the pores on the surface of the coating shells. The siloxane was grafted on the surface of coating shells to improve its hydrophobicity and thus delayed water entry. The nitrogen release experiment indicated that the synergistic effects of LS and siloxane improved the nitrogen controlled-release performance of bio-based coated fertilizers. Nutrient released longevity of SSPCU with 7 % coating percentage reached >63 days. Moreover, the nutrient release mechanism of coated fertilizer was further revealed by the analysis of the release kinetics analysis. Therefore, the results of this study provide a new idea and technical support for development of efficient and environment-friendly bio-based coated controlled-release fertilizers.

ABSTRACT

In this paper, a cationic photoinitiator (TAS) was used as a catalyst for the ring opening reaction of carboxymethyl chitosan (CMCS) and epoxidized soybean oil (ESO) under UV light to prepare CMCS-g-ESO conjugate, and the structure of the product was characterized by FT-IR, 1H NMR and GPC. Then, the spinosad-loaded microcapsules (SSD@CMCS-g-ESO) were prepared by ultrasonic self-assembly method. The results showed that TAS could catalyze the ring opening reaction of CMCS and ESO under UV-irradiation and the optimum reaction time was 1 h, with the molecular weight of 15,745. The average particle size of SSD@CMCS-g-ESO was about 2.16 µm, and the encapsulation efficiency (EE) and drug loading content (LC) of SSD@CMCS-g-ESO were 85.39 ± 2.05% and 20.17 ± 1.84%, respectively. In vitro release revealed that SSD@CMCS-g-ESO exhibited sustained-release and pH-responsive property, and the accumulative release in the buffer solution of pH = 6.5 and 7.4 was higher than in pH = 9.0. Furthermore, SSD@CMCS-g-ESO had a good antifungal properties against Fusarium oxysporum f. sp. cubense (Foc) compared with the unencapsulated SSD at the same drug dose. This work indicated that photo-chemical reactions could be used to prepare bio-based carrier materials to construct drugs delivery system for targeted treatment of fusarium wilt.

ABSTRACT

The EtOH extract of the roots and rhizomes of Clematis mandshurica afforded two new lignans clemomanshurinane C (1) and clemomanshurinane D (2), and three known compounds (3-5). Their structures were elucidated on the basis of spectroscopic means and hydrolysis products. All compounds displayed moderate inhibitory activity against enzyme lipoxygenase.

ABSTRACT

OBJECTIVE: To compare the efficacy of different points combination in the treatment of menopausal insomnia. METHODS: Ninety-six cases of menopausal insomnia were randomized into 3 groups, Xinshu (BL 15), Shenshu (BL 23), Sishencong (EX-HN 1), Shenmen (HT 7), Sanyinjiao (SP 6) were chosen in the restore interaction between the heart and the kidney group (group A, 32 cases); Zhaohai (KI 6), Jiaoxin (KI 8), Shenmai (BL 62), Pucan (BL 61) were chosen in the acupuncturing qiao mai group (group B, 32 cases); auricular Shenmen (TF4) and sensitive spot at the distribution area of auricular vagus nervus were chosen in the ear acupuncture group (group C, 32 cases). Six days made one session and the treatments were finished after 4 courses. The polysomnography (PSG) and Pittsburgh sleep quality index (PSQI) were employed before and after treatment to evaluate the alleviation of insomnia. RESULTS: The parameters of the sleep latency (SL), rapid wave sleep latency (RL) and sleep efficiency (SE) were significantly improved in the three groups, and the differences were statistically significant (P < 0.05, P < 0.01). The SL and awaking time (AT) in group C [SL (401.08 +/- 16.54) min and AT (4.87 +/- 2.64) times] were significantly superior to those in the other two groups [SL (50.36 +/- 18.47) min, (54.87 +/- 20.92) min, AT (5.98 +/- 2.11) times, (6.13 +/- 3.04) times, all P < 0.05]. The S(3+4) (%) in group C was also significantly higher than those in the other two groups (both P < 0.05). It was indicated by PSQI that the sleep quality of group C (0.78 +/- 0.12) was significantly superior to that in group B (1.32 +/- 0.29), the total score and cured and markedly effective rate in group C [(4.34 +/- 1.43), 68.8% (22/32)] were superior to those in group A [(7.48 +/- 3.09), 53.1% (17/32), both P < 0.05]. CONCLUSION: Ear acupuncture has a better curative effect than the restore interaction between the heart and the kidney group and acupuncturing qiao mai group, it is worth of being promoted.

ABSTRACT

Oridonin is a natural compound purified from Rabdosia rubescens that has remarkable anti-inflammatory and antitumor activities. Although oridonin has been used in traditional Chinese medicine for many years to treat inflammatory diseases, the underlying mechanisms of these activities are not well understood. In this paper, we explored whether oridonin could be used in transplantation and the mechanisms of its immunosuppression. Oridonin efficacy in transplantation is manifested by prolonged graft survival and decreased graft infiltration. Oridonin induces T cell apoptosis in a concentration- and time-dependent manner. In vivo, oridonin depleted large numbers of T cells from the spleen and peripheral blood. Decreased T cell numbers resulted from apoptosis, which was accompanied by increased phagocyte numbers in the periphery. Reduction in the number of thymocytes was observed in mice treated for 8days, and CD4+CD8+ cells were more sensitive to apoptosis induced by oridonin. Additionally, successive treatment with oridonin for 16days resulted in a considerable reduction in the total number of spleen cells and spleen volume. Thus, T cell depletion may play an essential role in prolonged graft survival and immunosuppression induced by oridonin.

ABSTRACT

OBJECTIVE: To explore the better therapy for peripheral facial paralysis. METHODS: One hundred and twenty patients were randomized into three groups: a common acupuncture group: acupuncture at Yangbai (GB 14), Sibai (ST 2) and Yingxiang (LI 20) as main acupoints, a ST 9 group: acupuncture at Renying (ST 9) as main and a ST 9 plus SGB group: acupuncture at Renying (ST 9) as main cooperated with stellate ganglion block (SGB). Once daily, 7 treatments made one session. After three sessions of treatment, the latency period and amplitude of evoked potential in ENoG, R1 value and R2 value of blink reflex were compared before and after the treatment in different groups separately. The total therapeutic effect was evaluated after treatment. RESULTS: All the treatments shortened the latency period of ENoG, and elevated the amplitude evoked potential significantly. After treatment, the latency period in ST 9 plus SGB group was reduced significantly as compared with common acupuncture group (P < 0.05). The amplitude of evoked potential in ST 9 group was increased significantly as compared with the other two groups (both P < 0.05). After treatment, in each group, R1 and R2 values were shortened significantly. The difference values of R1 and R2 in ST 9 group and ST 9 plus SGB group were all significantly higher as compared with common acupuncture group (both P < 0.05). Additionally, the difference value of R1 in ST 9 plus SGB group was higher significantly than that in ST 9 group (P < 0.05). The clinical cured and remarkably effective rate was 87.5% (35/40) in ST9 plus SGB group, which was higher than 77.5% (31/40) in ST 9 group, and 65.0% (26/40) in common acupuncture group (P < 0.05). CONCLUSION: As compared with common acupuncture group, ST 9 group and ST 9 plus SGB group achieve the much superior efficacy on peripheral facial paralysis. The treatment with ST 9 acupuncture and SGB can better repair the early reflex induced by the injury of facial nerve.
