RESUMEN

Granulomas are inflammatory tissue responses directed to a set of antigens. Trapped Schistosoma mansoni eggs promote productive granulomas in the tissues, and they are the main damage caused by schistosomiasis. Some S. mansoni antigenic proteins may have a direct involvement in the resolution of the granulomatous response. The ATP diphosphohydrolases isoforms of this parasite are immunogenic, expressed in all phases of the parasite life cycle and secreted by eggs and adult worms. Potato apyrase is a vegetable protein that cross-reactive with parasite ATP diphosphohydrolases isoforms. In this study, the vegetable protein was purified, before being inoculated in C57BL/6 mice that were later infected with cercariae. Sixty days after infection, adult worms were recovered, antibodies and cytokines were measured, and morphological granuloma alterations evaluated. Immunization of the animals induced significant levels of IgG and IgG1 antibodies and IFN-γ, IL-10 and IL-5 cytokines, but not IL-13, suggesting that potato apyrase is an immunoregulatory protein. Supporting this hypothesis, it was found that liver damage associated with schistosomiasis was mitigated, reducing the size of the areas affected by granuloma to 35% and increasing the presence of multinucleated giant cells in this environment. In conclusion, potato apyrase was found to be effective immunomodulatory antigen for murine schistosomiasis.

RESUMEN

The antischistosomal activity of clonazepam, when administered alone or in association with oxamniquine and praziquantel, was experimentally evaluated in mice infected with Schistosoma mansoni. The animals were treated 45 days post-infection with a single dose, by oral route, according to three treatment schedules: clonazepam 25 mg/kg and sacrificed 15 min, 1h or 4 h after treatment; clonazepam 1.0, 2.5 or 10.0 mg/kg and sacrificed 15 days post-treatment or with the dose of 10 mg/kg in association with oxamniquine 50 mg/kg or praziquantel 200 mg/kg, single dose, orally, every schedule with a control group. The efficacy of the drugs in vivo was assessed by means of worm counts and their distribution in mesentery and liver, mortality and oogram changes. In the chemotherapeutic schedules used, clonazepam did not present antischistosomal activity and the result of the association of this drug with oxamniquine or praziquantel was not significantly different from the one obtained when these two last drugs were administered alone. In the in vitro experiments, the worms exposed to 0.6 mg/mL clonazepam remained motionless throughout the 8-day-period of observation, without egg-laying, whereas the worms of the control group showed normal movements, egg-laying and hatching of miracidia on the last day of observation. The results obtained in the present study confirm the action of clonazepam on S. mansoni adult worm, in vitro, causing total paralysis of males and females. However, no additive or synergistic effects were observed when clonazepam were used in association with oxamniquine or praziquantel.

RESUMEN

Suspensoes aquosas do pericarpo do fruto, da casca da raiz, das folhas e das sementes de Guaiacum officinale foram testadas como moluscicida, cercaricida e piscicida em diferentes concentracoes. Em laboratorio, a suspensao do pericarpo do fruto apresentou 100 por cento de mortalidade a 100ppm para desovas de B. glabrata, a 20 ppm para caramujos adultos de Biomphalaria glabrata, B. straminea e B. tenagophila, a 5 ppm para Lebistes reticulatus (peixes) e a 1 ppm para cercarias de Schistosoma mansoni....

RESUMEN

The action of the ether artemisinin (artemether) on Shistosoma mansoni in mice and the hansters experimentally infected with the LE strain was studied. In mice, the drugs showed high schistosomicidal activity using a single intramuscular dose of 100 mg/Kg/day. By the oral route, this dose showed a low activity. Mice treated with a single intramuscular dose of 200 mg/Kg/day, and examined 15 days after treatment, presented 100% alteration of the oogram; when examined 45 days after treatment, the oogram was normal. With doses of 100 mg/Kg/day, i.m., during 3 or 5 consecutive days, the death rate of mice was very high. Morphologic analysis of the worms collected by perfusion of mice treated with a single dose of 100 mg/Kg/day, i.m., detected a marked decrease in the length of male and female forms, degenerative alterations in the parenchyma and in the reproductive system of the females, with reduction of vitellinic material and in ovary volume; the intestinal contents presented a marked despigmentation. In the male worms signifcant alteration was not apparent by optical microscopy.

RESUMEN

Os hidrolatos e óleos essenciais de diferentes espécies de Eucalyptus foram testados sobre caramujos adultos e desovas de Biomphalaria glabrata e cercárias de Schistosoma mansoni. Esses produtos foram obtidos por arraste de vapor. Dos 21 hidrolatos ensaiados, oito foram ativos sobre caramujos, nove demonstraram açäo sobre desovas e três apresentaram atividade sobre cercárias a partir da diluiçäo de 1:4 (V/V). Dos óleos estudados, 11 tiveram atividade para planorbídeos e desovas nas concentraçöes de 20ppm (p/V). O hidrolato do Eucalyptus deanei foi ativo sobre caramujos, desovas e cercárias a partir da diluiçäo de 1:4 e o óleo na concentraçäo de 20 ppm

RESUMEN

Foi avaliada, em laboratório, a açäo moluscicida de extratos aquosos (macerado e fervido), hexânico e etanólico de Aristolochia brasiliensis, Caesalpinia peltophoroides, Caesalpinia pulcherrima, Delonix regia, Spathodea campanulata e Tibouchina scrobiculata. As soluçöes dos extratos obtidos foram testadas sobre caramujos adultos e desovas de Biomphalaria glabrata, criados em laboratório, nas concentraçöes de 1, 10, 20, 200 e 1000ppm. Dos extratos testado o mais ativo foi o etanólico das flores da D. regia (flamboyant) que apresentou atividade moluscicida sobre caramujos adultos na concentraçäo de 20ppm
