RESUMEN

A multi-hyphenated analytical method that was successfully established in previous research was applied to quality evaluation of traditional Chinese medicine (TCM) to verify its feasibility in complex systems. Scutellariae Radix (SR), which significantly protects against oxidative damage from ischemia and reperfusion, was selected as the TCM for this study. A dual-activity detection system based on xanthine oxidase (XOD) inhibition and superoxide anion (O2-) scavenging activity was used to generate a multi-dimensional-multi-informational (MD-MI) integrated fingerprint of SR. Combined with HPLC-ESI-Q-TOF-MS analysis, 17 active compounds in SR were tentatively identified by comparison with reference substances or literature data. The quality of SR from different habitats was comprehensively and systematically evaluated in respect of chemical composition, XOD inhibition and O2- scavenging activity. It was confirmed that SR contains many antioxidants and XOD inhibitory substances with diverse functions. Among them, baicalin, norwogonin-7-O-glucuronide and baicalein are the main contributors to direct antioxidant activity. Acteoside, 5,7,2',5'-tetrahydroxy-8,6'-dimethoxy flavone, baicalin and baicalein are the main XOD inhibitory components of SR. Comprehensive analysis found that the antioxidant activity of SR from Gansu Province was superior to that from other provinces in terms of both XOD inhibition and O2- scavenging activity. It has been demonstrated that the method is capable of analyzing complex TCM matrices, and can provide a useful reference for establishing quality control of TCM from the perspective of MD-MI.

RESUMEN

DT-13 is an isolated compound from Dwarf lillytruf tuber and currently among active research drugs by National Natural Science foundation of China for its several potential effects. The drug has been reported for its multiple pharmacological actions however no thorough review studies are available on it. Our present study is highlighting the pros and cons of DT-13 focusing on its potential pharmacological actions, therapeutic utilization and further exploration for novel targets. The drug possesses very low toxicity profile, quick onset and long duration of action with slow elimination that combinely makes it favorable for the clinical studies. In vivo and in vitro studies show that the drug regulates multiple cellular functions for its several pharmacological effects including, anti-adhesive effects via regulation of tissue factor and transforming growth factor; anti-migratory effects through indirect regulation of NM-IIA in the tumor microenvironment, Tissue factor, down-regulation of CCR5-CCL5 axis and MMP-2/9 inhibition; anti-metastatic effects via regulation of MMPs and tissue factor; pro-apoptotic effects by modulation of endocytosis of EGF receptor; anti-angiogenic effects via regulation of HIF-1α,ERK, Akt signalling and autophagy inducing characteristics by regulating PI3K/Akt/mTOR signalling pathway. In addition to anti-tumor activities, DT-13 has significant anti-inflammatory, cardioprotective, hepatoprotective and immunomodulating effects. Pharmaceutical dosage form and targeted drug delivery system for DT-13 has not been established yet. Moreover, DT-13, has not been studied for its action on brain, colorectal, hepatic, pancreatic, prostate and blood cancers. Similarly the effects of drug on carbohydrate and glucose metabolism is another niche yet to be explored. In some traditional therapies, crude drug from the plant is used against diabetic and neurological disorders that are not reported in scientific literature, however due to profound effects of DT-13 on blood and cerebral ischemic disorders, it is reasonable to hypothesize that there could be an association of DT-13 that require further exploration.

RESUMEN

Osteoarthritis (OA),a common chronic degenerative joint disease,rises gradually with age,which seri-ously affects the quality of life of middle-aged and elderly patients. Currently,the therapeutic medications such as nonsteroidal anti-inflammatory drugs (NSAIDs)and analgesics might improve OA symptoms,but cannot prevent the development of OA. The active ingredients of traditional Chinese medicine (TCM)have unique advantages in the treatment of OA. This article reviews the research progress of active ingredients of TCM in the prevention and treatment of OA reported by domestic and foreign journals in the past five years from the aspects of inhibition of the secretion of inflammation-related factors,improvement of cartilage matrix synthesis and catabolic imbalance, inhibition of chondrocyte apoptosis,promotion of chondrocyte proliferation,and regulation of estrogen levels,with an attempt to provide a theoretical basis for the development of new drugs for OA.

The traditional efficacy of traditional Chinese medicine (TCM) is to summarize the clinical efficacy of TCM under the guidance of TCM theory,and it possess important guiding significance for the clinical use of TCM.Comparing with the modern medical system,the TCM system was different from western medicine in diagnosis and treatment modes,and the disease targets of TCM maybe more extensive than that of western medicine.Based on the traditional efficacy of TCM,combined with modern means of science and technology,the research on material basis and pharmacological mechanism of TCM was beneficial to discovery the new targets,new mechanism,new material and the secondary development of new drug of TCM.Furthermore,it also has far-reaching significance on interpret the modern scientific connotation of TCM efficacy.Therefore,deriving from the traditional efficacy of TCM,the research of modern pharmacodynamic material basis is the source for original research.

RESUMEN

Oxidative stress-induced neuronal apoptosis plays a vital role in the development of neurodegenerative disorders;while at present its regulation pathways mainly include the NF-κB pathway;p53 pathway;MAPK path-way;PI3 K/Akt pathway;Nrf2 pathway;as well as other signaling pathways.Research findings suggest that the active components in Chinese materia medica exert significant biological activities in the treatment of oxidative stress-induced neuronal apoptosis.In the present paper;we review the recent research advances on the relative mechanisms of active components in Chinese materia medica against oxidative stress-induced neuronal apoptosis;so as to provide some references or clues for the clarification of possible mechanisms of active components in Chinese materia medica and the treatment of related neurodegenerative diseases.

The application of the compound preparation of traditional Chinese medicine (TCM) and chemical drug in clinical has a long history. Drug regulatory requirements of the compound preparation of TCM and chemical drug in various periods were compared and analyzed, and the history and status of the way of scientific supervision of this preparation were summarized. This paper discussed the problems and deficiencies of the compound preparation of TCM and chemical drug, and put forward countermeasures and suggestions about to strengthen the scientific supervision of the compound preparation of TCM and chemical drug.

The author analyzed the importance of formulate national processing procedures of prepared slices of Chinese crude drugs combined actual work, proposed the overall objectives and tasks for the formulation, and emphasized to need to correctly deal with several important factors during the process of formulate "National processing procedures of prepared slices of Chinese crude drugs", unified the national standards of prepared slices, solved the real problems that the prepared slices of Chinese crude drugs industry faced.

<p><b>OBJECTIVE</b>To develop an HPLC-ELSD method for the determination of 25 (R, S) ruscogenin 1-O-[beta-D-glucopyranosyl (1 --> 2)] [beta-D-xylopyranosyl (1 --> 3)] beta-D-fucopyranoside in the tuberous roots of Liriope muscari from different habitats and different harvest time.</p><p><b>METHOD</b>A Shimadzu C18 column (4.6 mm x 150 mm, 5 microm) with a solvent system consisting of acetonirile-water (46: 54) was used, and detected by ELSD. The temperature of drift tube was 94 degrees C and the nebulizer nitrogen flow rate was 2.5 L x min(-1).</p><p><b>RESULT</b>The calibration curve of 25 (R, S) ruscogenin 1-O-[beta-D-glucopyranosyl (1 --> 2)] [beta-D-xylopyranosyl (1 --> 3)] beta-D-fucopyranoside showed good linearity in the range of 1.02-12.228 microg and the average recovery was 100.80%, with RSD of 1.8%. 10 batches of L. muscari from different habitats were analyzed, and the contents were 0.25% - 0.41%. The contents of 15 batches from different harvest time were 0.13%-0.38%.</p><p><b>CONCLUSION</b>The method is simple, rapid and sensitive, and can be used for determination of 25 (R, S) ruscogenin 1-O-[beta-D-glucopyranosyl (1 --> 2)] [beta-D-xylopyranosyl (1 --> 3)] beta-D-fucopyranoside in L. muscari. It provides the valuable basis for quality assessment of L. muscari.</p>

Combination disease and syndrome to construct animal model must be accord with the diagnosis and treatment mode of tradition Chinese medicine (TCM). The construct ion method should abide by the mode of combination prescription, syndrome, disease and efficacy. Prescription refers to those classic and effective formula. Syndrome refers to the symptoms and signs of prescription. Disease refers to model medical disease. Efficacy refers to the results of treatment. Constructing animal model should choose the classical prescription and formulate the clinical signs and symptoms. According to the main pathological process of modem diseases related to the syndrome to mine the methods of constructing animal models to express the main signs and symptoms, at last to testify the efficacy through the corresponding prescription. This method of constructing animal model is not only accord to the mode of diagnosis and treatment of TCM, but also affords a certain example of constructing animal model of combination model disease and syndrome.

AIM: To study the scavenging activities of Xiaoaiping Injection (Marsdenia tenacissima (Roxb) wight et Arn) for H2O2 and O2?- radicals by HPLC coupled with chemiluminescence and to trace the active constituents on line. METHODS: Solution of Xiaoaipin Injection was passed through HPLC to enter into the mixing tees (contained the luminol hydrogen peroxide solution and luminol pyrogallol solution,respectively),and recorded chemiluminescence signals were recorded,and their chemical structures were identified in combination with LCMS/MS. RESULTS: Three phenolic acids were identified,3-Caffeoylquinic acid,5-Caffeoylquinic acid,4-Caffeoylquinic acid,having scavenging activities for free radicals. CONCLUSION: Phenolic acids in Xiaoaiping Injection make a significant contribution to its scavenging activities for H2O2 and O2?-. It is shown that the method for on-line detection is an advantageous tool to rapidly determine antioxidants from traditional Chinese medicine.
