RESUMEN

We report a rapid, efficient, and scope-extensive approach for the late-stage electrochemical diselenation of BODIPYs. Photophysical analyses reveal red-shifted absorption - corroborated by TD-DFT and DLPNO-STEOM-CCSD computations - and color-tunable emission with large Stokes shifts in the selenium-containing derivatives compared to their precursors. In addition, due to the presence of the heavy Se atoms, competitive ISC generates triplet states which sensitize 1 O2 and display phosphorescence in PMMA films at RT and in a frozen glass matrix at 77 K. Importantly, the selenium-containing BODIPYs demonstrate the ability to selectively stain lipid droplets, exhibiting distinct fluorescence in both green and red channels. This work highlights the potential of electrochemistry as an efficient method for synthesizing unique emission-tunable fluorophores with broad-ranging applications in bioimaging and related fields.

RESUMEN

Aims: Delphinidin (DEL) is a plant-derived antioxidant with clinical potential to treat inflammatory pain but suffers from poor solubility and low bioavailability. The aim of the study was to develop a well-tolerated cyclodextrin (CD)-DEL complex with enhanced bioavailability and to investigate the mechanisms behind its antinociceptive effects in a preclinical model of inflammatory pain. Results: CD-DEL was highly soluble and stable in aqueous solution, and was nontoxic. Systemic administration of CD-DEL reversed mechanical and heat hyperalgesia, while its local application into the complete Freund's adjuvant (CFA)-induced inflamed paw dose-dependently reduced mechanical hyperalgesia, paw volume, formation of the lipid peroxidation product 4-hydroxy-2-nonenal (4-HNE), and tissue migration of CD68+ macrophages. CD-DEL also directly prevented 4-HNE-induced mechanical hyperalgesia, cold allodynia, and an increase in the intracellular calcium concentration into transient receptor potential ankyrin 1 expressing cells. Both 4-HNE- and CFA-induced reactive oxygen species (ROS) levels were sensitive to CD-DEL, while its capacity to scavenge superoxide anion radicals (inhibitory concentration 50 [IC50]: 70 ± 5 µM) was higher than that observed for hydroxyl radicals (IC50: 600 ± 50 µM). Finally, CD-DEL upregulated heme oxygenase 1 that was prevented by HMOX-1 siRNA in vitro. Innovation:In vivo application of DEL to treat inflammatory pain is facilitated by complexation with CD. Apart from its antioxidant effects, the CD-DEL has a unique second antioxidative mechanism involving capturing of 4-HNE into the CD cavity followed by displacement and release of the ROS scavenger DEL. Conclusion: CD-DEL has antinociceptive, antioxidative, and anti-inflammatory effects making it a promising formulation for the local treatment of inflammatory pain.

The heteroleptic sandwich complex [Cr(eta(5)-C5H5)(eta(7)-C7H7)] (trochrocene) was prepared by subsequent treatment of CrCl3 with NaCp and Mg in the presence of cycloheptatriene in yields of 40%. Selective dimetalation employing tBuLi/tmeda (N, N, N', N'-tetramethylethylenediamine) afforded the highly reactive species [Cr(eta(5)-C5H4Li)(eta(7)-C7H6Li)] x tmeda. An X-ray crystal-structure determination of its thf solvate revealed a symmetrical, dimeric composition in the solid state, that is, a formula of [Cr(eta(5)-C5H4Li)(eta(7)-C7H6Li)]2 x (thf)8, where the C5H4 moieties of both units are connected by two bridging lithium atoms. Addition of different element dihalides to the dilithio precursor facilitated the isolation of ansa complexes with boron and germanium in the bridging position. Structural characterization by X-ray diffraction studies on [Cr(eta(5)-C5H4)-BN(SiMe3)2-(eta(7)-C7H6)] and [Cr(eta(5)-C5H4)-GeMe2-(eta(7)-C7H6)] emphasized the strained character with tilt angles of 23.87(13) degrees and 15.07(17) degrees , respectively. In contrast, the isolation of the appropriate [1]stannatrochrocenophane failed because of the thermal lability of the resulting product. However, the corresponding 1,1'-disubstitued derivatives [Cr(eta(5)-C5H4R)(eta(7)-C7H6R)] (R = B(Cl)NiPr2, SiMe3, GeMe3, SnMe3) were obtained by reverse addition of the dilithio precursor to an excess of the element (di)halide. The unstrained nature was proven by a crystal structure analysis of the 1,1'-diborylated species. The electronic structure of these substituted trochrocene derivatives, as well as of the [2]bora and [n]sila congeners (n = 1, 2), was investigated by means of UV-vis spectroscopy and DFT methods. As a consequence of the strong electronic influence of the B-N pi-system on the LUMOs, the UV-vis studies revealed a complementary correlation of the lowest energy band maxima as a function of molecular distortion for the boron containing species on the one hand, and the boron-free compounds on the other hand. These trends were reproduced fairly well by time dependent DFT calculations.
