Choline is an essential nutrient that is necessary for both fetal development and maintenance of neural function, while its effect on female ovarian development is largely unexplored. Our previous study demonstrated that choline supplementation promotes ovarian follicular development and ovulation, although its underlying mechanism was unclear. To uncover the potential regulation pathway, eighteen female Yorkshire × Landrace gilts were fed with either standard commercial diet (Control group, n = 9) or choline supplemented diet (Choline group, additional 500 mg/kg of control diet, n = 9) from day 90 of age to day 186. At day 186, feces samples were analyzed for effects on the gut microbiome using 16S ribosomal RNA gene V3-V4 region sequencing with Illumina MiSeq, serum samples were analyzed for trimethylamine (TMA) and trimethylamine-N-oxide (TMAO) using HILIC method, and jejunum tissues were analyzed for immune related gene expression using qRT-PCR. Our results show that choline supplementation did not alter the circulating level of TMA and TMAO (P > 0.05), but rather increased gut microbiome alpha diversity (P < 0.05). Beta diversity analysis results showed that the choline diet mainly increased the abundance of Firmicutes, Proteobacteria, and Actinobacteria, but decreased the abundance of Bacteroidetes, Spirochaetes, and Euryarchaeota at the phyla level. Meta-genomic analysis revealed that choline supplementation activated pathways in the gut microbiota associated with steroid hormone biosynthesis and degradation of infertility-causing environmental pollutants (bisphenol, xylene, and dioxins). To further verify the effect of choline on intestinal activity, a porcine intestine cell line (IPEC-J2) was treated with serial concentrations of choline chloride in vitro. Our data demonstrated that choline promoted the proliferation of IPEC-J2 while inhibiting the apoptotic activity. qRT-PCR results showed that choline significantly increased the expression level of Bcl2 in both IPEC-J2 cells and jejunum tissues. The expression of IL-22, a cytokine that has been shown to impact ovarian function, was increased by choline treatment in vitro. Our findings reveal the beneficial effect of choline supplementation on enhancing the gut microbiome composition and intestinal epithelial activity, and offer insights into how these changes may have contributed to the ovarian development-promoting effect we reported in our previous study.

Given the globally increased waste stream of textile dyeing sludge (TDS), its co-combustion with agricultural residues appears as an environmentally and economically viable solution in a circular economy. This study aimed to quantify the migrations and chemical speciations of heavy metals in the bottom ashes and gas emissions of the co-combustion of TDS and waste tea (WT). The addition of WT increased the fixation rate of As from 66.70 to 83.33% and promoted the chemical speciation of As and Cd from the acid extractable state to the residue one. With the temperature rise to 1000 °C, the fixation rates of As, Cd, and Pb in the bottom ashes fell to 27.73, 8.38, and 15.40%, respectively. The chemical speciation perniciousness of Zn, Cu, Ni, Mn, Cr, Cd, and Pb declined with the increased temperature. The ash composition changed with the new appearances of NaAlSi3O8, CaFe2O4, NaFe(SO4)2, and MgCrO4 at 1000 °C. The addition of WT increased CO2 and NOx but decreased SO2 emissions in the range of 680-1000 °C. ANN-based joint optimization indicated that the co-combustion emitted SO2 slightly less than did the TDS combustion. These results contribute to a better understanding of ash-to-emission pollution control for the co-combustion of TDS and WT.

Retinol-binding protein 4 (RBP4) is known as a highly conserved adipokine for immune activation. Aeromonas hydrophila (A. hydrophila) is the most common zoonotic pathogen in aquaculture, which causes serious economic losses to aquaculture, especially to bighead carp (Hypophthalmichthys nobilis, H. nobilis) and silver carp (Hypophthalmichthys molitrix, H. molitrix). Recent studies along with our previous findings have shown that synthetic oligodeoxynucleotides containing CpG motifs (CpG ODN) can play a good role in aquatic animals against infection. In order to clarify the relationship between CpG ODN and RBP4 under A. hydrophila infection, firstly, full-length RBP4 cDNAs from H. nobilis and H. molitrix were cloned. And characteristics of RBP4, including sequence and structure, tissue distribution and genetic evolution were analyzed. In addition, mRNA expression levels of RBP4, cytokine, toll-like receptors (TLRs), morbidity and survival rates of H. nobilis and H. molitrix were observed post CpG ODN immunization or following challenge. The results indicated that hn/hm_RBP4 (RBP4 genes obtained from H. nobilis and H. molitrix) had the highest homology with Megalobrama amblycephala. Distribution data showed that the expression level of hn_RBP4 mRNA was higher than that of hm_RBP4. After CpG ODN immunization followed by A.hydrophila challenge, significantly higher survival was observed in both carps, together with up-regulated RBP4 expression. Meanwhile, hn/hm_IL-1ß level was relatively flat (and decreased), hn/hm_IFN-γ, hn/hm_TLR4 and hn/hm_TLR9 levels increased significantly, but hn/hm_STRA6 showed no significant change, compared with control. Moreover, CpG ODN immunization could induce stronger immune protective responses (higher IFN-γ/gentle IL-1ß level and lower morbidity/higher survival rate) against A. hydrophila in H. nobilis, along with higher RBP4 level, when compared with that in H. molitrix. These results demonstrated that RBP4 was well involved in the immune protection of CpG ODN. Based on the results, we speculated that in the case of A. hydrophila infection, TLR9 signaling pathway was activated by CpG ODN. Subsequently, CpG ODN up-regulated RBP4, and RBP4 activated TLR4 signaling pathway. Then TLR4 and TLR9 synergistically improved the anti-infection responses. Our findings have good significance for improving resistance to pathogen infection in freshwater fish.

The present study characterized the kinetic, thermodynamic and performance parameters, products, factorial interactions, and optimal conditions of combustions of waste tea (WT) and tea leaves (TL) in N2/O2 and CO2/O2 atmospheres through a thermogravimetric/Fourier transform infrared spectrometry (TG-FTIR). The main combustion occurred in the range of 200-600 °C. The increased heating rate increased all the combustion parameters regardless of the fuel and atmosphere type. Activation energy was shown different change tendency with the increasing conversion (α). CO2, H2O, CH4, CO, CO, NH3, and HCN were the main gas products of WT and TL combustions. A three-way interaction among fuel type, atmosphere type and heating rate was found to be significant. The joint optimization of mass loss, derivative TG, and differential scanning calorimetry was achieved using 1049.3 °C, TL, 40 °C/min, and CO2/O2 atmosphere for the operational settings of temperature, fuel type, heating rate, and atmosphere type, respectively.
